The objective of this study is to investigate the contamination levels and dispersion patterns of arsenic and heavy metals and to estimate the bioaccessible fraction of the metals in soil and plant samples in the vicinity of the abandoned Songcheon Au-Ag mine. Tailings, soils, plants (Chinese cabbage, red pepper, soybean, radish, sesame leaves, green onion, lettuce, potato leaves, angelica and groundsel) and waters were collected around the mine site. After appropriate preparation, all samples were analyzed for As, Cd, Cu, Pb and Zn by ICP-AES and ICP-MS. Elevated levels of As and heavy metals were found in tailings. Mean concentrations of As in agricultural soils were higher than the permissible level. Especially, maximum level of As in farmland soil was 513 mg/kg. The highest concentrations of As and Zn were found in Chinese cabbage (6.7 mg/kg and 359 mg/kg, respectively). Concentrations of As, Cd, and Zn in most stream waters which are used for drinking water around this mine area were higher than the permissible levels regulated in Korea. Maximum levels of As, Cd and Zn in stream waters were 0.78 mg/L, 0.19 mg/L and 5.4 mg/L, respectively. These results indicate that mine tailings can be the main contamination sources of As and heavy metals in the soil-water system in the mine area. The average of estimated bioaccessible fraction of As in farmland soils were $3.7\%$ (in simulated stomach) and $10.8\%$ (in simulated small intestine). The highest value of bioaccessible fraction of metal in farmland soils was $46.5\%$ for Cd.
The aim of this study is to explores the possibility of applying new aquaponics using guppies, a tropical fish breeding as companion fish at home, different from the aquaponics system using fish species such as loach, carp, and catfish for commercial purposes. To facilitate the application of Aquaponics at home, a system was established by connecting a water tank, water plants, hydroponic pots, plant growth LEDs, and Arduino sensors using Internet of Things(IoT) technology. As a hydroponic crops, lettuce that can be easily obtained and consumed at home was selected. In order to confirm the applicability of aquaponics using tropical fish, the growth rates of hydroponic crops in the same environment were compared as a control. The growth rate of aquaponics crops using tropical fish was about 77.4% of that of hydroponic crops. This will produce the same effect as hydroponic cultivation if conditions correspond with enough fish quantity to feed plant and appropriate pH control for growth are met. It can be seen that, and in the future, it can be used to develop an Aquaphonics standard system applicable at home.
The physico-chemical characteristics of water and sediment, and structures of vegetation of the vascular hydrophytes were investigated in the West Nakdong River. Water quality was eutrophic according to the mean values and the ranges of water properties such as pH, DO, BOD, chlorophyll a, total nitrogen and phosphate, and other nutrients. A few cases were hypereutrophic for chlorophyll a level in summer. Soil reaction was weak acid. Composition of sediment was mainly sand except in SI(Sinan chideung) of which was mainly clay, and SU(Suan chideung) of which was mainly silt. Flora of vascular hydrophyte had 26 species and 1 variety comprising 16 families. Trapa japonica was dominant species in the sites of DU(Dunchido), GA(Garak chideung) and SU. Nymphoides peltata and Hydrocharis dubia dominated in DA and SI, respectively. Species diversity and evenness were relatively high in SI and SU but dominance was high in DA. After June, water lettuce(Pistia stratiotes) and water hyacinth(Eichhornia crassipes) were flowed from tributary to the river. Standing crop of macrohydrophytes was high in DA from April to August, but it showed maximum standing crop (445g·dw/㎡) in DU after disturbance by explosive growth of exotic plants in October. In comparison with those in 1985, total productivities in DU and GA decreased to 33.5%, and the reduction ratio of dominant species, Trapa japonica was 56.7%. Najas marina, N. minor, Myriophyllum spicatum and Nymphoides indica have disappeared ever since the Nakdong barrage was constructed in the Nakdong river. They were divided into three groups (GA-SU-DU, DA, SI) by cluster analysis. Introduction of the exotic species in this river caused decreasing of endemic plants including endangered species Euryale ferox and rare species Hydocharis dubia, and food plants for waterfowl such as Trapa japonica, Vallisneria asiatica and Potamogeton crispus.
Allelopathic inhibition by substance(s) originated from sicklepod(Cassia tora L.) seed on germination of other plant seeds, was confirmed and used to elucidate general mechanism of allelopathy which was occurred widly in natural and/or agricultural ecosystems. The mechanism was deduced from changes in water contents, ${\alpha}$-amylase activities, protease activities, concentrations of abscisic acid and total phenolic compounds during germination of rice and radish seeds treated with water-soluble extracts of sicklepod seeds. The results obtained were summarized as follows. 1. The % germinations of radish, rice, lettuce, barley seeds were decreased by substances originated from crushed sicklepod seeds. 2. By the treatment of water-soluble extracts of sicklepod seeds, the inhibition of germination of radish seeds was occured with the sequential phenomena of increase of protease activity(synthesis), decrease of water content and increase of total phenolic compounds content. 3. In rice, the inhibition of seed germination by the water-slouble extracts of sicklepod seeds was related to increase of abscisic acid concentration and then decrease of ${\alpha}$-amylase activity(synthesis).
To enlarge application field of electrolyzed acid-water(EAW) on food industry, the changes of EAW properties by storage conditions and heating were investigatet. It was showed that storing EAW in closed container is mon effective to keep up the oxidation-reduction potentials(ORP), hyperchloride content and pH than stored in opened ones. ORP of EAW stored in closed container could be kept mon than 1 month as 1,150 mV levels. Ruing heating from 2$0^{\circ}C$ to 95$^{\circ}C$, ORP was increased to 1,150 mV levels at 95$^{\circ}C$ after gradual decrease to 5$0^{\circ}C$. Tyrosinase activity was decreased approximately to 26%~35% in EAW having a 950 mV~1,140 mV ORP. Also it was confirmed that EAW has anti-browning effect as sliced apple and potato, and their juices treated with EAW had conspicous difference in their $\Delta$E value. 12 kinds of pesticides such as aldrine, capful diazinon, diedrin, $\alpha$-endosulfan $\beta$-endosulfan, endosulfan sulfate, endrin, $\alpha$-BHC, o,p'-DDT, procymidone, PCNB added in EAW were recovered from ND~73.6% comparing to ones added in distilled water. The recovered amounts of pesticides, procymidone and diazinon in lettuce after soaking in EAW were 1.12 ppm and ND, compared with those of amounts soaked in distilled water were 3.67 ppm and 3.05 ppm respectively. So, it seems that EAW has potentials to promote the degradation of pesticides.
This study was conducted to investigate the influences of bicarbonate ($HCO_3^-$) concentrations in irrigation solution on growth of lettuce and change in chemical properties of root media. The blue leaf and red leaf lettuces with two true leaf stages were transplanted into 10 cm diameter plastic pots filled with perlite. The five treatments were made by dissolving $NaHCO_3$ into irrigation solution to reach 30, 70, 110, 150 and $180mg{\cdot}L^{-1}$$HCO_3^-$. The crops were fed with fertilizer solution contained $HCO_3^-$ with various concentrations and controlled to $100mg{\cdot}L^{-1}$ in nitrogen concentration. The pH in soil solution of root media 10 weeks after transplant of blue lettuces were 7.04 and 7.10 in the treatments of 30 and $70mg{\cdot}L^{-1}$ of $HCO_3^-$, respectively. But those rose gradually after week 3 and finally reached 7.39, 7.48 and 7.56 at week 10 in the treatments of 110, 150 and $180mg{\cdot}L^{-1}\;HCO_3^-$, respectively. The pH in the treatments of 30 and $70mg{\cdot}L^{-1}\;HCO_3^-$ in cultivation of red leaf lettuce were around 6.65 during week 4 to week 8, but this rose abruptly and reached 6.92 and 7.01 at week 10, respectively. Those in the treatments of 110, 150, and $180mg{\cdot}L^{-1}\;HCO_3^-$ rose gradually and finally reached to 7.49, 7.53, and 7.58, respectively. The EC rose gradually after week 2 in all treatments of blue and red leaf lettuces. The change of macro ion concentrations in both blue and red leaf lettuces showed similar trends. The concentrations of $PO_4-P$, $Ca^{2+}$ and $Mg^{2+}$ increased gradually in all treatments of $HCO_3^-$ during cultivation of blue and red leaf lettuces. As the concentrations of $HCO_3^-$ in irrigation solution were elevated, the concentrations of $PO_4-P$, $Ca^{2+}$ and $Mg^{2+}$ became higher and that of ${SO_4}^{-2}$ became lower in soil solution of root media. The main reason of concentration changes were that $HCO_3^-$ influenced pH and the pH changes also affect the activities of the ions in soil solution of root media.
The objective of this study was to isolate pathogenic Escherichia coli from fresh vegetables with selective media and Petrifilm, and identify a suspicious colony using biochemical identification. Twenty gram of lettuce, twenty gram of cabbage and ten gram of sprout were prepared, and a 5-strain mixture of pathogenic E. coli (Enterohemorrhagic E. coli NCCP11142, Enterotoxigenic E. coli NCCP14037, Enteropathogenic E. coli NCCP14038, Enteroaggregative E. coli NCCP14039, Enteropathogenic E. coli NCCP15661) was inoculated to obtain 1, 2 and 3 log CFU/g. Eighty to ninety milliliter of buffered peptone water (BPW) was placed and pummeled for 60 s. As a results, the Petrifilm method was all positive, but enrichment method of qualitative analysis was negative except for 3-log CFU/g inoculated lettuce. Regarding biochemical identification of pathogenic E. coli, the identification rates were dependent on type of methods and vegetables; lettuce: API 20E 100% (44/44), Microgen GNA 100% (44/44) and Food System 66.7% (10/15), cabbage: API 20E 64.7% (22/34), Microgen GNA 50% (16/32) and Food System 60% (9/15), sprout: API 20E 65.1% (28/43), Microgen GNA 62.3% (27/43) and Food System 53.3% (8/15). These results could be useful in determining an appropriate method to detect pathogenic E. coli in fresh vegetables.
To compare germinability of primed seeds with that of water imbibed and nonprimed seeds at different temperatures, percent germination, number of days to attain 50% of the final germination percentage (T50) and mean number of days to germination (MDG) were evaluated at various temperatures ranged from $10^{\circ}C$ to $35^{\circ}C$. Primed carrot seeds in -0.50 MPa PEG 8000 showed higher percent germination at $10^{\circ}C$ or $35^{\circ}C$. Reductions in T50 and MDG values were observed at all germination temperatures when primed with the PEG. The PEG combined with 100 mM $K_3PO_4$ was not as effective as the PEG alone in improving the germinability of carrot seeds. Nonprimed lettuce seeds germinated only 2% at $30^{\circ}C$ or $35^{\circ}C$. However, priming with 50 mM $K_3PO_4$ increased germination to 40% at $30^{\circ}C$ and to 21% at $35^{\circ}C$. In contrast to the single treatment of 50mM $K_3PO_4$, priming lettuce seeds in a mixture of 50mM $K_3PO_4$ and 200 mM $KH_2PO_4$ resulted in a lower percentage of germination, but a significant greater reduction of T50 and MDG. Percent germination was increased when 200 mM $KH_2PO_4$-primed onion seeds and 100 mM $Ca(NO_3)_2$-primed Welsh onion seeds were germinated at $10^{\circ}C$ or $35^{\circ}C$. Combined treatments with $Ca(NO_3)_2$ for onion and with $KH_2PO_4$ for Welsh onion seeds did not improve percent germination, but they significantly reduced T50 and MDG.
This study was conducted to extract the factors affecting the microbial safety of leaf and stem vegetables in the high school foodservice and to provide information for supplying the safe foodservice menu. The lunch and dinner menu (1,945 data) of the total 6 high schools at the Central and the South Region in March, June, September, and December were collected. The frequency analysis and the multiple correspondence analysis (MCA) based on the 3 factors (potentially hazardous food (PHF), leafy and stem vegetables in the menu, the cooking methods) were conducted. The most frequent PHF was the menu of blanched vegetables, salads, seaweeds and fried chicken. The most frequent consumed leaf and stem vegetables were spinach, chive, lettuce, Western cabbage, perilla leaf, iceberg lettuce, chicory, leek and broccoli. MCA based on the leaf and stem vegetables, the region, and the cooking method (cooked/non-cooked) showed that garlic stem and spinach were more used in the Central Region, while water drop-wort were more used in the South Region. Iceberg lettuce, Bok choy and leek were included frequently in the PHF menu. Plant products frequently used in PHF menu requires the food safety system such as Good Agricultural Practice (GAP) to reduce the microbial risk. The menu database according to raw materials based on cooking methods (heating or mixing) as well as the development and verification of menu based on the microbial safety will be contributed to provide the safer foodservice menu.
Water extracts of polygonum hydropiper and Polygonum aviculare completely inhibited the germination of lettuce seeds. Methanol extracts from these two species also inhibited the seed germination of lettuce (Lactuca sativa) and Oenothera odorata. Fifteen phenolic acids in total were identified by GLC from P. hydropiper and eighteen from P. aviculare. The most common phenolic acids identified from P. hydropiper were sinapic, salicylic+vanillic and ferulic acid presented in all the fractions. In addition, salicylic+vanillic, tannic+gallic, sinapic, ferulic and p-coumaric acid seemed to be important phenolic compounds in terms of quantity. However, salicylic+vanillic acids were the unique phenolic acids occurred in all the fractions of P. aviculare. The others such as tannic+gallic, sinapic, ferulic, p-coumaric acid, p-cresol and catechol present in large amount appeared also the important phenolic substances influencing allelopathic effects of P. aviculare. Linolenic acid and oxalic acid were the major fatty and organic acids in both plant species, presented in 2.38mg/g and 20.588mg/g in P. hydropiper, 3.70mg/g and 14.288mg/g in P. aviculare, respectively, which seem to be exhibiting allelopathic effects of these plants. Total alkaloids were presented in low amount such as 0.20% in P. hydropiper arid 0.22% in P. aviculare which may not be important elements. Pet. ether extracts were 2.42% in P. hydropiper and 1.65% in P. aviculare, which exhibit another potential for allelopathic effects that need further investigation. Various authentic phenolic compounds at different concentrations inhibited the germination of lettuce seed, indicating that the phenolic substances identified here may be directly related to biologically active substance.
