Recently, we reported a satellite RNA (Paf-satRNA) which is encapsidated in a pepper isolate of Cucumber mosaic virus (CMV-Paf) regulated symptom attenuation of the helper virus. To characterize mild symptom domain of Paf-satRNA, a series of chimeric cDNAs of satRNAs were created by using full-length cDNA clones of Paf-satRNA and a Pep-satRNA, chlorosis-inducing satRNA in pepper plants, and analyzed for determinants of symptom attenuation. When compared the nucleotide sequences, the 3' and 5' terminal sequences of the two wild-type (wt) satRNAs contained relatively conserved sequences which are the typical to CMV satRNA. Ten bases insertions were found in PepY-satRNA, and two variable regions, 81st to 113th and 183rd to 265th from the 5'-end, were located in the middle parts of the satRNAs. To delineate the attenuated symptom-related domain for the Paf-satRNA, in vitro transcripts RNAs transcribed from the wt cDNAs and constructed chimeric cDNAs were combined with genomic RNAs, RNA1, RNA2 and RNA3, of CMV-Fny and inoculated onto Nicotiana benthamiana plants. These transcripts were fully infectious onto the N. benthamiana and infectivity was confirmed by the RT-PCR. Chimeric Paf(H/N)-satRNA and PepY(N/A)-satRNA as well as Paf-satRNA induced very mild mosaic or symptomless infection on N. benthamiana. By contrast, typical mosaic symptom and stunting of infected plants were induced when PepY-satRNA, PepY(H/N)-satRNA and Paf(N/A)-satRNA were infected to N. benthamiana. Paf-satRNA coinfected with CMV-Fny RNAs induced very mild to sympomless on pepper plants whereas PepY-satRNA-infected pepper expressed typical chlorosis mosaic symptom. Two kinds of chimeric mutants, Paf(H/N)-satRNA and PepY(N/A)-satRNA, induced mild mosaic or symptomless infection onto pepper plants, while PepY(H/N)-satRNA and Paf(N/A)-satRNA showed typical chlorosis and mosaic symptom with stunting. This results suggest that mild symptom-related domain for the Paf-satRNA was located on HpaI-NarI region.

The purpose of this study is to provide data for the restoration of 'Yongyun(龍淵)' and 'Hwahongmun(華虹門)' through an investigation of the vicinity of their original landscapes at the time of construction of Hwaseong in Suwon and through tracing the transformative process of the environments of this vicinity. The results are as follows; As identified by 'Yongyunjung(龍淵亭)' and 'Yongdugak(龍頭閣)', other names of Yongyun, 'Banghwasuryujung(訪花隨柳亭)', which was built on 'Yongduam', is a facility whose place identity is highlighted with a sense of unity with Yongyun. The south lakefront of Yongyun, bordering Banghwasuryujung, has boundaries that make the best use of the natural geographical features of Yongduam while the current circumference of Yongyun is comparatively shorter than its original state. The size of 'Joongdo(中島)', however, seems to be an example of apparent over-design complement and reorganize 'Joongdo', which had been restored larger than its original state at the time of restoration in the 1970s. The depth of 'Yongyun' was created to be lower than the actual depth, without consideration for its initial depth, as soil was accumulated through continuous flooding after it was created. It is assumed that the original drains which were installed about 10m inside the lake were created facing the stream. As regards the planting environment, a circular planting of willows was made in the outskirts of 'Yongyun', except the 'Yongduam' which is a pure forest, and a mix of 'Pinus densiflora', shrubbery and deciduous broad leaf trees was planted in 'Joongdo'. Of the plants growing in the area of this study, plant species introduced to Korea after Hwaseong was constructed are found, most of which provide interest and attraction. The old pine trees growing in a group once grew in the castle areas of the vicinity even in the 1920s, the period of Japanese occupation, but they disappeared from the area in the aftermath of subsequent urban development and the Korean War. Although restored to the site, the number and space taken up by these trees are insignificant compared to those of the original environment. On the basis of these results, the following is considered necessary for the true restoration of the vicinity of 'Yongyun' and 'Hwahongmun': First, the grounds of 'Yongyun' should be dredged deeply enough to expose the bedrock and should be recreated in the rough outline of a half moon by extension to the southwest toward 'Yongduam' and 'Hwahongmun', and the size of 'Joongdo' should be significantly reduced. Secondly, considering that most plant species, except the pine trees and wild trees in 'Yongduam', are non-native plants introduced in order to provide such attractions such as the appreciation of scenic areas, they should be replaced with native species, mainly with the pine trees which were utilized during the construction of Hwaseong. The weeping willows planted in the 'middle-island' should be relocated to the outskirts of 'Yongyun', and replaced with pine trees as the major trees and maple trees or deciduous broadleaf trees to fill in the gaps. Thirdly, exotic species such as the 'Pinus rigida' planted in a group around 'Banghwasuryujung' and 'Bugammun' and 'Pinus strobus' planted in the vicinity of Hwahongmun' should be removed.

Fusaric acid (FA) is a mycotoxin produced by Fusarium species. Its toxicity is relatively low but often associated with other mycotoxins, thus enhancing total toxicity. To date, biosynthetic genes or enzymes for FA have not been identified in F. oxysporum. In order to explore the genetic element(s) for FA biosynthesis, restriction enzyme mediated integration (REMI) procedure as an insertional mutagenesis was employed using FA producing-F. oxysporum strains. Genetic transformation of two F. oxysporum strains by REMI yielded more than 7,100 transformants with efficiency of average 3.2 transformants/${\mu}g$ DNA. To develop a screening system using phytotoxicity of FA, eleven various grains and vegetable seeds were tested for germination in cultures containing FA: Kimchi cabbage seed was selected as the most sensitive host. Screening for FA non-producer of F. oxysporum was done by growing each fungal REMI transformant in Czapek-Dox broth for 3 weeks at $25^{\circ}C$ then observing if the Kimchi cabbage seeds germinated in the culture filtrate. Of more than 5,000 REMI transformants screened, fifty-three made the seeds germinated, indicating that they produced little or fewer FA. Among them, twenty-six were analyzed for FA production by HPLC and two turned out to produce less than 1% of FA produced by a wild type strain. Sequencing of genomic DNA regions (252 bp) flanking the vector insertion site revealed an uncharacterized genomic region homologous (93%) to the F. fujikuroi genome. Further study is necessary to determine if the vector insertion sites in FA-deficient mutants are associated with FA production.

This experiment was carrid out to develop a culture method by sequential highland-lowland cultivation for year-round production of fresh leaves in Ligularia stenocephala. Rootstocks of Ligularia stenocephala cultivated in Daegwallyeong openfield were digged up on late October 2007 and then stored in $-2^{\circ}C$ refrigerator. The rootstocks were monthly transplanted in protected greenhouse in Gangneung from November 2007 to June 2008 for lowland cultivation, and also they were monthly transplanted in rainshelter in Daegwallyeong from June to August 2008 for highland cultivation. The early growth of Ligularia stenocephala transplanted on January 2008 was faster. Most plant of Ligularia stenocephala regardless of transplanting time of rootstock grew over 30cm at the time of first harvesting of leaves till May, while the yield decreased during the summer season. The average plant height of Ligularia stenocephala transplanted on July and August in Daegwallyeong highland was lower than that cultivated in Gangneung lowland during spring season. Using of rootstock stored made it possible to produce in highland from Apr. to Oct. during summer season, and also to produce in lowland from Nov. to May next year. Therefore, the results indicates that sequential highland-lowland cutivation by using of stored rootstock will be capable of yearround production of fresh leaves in Ligularia stenocephala.

The recent global climate change induced the drought, flooding, and insect pest outbreaks. These caused the severe damage to crop yield in the domesticated field and occurrence change of insect pest species. The sap-sucking insect pest, aphids are common in soybean [Glycine max (L.) Merr.] and cause serious yield losses. Thus, developing resistance cultivars is promising and efficient strategy to prevent the significant yield losses by aphid and screening germplasm is the essential procedure to achieve this goal. We tried to establish a resistance test indicator for foxglove aphid, Aulacorthum solani (Kaltenbach), in soybean and found that plant damage degree or infested plant damage is most suitable one. Also we screened around 1,200 of soybean germplasm including wild and cultivated species for its resistance to foxglove aphid from the various origins, and 67 soybeans, including PI 366121, showed antixenosis resistance, 31 germplasms among 67 antixenosis germplasms were showed antibiosis to foxglove aphid with non-choice test. The identified foxglove aphid resistant soybean resources showed significantly low rate in survival test. Furthermore, resistance type, (i.e., antibiosis or antixenosis) of each candidate were varied. In this research, we established the screening index for foxglove aphid resistance in soybean, and identified the resistance varieties. This result could be useful resources in breeding for new foxglove aphid resistance soybean cultivars, and provide fundamental information to investigate the resistance mechanism in soybean.

This study was designed to investigate variation of pinitol content in different parts of seventeen legume species including silk tree (Albizia julibrissin). D-pinitol has been demonstrated to exert insulin-like and anti-inflammatory effects. These legumes were collected from Gyeongsangbuk-Do in Korea. Significant difference in pinitol content was observed among 17 different legume species. However, it was the highest in sericea lespedeza (Lespedeza cuneata). The highest pinitol content was observed in leaf followed by stem, pod shell, seed and root among plant parts. Legume plants which had higher pinitol content in leaves were chinese pea shrub, bastard indigo, wild cowpea and sericea lespedeza, having 59.9 mg/g, 62.2 mg/g, 69.6 mg/g and 65.4 mg/g, respectively. Stem of astragali radix showed the highest pinitol content among all leagumes, which was 34.0 mg/g. In case of root, kudzuvine showed the highest pinitol content followed by licorice and chinese pea shrub, which were 24.6 mg/g, 16.9 mg/g, and 16.5 mg/g, respectively. However, in the case of pod shell and seed, only lablab purpureus and sericea lespedeza showed the highest pinitol content 52.5 mg/g (for pod shell) and 24.9 mg/g (for seed), respectively. Concluding, the sericea lespedeza showed the highest total pinitol content of whole plant followed by chinese redbud, chinese pea shrub, bastard indigo and silk tree. Considering the pinitol content and harvesting yield, sericea lespedeza can be used as a practical medicinal herb.
The cytochrome P450s (P450s) metabolizing natural products are among the most versatile biological catalysts known in plants, but knowledge of the structural basis for their broad substrate specificity has been limited. The activity of p-coumarate 3-hydroxylase (C3H) is thought to be essential for the biosynthesis of lignin and many other phenylpropanoid pathway products in plants however, all attempts to express and purify the protein corresponding C3H gene have failed. As a result, no conditions suitable for the unambiguous assay of the enzyme are known. The detailed understanding of the mechanism and substrate-specificity of C3Hdemands a method for the production of active protein on the milligram scale. We have developed a bacterial expression and purification system for the plant C3H, which allows for the quick expression and purification of active wild-type C3H via introduction of combinational mutagenesis. The modified cytochrome P450 C3H ($C3H_{mod}$) could be purified in the absence of detergent using immobilized metal affinity chromatography and size exclusion chromatography following extraction from isolated membranes in a high salt buffer and catalytically activated. This method makes the use of isotopic labeling of C3H for NMRstudies and X-ray crystallography practical, and is also applicable to other plant cytochrome P450 proteins.
Chrysanthemum boreale M. (hereafter, C. boreale M.), a perennial flower, has been historically used as a natural medicine in Korea. With increasing concerns for health-improving foods, the demand for C. boreale M. has become higher than ever. Howevr, the amount of wild C. boreale M. collected from mountainous areas is not enough to cover all demands. The cultivation system and fertilization strategy are required to meet increasing demand on C. boreale M. with a good quality. We investigated the effects of nitrogen application on plant growth and effective components of C. boreale M. to suggest optimum rate of nitrogen fertilization. C. boreale M. was cultivated in a pot scale (1/2000a scale), and nitrogen applied with rate of 0(N0), 50(N50), 100(N100), 150(N150), 200(N200), and $250(N250)kg\;ha^{-1}$. Phosphate and potassium were applied at the same level ($P_2O_5-K_2O=80-80kg\;ha^{-1}$) in all treatments. Maximum yield achieved in 246 and $226kg\;ha^{-1}$ N treatment on the whole plant and the flower part, a valuable part as a herbal medicine, respectively. Proline was the most abundant amino acid in the flower of C boreal M. and the contents of amino acids increased with increasing nitrogen application rate in flower. Nitrogen recovery efficiency was high more than 41% in all nitrogen treatments and increased to 61.8% in nitrogen N100 treatment. From the nitrogen content, the high nitrogen uptake, the low residue of mineral N and the reasonably good apparent fertilizer recovery, it can be inferred that C. boreale M. made efficient use of the available nitrogen. In flower, contents of Cumambrin A. which is a sesquiterpene compound and has the effect of blood-pressure reduction, decreased with increasing nitrogen application. However, the amount of Cumambrin A in flower increased as nitrogen rate increased, because of increasing flower yield. Conclusively, nitrogen fertilization could increase yields and enhance quality. The optimum nitrogen application rate might be on the range of $225{\sim}250kg\;ha^{-1}$ in a mountainous soil.
Heteropappus arenarius Kitam., an autumn-flowering biennial belonging to wild chrysanthemums, is found to be native in southeastern coastal area and Jeju island of Korea. It could play a good role for ground cover plants on a large-scale landscape area, especially, barren soil or sloping hillside. This study was initiated to screen optimum germination temperature influenced by local strain and harvesting stage of H. arenarius. The following was the response of seed germination between local strain and temperature. The average of final germination percentage (FG) was the highest in 'Guryongpo' (89.7%) among four local strains, followed by 'Gujwa' (87.3%), 'Gampo' (87.3%), and 'HKNU-I' (71.5%). The average of $T_{50}$ was shorter in 'Gujwa' (3.6 d) and 'Guryongpo' (4.0 d) than the others. The average of FG and $T_{50}$ was the highest as 76.2% and shortest as 3.6 d in $20^{\circ}C$, respectively, followed by $30^{\circ}C$, $25^{\circ}C$, and $15^{\circ}C$. In case of 'Gujwa', however, FG and T50 was higher in $20^{\circ}C$ and shorter in $15^{\circ}C$ than others. In the relationship between harvesting stage and temperature, the average of FG was greatly higher in Stage III (90.7%) and Stage IV (88.6%) than the others including Stage II (35.7%) and Stage I (26.0%). The average of $T_{50}$ was shorter in Stage IV (3.7 d) and Stage III (4.3 d) than the others, which showed less than 50% of FG. Nevertheless, the available range of seed harvesting stage was from Stage I to Stage IV because H. arenarius seeds could germinate at all stages. In conclusion, it was recommended that the optimum temperature and harvesting stage was $20^{\circ}C$ and Stage $III{\sim}IV$, respectively, for seed germination of H. arenarius.
Owing to the lack of consistent research on endangered plant species in Korea, there are insufficient data to preservespecies and expand habitats. This study analyzed the preferred habitat and threats to the survival of Aconitum austrokoreense, found on Baekwun Mountain in Gurye-gun, Gwangyang-si, Jeollanam-do Province, and classified as a level two endangered wild plant by the Ministry of Environment, by investigating major environmental factors such as climate, location, soil, and stand structure. By examining five selected sites inhabited by Aconitum austrokoreense on BaekwunMountain, this study found that the habitat had an altitude of 420 to 675 m above sea level and showed a northeast tendency, spreading over a range of inclination angles between 15° and 37°. The average number of plants across the five sites was 156. Site 4 (550 m) had the highest density of 372 plants, with an average height of 0.6 m. The average soil moisture and relative light intensity were 20.48% and 7.34%, respectively. Layer soil was presumed to be sandy loam, characterized by high sand content and good drainage. The habitat had average soil pH of 5.2, average organic matter of 16.46%, average nitrogen of 0.86%, average available phosphate of 11.86 mg/kg, average electrical conductivity of 0.44 dS/m, and average cation exchange capacity of 37.04 cmolc/kg. The total carbon in soil averaged 10.68%. From the analysis of the vegetation structure of sites inhabited by Aconitum austrokoreense, the dominant populations were Pinus koraiensis and Lindera erythrocarpa in Site 1, Magnolia obovata and Carpinus laxiflora in Site 2, Zelkova serrate and Quercus variabilis in Site 3, Staphylea bumalda and Lindera erythrocarpa in Site 4, and Morus bombycis,Styrax japonicus, and Carpinus laxiflora in Site 5. With most habitats located near trails and sap collection sites of Acer pictum, the species were exposed to artificial damage and interference threats.

제 5 장 계약 해지 및 이용 제한

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
