Antioxidant activity and total polyphenolics of Shiitake Mushroom extracts(Lentinus edodes)

  • Methanol and water crude extracts, crude polysaccharides and crude polysaccharides-free of water crude extracts from Shiitake mush-room (Lentinus edodes) were investigated for their antioxidant capacity in total polyphenolics and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. Total polyphenolics in the crude polysaccharides-free of water crude extracts were higher than that of the other ones. And crude polysaccharides-free of water crude extracts and water crude extracts show high on DPPH free radical scavenging activity. Positive correlations were found between total polyphenolics and their antioxidant activity.

  • The aim of the present experiments was to investigate the effect of Yukmijihwangtang water extracts on the plasma renin activity and plasma levels of atrial natriuretic peptide and aldosterone in rats. The results of this study were as follows: 1. Plasma renin activity decreased significantly after the administration of Yukmijihwangtang water extracts 1.5ml/kg. 2. Plasma levels of atrial natriuretic peptide (ANP) decreased significantly after the administration of Yukmijihwangtang water extracts. 3. Plasma levels of aldosterone increased significantly after the administration of Yukmijihwangtang water extract.

Effects of Hot Water Soluble Extract from Angelicae Gigantis Radix on the Lipid Metabolism and Antioxident Defense System in rats fed Hypercholesterol Diet (당귀 열수추출물이 고콜레스테롤 식이를 공급한 흰쥐의 지질대사와 황사화계에 미치는 영향)

  • To investigate the effects of hot water soluble extract from Angelicae Gigantis Radix on lipid metabolism and antioxidant defense system in rats fed hypercholesterol diet, Sprague-Dawley male rats were fed either hypercholesterol diet with water or hot water soluble extract from Angelicae Gigantis Radix. Hypercholesterol diet contained 18 % beeftallow and 5 % cholesterol. After 4 weeks of experimental periods serum and liver obtained. The level of serum lipid and the activities of GOT and GPT was measured. The level of lipid and TBARS and the activity of GSH-Px was measured also. The results obtained were: 1) Food intake in the experimental group supplied hot water soluble extract from Angelicae Gigantis Radix was low significantly. 2) In the experimental group supplied hot water soluble extract from Angelicae Gigantis Radix of serum total lipid and total cholesterol was significantly low. 3) Serum GOT activity was significantly low in the experimental group supplied hotwater soluble extract from Angelicae Gigantis Radix. There was no difference of serum GPT activity between control and experimental group supplied hot water soluble extract from angelical Gigantis Radix. 4) There was no difference of lipid composition, TBARS level and GSH-px activity in liver between control and experimental group. The result of this study indicates that the hot water soluble extract from Angelicae Gigantis Radix was effective in decreasing the food intake of the experimental animals in the hypercholesterol diet and also effective in decreasing the levels of serum total lipid, total cholesterol and GOT activity. However it was not effective to change the level of lipid, TBARS and GSH-Px activity of liver.

  • Effect of water activity $(0.32{\sim}0.89)$ on the physicochemical properties of sweet potato starch during the storage for 5, 15 and 30 days at $40^{\circ}C$ was investigated. Shapes and sizes of starch granules were not changed. X-ray diffraction patterns of the starches appeared equally Ca-crystal structure. Sorption isotherm with storage day was sigmoidal. A slight loss of iodine affinity, increase in water binding capacity, and decrease in swelling power at $80^{\circ}C$ occurred as water activities increased. Viscosity pattern under Brabender Amylogram was not significantly changed with water activity, but initial pasting temperature decreased as water activity increased. The viscosity at $50^{\circ}C$, consistency and setback were increased with increasing storage day and water activity.

Pharmacetical Characteristics of Solvent Fractions Isolated from Glycyrrhiza uralensis

  • In this study, the biological activities of Glycyrrhiza uralensis were investigated, including antioxidative, fibrinolytic, thrombin inhibitory, and a-glucosidase inhibitory activity. The hot water extract of Glycyrrhiza uralensis was fractionated into hexane, $CHCl_3$, ethyl acetate, butanol, and water fractions, and each of these fractions were assayed individually. The water fraction showed the highest extraction yield of 19.45% (w/w). Using the DPPH method, the free radical scavenging activity was to be the strongest in the $CHCl_3$ fraction at 89.3%. Using the fibrin plate method, only the butanol fraction showed a substantial plasmin activity of 0.62 units/ml. In thrombin inhibitory activity tests, a 100-fold dilution of the ethyl acetate fraction showed the strongest activity of 46.9%. In the a-glucosidase inhibitory activity assay, a 100-fold dilution of the $CHCl_3$ fraction showed the strongest activity of 80.6%. In conclusion, the combined results of this study demonstrate that the extracts of Glycyrrhiza uralensis can be used as a material for the development of biofunctional foods for diabetics.

Biological Activity of Aloe Vera Gel and Skin Extracts (알로에 베라 젤 및 껍질 추출물의 생리활성 평가)

  • In vitro biological activities of Aloe vera gel and skin extracts were evaluated. Total polyphenol contents of Aloe vera skin were measured 41.12 mg/g. DPPH radical scavenging activity of Aloe vera skin-70% EtOH extract, Aloe vera skin-water extract, Aloe vera gel-70% EtOH extract and Aloe vera gel-water extract were 55%, 38%, 11% and 10%, respectively. In addition, 70% EtOH extract and water extract were compared with respect to SOD-like antioxidant activity of Aloe vera-70% EtOH extract has higher activity than Aloe vera water extract. Tyrosinase inhibition rate of Aloe vera gel extract was higher than Aloe vera skin extract. Alcohol dehydrogenase (ADH) and Acetaldehyde dehydrogenase (ALDH) relative percentage activity of Aloe vera gel extract were 126% and 216%, respectively. It was suggested that Aloe vera gel and skin extracts could be used as a functional biomaterial for functional food and cosmetics.

Inhibitory Effects on Melanin Biosynthesis and Tyrosinase Activity; Cytotoxicity in Clone M-3 and Antioxidant Activity by Cuscuta japonica, C. australis, and C, chinensis Extracts (새삼, 실새삼 및 갯실새삼 추출물이 Clone M-3 세포주의 Melanin 생합성 및 Tyrosinase 활성에 미치는 영향과 세포독성 및 항산화효과)

  • Water extracts, ethanol extracts, and juice of Cusuta japonica, C, australis, and C. chinensis were prepared, and their cytotoxicity, antioxidant activity and inhibitory effects on tyrosinase activitiy and melanin biosynthesis were estimated by using melanoma Clone M-3. From this study; the following conclusions were attained. Extracts of Cuscuta japonica, C. australis, and C. chinensis showed noticeable cytotoxicity except ethanol extracts from the stem of C. australis. A maximual cytotoxicity was observed with tile ethanol extract from the seed of C, australis (87.39%). While the ethanol extract from the seed of C. japonica (91.88%) showed the most pronounced inhibitory effect on melanin biosynthesis, the water extract from the stem of C. japonica (1.05%) possessed very little inhibitory effect. The most inhibitory effect on tyrosinase activity was observed with the water extract from the stem of C. australis (76.67%). Howeverr the water extract from the stem of C. japonica showed a very poor effect on the inhibition of tyrosinase activity All the preparations, except extracts from the seed of C. australis were able to remove reactive oxygen species (ROS) in a dose-dependent manner. The juice of C. japonica demonstrated the strongest activity (59.02%).

Anti-oxidization Effect of Extracts from Oriental Medicine and Cereal Medium Where Tricholoma matsutake Mycelia were Cultured

  • In order to explore the anti-oxidization effect of oriental medicine and cereal medium(OCM) where Tricholoma matsutake mycelia were cultured, measurement of hot water extract and UMPM(extraction method using ultra sonic waves, micro waves, micro bubble) extract, the total polyphenol content of crude polysaccharide from each extract, SOD-like activity, electron donating ability(EDA), xanthine oxidase inhibitory activity, and tyrosinase inhibitory activity was conducted. The total polyphenol content of each extract was found to be 16.36% for hot water extract(HE) group and 15.73% for UMPM extract(UE) group and the amount of crude polysaccharide precipitated into ethanol of extracts was found to be 8.79% for UMPM ethanol extract(UEE) group and 6.48% for hot water ethanol extract(HEE) group. As a result of measurement of SOD-like activity by concentration of each extract, it was found to be 96.17% for UE group, 91.23% for HE group, 91.33% for UEE group, and 87.11% for HEE group at 20 mg/mL. In the case of EDA, it was found to be 47.55% for UE group, 44.93% for HE group, 25.38% for UEE group, and 18.36% for HEE group. And in the cases of the rates of xanthine oxidase inhibitory activity and tyrosinase inhibitory activity, as the concentration of each extract increased, the inhibition rate increased accordingly. As a result of comparison between hot water extract method and UMPM extract method using extracts obtained from oriental medicine compound medium where Tricholoma matsutake mycelia were cultured, all of the extracts were judged to have a high anti-oxidization effect. In particular, UMPM extracts were found to have higher polyphenol content, SOD-like activity, EDA, xanthine oxidase inhibitory activity and tyrosinase inhibitory activity compared to hot water extract method. In this regard, extracts obtained from OCM where Tricholoma matsutake mycelia were cultured are considered to have high availability as functional material when and if they are prepared using UMPM extract method.

Antioxidative and Free Radical Scavenging Activity of Water Extract From Dandelion (Taruaxacum officinale) (민들레 물추출물의 항산화 및 자유라디칼 소거활성)

  • The antioxidative and free radical scavenging activity of water extracts of dandelion were investigated. Antioxidative and radical scavenging activity were assessed by means of different tests; inhibition of peroxidation on linoleic acid model system, scavenging DPPH radical, scavenging of hydroxyl radical by chemiluminescence assay, scavenging of superoxide anion radical by EPR spectroscopy and scavenging of hydrogen peroxide. The leaf extract showed strong antioxidant activity in linoleic acid system. The antioxidant activity of water extracts of dandelion increased with increasing concentrations of extracts. The scavenging activity of the dandelion extracts, on inhibition of the DPPH radical, was related to the reaction time. Hydroxyl radical were generated by lenten reaction and dandelion extract was found to scavenge OH˙in a concentration-dependent manner. The water extract of leaf had effective scavenging activities on hydrogen peroxide and superoxide anion radical. From the these data, it is evident that water extract of dandelion leaf is an effective scavenger for OH˙, O$_2$¨, DPPH˙, hydrogen peroxide. And, the antioxidative effect observed is believed to be partly due to this radical scavenger activity.

Screening of Antimicrobial Activity from Differential Extracts of Allii sativi Bulbus (대산의 분획별 추출물에서 항균활성 검색)

  • Allii sativi Bulbus(garlic) have been shown to possess medicinal value, in particular, antimicrobial activity. In this study, we compared the efficacy on some pathogenic bacteria and fungus among several different extracts(water, hexane, ethyl acetate, methanol, chloroform) of Allii sativi Bulbus. Animal pathogenic bacteria and fungus(S. gallinarium: KCTC 2441, S. flexneri: KCTC 2361, E. cloacae: KCTC 2006, K. pneumonia: KCTC 2208, C. albicans: KCTC 1940) were used to test by measurement of minimum inhibitory concentrations(MIC) and disc diffusion. Allii sativi Bulbus were cut and mixed with water at 37℃ about 24 h and filtered, and several different solvents(hexane, chloroform, ethyl acetate, methanol) were respectively added to separate the fraction of each solvent. The antimicrobial(bacteriocidal) and antifungal effect were apparently shown from water extract, hexane and ethyl acetate extract against using strains(Staphylococcus gallinarium, Shigella flexneri, Enterobacter doacae, Klebsiella pneumonia, Candida albicans). Especially, the water extract showed the superior efficacy. And the clear zone size of water extract (11~27 mm) was greater than that of gentamycin, hexane extract and ethyl acetate extract against S. gallinarium. S. flexneri, K. pneumonia and C. albicans. Minimum inhibitory concentrations(MIC) of water extract appeared to around 2.0~7.5 ㎎/㎖ against S. gallinarium, S. flexneri, E. cloacae and K. pneumonia. The greater activity was shown by water extract because the MIC of water extract for C. albicans observed in very low concentration(<1.0 ㎎/㎖) compared to hexane(5.0 ㎎/㎖) and ethyl acetate(10.0 ㎎/㎖). Therefore, these results exhibited that water extract of Allii sativi Bulbus have stronger antimicrobial activity than hexane and ethyl acetate extract, and may be useful as topical medicine of superficial infections causing C. albicans as well as antifungal agents.
