Studies on the Antioxidative Ability of Methanol and Water Extracts from Orostachys japonicus A. Berger According to Harvest Times (채취 시기에 따른 와송 추출물의 항상화 활성 연구)

  • This study examined the antioxidant activity of methanol and water extracts from Orostachys japonicus A. Berger according to harvest times, by measuring electron donating ability(EDA), reducing power, superoxide dismutase(SOD)-like activity, thiobarbituric acid reactive substances(TBARS) and antioxidant activity within a linoleic acid emulsion. EDA increased proportionally with the extract concentration and the methanol extract had slightly stronger effects than the water extract. And reducing power and SOD-like activity were highest in the methanol extract. Overall, based on the data, the methanol extract of O. japonicus A. Berger harvested during $August{\sim}October$ presented the highest level of antioxidative activity and may be a good candidate as a natural antioxidant source.

  • Paulownia coreana is a medicinal, edible and industrial plant with the largest leaf, and is native to Korea. We evaluated the reducing power activities, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities (RSAs), nitrite scavenging activities (NSAs) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation RSAs by solvent extraction of P. coreana bark by using $50^{\circ}C$ hot water and $25^{\circ}C$ methanol. The antioxidative effect of P. coreana bark extract on corn seed oil was evaluated using the Rancimat test. The extraction yields on dry weight basis with 15 folds of hot water and methanol were 23.88% and 5.30%, respectively; further the flavonoid content in the hot water extract was over 2.5 times more than that in the methanol extract. The DPPH RSA of the methanol extract was substantially higher than that of the water extract, whereas the NSA of the water extract was higher than that of the methanol extract at pH 1.2. The ABTS RSAs of the two extracts were almost the same as that of ascorbic acid and 2,6-ditertiarybutyl hydroxytoluene. The two extracts of P. coreana bark in this study were found to slightly improve the oxidation stability of corn seed oil.

  • For the purpose of developing a new drug against cerebral infarction, we chose Ijintang which is frequently applied in the field of Jung Pung of traditional medicine. From this formula, water extract and methanol extract were prepared respectively. Animals were divided into three groups; control group, water extract medicated group, methanol extract treated group. water extract was given 195mg/kg orally and methanol extract, 165mg/kg for 2 weeks in case of cerebral edema and 1 month in regional cerebral blood flow. On the regional cerebral blood flow, the drug medicated groups showed no significant changes as compared gith control group. There was no significant difference on the cerebral edema among the groups. According to the results above, it is suggested that oral medication of water extract and methanol extract from Ijintang have no effect on the change of rCBF and edema after acutely induced cerebral infarction. However, it is thought that modification of administration method and development of new animal model for traditional medicine are needed.

Antioxidant and Neuronal Cell Protective Effects of Eugenia caryophyllata Thunb. by Extraction Solvent (용매별 정향 추출물의 항산화 및 신경세포 보호 효과)

  • This study examined the antioxidant and neuronal cell protective effects of the water and methanol extracts of Eugenia caryophyllata Thunb. The total polyphenol content was significantly higher in the methanol extract than in the water extract. The DPPH radical scavenging activity in the water extract was similar to Vit. C at a concentration of $100{\sim}200{\mu}g/mL$. The ABTS radical scavenging activity in the water and methanol extract was similar to Vit. C at a concentration of $800{\sim}1,000{\mu}g/mL$. The superoxide dismutase (SOD)-like activity in the methanol extract was similar to Vit. C at a concentration of $800{\sim}1,000{\mu}g/mL$. The DPPH, ABTS radical scavenging and (SOD)-like activity increased with increasing extract concentration. In a cell viability using MTT, the water extract (50 and 100 ppm) and methanol extract (100 ppm) had a protective effect against $H_2O_2$-induced neurotoxicity.The result ssuggest that the extract of E. caryophyllata Thunb. has antioxidant activities and may be useful for treating neurodegenerative disorders.

Study on Synthesis of Dimethyl Ether Using Silica Membrane Reactor (Silica막 반응기를 이용한 Dimethyl Ether 합성에 관한 연구)

  • Water selective silica membranes were prepared fur use as membrane reactor for synthesis of dimethyl ether (DME) by methanol dehydration. Silica membranes formed on a Porous SUS tube by ultrasonic spray Pyrolysis (USP) and chemical vapor deposition (CVD) using tetraethoxysilane (TEOS) as precursor. The CVD-derived membranes formed higher level of trade-off line between water permeance and water/methanol selectivity than that of the USP-derived membranes. The membrane reactor possessing water permeance of $1.2\times10^{-7}\;mol\;{\cdot}\;m^{-2}\;{\cdot}\;S^{-1}\;{\cdot}\;Pa^{-1}$ and water/methanol selectivity of 10 exhibited increase in methanol conversion of about $20\%$ comparing to conventional reactor system. These findings led us to conclude that the dehydration membrane reactor simultaneously separating the water vapour produced in the reaction zone was effective in increasing the reaction conversion.

Selection of an Optimal Entrainer for Extractive Distillation of Azeotropic Acetone/Methanol System (Acetone/Methanol계 공비물의 추출증류를 위한 최적 Entrainer 선정)

  • A study on the selection of an optimal entrainer as the third component among water, aniline, 1,3-diethylbenzene, furfural, and MEK, for the extractive distillation of an azeotropic acetone/methanol system was performed using both the entrainer effect vapor-liquid equilibrium (VLE) and the relative volatility. In the case of water as the entrainer, a VLE curve without azeotropic point in the range of water composition from 0.3 up to 0.7 mole fraction could be obtained by both the experiment and the calculation using modified-UNIFAC model. For aniline and 1,3-diethylbenzene, however, VLE curve without azeotropic point could be obtained only at compositions above 0.7 mole fraction, which exhibited that they could be hardly utilized as the entrainer. Moreover, both furfural and MEK were verified to be improper entrainer since they formed an azeotropic phase. Relative volatility of water showed greater than 1.0 and increased with compositions, while those of the others decreased non-linearly, exhibiting that only water could be utilized as the proper entrainer for the extractive distillation of azeotropic acetone/methanol system.

  • The purpose of this study is to investigate optical behaviour of coloring matter of gromwell -extracted by water, acetic acid/methanol, and the other solvents. The results were as following: UV, VIS spectra of the solution of coloring matter of gromwell extracted by water at below $80^{\circ}C$ did not reveal in the range of the visible light wavelength and coloring matter of gromwell decomposed to blue particle at $95^{\circ}C$. The solution extracted by acetic acid/methanol was tinged with reddish purple and its spectra shifted to the longer wavelength according to increase of the solvent temperature. The color of the solution extracted by water became extinct as time went by, but that extracted by acetic acid/methanol kept up original color. In case of dried gromwell, extraction by acetic acid/methanol was easier than by water, and variation of spectra did not appear but showed hypochromic shift.

Negative Effects of Water Extracts from Pinus densiflora Sawdust on Mycelium Growth of the Shiitake Mushroom Lentinula edodes

  • Extracts from sawdust of Pinus densiflora were showed antifungal activity against Lentinula edodes. It was extracted by hot water and then successively extracted by n-hexane, ethyl acetate, and methanol. The yields of the n-hexane-soluble, ethyl acetatesoluble, methanol-soluble and methanol-insoluble fractions of water extracts were 8.2%, 10.6%, 32.0%, and 49.2%, respectively. The ethyl acetate-soluble fraction showed the greatest antifungal activity against L. edodes: 41.5% inhibition at 1,000 ppm. However, there were not significant differences of antifungal activities between n-hexane-soluble fraction and methanol-soluble fraction at a concentration of 1,000 ppm. The hot water extracts showed 23.5% of antifungal activity against L. edodes at a concentration of 1000 ppm. The four antifungal compounds were separated from ethyl acetate fraction by thin layer chromatography.

Antioxidative Capacity and Total Phenolic Compounds of Methanol Extract from Zizyphus jujuba (대추 메탄올 추출물의 페놀성 화합물 함량과 항산화 효능)

  • The antioxidative capacity of methanol extracts prepared from jujubes (Zizyphus jujuba) were investigated by measuring total phenolic compounds, electron donating ability and induction period on lard. The extracts of large, medium and small jujubes of $80\%$ methanol extracts (methanol extracts) and soaking in the water before $80\%$ methanol extracts (water-methanol extracts) were Iyophilized. Total phenolic compounds of small, medium and large jujube of methanol extracts were 326.46 mg$\%,223.13\\;mg\%,\;158.06\;mg\%$ and that of water-methanol extracts were $113.23\;mg\%,\;81.45\;mg\%,\;72.78\;mg\%$, respectively. The Electron donating ability for the water-methanol extracts added samples of large (1.5$\~$3.0$\%$), medium and small jujube $(1.0\~3.0\%)$ were superior to the $0.02\%$ BHT added sample. On the induction period, all samples with jujube extracts were exhibited higher Antioxidant Index (AI: Induction period of lard with jujube extracts divided by induction period of lard) than that of the control and the water-methanol extracts were more effective than the methanol extracts on the antioxidative capacity for the lard. This result indicated that two lyophilized extracts of jujubes showed excellent free radical scavenging ability and long induction period on lard.

Solumycin : A water-soluble antifungal antibiotic from Streptomyces sp. LAM-593 (Streptomyces sp. LAM-593이 생산하는 수용성 항진균성 항생물질)

  • A water soluble antifungal antibiotic, Solumycin, was separated from the culture broth of Streptomyces sp. LAM-593, isolated from soil, by butanol extraction, alumina-, 1st and 2nd Sephadex LH-20 column chromatography. The substance was pale yellow crystal which gave a single spot at Rf value 0.24 with ethanol-ammonia water-water (8:1:1), 0.46 with butanol-ethanol-water (5:1:4), 0.84 with 50% methanol on silica gel TLC. It was dissolved well in water, methanol and acidic aq. butanol but not in ethanol, acetone, ethyl acetate, chloroform. acetic acid etc., and gave positive Fehling and Molish reaction. The UV spectrum in methanol showed absorption at 342, 361, 380, and 404 nm. The antibiotic was active against fungi such as Candide, Cryptococcus, Saccharomyces, Trichophyton and Trichosporon, but not to bacteria such as Bacillus, Escherichia and Staphylococcus.
