Protective Effects of Chrysanthemi Indici Flos Extract and Flaxseed Oil Mixture on HCl/ethanol-induced Acute Gastric Lesion Mice (급성 위염 동물 모델에서 감국(甘菊) 추출물과 아마인유(亞麻仁油) 혼합물의 위 점막 보호 효과)

  • Objectives : The objective of this study was to investigate the protective effect of Flaxseed oil and Chrysanthemi Indici Flos 50% ethanol extract in an HCl/ethanol induced acute gastritis model. Methods : ICR mice were divided into 6 groups; normal mice (Nor), gastritic mice with distilled water (Veh), gastritic mice with 10 mg/kg sucralfate (SC), gastritic mice with 16 g/㎏ Flaxseed oil (FO), gastritic mice with FO + 50 mg/kg Chrysanthemi Indici Flos (FCL), and gastritic mice with FO + 100 mg/kg Chrysanthemi Indici Flos (FCH). Then, mice were orally administered with 150 mM HCl/60% ethanol and caused acute gastritis. After 1 hr, mice were sacrificed, and blood and stomach tissue were collected. Results : Administration of FCL and FCH to mice prior to the induction of gastritis was found to reduce gastric injury. reactive oxygen species (ROS) and peroxy nitrite ($ONOO^-$) levels of stomach tissues were significantly decreased in FO, FCL, and FCH compared to Veh group. As results of stomach protein analyses, FCL and FCH effectively reduce inflammatory-related factors such as inducible nitric oxide synthase (iNOS), interleukin-6 (IL-6), and interleukin 1 beta ($IL-1{\beta}$) in gastric lesion mice. In addition, nuclear factor kappa B p65 ($NF-{\kappa}B$ p65) and phosphorylation inhibitor of nuclear factor kappa $B{\alpha}(p-I{\kappa}B{\alpha})$ were down-regulated in FCL and FCH administrated gastric lesion mice. Conclusions : These results suggest that FCL and FCH has an inhibitory effect against gastric injury. Therefore, FCL and FCH has the potential to be used as a natural therapeutic drug.

Epigallocatechin-3-gallate suppresses hemin-aggravated colon carcinogenesis through Nrf2-inhibited mitochondrial reactive oxygen species accumulation

  • Background: Previous studies have presented evidence to support the significant association between red meat intake and colon cancer, suggesting that heme iron plays a key role in colon carcinogenesis. Epigallocatechin-3-gallate (EGCG), the major constituent of green tea, exhibits anti-oxidative and anti-cancer effects. However, the effect of EGCG on red meat-associated colon carcinogenesis is not well understood. Objectives: We aimed to investigate the regulatory effects of hemin and EGCG on colon carcinogenesis and the underlying mechanism of action. Methods: Hemin and EGCG were treated in Caco2 cells to perform the water-soluble tetrazolium salt-1 assay, lactate dehydrogenase release assay, reactive oxygen species (ROS) detection assay, real-time quantitative polymerase chain reaction and western blot. We investigated the regulatory effects of hemin and EGCG on an azoxymethane (AOM) and dextran sodium sulfate (DSS)-induced colon carcinogenesis mouse model. Results: In Caco2 cells, hemin increased cell proliferation and the expression of cell cycle regulatory proteins, and ROS levels. EGCG suppressed hemin-induced cell proliferation and cell cycle regulatory protein expression as well as mitochondrial ROS accumulation. Hemin increased nuclear factor erythroid-2-related factor 2 (Nrf2) expression, but decreased Keap1 expression. EGCG enhanced hemin-induced Nrf2 and antioxidant gene expression. Nrf2 inhibitor reversed EGCG reduced cell proliferation and cell cycle regulatory protein expression. In AOM/DSS mice, hemin treatment induced hyperplastic changes in colon tissues, inhibited by EGCG supplementation. EGCG reduced the hemin-induced numbers of total aberrant crypts and malondialdehyde concentration in the AOM/DSS model. Conclusions: We demonstrated that EGCG reduced hemin-induced proliferation and colon carcinogenesis through Nrf2-inhibited mitochondrial ROS accumulation.

Analysis on Results and Changes in Recent Forecasting of Earthquake and Space Technologies in Korea and Japan (한국과 일본의 지진재해 및 우주이용 기술예측에 대한 최근의 변화 분석)

  • This study analyzes emerging earthquake and space use technologies from the latest Korean and Japanese scientific and technological foresights in 2022 and 2019, respectively. Unlike the earthquake prediction and early warning technologies presented in the 2017 study, the emerging earthquake technologies in 2022 in Korea was described as an earthquake/complex disaster information technology and public data platform. Many detailed future technologies were presented in Japan's 2019 survey, which includes largescale earthquake prediction, induced earthquake, national liquefaction risk, wide-scale stress measurement; and monitoring by Internet of Things (IoT) or artificial intelligence (AI) observation & analysis. The latest emerging space use technology in Korea and Japan were presented in more detail as robotic mining technology for water/ice, Helium-3, and rare earth metals, and manned station technology that utilizes local resources on the moon and Mars. The technological realization year forecasting in 2019 was delayed by 4-10 years from the prediction in 2015, which could be greater due to the Corona 19 epidemic, the declaration of carbon neutrality in Korea and Japan in 2020 and the Russo-Ukrainian War in 2022. However, it is required to more active research on earthquake and space technologies linked to information technology.

Coix lacryma-jobi var. mayuen Stapf Sprout Extract Ameliorates High-Fat Diet-Induced Obesity by Upregulating LKB1/AMPK Signaling (LKB1/AMPK 신호 전달 경로의 활성화로 인한 새싹율무 열수 추출물의 항비만 효과)

  • Objectives : AMP-activated protein kinase (AMPK) is a key metabolic regulator that reduces lipogenesis. AMPK is mainly activated via phosphorylation of liver kinase B (LKB) 1 under energy stress. Here, we highlighted the anti-obesity effect and underlying mechanism of Coix lacryma-jobi var. mayuen Stapf sprout water extract (CSW) sprout extract in connection with the LKB1/AMPK signaling pathway. Methods : C57BL/6 mice (20~25 g) fed HFD to induce obesity and at the same time administered CSW 100 mg/kg (CSWL; (CSWL; CSW low concentration) or CSW 200 mg/kg (CSWH; CSW high concentration) or Garcinia extract (Garcinia) 200 mg/kg orally for 6 weeks. Body weight and food intake were measured at the same time each day. After 6 weeks of CSW administration, liver tissue and serum were obtained through an autopsy. After the end of the experiment, biochemical analysis (triglycerides (TG), total cholesterol (TC), HDL-cholesterol, and LDL-cholesterol) was performed on the serum. And then, protein levels related to TG and TC synthesis were measured through western blot analysis in liver tissue. Results : As a result, serum TG, TC, and LDL-cholesterol levels were significantly increased in the control group and significantly decreased in the CSW administration group. On the other hand, the HDL-cholesterol level was increased in the CSW-administered group. And as a result of Western blot analysis, CSW significantly increased the phosphorylation of LKB1 & AMPK, and remarkably decreased the expression of factors related to TG and TC synthesis. Conclusions : Our findings suggest that CSW influences the TG and TC synthesis to positively affect HFD-induced obesity in C57BL/6 mice.

Fracture Behaviors of Jointed Rock Model Containing an Opening Under Biaxial Compression Condition (이축압축 조건에서 공동이 존재하는 유사 절리암반 모델의 파괴 거동)

  • Underground construction such as tunneling can induce damages on the surrounding rock mass, due to the stress concentration of in situ stresses and excessive energy input during construction sequence, such as blasting. The developed damage on the rock mass can have substantial influence on the mechanical and hydraulic behaviors of the rock masses around a tunnel. In this study, investigation on the generation of damage around an opening in a jointed rock model under biaxial compression condition was conducted. The joint dip angles employed are 30, 45, and 60 degrees to the horizontal, and the synthetic rock mass was made using early strength cement and water. From the biaxial compression test, initiation and propagation of tensile cracks at norm to the joint angle were found. The propagated tensile cracks eventually developed rock blocks, which were dislodged from the rock mass. Furthermore, the propagation process of the tensile cracks varies with joint angle: lower joint angle model shows more stable and progressive tensile crack propagation. The development of the tensile crack can be explained under the hypothesis that the rock segment encompassed by the joint set is subjected to the developing moment, which can be induced by the geometric irregularity around the opening in the rock model. The experiment results were simulated by using discrete element method PFC 2D. From the simulation, as has been observed from the test, a rock mass with lower joint angle produces wider damage region and rock block by tensile cracks. In addition, a rock model with lower joint angle shows progressive tensile cracks generation around the opening from the investigation of the interacted tensile cracks.

  • In the preliminary investigation (Park et al., 2009), the use of compressional wave velocity and its measurement techniques were proposed as a new quality control measure for trackbed fills. The methodology follows exactly the same procedure as the density control, except the density being replaced by the compressional wave velocity involving consistently with resilient modulus of design stage. The specifications for the control also include field compaction water content of optimum moisture content ${\pm}2%$ as well as the compressional wave velocity. In this sequel paper, crosshole and resonant column tests were performed as well direct-arrival method and laboratory compressional wave measurements to verify the practical applicability of a methodology far the new quality control procedure based upon compressional wave velocity. The stress-modified crosshole results reasonably well agree with the direct-arrival values, and the resonant column test results also agree well with the field crosshole results. The compressional wave velocity turned out to be an excellent control measure for trackbed fills both in the theoretical and practical point of view.

Evaluation of Future Hydrologic Risk of Drought in Nakdong River Basin Using Bayesian Classification-Based Composite Drought Index (베이지안 분류 기반 통합가뭄지수를 활용한 낙동강 유역의 미래 가뭄에 대한 수문학적 위험도 분석)

  • Recently, the frequency and intensity of meteorological disasters have increased due to climate change. In South Korea, there are regional differences in vulnerability and response capability to cope with climate change because of regional climate characteristics. In particular, drought results from various factors and is linked to extensive meteorological, hydrological, and agricultural impacts. Therefore, in order to effectively cope with drought, it is necessary to use a composite drought index that can take into account various factors, and to evaluate future droughts comprehensively considering climate change. This study evaluated hydrologic risk(${\bar{R}}$) of future drought in the Nakdong River basin based on the Dynamic Naive Bayesian Classification (DNBC)-based composite drought index, which was calculated by applying Standardized Precipitation Index (SPI), Streamflow Drought Index (SDI), Evaporate Stress Index (ESI) and Water Supply Capacity Index (WSCI) to the DNBC. The indices used in the DNBC were calculated using observation data and climate scenario data. A bivariate frequency analysis was performed for the severity and duration of the composite drought. Then using the estimated bivariate return periods, hydrologic risks of drought were calculated for observation and future periods. The overall results indicated that there were the highest risks during the future period (2021-2040) (${\bar{R}}$=0.572), and Miryang River (#2021) had the highest risk (${\bar{R}}$=0.940) on average. The hydrologic risk of the Nakdong River basin will increase highly in the near future (2021-2040). During the far future (2041-2099), the hydrologic risk decreased in the northern basins, and increased in the southern basins.

  • In this study, a series of column test as a way in order to make up for the weakness point of the conventional acceleration method were conducted to both propose the suction board drain method and grapes the specific improvement character of this method as a result of a sort of plastic drain board and a phase of vacuum pressure conditions. On this occasion, the study focused on computing the effective factors of the fittest Suction board drain method affected by each condition through confirming the settlement generated during the test, the water content reduction and stress increase effect occurred arising from the test, and the ratio of consolidation related to the improvement period. In accordance with the shape of core and that whether the core is attached to the filter(pocket or adhesion), the castle type of adhesion and the column type of pocket are more efficient than the others as a consequence of the test to find out the improvement effect depending on each drainage such as a castle type, coil type, harmonica type, column type of pocket and a castle of the adhesion. In case of the step suction pressure, the shorter the period of $-0.8\;kg/cm^2$ as a final step of the suction pressure is, the better the improvement is. In addition, the correlation between degree of consolidation per each suction pressure level and duration of application was drawn as a curve and the point of inflection on this curve was provided to determine the duration period to maximize the consolidation.

Evaluation of Salt Tolerance of Three Foliage Plant as affected by Salinity Concentration in Indoor Ornamental Hydroculture (실내 관상용 물재배에서 염분농도에 따른 3가지 관엽식물의 내염성 평가)

  • This study investigated the growth characteristics of Euonymus japonicus, Hedera helix, and Peperomia puteolata treated with different calcium chloride (CaCl2) concentrations to evaluate salt tolerance limits in hydroculture cultivation. Six concentrations of CaCl2 (0, 1, 2, 5, 10, and 15 g·L-1 referred to as Cont., C1, C2, C5, C10, and C15) were applied to solution - grown plant species. The survival rate, growth index, plant height, plant width, leaf width, leaf length, number of leaves, and relative chlorophyll contents were measured at monthly intervals. Euonymus japonicus, Hedera helix, and Peperomia puteolata survived up to C2, C5, and C10 at each CaCl2 concentration. The Euonymus japonicus was higher in the C1 treatment than in the Cont. for most growth characteristics. Hedera helix had the highest leaf width, leaf length, and number of leaves in the Cont., a significant difference was observed compared with the C1 treatment. The chlorophyll content did not differ significantly between the C5 and Cont. treatments. The leaf width and length of Peperomia puteolata were greater in the C2 and C1 treatments than in the Cont., whereas the number of leaves and chlorophyll content were the highest in C5. Dry weight analysis revealed that Euonymus japonicus, Hedera helix, and Peperomia puteolata were the lowest in the Cont. treatments. Euonymus japonicus was 74% in C15, and Hedera helix, and Peperomia puteolata were analyzed at approximately 37%- 50% and 9%-14%, respectively, regardless of the concentration in the CaCl2 treatment groups. In indoor hydroponic cultivation, the salt tolerance limit concentrations of Euonymus japonicus, Hedera helix, and Peperomia puteolata are 2, 5, and 10 g·L-1, respectively, indicating that hydroculture management techniques should be applied at higher concentrations.

  • This study aimed to evaluate the swelling behavior characteristics of KJ-II buffer blocks by performing numerical analysis of swelling pressure measurement experiments using the nonlinear elasticity model of COMSOL Multiphysics. The analysis was conducted under boundary conditions that included isotropic constraints and water injection pressure, mirroring the experimental settings. Validation of the numerical model was achieved by comparing its outputs with experimental results. The validated model was then used to simulate swelling deformations under unconfined conditions and to analyze swelling pressure as influenced by dry density and the geometric shape of the buffer material. The results accurately represented the swelling deformation observed during the saturation process and demonstrated that swelling pressure increases with higher dry density. Moreover, simulations concerning the geometric shape of the buffer material indicated a markedly faster rate of pressure increase in U-shaped samples compared to cylindrical ones. Analysis suggested that stress manifested preemptively near the internal edges of U-shaped samples during saturation. To enhance the simulation's fidelity to actual buffer material behavior, further refinement of the analysis model using a nonlinear elasticity model is recommended.
