Studies on the Extraction Efficiency of Polyacetylene from Korean Ginseng (추출방법에 따른 인삼의 Polyacetylene 성분 회수율 비교 연구)

  • The extraction efficiencies of panaxynol and panaxydol were optimal at 80$^{\circ}C$ with soxhlet method. The extraction efficiencies increased up to 45$^{\circ}C$ with shaking method. Amounts of panaxynol and panaxydol were determined by gas chromatography. Extracted quantities of panaxynol and panaxydol using the shaking method increased over a period of 14 hours. The efficiencies of panaxynol and panaxydol extraction by soxhlet and shaking methods were higher for smaller particle sizes. Upon water swelling treatment, extraction efficiencies of panaxynol and panaxydol decreased gradually with time for both methods.

Comparison of Free Amino Acids in Soybean Paste (Doenjang) by Different Extraction Solvents and Analytical Methods (추출 용매와 분석 기법에 따른 된장의 유리아미노산 비교)

  • This work was conducted to obtain a rapid, accurate, and precise procedure for free amino acids analysis in Doenjang with HPLC-OPA (high performance liquid chromatography using-phthalaldehyde) and AAA (automatic amino acid analyzer) methods. Different sample extraction procedures among water, 0.1 M perchloric acid, and 0.1% meta-phosphoric acid were also compared. The optimal extraction solvent was 0.1% meta-phosphoric acid for both the HPLC-OPA and AAA methods. Good recoveries for glycine and methionine were observed using the 0.1% meta-phosphoric acid extraction with HPLC-OPA method. Method precisions (% relative standard deviation) for the free amino acids ranged for 1.62% to 8.27%, in which the HPLC-OPA method with water extraction showed the lowest value at 1.62%. Inhibition rates of the free amino acids in Doenjang were greatest with an addition of NaCI at a 1% concentration.

Hot Water Extraction of Crude Chondroitin Sulfate from Shark Cartilage Treated with Ozone Water (오존수로 세척한 상어연골로부터 Crude Chondroitin Sulfate의 열수추출)

  • This study was conducted to investigate the effect of ozone treatment on removal of brown color and ammonia-like odor produced from shark cartilage (SC) and to establish the optimal extraction conditions of chondroitin sulfate (CS) by NaOH and hot water. Treatment of SC with 0.3 ppm ozone water resulted in higher color $L^*$ value but lower $a^*$ and $b^*$ values compared with those of non-treated SC, without affecting its proximate compositions (moisture, protein, lipid, carbohydrate, and ash). Ozone treatment also increased the overall acceptability of SC by reducing ammonia-like odor. The optimum concentration and time for extraction of CS from SC using NaOH at $100^{\circ}C$ were 1 N and 120 min, respectively, with 10.02% of extraction yield. The total yield (10.01%) of CS obtained by 7 times repeated extraction with hot water at $100^{\circ}C$ was comparable to that of the 1 N NaOH extraction for 120 min at $100^{\circ}C$.

Determination of Optimal Conditions by Response Surface Methodology and Quality Characteristics of Water Extracts of Phellinus linteus (반응표면분석을 이용한 상황버섯 열수추출액의 최적 추출조건과 품질 특성)

  • Response surface methodology was implemented to determine an optimal extraction condition in Phellinus linteus water extract. Extraction was performed on 10 experimental conditions including independent variables such as extraction time $(1{\sim}5\;hrs)$ and water volume over sample (sample : $H_2O$ = 1 : $40{\sim}200$, W/V), color browning, reducing and total sugar, that were based on the significant levels of 10% of central composition design. Color browning, reducing and total sugar contents were found to be more affected when the water volume was increased rather than extraction time. Maximum extraction condition was acquired at extraction time of $3.0{\sim}4.5\;hrs$ and water volume of $40{\sim}58.2\;ml$. Being extracted at the optimal extraction condition two of the free sugars, sucrose (0.126%) and glucose (0.012%), were detected. Total content of the free amino acids was found to be $503.26\;{\mu}g%$. Among them, essential amino acid contents were revealed as 5.4%. One major peak from gel permeation chromatography contained polysaccharide(s) with the molecular weights of 10 KDa.

  • To determine the concentrations of microcystins present in lake water or in tap water using high performance liquid chromatography, it is necessary to concentrate a large volume of water samples (about 20 L) into very small volume (0.1-0.3 mL). Concentration can be conveniently done when disc type solid phase extraction (SPE) apparatus is used. Using this apparatus we have investigated the recovery rates of three kinds of microcystins, RR, YR, LR. The recovery rates were relatively low and the reproducibilities were not good either. It is expected, however, that the appropriate selection of the disc conditioning and eluting solvents and reproducible reconcentration process after SPE will improve both the recovery rates and the reproducibilities.

  • An analytical method for the simultaneous measurement of trace Cu, Sn, and Bi in sea water has been investigated by Inductively Coupled Plasma-Mass Spectrometry. Amberlite IRC-718 resin was used as a solid phase in solid-liquid extraction technique for the removal of matrix interferences such as Na, S, P, and other polyatomic ion species. Recoveries of 99.8% for Cu, 99.6% for Sn, and 97.9% for Bi were obtained for the standard spiked sample. The developed method was applied to analysis of trace metals in sea water.

  • The use of the microwave-assisted process for the extraction of effective constituents from ginseng was investigated at various operating conditions. The influence of solvent (ethanol-water, 50% v/v) to ginseng ratio, particle size and applied microwave power on the efficiency of extraction was examined. The microwave extraction system used was custom manufactured so that the intensity of microwave may be varied by using anode-voltage controller. It was found that the ratio of 6 : 1 (vol/mass) gave a good extraction efficiency. Small particle size gave high yield but it caused difficulties in the separation of solvent from the sludge. The higher power was no guarantee of the efficient extraction yield. The more important factor than the employed power was the adequate temperature under sufficient contact time. Using deionized-water as swelling agent, the degree of swelling of ginseng by microwave heating and conventional heating in water-bath was also studied. It was observed that the microwave heating enhanced the swelling much more than the conventional heating. It is believed that this enhanced swelling was responsible for the rapid microwave-assisted extraction rate.

The Solubility Characteristics of Organic Compounds in Urban Aerosol Samples

  • The solubility characteristics of organic compounds were studied in terms of the extraction efficiency as a function of the polarity of the organic solvent, and the acidity of water in urban aerosol samples collected in University of East Anglia (UEA), Norwich, England. The extraction efficiency of organic compounds were evaluated with respect to the organic carbon, -nitrogen and -hydrogen by means of a wide range of solvent which include polar and nonpolar organic solvents as well as acids and alkaline water. In addition, after being dissolved in aqueous solution, the aqueous chemistry of organic compounds were studied in terms of the organic metal complexes in aerosol, which were studied with oxalic acid, copper, and zinc. The results of this study indicate that solubility characteristics of organic compounds depend on the polarity of the solvents and the acidity of the solvents. In particular, some organic compounds are water soluble, even though they are much smaller than acetone soluble fractions. In the comparison between polar organic solvent extraction and non- polar organic solvent extraction, it can be thought that significant fraction of organic compounds analysed in the aerosol samples, are polar organic compounds because of the higher extraction efficiencies of organic compounds in polar organic solvent extraction than in nonpolar organic solvent extraction. Regarding the study of the oxalic -metal complexes, it can be thought that most oxalic acids are present in the form of oxalic -copper complexes in the aerosols collected at UEA.

Comparison of Biological Activities of Acer mono and A. okamotoanum Extracts by Water Extracton and Low Temperature High Pressure Extraction (고로쇠와 우산고로쇠 저온고압 수용성 추출물의 유용생리활성 비교)

  • Hepatoprotective and antioxidant activities of Acer mono and A. okamotoanum were compared after being extracted by low temperature and high pressure (LTHP) at 20 MPa and $60^{\circ}C$ for 15 minutes. Extraction yield of both A. mono and A. okamotoanum was increased about 40~43% by this extraction process. On scavenging activities, the bark of A. okamotoanum from this extraction process showed the highest activity as about 97%. This value was higher than that from conventional water extraction and A. mono extracts. Both of A. mono and A. okamotoanum showed high ability on nitrite scavenging, but decreasing tendency according to increasing of pH. On SOD-like test, A. okamotoanum had the highest activity as 46.28% at $1.0\;mg/m{\ell}$ concentration. A. okamotoanum extracted by LTHP also showed the highest activity as 197.38% in adding $1.0\;mg/m{\ell}$ concentration. Generally, the extracts from low temperature and high pressure extraction process are higher hepatoprotective and antioxidant activities than that from conventional water extraction. It can conclude that the bark of A. okamotoanum has better biological activities than other parts of A. mono.

Analysis of the Extraction Condition of Soluble Acidic Polysaccharides from Ginseng Marc (인삼박으로부터 수용성 산성다당체의 추출 조건 분석)

  • This study was carried out to investigate the optimum conditions for extraction of soluble acidic polysaccharides from ginseng marc. Method of carbazole-sulfuric acid was applied to determine the amount of acidic polysaccharides in ginseng marc. The amounts of soluble acidic polysaccharides in water extract of ginseng marc were increased with increasing extraction temperature. The contents of acidic polysaccharides were not significantly different despite the extraction time increasing from 0.5 hours to 6 hours. To estimate the rehydration rate of the freeze dried polysaccharide, the extracted acidic polysaccharide fraction powder was determined the amount of soluble acidic polysaccharides by carbazole-sulfuric acid method again. The rehydration rate of acidic polysaccharides from water-extract of red ginseng marc at room temperature was 100%. On the other hand, the rehydration rate of acidic polysaccharide of red ginseng marc at boiling temperature was about 50%. The rehydration rate of acidic polysaccharides from water-extract of white ginseng marc at room temperature was 50%. The rehydration rate of acidic polysaccharide of red ginseng marc at boiling temperature was about 40%. The rate of soluble acidic polysaccharide of Red Ginseng is higher than that of White Ginseng. We can find out the maximum extraction method of soluble acidic polysaccharide from ginseng marc.
