IDENTIFICATION AND CHARACTERIZATION OF PHOSPHOLIPASE $A_2$ IN OAT CELLS

  • The activity of phospholipase A$_2$ (PLA$_2$) was identified and characterized from cytosolic and membrane fractions of oat cells, respectively. PLA$_2$ activity was determined fluorometrically in the presence of serum albumin using phospholipids labeled at sn-2-acyl position with 10-pyrenyldecanoic acid. When the cell-free extracts of oat tissues were fractionated by ultracentrifugation at 100,000 x g and the PLA$_2$ activity was assayed, we found that most of the PLA$_2$ activity was revealed from the cytoplasmic fraction rather than from the membrane fraction. The activity of cytosolic PLA$_2$ was dependent on Ca$^{2+}$ concentration and the optimum concentration of Ca$^{2+}$ was found to be 100 $\mu$M. It was also found that PLA$_2$ could be translocated toward the membrane site from the cytosol upon increasing Ca$^{2+}$ concentration. These results might suggest that an increased [Ca$^{2+}$]$_i$ by phytochrome action could promote the translocation of the cytosolic PLA$_2$ toward the membrane site.

  • This research has been produced by asking 274 department of health students and 232 students majoring industrial department from 27. Aug. 1997 to 05. Sep. 1997 and the results are as follows. 1. Satisfaction level on the contents was good, as you see the department of health part's level was 3.22 and the industrial part's level 3.14. 2. Satisfaction level on the practice circumstances was of low, department of health part's level was 3.25 and the industrial part's level was 2.82. 3. Satisfaction level on the length of practice time was as follows: Department of health part's level was 3. 25 and the industrial part's level was 3.08. 4. Satisfaction level on the teaching skill during practice was totally different each other: Department of health part's level was 2.10 and the industrial part's level was 2.76. 5. Satisfaction level on the evaluation and personal relationship was as below. Evaluation-department of health part's level was 2. 57 and the industrial part's level was 3.17. Personal Relationship-department of health part's level was 2.60 and the industrial part's level was 3.15. 6. Total satisfaction level was as follow: department of health part's level was 2.74 and the industrial part's level was 3.04. The satisfaction level for the student of industrial part was higher them that of health part.

  • This thesis compared and analyzed the characteristics, centered on a characteristic curve, about X-ray computerized tomographic film generally used in Korea recently. The results were as follows: 1. Maximum gradient (gamma) was film A-2.19, film B-2.00, film C-1.92. 2. Latitude was confined within the limits of density 0.6-2.0 in all. 3. When we changed window center from 0 to 40, the difference of density, which was in 1.0, was film A-0.12, film B-0.16, fil m C-0.14. 4. When we changed window width from 0 to 500, the difference of density, which was in 1.0, was film A-0.06, film B-0.08, film C-0.05. 5. In the case of window width 50 and window center 20, latitude was 0.5-2.0, 0.5-2.2, 0.5-1.9 and gamma value was 1.74, 1.82, 1.72 respectively, 6. In the case of window width 80 and window center 30, latitude was 0.6-2.2, 0.6-2.3, 0.8-2.2 and gamma value was 2.0, 2.13, 1.95 respectively.

Purification and Characteristics of Amylase from Haloarcular sp. EH-1 (Haloarcular sp. EH-1이 생산하는 Amylase의 정제 및 특성)

  • EH-1 was highest at 9 days of incubation. This regrowth and enzymatic activity of Haloarcular sp. EH-1 was highest at 9 days of incubation. This amylase was purified by acetone fractionation, DEAG-Cellulose column chromatography, 1st Sephadex G-75 gel filtration, CM-Cellulose column chromatography and 2nd Sephadex G-75 gel filtration. The amylase was purified about 98.64 fold with a yield of 11.75%. The molecular weight of amylase was estimated to be about 43,000and 40,000 by gel filtration and SDS-polyacrylamide gel electrophoresis, respectively, suggesting that the enzyme was a monomer. Amylase had an optimal temperature of 4$0^{\circ}C$, and an optimum pH of 7.0, and the thermal stability was observed the above 50% at 10$0^{\circ}C$ after 1 hour, and the stable range of pH was 6.0 to 8.0. The enzymatic activity was increased in the presence of 10 mM 2-mercaptoethanol, slightly by 10 mM SnCl2.2H2O.FeCl2.4H2O.CuCl2.2H2O.HgCl2.6H2O and SDS. End products from soluble starch were glucose, maltose and maltotriose, and Km value for soluble starch was 2.5mg/ml.

  • The degree of The degree of The degree of ${Zn}^{2+}$ effect on the photosynthetic electron transport and photophosphorylation activities in barley chloroplasts has been tested.${Zn}^{2+}$treatment was done in the 2 ways. One was that it was added into the chloroplasts suspensions isolated from the plants grown under the normal ${Zn}^{2+}$level (10$^{-6}$ M). The other was that the different concentrations of ${Zn}^{2+}$was applied in each growth medium. Then, it was not added into the chloroplasts suspensions isolated from the plants. PS II activity in both way of the treatments was more severely inhibited than PS I by the increment of ${Zn}^{2+}$ concentration. The photophosphorylation activity measured by pH measurement was gradually decreased with the increase of ${Zn}^{2+}$concentration in both ways, too. However, it was shown that M $n^{2+}$ could be near fully overcome the inhibitory effect of ${Zn}^{2+}$in PS II, and $Mg^{2+}$ could also reduce the Z $n^{2+}$ inhibition in the photophosphorylation. In the low concentrations of $Mg^{2+}$ (3 to 5$\times$10$^{-3}$ M) in the suspension, ${Zn}^{2+}$(2$\times$10$^{-5}$ M) could increase the activity of photophosphorylation. As compares to other cations, Z $n^{2+}$ caused less inhibitory effect on the photophosphorylation activity than Cu, Cd, but more than Pb and Ni. Some Physical and Chemical Properties of Synthesized Dawsonite (합성 Dawsonite의 물리적, 화학적 성질)

  • $NaAl(OH)_2CO_3$was synthesized using colloidal earth (Allophane) as the starting material and some of its were studied in detail. It was found that Dawsonite was formed in the pH range (pH 12.5~12.0) that the concentration of $HCO_3^-$ was just begun to increase and the presence of $HCO_3^-$ in the product was clarified from the infrared absorption spectrum. The chemical formular of Dawsonite was therefore presumed as $NaAlO (OH) HCO_3$. From toahhe results of X-ray powder diffraction, both peaks at 5.7 $\AA$ and 2.8 $\AA$ were observed, and fibrous crystalline structure was observed from electron micrograph and also found from the microscopic electron diffraction at 5.7 $\AA.$ Therefore the fibrous axis was considered as =Al=O2=Al=O2=Al=(*image) direction. True specific gravity of Dawsonite was 2.44 and its porosity was 91.4%. It was practically insoluble in water, but decomposed in the boiling water to form Pseudo Boehmite. Stable pH range of Dawsonite was about 4.5~11.5. From the results of D.T.A. and T.G.A., it was observed that $CO_2$was liberated at $350^{\circ}C$, and $H_2O$ at $650^{\circ}C$, and converted into strongly hygroscopic $NaAlO_2$, which was easily decomposed in water into $\beta-Al(OH)_3(Bayerite)$ and NaOH.

A Study on Airborne Concentrations of $SO_2$, TSP and Air Quality Standards of a Subway Stations (지하철 구내의 대기 중 $SO_2$, TSP 농도와 대기 허용기준에 관한 연구)

  • The airborne concentrations of Sulfur Dioxide and Total Suspended Particulates were investigated in 4 subway lines in Seoul at early summer, 1990 and early summer, 1991. The results of the study were as follows: 1. The airborne concentrations of SO$_2$ and TSP were 0.022 ppm, 445.7 $\mu$g/m$^3$ respectively. And percents of over annual air quality standard of WHO was that SO$_2$ and TSP were 41.7%, 100% respectively. 2. Airborne SO$_2$ concentrations by subway lines were that line 1 was 0.025 ppm, line 2 was 0.023 ppm, line 3 was 0.020 ppm, and line 4 was 0.017 ppm. And TSP concentrations by subway lines were that line 2 was 533.8 $\mu$g/m$^3$, line 1 was 516.5 $\mu$g/m$^3$, line 4 was 371.6 $\mu$g/m$^3$, and line 3 was 369.3 $\mu$g/m$^3$ 3. Annual variation of concentration of TSP was not significant statistically (t=0.327), and that of SO$_2$, in 1990 was slightly higher than that in 1991 (t=1.433, p<0.1). 4. Coefficients of correlation between TSP and SO$_2$ by years were that early summer, 1990 was r=0.277 (p>0.1), and early summer, 1991 was r=0.32 (p>0.1).

  • The hydration for the system of fluorogypsum and 20wt% blast furnace slag was investigated at liquid/soild ratio of 0.45 for 1, 3, and 7 days by using 3 kinds of accelerators such as K2SO4 Al2(SO4)3 $.$16-18H2O and AlK(SO4)2$.$12H2O After curing the hardened specimen was characterized by the compressive strength the content of combined water XRD, DTA and SEM It was found that the activating effect was increased in the order of K2SO4

  • The study was aimed at investigating sintering characteristics, the overall properties from the ternary Li2O-Al2O3-SiO2 system. This system, which was considered without additives and selected five compositions, was studied with variation of the amount of SiO2. The reactivity of the system was observed by D.T.A.. Characteristics of sintering, microstructure, property and stability of produced phase were studied. It was found that because the range of sintering was narrow and the temperature of sintering was close to the melting temperature, the sintering by the general method was difficult. Also, these linear thermal expansion coeffecients were measured.

  • The composition of base glass was selected as MgO 8, Al2O3 24, SiO2 68 in weight percent. TiO2 and ZrO2 were added to the base glass to investigate their effects as nucleating agents. In the case of ZrO2 addition, the optimum temperature for nucleation, which was related to the precipitation of tetragonal ZrO2, was 80$0^{\circ}C$. The optimum growth condition for the crystal was 87$0^{\circ}C$ for 8 hrs, and the major crystal phases precipitated in the samples were $\beta$-quartz ss. and mullite. The light transmissivity turned out to be around 80 per cent. On the other hand, when the TiO2 was added, it was difficult to determine the nucleating temperature, because the samples turned easily into translucency during the heat treatment. Therefore, it was almost impossible to retain transparency in the samples. The light transmissivity was below 30 per cent.
