Production of chickens with green fluorescent protein-knockin in the Z chromosome and detection of green fluorescent protein-positive chicks in the embryonic stage

  • Objective: The clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated protein 9 (Cas9) system, which is the most efficient and reliable tool for precisely targeted modification of the genome of living cells, has generated considerable excitement for industrial applications as well as scientific research. In this study, we developed a gene-editing and detection system for chick embryo sexing during the embryonic stage. Methods: By combining the CRISPR/Cas9 technical platform and germ cell-mediated germline transmission, we not only generated Z chromosome-targeted knockin chickens but also developed a detection system for fluorescence-positive male chicks in the embryonic stage. Results: We targeted a green fluorescent protein (GFP) transgene into a specific locus on the Z chromosome of chicken primordial germ cells (PGCs), resulting in the production of ZGFP-knockin chickens. By mating ZGFP-knockin females (ZGFP/W) with wild males (Z/Z) and using a GFP detection system, we could identify chick sex, as the GFP transgene was expressed on the Z chromosome only in male offspring (ZGFP/Z) even before hatching. Conclusion: Our results demonstrate that the CRISPR/Cas9 technical platform with chicken PGCs facilitates the production of specific genome-edited chickens for basic research as well as practical applications.

Identification and Phylogeny of the Human Endogenous Retrovirus HERV-W LTR Family in Human Brain cDNA Library and Xq21.3 Region

  • Human endogenous retroviral long terminal repeats (LTRs) have been found to be coexpressed with sequences of genes located nearby. It has been suggested that the LTR elements have contributed to the structural change or genetic variation of human genome connected to various diseases. The HERV-W family has been identified in the cerebrospinal fluids and brains of individuals with schizophrenia. Using a cDNA library derived from a human brain, the HERV-W LTR elements were examined and five new LTR elements were identified. These elements were examined using a YAC clone panel from the Xq21.3 region linked to psychosis that was replicated on the Y chromosome after the separation of the chimpanzee and human lineages. Fourteen elements of the HERV-W LTR were identified in that region. Those LTR elements showed a high degree of sequence similarity ($91.8-99.5\%$) with previously reported HERV-W LTR. A phylogenetic tree obtained from the neighbor-joining method revealed that new HERV-W LTR elements were closely related to the AXt000960, AF072504, and AF072506 from the GenBank database. The data indicates that several copy numbers of the HERV-W LTR elements exist on the Xq21.3 region and are also expressed in the human brain. These LTR elements need to be further investigated as potential leads to neuropsychiatric diseases.

  • 한국산 박새속 조류 Porus major(박새), Porus atter(진박새), Porus poiustris(쇠박새), Parus varius(곤줄박이)의 핵형을 일반염색 방법으로 분석한 결과 4종의 염색체 수는 모두 2n=78∼80으로 나타났고, 성 염색체를 포함한 7쌍이 macrochromosome, 그 외 32∼33쌍이 microchromosome이었다. 종간 차이를 나타내는 염색체는 5번째 염색체와성염색체인 Z·W-염색체였다. 이러한 핵형의 차이는 5번째 염색체에서는 pericentric inversion, 성 염색체에서는 전좌에 의한 것으로 생각된다. The chromosomal analysis of Pows major, Paws after, Paws palustris and Paws vorius of the genus Paws in Korea were performed by conventional Giemsa staining method. The diploid number of four species were 2n=78-80, and there were 7 pairs of macrochromosomes and 32 or 33 pairs of microchromosomes. The 5th and Z·W-chromosomeswere distinctly different between interspecies. Probably these karyological differences were speculated by pericentric inversion in 5th chromosome and translocation in Z·W-chromosomes.

Analysis of Single Nucleotide Polymorphism of MMP3 Gene in Korean Genome

  • MMP3 (Matrix metalloproteinase-3) is an important gene in the development of cardiovascular and metabolic diseases. It is also reported that the genotype of MMP3 could be a factor for disease conditions. So, SNP analysis is a prerequisite to study MMP3 related diseases. However, statistical data or analytical reports of this gene in the Korean population is not available. We have employed PCR and ARMS technique to amplify the position of Lys45Glu which is located within chromosome 11q22.3 and exon 2. Genomic DNA were extracted from 201 people. We found that, 17 individuals had the wild homozygote type (W/W, 8%), 98 individuals had the SNP homozygote type (S/S, 49%), 86 had the heterozygote type (W/S, 43%). This study should facilitate research on the cause of cardiovascular diseases due to polymorphisms in the MMP3 gene and to develop further therapy at the genetic level.

  • Genetics has played a pivotal role in increasing the world food production through revolutions in plant and animal sciences. Though the attention on fisheries has been inadequate but the growing importance of modern genetic manipulations and biotechnological innovations to aquaculture has been realized. Recent advances in fish genetics and molecular biology have provided a suite of useful techniques, which have several applications in aquaculture. This paper reviews the advancement in the applications of selection, hybridization, chromosome engineering, sex control, gene transfer and molecular technologies for enhanced aquaculture productivity.

FREQUENCY OF GIEMSA C-BAND CHROMOSOMES IN THREE INBRED LINES OF CHICKENS

  • Giemsa C-banded mitotic chromosome prepatations from White Leghorn, New Hampshire and Rhode Island Red inbred lines were compared for frequency of C-band regions on individual chromosomes. Except for autosomes 3, 6, 8 and 9 and W sex chromosomes, C-banding was extremely variable in other macrochromosomes. No divergence for C-band difference between homologous chromosomes of these lines was detected. Approximately 75% of the mitotic metaphase microchromosomes have recognizable C-band regions with the current technique.
