Projections from the Prefrontal Cortex to the Dorsal Raphe Nucleus of the Rat

  • Projections from the prefrontal cortex to subdivisions of the dorsal raphe nucleus were investigated in the rat using retrograde and anterograde tracing methods. A retrograde tracer, gold-conjugated horseradish peroxidase (WGA-apo-HRP-gold), was injected into each subdivision of the dorsal raphe including lateral wing, dorsomedial, and ventromedial areas. The majority of retrogradely labeled cells were located in the prelimbic, infralim-bic, and dorsal peduncular areas of the medial prefrontal cortex. A few cells were also identified in the cingulate, various regions of the orbital, and agranular insular cortices. Secondly, an anterograde tracer, Phaseolus vulgaris leucoagglutinin (PHA-L), was injected into the medial prefrontal cortex involving the prelimbic or infralimbic areas. Axonal fibers with varicosities were identified in all subdivisions of the DR including the lateral wing, dorsomedial, and ventromedial areas. Projections were bilateral, with ipsilateral predominance. Axonal fibers were observed at the lateral border of medial longitudinal fasciculus or in the interfascicular region at the midline. The present findings demonstrate that both the midline and lateral wing regions of the dorsal raphe nucleus receive excitatory input from cognitive and emotional centers of the cerebral cortex.

Descending Projections from the Prefrontal Cortex to the Locus Coeruleus of the Rat

  • The fiber projection from the prefrontal cortex to the locus coeruleus (LC) in the periventricular region was analyzed in rat using anterograde and retrograde tracing methods. Following injection of an anterograde tracer, Phaseolus vulgaris leucoagglutinin (PHA-L), into prelimbic and infralimbic regions of the medial prefrontal cortex, labeled axonal fibers with varicosities were observed bilaterally within the LC, with ipsilateral predominance. Terminal labeling was also observed in the region medial to the nucleus at rostral to middle levels of the LC, whereas axonal labeling in the caudal LC was minimal. Anterogradely-labeled axonal fibers were not found in the subcoerulear region. A retrograde tracer, gold-conjugated and inactivated wheatgerm-agglutinin horseradish-peroxidase (WGA-apo-HRP-gold), was injected into several rostro-caudal levels of the LC. Majority of retrogradely-labeled cells were observed in the prelimbic or infralimbic regions of the medial prefrontal cortex when the injections were made into rostral to middle levels of the LC. Only a few cells were observed in cingulate, dorsal peduncular, orbital, or insular cortices. The present findings suggest that the nucleus LC receives restricted, excitatory inputs from cognitive, emotional, and autonomic centers of the cerebral cortex and might secondarily have influences on widespread brain regions via its diversified monoaminergic innervation.

MORPHOLOGICAL ANALYSIS OF DEVELOPMENTAL CHANGES IN SOMA AREA OF DIGASTRIC MOTONEURONS IN THE RAT TRIGEMINAL MOTOR NUCLEI (흰쥐의 두힘살근 운동신경세포의 발달과정에 관한 형태학적 분석)

  • To analyze the developmental changes in soma diameters of digastric motoneurons, wheat-germ agglutinin conjugated horseradish peroxidase (WGA-HRP) was injected into the digastric muscle and visualized the retrogradely HRP-labeled motoneurons through tungstate/tetramethylbenzidine (TMB) and following diaminobenzidine (DAB) reactions. The results obtained from Sprague-Dawley rats at postnatal days 1 (P1), 10 (P10) and 30 (P30) indicated as follows: firstly, soma diameters of digastric motoneurons showed unimodal distribution in all postnatal days examined; secondly, the period of P1 to P10 (period 1) showed about 2 times faster growth rate than that of P10 to P30 (period 2); thirdly, the smallest soma examined in each postnatal day exhibited slower growth rate with that of the largest one (increase ratio in soma diameters from P1 to P30, smallest vs. largest = 1.62 : 1.93); Finally, relative growth rates a day showed again that period 1 had faster growth rate than that of period 2. Consequently, developmental changes in soma diameters of digastric motoneurons resulted in very different growth rates between both periods. This implies that the growth of the soma is almost completing within P10 and thereafter growing slowly. The period 1 and 2 are corresponding to sucking and sucking/masticatory period, respectively. Therefore present study providing morphological changes in soma diameters of digastric motoneurons suggests that both periods and their different growth rates of the motoneurons in each period may closely be related with each other.

  • We developed a new panel of markers for the characterization of chicken PGCs. The results of immunostaining demonstrated that anti-SSEA-3, anti-SSEA-4, anti-integrin 6, and anti-integrin 1 antibodies. and STA and DBA bound specifically to chicken PGCs. These reagents could be used to characterize chicken PGCs together with conventional marker reagents such as PAS and anti-SSEA-1 antibody. We also showed that double staining of PGCs with the newly developed markers was feasible, which might contribute to rapid detection and accurate characterization of chicken PGCs.

Development of Multiplex Polymerase Chain Reaction Assay for Identification of Angelica Species (Multiplex Polymerase Chain Reaction을 이용한 당귀 종 판별)

  • Background: Angelica gigas, A. sinensis, and A. acutiloba are commercially important in the herbal medicine market, and among them, A. gigas has the highest economic value and price. However, their similar morphological traits are often used for fraud. Despite their importance in herbal medicine, recognition of the differences between Angelica species is currently inadequate. Methods and Results: A multiplex polymerase chain reaction (PCR) method was developed for direct detection and identification of A. gigas, A. sinensis, and A. acutiloba. The gene for the distinction of species was targeted at ITS in the nucleus and trnC-petN gene in chloroplasts. The optimized multiplex PCR in the present study utilized each Angelica species-specific primer pairs. Each primer pair yielded products of 229 base pairs (bp) for A. gigas, 53 bp for A. sinensis, 170 bp for A. acutiloba. Additionally non-specific PCR products were not detected in similar species by species-specific primers. Conclusions: In the present study, a multiplex-PCR assay, successfully assessed the authenticity of Angelica species (A. gigas, A. sinensis, and A. acutiloba). and whole genome amplification (WGA) was performed after DNA extraction to identify, the species in the product. The detection method of raw materials developed in the present study could be applied to herbal medicine and health functional food management.

Glycoantigen Biosyntheses of Human Hepatoma and Colon Cancer Cells are Dependent on Different N-Acetylglucosaminyltransferase-III and -V Activities

  • UDP-N-Acetylglucosamine(GlcNAc):$\beta$1,4-D-mannoside$\beta$-l ,4N-acetylglucosaminyltransferase-III (GnT-III) and UDP-N-GlcNAc:$\alpha$-6-D-mannosid$\beta$-1,6N-acetylglucosaminyltransferase-V(GnT - V) activities were determined in human hepatoma cell lines and metastatic colon cancer cells, and their activities were compared with those of normal liver cells and fetal hepatocytes. GnT-III activities were higher than those of GnT-V in hepatic carcinoma cells. When the two enzyme activities were assayed in highly metastatic colon cancer cells, GnT - V activities were much higher than those of GnT-III. When GlcN, GlcN-biant-PA and UDP-GlcNAc were used as substrates, the enzymes displayed different kinetic properties between hepatic and colon cancer cells, depending on their metastatic potentials. Normal cells of two origins had characteristically very low levels of GnT-III and -V activities, whereas hepatoma and colon cancer cells contained high levels of activities. These data were supported by RT-PCR and Northern blot analyses, showing that the expression of GnT-III and -V mRNAs were increased in proportion to the enzymatic activities. The increased GnT-III, md -V activities were also correlated with increased glycosylation of the cellular glycoproteins in hepatoma and colon cancer cells, as examined by lectin blotting analysis by using wheat germ glutinin (WGA), erythroagglutinating phytohemagglutinin (E-PHA), leukoagglutinating phytohemagglutinin (L-PHA), and concanavalin A (Con A). Treatment with retinoic acid, a differentiation agent, resulted in decreases of both GnT-III and -V activities of HepG2 and HepG3 cells. In colon carcinoma cells, however, treatment with retinoic acid resulted in a reduction of GnT-V activity, but not with GnT-III activity. Although the mechanism underlying the induction of these mzymes is unclear, oligosaccharides in many glycoproteins have been observed of cancer cells.

Identification of Faulty Red Pepper Powder Containing Seasoned Red-pepper Sauce (고추다대기 혼입 불량고춧가루 판별법 개발)

  • In this study, the experimental method has been investigated using molecular biological way to identify raw materials from seasoned red-pepper sauce which is one of the most popular spices in Korea. 6 kinds of seasoned red-pepper sauces were chosen as a sample containing chilli pepper, garlic, onion as a major ingredient and species specific primers were used for the identification of the raw material of processed food. Selected samples were pre-treated to remove salt (samples were washed with distilled water 3~4 times for desalting), after that, to amplify the extracted genes, whole genome amplification (WGA) kit was performed. Afterwards, PCR products were confirmed through the electrophoresis. As a result, 102, 180, 280 bp of specific PCR products were confirmed for each major ingredients such as chilli pepper, garlic, onion. From this study, the gene extraction method was validated for the identification of ingredients from the spices and it would be applied to distinction of low quality chilli pepper powder including seasoned red-pepper sauce illegally.

Effects of Electroacupuncture on the Alteration of Inflammation-related Proteins and Glycoconjugates in the Ankle Joint of Complete Freund's Adjuvant-injected Rats (전침이 Complete Freund's Adjuvant를 주사한 흰쥐의 거퇴관절내 염증관련 단백질 및 복합당질 변화에 미치는 연구)

  • 목적 : 만성 염증성 질환에 대한 전침효과를 알아보기 위해 complete Freund's adjuvant(CFA) 유발 관절염 모델의 거퇴 관절 내 염증관련 단백질 및 복합당질의 변화를 살펴보았다. 방법 : Sprague-Dawley계 흰쥐의 족부에 CFA를 주사한 다음 3일 간격으로 2Hz, 15Hz 및 120Hz 전침자극을 주며 부종 형성여부를 plethysmometer로 측정하여 판정하였으며 30일째 거퇴관절을 취하여 4% paraformaldehyde에 고정하고 EDTA용액에서 탈회시켜 파라핀연속 절편을 얻어 $NF-{\kappa}B$를 비롯한 5종의 염증관련 단백질의 발현 및 복합당질 변화를 살펴보았다. 결과 : 관절연골 내 면역반응 중 연골기질은 반응이 없거나 약하고 연골세포는 $NF-{\kappa}Bp65$, $I-{\kappa}B{\alpha}$, iNOS 반응이 강하며 특히 유리연골층에서 더 현저하였으나 염증 및 전침자극에 따른 변화는 없었다. 관절낭에서 면역반응을 살펴보면 염증유발시 활액세포의 면역반응세포는 $I-{\kappa}B{\alpha}$가 감소한 반면 iNOS, $IL-1{\beta}$는 증가하며 특히 iNOS 증가가 현저하였으며 전침자극에 의해 iNOS 가 감소하였다. 활액막조직에서 모든 면역반응이 증가하며 특히 $NF-{\kappa}Bp65$, $I-{\kappa}B{\alpha}$, iNOS 반응이 현저한데 전침자극에 의해 $IL-1{\beta}$를 제외한 모든 반응이 감소하였다. 복합당질 염색성은 CFA를 주사한 염증유발 흰쥐군이 정상군에 비해 감소하였다. 관절연골 중 구역간질의 중성복합당질 및 연골세포피막의 산성복합당질이 현저히 감소하였다 Lectin반응도 DBA을 제외한 모든 발현이 염증유발시 감소하였다. 그러나 전침처리에 의해 정상군과 유사한 염색성과 lectin반응을 유지하였다. 특히 구역간질의 중성복합당질과 연골세포의 sWGA와 RCA-1 반응이 현저하였다. 결론 : 만성 염증성 동물모델의 거퇴 관절 내 염증관련 단백질은 관절연골보다 관절낭에서, 복합당질의 변화는 관절연골에서 큰 변화를 보였으며 전침의 자극에 의해 이들 변화가 억제되는 것을 알 수 있다. 이상의 결과로 보아 전침처치는 염증관련 단백질 발현 및 복합당질의 변화 억제를 통해 만성 관절염 질환에 효과적임을 알 수 있다.

Whole Genome Association Study to Detect Single Nucleotide Polymorphisms for Body Conformation Traits in a Hanwoo Population

  • A whole genome association (WGA) study was conducted to identify quantitative trait loci (QTL) for body conformation traits in Hanwoo cattle. The phenotypes of 497 steers were recorded from the Hanwoo Improvement Center of National Agricultural Cooperative Federation, Seosan, Korea, and analyzed using the Illumina Bovine 50 k SNP chip. A set of 35,987 SNPs that were available in the Hanwoo population was selected from the chip. After adjustments for the effects of year-season of birth, region and sire, phenotypes were regressed on each SNP using a linear regression model. Three hundred nineteen SNPs were detected for the ten conformation traits (p<0.003). For the significant SNPs, stepwise regression procedures were applied to determine best sets of markers. A total of 72 SNPs were selected (p<0.001), for which the sets of 5, 9, 10, 9, 8, 11, 4, 6, 3 and 7 SNPs were determined for height at withers, rump height, body length, chest depth, chest width, rump length, hip width, thurl width, pinbone width and heart girth, respectively. About 7-26% of the total phenotypic variation was explained by the set of SNPs for each trait. QTL for the conformation traits were harbored on most bovine chromosomes (BTAs). Four SNPs with pleiotropic effects on height at withers and rump height were detected on BTAs 3, 4, 6 and 16. A SNP with pleiotropic effects on chest width and rump length was also detected on BTA10. Two QTL regions, i.e. between 87 and 97 Mb in BTA3 and between 41 and 44 Mb in BTA7, were found, in which SNPs were detected for the five and three conformation traits, respectively. The detected SNPs need to be validated in other Hanwoo populations for commercial application to the genetic improvement of conformation characteristics in Hanwoo via marker-assisted selection (MAS).

The Shape of Synaptic Vesicles in the Tooth Pulp Afferent Terminals and P-endings in the Rat Trigeminal Nucleus Principalis (흰쥐의 삼차신경주감각핵에서 치수유래 일차들신경종말 및 이와 연접하는 연접이전신경종말의 연접소포 형태 비교)

  • In order to investigate shape of synaptic vesicles of the tooth pulp afferent boutons and their presynaptic endings (p-endings), and the neuroactive substance of the p-endings in the trigeminal nucleus principalis, rat incisor tooth pulp afferents were labeled by the horseradish peroxidase (HRP) and quantitative ultrastructural analysis and postembedding immunogold labeling were performed. Labeled tooth pulp afferent boutons contained clear, spherical synaptic vesicles (diameter: $45{\sim}55\;nm$) and occasionally dense core vesicles(diameter: $80{\sim}120\;nm$). They formed symmetrical synapses with unlabeled axon terminals (p-endings) containing pleomorphic synaptic vesicles. The ratio of short to long diameter (form factor) of synaptic vesicles of pulp afferent boutons was 0.6 to 0.99, whereas that of p-endings was 0.25 to 0.99. In addition, most of the p-endings showed GABA-like immunoreactivity. These results indicate that the shape of synaptic vesicles is quite different between the tooth pulp afferent boutons and p-endings, and the p-endings may contain GABA as a neuroactive substance in the trigeminal nucleus principalis.
