불활성 가스하 열건조에 따른 금속시험편의 수소잔류물 거동 분석 (Variation of Hydrogen Residue on Metallic Samples by Thermal Soaking in an Inert Gas Environment)

  • Hydrogen penetration into a metal leads to damages and mechanical degradations and its content measurement is of importance. For a precise measurement, a sample preparation procedure must be optimized through a series of studies on sample washing and drying. In this study, two-step washing with organic solvents and thermal soaking in inert gas were tried with a rod-shaped, API X65 steel sample. The samples were machined from a steel plate and then washed in acetone and etyl-alcohol for 5 minute each and dried with compressed air. After then, the samples were thermally soaked in a home-made nitrogen gas chamber during 10 minute at different heat gun temperatures from 100 to $400^{\circ}C$ and corresponding temperature range in the soaking chamber was from 77 to $266^{\circ}C$ according to the temperature calibration. Hydrogen residue in the samples was measured with a hot extraction system after each soaking step; hydrogen residue of $0.70{\pm}0.12$ wppm after the thermal soaking at $77^{\circ}C$ decayed with increase of the soaking temperature. By adopting the heat transfer model, decay behavior of the hydrogen residue was fitted into an exponential decay function of the soaking temperature. Saturated value or lower bound of the hydrogen residue was 0.36 wppm and chamber temperature required to lower the hydrogen residue about 95% of the lower bound was $360^{\circ}C$. Furthermore, a thermal desorption spectroscopy was done for the fully soaked samples at $360^{\circ}C$. Weak hydrogen peak was observed for whole temperature range and it means that hydrogen-related contaminants of the sample surface are steadily removed by heating. In addition, a broad peak found around $400^{\circ}C$ means that parts of the hydrogen residue are irreversibly trapped in the steel microstructure.

Light Hydrocarbon에 오염된 토양의 용매추출조건에 관한 연구 (Determination of Soil Washing Condition for Light Hydrocarbon Contaminated Soils)

  • 본 실험은 석유의 주요성분인 benzene, toluene, p,m,o-xylene, ethylbenzene, n-propylbenzene, 1,2,4-trimethylbenzene, n-butylbenzene 등 light hydrocarbon에 오염된 토양의 회복에 필요한 최적 용매추출 조건을 밝히기 위하여 실시하였다. 토양의 수분함량이 증가할수록 추출효율은 감소하였으며, Methanol을 용매로 사용하였을때에 BTEX화합물은 1시간 추출시 최고 추출효율에 도달하였으며 고분자 hydrocarbon은 4시간이상 추출시 최고 추출효율에 도달하였다. 2-Propanol을 추출용매로 사용할때는 모든 light hydrocarbon은 추출 4시간 경과시 최고의 추출효율을 나타냈다. 추출용매와 토양의 비율이 2:1일때 가장 경제적이고 효율적이고 추출이 이루어 졌으며, 0.4mg/g이상의 토양오염 농도에서 최적의 추출효과가 나타났다. 위의 최적조건으로 light hydrocarbon 오염토양을 batch extraction을 이용하여 추출하였을 때 평균 66%의 light hydrocarbon이 회수되었으며, 회수율은 토양의 종류, 추출용매의 종류 그리고 hydrocarbon의 종류에 따라 좌우되었다. 본 연구의 결과는 석유에 오염된 토양의 회복을 위한 토양세척에 사용될 수 있을 것으로 사료된다.

역류추출(逆流抽出) 및 등전침전(等電沈澱)에 의한 유채박(油菜粕) 단백질(蛋白質)의 분리(分離) (Protein Isolates from Rapeseed: (Countercurrent Extraction and Isoelectric Precipitation))

  • 탈지(脫脂) 유채박중(油菜粕中)에는 필수(必須) 아미노산(酸)이 다수함유(多數含有)한 양질(良質)의 단백질(蛋白質)을 함유(含有)하고 있으나 독소(毒素)인 myrosinase이 활성(活性)을 억제(抑制)시키거나 효소(酵素) 가수분해물(加水分解物)인 isothiocyanate 및 oxazoolidinethione을 제거(除去)하여야 한다. 이와 같은 독소(毒素)를 myrosinase 활성(活性)을 감소(減少)시키는 방법(方法)으로 처리하여 전연무독(全然無毒)한 단백질(蛋白質)을 분리(分離)하였다. 즉(卽) pH11.0에서 냉시(冷時)에 추출(抽出)하고 $0^{\circ}C$에서 침전(沈澱)을 분리(分離)하였으며 식품화학적(食品化學的)인 성질(性質) 비교(比較)를 하였다. 유채단백질(油菜蛋白質)은 분자수(分子數)가 많아 침전시(沈澱時)에는 조제(助劑)인 혹(或)은 알긴 산(酸)소 다- 를 이용(利用)하여 좋은 결과(結果)를 얻었다. 또 pH 6.7, 5.6 및 5.0에 따라 색상(色相)을 단리하는 단백질(蛋白質)이 분리(分離)되었고 수세(水洗)와 acetone을 사용(使用)하여 색소(色素)를 제거(除去)시킬 수 있었으며 냉동건조(冷凍乾燥)하여 변색(變色)을 방지(防止)하였다. 역류(逆流) 추출법(抽出法)은 양산(量産)할 수 있는 단백질(蛋白質) 추출(抽出) 방법(方法)이다.

Complication of epiduroscopy: a brief review and case report

  • Epiduroscopy is defined as a percutaneous, minimally invasive endoscopic investigation of the epidural space. Periduroscopy is currently used mainly as a diagnostic tool to directly visualize epidural adhesions in patients with failed back surgery syndrome (FBSS), and as a therapeutic action in patients with low back pain by accurately administering drugs, releasing inflammation, washing the epidural space, and mechanically releasing the scars displayed. Considering epiduroscopy a minimally invasive technique should not lead to underestimating its potential complications. The purpose of this review is to summarize and explain the mechanisms of the side effects strictly related to the technique itself, leaving aside complications considered typical for any kind of extradural procedure (e.g. adverse reactions due to the administration of drugs or bleeding) and not fitting the usual concept of epiduroscopy for which the data on its real usefulness are still lacking. The most frequent complications and side effects of epiduroscopy can be summarized as non-persistent post-procedural low back and/or leg discomfort/pain, transient neurological symptoms (headache, hearing impairment, paresthesia), dural puncture with or without post dural puncture headache (PDPH), post-procedural visual impairment with retinal hemorrhage, encephalopathy resulting in rhabdomyolysis due to a dural tear, intradural cyst, as well as neurogenic bladder and seizures. We also report for first time, to our knowledge, a case of symptomatic pneumocephalus after epiduroscopy, and try to explain the reason for this event and the precautions to avoid this complication.

Diagnostic Aspects of Fine Needle Aspiration for Lung Lesions: Series of 245 Cases

  • Background: Transthoracic fine needle aspiration (FNA) is one of several methods for establishing tissue diagnosis of lung lesions. Other tissue or cell sources for diagnosis include sputum, endobronchial biopsy, washing and brushing, endobronchial FNA, transthoracic core needle biopsy, biopsy from thoracoscopy or thoracotomy. The purpose of this study was to compare the sensitivity and specificity of FNA and other diagnostic tests in diagnosing lung lesions. Materials and Methods: The population included all patients undergoing FNA for lung lesions at Meir Medical Center from 2006 through 2010. Information regarding additional tissue tests was derived from the electronic archives of the Department of Pathology, patient records and files from the Department of Oncology. Sensitivity, specificity, diagnostic accuracy, and positive and negative predictive values were calculated for each test. Results: FNA was carried out in 245 patients. Malignant tumors were diagnosed in 190 cases (78%). They included adenocarcinoma (43%), squamous cell carcinoma (15%), non-small cell carcinoma, not otherwise specified (19%), neurondocrine tumors (7%), metastases (9%) and lymphoma (3%). The specificity of FNA for lung neoplasms was 100%; sensitivity and diagnostic accuracy were 87%. Conclusions: FNA is the most sensitive procedure for establishing tissue diagnoses of lung cancer. Combination with core needle biopsy increases the sensitivity. Factors related to the lesion (nature, degenerative changes, location) and to performance of all stages of test affect the ability to establish a diagnosis.

Development of a CAN-based Real-time Simulator for Car Body Control

  • This paper presents a developing procedure of the CAN-based real-time simulator for car body control, aiming at replacing the actual W/H (Wiring Harness) and J/B(Junction Box) couple eventually. The CAN protocol, as one kind of field-bus communication, defines the lowest 2 layers of the ISO/OSI standard, namely, the physical layer(PL) and the data link layer(DLL), for which the CSMA/NBA protocol is generally adopted. For CPU, two PIC18Fxx8x's are used because of their built-in integration of CAN controller, large internal FLASH memory (48K or 64K), and their costs. To control J/B's and actuators, 2 controller boards are separately implemented, between which CAN lines communicate through CAN transceivers MCP255. A power motor for washing windshield, 1 door lock motor, and 6 blink lamps are chosen for actuators of the simulator for the first stage. For the software architecture, a polling method is used for the fast global response time despite its slow individual response time. To improve the individual response time and to escape from some eventual trapped-function loops, High/Low ports of the CPU are simply used, which increases the stability of the actuator modules. The experimental test shows generally satisfactory results in normal transmitting / receiving function and message trace function. This simulator based on CAN shows a promising usefulness of lighter, more reliable and intelligent distributed body control approach than the conventional W/H and J/B couple. Another advantage of this approach lies in the distributed control itself, which gives better performance in hard real-time computing than centralized one, and in the ability of integrating different modules through CAN.

농업기계 단기임대사업의 효율적 운영을 위한 실태조사 연구 (Status survey on short-term agricultural machinery rental system for efficient operation)

  • Status survey on short-term agricultural machinery rental business was conducted to provide basic data for effective and sustainable implementation of the rental system. Selected survey samples were 34 rental management institutions such as city and county level government offices and agricultural technology development centers, and Primary Agricultural Cooperatives. Survey was conducted through mailing of questionnaire papers and direct interviews with the officers in charge of the agricultural machinery short-term rental management. Number of agricultural machinery retained by the 34 management institutions for the machinery rental business was 3,699, and numbers of the machinery were 1630 for upland crops, 929 for rice, 542 for orchard farming, 274 for animal husbandry, and 324 for common use. Regarding size of warehouse for rental agricultural machinery, 50% of the institutes were less than 660 $m^2$, 26.5% were greater than 993 $m^2$, and 23.5% were between 663 and 990 $m^2$. Institutes maintaining machinery washing facilities were only 10 (29%) among the 34 rental management institutions. Agricultural machinery rental business was advertised to farmers by 91% of the institutes, and the methods were leaflet (35.2%), village broadcasting (26.5%), call-up education (23.6%), and TV and radio (14.7%). Major contents of the advertisement were rental procedure (52.9%), rental machinery (26.5%), and rental cost (20.6%).

고구마 가공 슬러지로부터 회수된 고구마 전분의 물리화학적 특성 (Physicochemical Properties of Sweet Potato Starch Reclaimed from Sweet Potato Processing Sludge)

  • 고구마의 가공공정(절단, 성형, 세척) 중 발생하는 고구마 가공슬러지로부터 고구마전분을 회수하여 전분소재로서의 활용가능성을 재고하기 위해 회수된 고구마전분들의 물리화학적 특성을 조사하였고 고구마 가공 슬러지의 냉동저장에 따른 고구마전분의 특성 변화를 비교하였다. 고구마 가공 슬러지로부터 회수된 전분들의 총전분 함량은 94.1-95.6%의 범위에 있었으나 겉보기 아밀로오스와 인 함량은 냉동 고구마 가공 슬러지로부터 회수된 고구마 전분이 다른 전분들에 비해 유의적으로 낮았다. 냉동저장 처리는 고구마전분의 아밀로펙틴 분지사슬 분포에 큰 영향을 미치지 않았으나 고구마전분 입자의 결정구조를 변형시켰으며 무정형 영역을 감소시켰다. 이로 인해 고구마전분의 팽윤력과 페이스팅 점도특성은 감소하였으며 최대호화온도와 호화종결온도는 상승하는 결과를 초래하였다. 전체적인 결과를 고려할 때 고구마 가공 슬러지를 획득한 즉시 회수한 고구마전분은 고구마로부터 직접 분리한 고구마전분과 물리화학적 특성이 유사하여 상업적인 전분소재로 사용가능할 것으로 기대되지만 냉동저장된 고구마 가공 슬러지로부터 분리 정제한 고구마전분을 상업적으로 이용하기 위해서는 가공식품에 있어 이의 가공적성 및 최종품질에 대한 영향 조사가 선행되어야 할 것 같다.

어류 병원성 세균 Edwardsiella tarda의 형질전환 및 재조합 ghost 세균에서의 E-lysis 유전자의 전사 발현 특징 (Transformation of Edwardsiella tarda and Transcriptional Characteristics of E-lysis Gene in Recombinant Bacterial Ghosts)

  • 어류 에드워드감염증에 대한 예방 재조합 ghost 백신을 개발하기 위한 연구의 일환으로 어류 병원성 세균인 Edwardsiella trada를 대상으로 플라스미드 형질전환을 실시하고 형질전환 안정성을 평가하였으며, 형질 도입된 재조합 ghost 세균의 E-lysis 유전자 발현을 분석하였다. E. tarda를 대상으로 한 ghost 유도는 대장균에 비해 상대적으로 장시간의 반응 시간이 요구되며 lysis의 개시가 지연된 점을 고려 시 발현된 E protein의 용해 능력 또는 E-gene의 전사발현 양이 E. tarda에서 다소 약화되는 것으로 나타났다. 그러나 대장균에 비해 ghost 유도 속도가 다소 낮음에도 불구하고 반응이 완성되었을 시점에서의 E. tarda의 ghost 효율은 대장균과 전혀 차이가 없이 99.99% 이상의 유도효율을 나타내었다.

  • 법적시료(Forensic Evidences)로 사용되는 혈흔, 정액반, 타액반 및 모발을 DNA의 분리과정 없이 FoLT (Formamide Low Temperature) PCR법으로 DNA의 특정부위를 분석하고자 하였다. FoLT PCR법으로 3종류의 STR(Short Tandem Repeat) 좌위와 성별을 확인하는 Amelogenin gene을 PCR법으로 동시에 분석하기 위하여 formamide농도와 annealing온도를 검토한 바 최적의 formamide농도는 8%(v/v)이었고 annealing온도는 $48^{\circ}C$이었다. 그리고 1% Triton X-100을 이용하여 시료를 세척한 경우에 증폭 효율이 증가하였다. 따라서 FoLT-PCR법을 이용한 경우 DNA를 분리하기 위한 전처리없이 소량의 시료를 기존의 PCR법보다 빠른 시간내에 한번의 PCR 및 전기영동으로 HumTH01, HumTPOX, HumCSF1PO 및 Amelogenin을 동시에 분석할 수 있었다.
