Mume fructus is the roasted fruits of Prunus mume and has been used as traditional chinese medicine. In this study, mume fructus extracts were prepared by three different methods including hot water extract (sample1), fermentation extract using Lactobacillus (Sample2-LP and sample-LA) and ethanol extract (sample3). Total polyphenol and flavonoid contents were improved by fermentation process, compared to water extract. Sample 3 showed the highest activity in DPPH radical scavenging. The cytotoxicity of the sample 2-LP was in the range of 83.3% cell viability at $300{\mu}g/m{\ell}$ concentration. Mume fructus extracts in a concentration-dependent manner inhibited melanogenesis and NO synthesis was inhibited. Specifically, the extracts fermented by L. plantarum (sample2-LP) showed higher anti-oxidant activity, anti-inflammatory and skin whitening activity than other extracts. It suggests that sample 2-LP could be potentially used as a resource of materials for functional cosmetics.

The effect of structured water by dissolution of xenon was examined from the view point of the suppression of both browning and respiratory metabolism of broccoli. The structured water is formed duet to hydrophobic interaction when xenon gas dissolves into water. NMR measurements were carried out to determine proton spin-spin relaxation time, T2, for water. There was a difference in proton T2 between distilled water and structured water. This can be interpreted as the change of water structure. Fro the broccoli cut in half stored for 16 days at 279K, the section color did not change appreciably for the sample whose water was structured by dissolution of xenon whose initial partial pressure was 0.39MPa. In contrast to this, the browning of section surface was observed for the sample stored under the condition of nitrogen gas at the same partial pressure as xenon and for the sample stored under atmospheric condition . These results led to the conclusion that the suppression of b owning by oxidation was due to structured water but not to applied pressure. Adding to this, the water structured by xenon has resulted in suppression of respiratory metabolism of broccoli.

The purpose of this study was to evaluate the changes on the characteristics of the oxidative stability of Tenebrio molitor larvae during cold storage at $4^{\circ}C$. Pretreatment for T. molitor larvae was designed into three methods: raw (R), freeze-dried (F.D.), and pan-fried (P.F.). The water content of the raw sample (61.46%) was higher than those of other samples (F.D.: 5.02%, P.F.: 3.67%) and its high water content was expected to facilitate the oxidation of the raw sample. In our results, the peroxide value and the carbonyl value of all of the samples increased and the raw sample, after storage for 18 day, showed the highest value. The pan-fried sample had no significant increase in its lactic acid content, acid value, and thiobarbituric acid value; whereas those values were increased in the raw sample and the freeze-dried sample (p<0.05). The browning reaction was more progressed in the pan-fried sample than other samples at 0 day, but there was no significant change during the storage. The raw sample and the freeze-dried sample had their browning indexes increase with the increasing storage period (p<0.05). The pan-fried sample produced less oxidation products than the freeze-dried sample, indicating that the unheated sample was more susceptible to oxidation than the heated samples. In conclusion, heating treatment and low water content would be effective for improving the safety and stability of T. molitor larvae during cold storage.
Algae bloom occurred in reservoir in summer can cause taste and odor in water and disturb the flocculation and sedimentation processes in water treatment plant and cause sand filter plugging. It was also reported that microcystins, anatoxin and saxitoxin released from cyanobacteria had acute toxic effects on liver and nervous system. For these reasons, many advanced countries inclusive of WHO set the guideline for these toxins and cyanotoxins have been managed with regular monitoring in Korea as well. However, complex sample preparation steps such as a solid phase extraction (SPE) and derivatization are required with an existing analysis method with HPLC. We needed to improve an analysis method for low extraction efficiency and long sample preparation time. In this study, we have established a new LC/MS/MS method which can simultaneously determine 6 cyanotoxins (Microcystins-LR, Microcystins-RR, Microcystins-YR, Anatoxin-a, Saxitoxin, Neosaxitoxin) with only simple filtration step. When $75{\mu}L$ filterated sample was injected onto the LC-MS/MS, the recovery ranged from 86% to 112% and the MDL was $0.025{\sim}0.581{\mu}g/L$. We can make the MDL be lower than the guideline ($1{\sim}3{\mu}g/L$) of advanced countries with simple preparation.
Methods of bedsoil analysis were difficult to be applied universally because use and material of bedsoil are diverse from country to country. Korean Standard Methods for Bedsoil Analysis was developed to measure the water content, water retention, and saturated hydraulic conductivity. Fifty-three samples for horticultural bedsoil and nine samples for paddy rice bedsoil in the current market were collected. Water content of bedsoil was determined using gravimetric method through $105^{\circ}C$ oven-dry for 16 hours, but different calculations between horticultural and paddy rice bedsoils were chosen according to different predominant component, plant residue or mineral. Water content percentage of horticultural bedsoil was calculated as [(weight of sample before oven-dry - weight of sample after oven-dry)/(weight of sample before oven-dry)]${\times}100$, while that of paddy rice bedsoil as [(weight of sample before oven-dry - weight of sample after oven-dry)/(weight of sample after oven-dry)]${\times}100$. Water retention was measured at water potential -0.5, -1, -3, -5, -7, -10 kPa by Sandbox method and saturated hydraulic conductivity was measured by constant head method using acryl cylinder (${\Phi}5cm{\times}L\;20cm$). By new 'Korean Standard Methods of Bedsoil Analysis', the average water content of horticultural bedsoil was obtained 46.34%(w/w) and that of paddy rice bedsoil 16.89%. For horticultural bedsoil, easily available water(EAW), water buffering capacity(WBC), and optimal matric potential(OMP) was estimated at 28.4%, 7.01%, and -5.60 kPa, respectively. Optimal moisture content was 44.41% and average saturated hydraulic conductivity for bedsoils was estimated at $28.4cm\;min^{-1}$.
Due to the fast response to algae bloom issue in drinking water treatment plant, very fast determination methodology for algal toxin is required. In this study, column switching technique based online preconcentration method was combined with high resolution full scan mass spectrometer to save sample preparation time and to obtain fast and accurate result. After parameter optimization of online preconcentration, 1mL filtered sample was directly injected to trap column with switching valve system. Next, target toxins are eluted by 98% acetonitrile and analysed with 150 - 1,100 amu scan range at 50,000 resolving power. Method detection limit (MDL) for microcystin-LR, the most toxic isomer, was 0.1 ng/mL and others such as microcystin-YR, microcystin-RR and nodularin were 0.08, 0.03 and 0.04 ng/mL, respectively. This is the best improved sensitivities with 1mL volume in the literature. Furthermore, due to the use of ultra pressure HPLC (UPLC), the whole method run was completed in 4 min. Real sample applications for 173 sample including 55 surface water and 118 treatment plant samples for raw and treated water could be done within 16 hours. In our calculation, this methodology is roughly 80% faster than the previous manual solid-phase extraction with LC-MS/MS method.
The peels of Carya cathayensis Sarg. (PCCS) were treated under subcritical water conditions ($130^{\circ}C$ to $280^{\circ}C$ for 0 to 120 min). The extract from PCCS included reducing sugar, proteins, and compounds with radical scavenging activity. Addressing the reducing sugar that is a resource of bioethanol, we could maximize the reducing sugar under the subcritical water ($190^{\circ}C$ for 60 min) and obtain 0.24 g/g-sample together with 9.7 units/mg-sample of radical scavenging activity. The obtained extract was estimated to correspond to 1 L of bioethanol/100 g-sample. It was therefore considered that the treatment by subcritical water could yield reducing sugar and natural compounds with radical scavenging activity.
This study was carried out to examine the behavior of THM formation in water treated with chlorine dioxide where humic acid was used as THM precursor. THM was not detected in bromide-free water, but formed in water containing bromide. When 10 mg/l of chlorine dioxide was added to water containing 5 mg/l of humic acid and bromide respectively, 20.46 ${\mu}$g/l of THM was formed. It is postulated that chlorine dioxide oxidize bromide to hydrobromous acid, which subsequently reacted with humic acids similar to chlorine reaction. The formation of THM could be reduced at low pH. Among THM formed, CHBr$_3$ was the predominant species in the alkaline solution, while CHCl$_3$ in the acidic solution. A sample pretreated with chlorine dioxide for 24h before addition of chlorine showed a reduction of 75.1% in THM formation, compared with a sample not pretreated with chlorine dioxide and a sample treated by chlorine for 24h prior to addition of chlorine dioxide also showed a reduction of 37.8% in THM formation, compared with a sample not added with chlorine dioxide. It may explain that chlorine dioxide oxidizes directly a fraction of THM.

Objectives : This study was performed to examine the effect of Sinchim on enhancing of memory in rat with water maze. Methods : Experimental animals(white rat) were classfied into normal, control, and aroma sample group. Then, they were injected .{\beta}-amyloid into control and aroma sample group rat's brain about $5{\mu}l$ to injure its brain. After rat smelt Sinchim about 12days, I did water maze test and anatomized its hippocampus. Sections were cut coronally at 30 ${\mu}m$.(XI00) Results: 1. In acquisition test of water maze learning, .{\beta}-amyloid injured group took more time than normal group to reach the escape platform noticeably and through the session of trial, Sinchim aroma sample group shortened time than .l3-amyloid injured group after 6 days. 2. In the acetyltransferase(AchE) immunostained method, it was shown that Sinchim aroma sample recovered tbe syntbesis of ChAT(Choloneacetyltransferase). Conclusion: Smelling Sinchim would be useful for enhancing of memory.
The Forsythiae Fructus is an oriental medicine containing various lignans. In this study, the Forsythiae Fructus were extracted by hot water (Sample 1), hot water after bio-conversion using Lactobacillus strain (Sample 2-LP2, 2-LA, 2-LC, 2-LL, 2-BL and 2-LM) and 70% ethanol (Sample 3). Total polyphenol and flavonoid contents were improved by bio-conversion process using Lactobacillus strain, compared to water extract. Especially, sample 2-LL and 2-LA which had shown the high total polyphenol and flavonoid content in antioxidant activity. Also, sample 2-LL and 2-LA showed higher melanin generation inhibitory activity as of 55%, 53% in maximum extract concentration of $100{\mu}g/m{\ell}$. In the anti-inflammation test of the Forsythiae Fructus extracts, nitric oxide (NO) synthesis was inhibited. Specially, both 70% Forsythiae Fructus ethanol extract and sample 2-BL which have shown the relatively higher 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging and superoxide dismutase (SOD) like activities. In conclusion, the Forsythiae Fructus extracts with bio-conversion process has effect of skin whitening and anti-inflammation activity than other extracts. It could be used as a valuable materials for functional cosmetics.
