공간 차원에 관한 시각적 패턴 연구 - 황금비, 피보나치 수열, 프랙털 이론을 중심으로 - (Study on Visual Patterns about Spatial Dimensions - Centered on the Golden Ratio, Fibonacci Sequence, and Fractal Theory -)

  • This study intended arousal of other viewpoints that deal with and understand spaces and shapes, by describing the concept of 'dimensions' into visual patterns. Above all, the core concept of spatial dimensions was defined as 'expandability'. Then, first, the 'golden ratio', 'Fibonacci sequence', and 'fractal theory' were defined as elements of each dimension by stage. Second, a 'unit cell' of one dimension as 'minimum unit particles' was set. Next, Fibonacci sequence was set as an extended concept into two dimensions. Expansion into three dimensions was applied to the concept of 'self-similarity repetition' of 'Fractal'. In 'fractal dimension', the concept of 'regularity of irregularity' was set as a core attribute. Plus, Platonic solids were applied as a background concept of the setting of the 'unit cell' from the viewpoint of 'minimum unit particles'. Third, while 'characteristic patterns' which are shown in the courses of 'expansion' of each dimension were embodied for the visual expression forms of dimensions, expansion forms of dimensions are based on the premise of volume, directional nature, and concept of axes. Expressed shapes of each dimension are shown into visually diverse patterns and unexpected formative aspects, along with the expression of relative blank spaces originated from dualism. On the basis of these results, the 'unit cell' that is set as a concept of theoretical factor can be defined as a minimum factor of a basic algorism caused by other purpose. In here, by applying diverse pattern types, the fact that meaning spaces, shapes, and dimensions can be extracted was suggested.

A study on Visual Expression to express Sound Characteristics of Public Places

  • The causes of noise generation according to the classification of indoor spaces are very diverse. Individual happiness is infringed by this noise. In this paper, We tried to visualize the spatial sound characteristics of public places using sound color to express them so that anyone can sympathize. The noise inside a conference room of a medical device company was measured for 100 minutes, and the frequency band was divided into three different types of existing sound pressure expression units. Because the size of the noise is expressed differently depending on the situation, There are cases where there is a difference of opinion between the measurer and the researcher. This noise measurement experiment was conducted, and the sound color was applied to classify it on a log scale considering auditory characteristics. As a result of comparing this with the result expression for different loudness expression units, A specific table in different units yielded almost similar results. In addition, the sound source section for 100 minutes was divided into three analysis sections, the analysis sections were different, and the size of the energy ratio for each analysis section was divided in the form of an envelope. The characteristics of the low-frequency region of the space have a high energy ratio, and the decrease in the energy ratio according to the increase in frequency is constant and regular. You can see that conversations are possible.

Electroporation을 이용한 잔디(Zoysia japonica Steud.) 및 벼(Oryza sativa L.) 배발생세포로의 DNA 도입 (DNA Delivery into Embryogenic Cells of Zoysiagrass(Zoysia japonica Steud.) and Rice(Oryza sativa L.) by Electroporation)

  • 간편하면서도 효율적인 단자엽 식물의 형질전환 기법을 개발하기 위하여 배발생 세포를 직접 electroporation하여 DNA를 도입하는 실험을 벼와 잔디에서 실시하였다. 잔디는 수정 후 3주된 미숙배에서 캘러스를 유도하였으며, 2.4-D가 1 mg/L 함유된 액체 MS배지로 옮겨 진탕배양한 것을 electroporation 실험에 이용하였다. 벼는 20 mm 정도의 미숙화서 유래의 캘러스를 액체 N$_{6}$배지(1 mg/L 2.4-D 함유)에서 진탕배양하여 획득한 세포주를 사용하였다. 액체 진탕배양한 세포괴를 GUS expression vector인 pGA1074 (30 $\mu\textrm{g}$/ml)와 함께 MS 액체 배지에서 Electroporation하였다. 세포벽과 세포막을 통한 세포로의 DNA 전이는 GUS 유전자의 발현 여부 및 정도에 따라 결정하였다. 400 volt, 800 $\mu$F capacitance로 electroporation 처리된 벼와 잔디의 세포괴들은 200 ${\mu}\ell$ (packed cell volume)의 세포괴 당 25 unit (1 unit=파란색을 띤 독립된 세포군) 이상의 빈도로 GUS 활성을 나타내었다. 반면에 무처리 세포주 및 처리한 비배발생 세포주에서는 GUS 발현이 일어나지 않음을 반복적으로 확인차였다. 따라서 electroporation에 의한 벼와 잔디의 형질전환실험에서 원형질체 대신 intact한 배발생 세포가 이용될 수 있음을 의미한다

  • Xanthomonas oryzae pv. oryzae (Xoo) is a plant pathogen, which causes a bacterial blight of rice. The bacterial blight is one of the most devastating diseases of rice in most of the rice growing countries and there is no effective pesticide against bacterial blight. The β-ketoacyl-acyl carrier protein synthase III (FabH) plays a key role in fatty acid synthesis (FAS) and is a promising drug target for the development of antibacterial agents. Xoo0878 gene, a fabH gene, from Xoo was cloned and its gene product Xoo0878 was expressed, purified and crystallized. Xoo0878 crystal diffracted to 2.1Å resolution and belonged to the triclinic space group P1, with unit-cell parameters a = 57.3Å, b = 64.7Å, c = 104.2Å and α = 81.6°, β = 84.7°, γ = 74.4°. There are four monomers in the asymmetric unit, with a corresponding crystal volume per protein weight of 2.65 Å3 Da-1 and a solvent content of 53.6%. Xoo0878 structure will be useful to develop new antibacterial agents against Xoo.

  • The nickel and cobalt resistance of Cupriavidus metallidurans CH34 is mediated by the CnrCBA efflux pump encoded by the cnrYHXCBAT metal resistance determinant. The products of the three genes cnrYXH transcriptionally regulate expression of cnr. CnrY and CnrX are membrane-bound proteins, probably functioning as anti-sigma factors, whereas CnrH is a cnr-specific extracytoplasmic functions (ECF) sigma factor. The periplasmic domain of CnrX (residues 29-148) was cloned as a N-terminal His-tagged protein, expressed in Escherichia coli, and purified using affinity chromatography and gel filtration. The molecular mass was estimated to be about 13.6kDa by size exclusion chromatography, corresponding to a monomer. The tetragonal bipyramid crystals were obtained by mixing an equal volume of protein in 50mM Tris-HCl, pH 7.5, 1% glycerol, 100mM NaCl, 1mM DTT, and the reservoir solution of 15% w/v PEG 2000, 100mM lithium chloride at 277K in 2-4 days using hanging drop vapor diffusion. The protein concentration was 24mg/ml. The crystal that diffracted to $2.42{\AA}$ resolution belongs to space group $P4_1\;or\;P4_3$ with unit cell parameters of $a=b=32.14{\AA},\;c=195.31{\AA},\;{\alpha}={\beta}={\gamma}=90^{\circ}$, with one molecule of CnrX in the asymmetric unit.

  • The Siuejunsuh is a book of Korean woman's life in the Yi-dynasty which published late in 18 century by unknown author. I have studied the food habits of the Yi-dynasty that wrote in siuejunsuh as following. The staple foods ae Bab (boiled cooked rice) 2, Juk(gruel) 6, Mieum(thin rice gruel) 3, Uee (liquid) 1, D'ockguk 2, Mandu3 Kinds I this book. the side dishes are Guk(soup) 24, Jochi 7, jungol 2, J'im 16, Sinsollo 1, Bockuem 5, Sun 6, Gui 16, Jock 10, jon 12, jorim 7, Namul 14, Sangchae 4, Hyae 17, Joban 2, Nooruemi 1, Sooranpoached egg) 2, Sondae 2, jockpyun 1, Choe 1, Muchim 11, Pyunuk 2, Jockgall 6, Po 9, Ssam 4, Jup 1, Jangaji 27, Kimchi 20, and other 27 Kinds. the desserts are D'ock 41, jabgwa 52, Beverages 17 kinds. the alcohol and frutie liquor are 18 kinds. The seasonings are soybean sauce, Jinjang, red peper bean paste, Zupjang, Dampukjang, Chunggukjang, honey, vinegar, mustard, Yunzup etc. Storage foods are vegetables,fruits and beans etc. There are 62 kinds of table ware and cooking kitchen utensiles, but many of them came to usefulness. 16 kinds of measuring unit are very non-scientific because that is not by weight but by bulk or volume. Thee are many food making terms which are 179 kinds of prepared cooking term, 22 kinds of cutting term and 16 kinds of boiling term. And 20 kinds of expression of taste can be seen in this book. Food serving methods are D'ockiserving method, fruits and dried food serving methode in chanhap (food packed in a nest of boxes) Guksoo serving method, Jock serving method, fruits serving method, Seup serving method etc. Table setting methods ae 9 sets of dinner, 7 sets of dinner, 5 sets of dinner, side table setting, liquor table setting, Sinsollo table setting, food's dab table setting etc.

"규호시의방"의 정리학적 고찰 (A Study on the Book "Gwngonsiuebang")

  • The Gwugonsiuebang is a book of Korean woman's life in the Yi dynasty which published I 1653 by Mme. Jang. I have studied the food habits of the Yi dynasty that wrote in Gwugonsiueband as following. The staple foods are Guksu (wheat vemicell as like western noodles) 5, Mandu (bun stuffed with seasoned meat and vegetables) 6 kinds in this book. the side dishes are Guk (soup) 8, J'm (steamed meat or fish) 6, chae6, Nooruemi 5, Hyae (sliced raw fish) 3, Jockpyun (jellied beef soup) 3, Jockgall (salted sea food) 2, Jock (skewer or broach) 2, jihee 2, Sun (Steaming of stuffed vegetable) 1, Bockuem (saute) 1, Jon (pan fried fish) 1, Gui (meat or fish grilled with seasoning) 1, and the other 13 kinds. The desserts are D,ock (Korean rice cake) 11, jabgwa 8, Beverages 5 kinds and Jungwa 1 kind. The alcohol and fruits wine are 51 kinds. The alcohol and fruits wine are 51 kinds. Now and then, there are no special difference of winter over pass for vegetables, fruits, dried beef, dried fish and salt fishes. In yi dynasty, there are 62 kinds of table ware and cooking kitchen utensils, but many of them come to uselessness. 19 kinds of measuring unit are very non-scientific because that is not by weight but by bulk or volume. There are many food making terms which are 198 kinds of prepared cooking term, 11 kinds of cutting term and 20 kinds of boiling term. And 10 kinds of expression of taste can see this book.

유수단면 흐름장에서 Shear 효과를 갖는 Stokes Drift의 계산법 (New Method of Computing the Stokes Drift Including Shear Effect in the Cross-Sectional Flow Field)

  • Stokes drift(SD) and Lagrangian discharge(LD) are important factors for analysis of flushing time, tidal exchange, solute transport and pollutant dispersion. The factors should be calculated using the approached method to flow phenomena. The aim of this paper re-examines the previous procedures for computing the SD and LD, and is to propose the new method approached to stratified flow field in the cross-section of coastal region, e.g. Masan Bay. The intensity of velocity near the bottom boundary layer(BBL) depends on the sea-bed irregularity in the coastal estuaries. So we calculated the depth mean velocity(DMV) considering that of BBL omitted in Kjerfve's calculation method. It revealed that BBL effect resulting in application of the bay acts largely on DMV in half more among 1l stations. The new expression of SD and LD per unit width in the cross-section using the developed DMV and proposed decomposition procedure of current were derived as follow : $$Q=u_0+\frac{1}{2}H_1{U_1cos(\varphi_h-\varphi_u)+U_3cos(\varphi_h-\varphi{ud})} LD ED SD$(Q_{skim}+Q_{sk2}) The third term, $Q_{sk2}$, on the right-hand of the equation is showed newly and arise from vertical oscillatory shear. According to the results applied in 3 cross-sections including 11 stations of the bay, the volume difference between proposed and previous SD was founded to be almost 2 times more at some stations. But their mean transport volumes over all stations are 18% less than the previous SD. Among two terms of SD, the flux of second term, $Q_{skim}$, is larger than third term, $Q_{sk2}$, in the main channel of cross-section, so that $Q_{skim}$ has a strong dependence on the tidal pumping, whereas third term is larger than second in the marginal channel. It means that $Q_{sk2}$ has trapping or shear effect more than tidal pumping phenomena. Maximum range of the fluctuation in LD is 40% as compared with the previous equations, but mean range of it is showed 11% at all stations, namely, small change. It mean that two components of SD interact as compensating flow. Therefore, the computation of SD and LD depend on decomposition procedure of velocity component in obtaining the volume transport of temporal and spacial flow through channels. The calculation of SD and LD proposed here can separate the shear effect from the previous SD component, so can be applied to non-uniform flow condition of cross-section, namely, baroclinic flow field.

거창 송계사(松溪寺) 목조여래좌상과 18세기 후반 조각승 상정(尙淨) 불상의 작풍(作風) 연구 (A Study on the Wooden Seated Buddha at Songgyesa Temple in Geochang and the Sculpting Style of Sangjeong, a Monk Sculptor in the Late 18th Century)

  • 상정(尙淨)은 18세기 중·후반에 활동한 조각승으로, 지금까지 알려진 수조각승 상정의 조상 활동 6건과 그가 조성한 기년 불상 중 마지막 작품으로 발견된 1767년 거창 송계사 <목조여래좌상>을 토대로 상정 불상의 전체적인 작풍을 설정하였다. 상정이 조성한 불상의 공통점은 귀의 생김새, 상·하반신 옷주름, 오른손이 놓인 위치, 하반신 표현 등에 나타나 있었다. 그 중 하반신 표현은 불상 하단에 표현된 연잎 사이로 하반신 옷주름이 흘러내리는 A유형, 연잎과 대좌 표현 없이 하반신에 'S'자형 옷주름만 형성된 B유형, 불신과 대좌를 일체형으로 조성하여 대좌에 표현된 연잎 사이로 하반신 옷주름이 흘러내리는 C유형 등 세 가지 유형으로 구분되는 양상을 확인할 수 있었다. 상정은 그가 유일하게 조각 수업을 받았던 스승 태원(泰元)의 작풍을 충실히 계승하였을 것으로 판단되었다. 이는 서울 봉은사 <목조석가여래삼존좌상 및 나한상> 조성 불사에서 수조각승 태원 아래에서 상정이 3위 조각승(3/12위)으로 참여한 기록과 봉은사 석가상에서 표현된 'S'자형 하반신 옷주름이 상정이 조성한 모든 불·보살상에 반영된 모습을 통해 확인할 수 있었다. 뿐만 아니라 상현좌 표현, 보발의 형상 또한 18세기 전반 조각승 진열(進悅)에서 태원을 거쳐 상정에게로 이어졌던 것으로 추정되었다. 상정과 태원 두 조각승의 작풍은 불상에서 나타나는 양감과 측면의 두께, 하반신 옷주름의 요철 강도, 오른쪽 가슴의 옷자락 형태, 귀의 세부적인 표현 등에서 차이점이 발견되었다. 이와 같은 과정을 통해 조각가 개인의 양식을 추출하였고, 상정의 작풍을 갖고 있는 불·보살상 10건 14점에 대해 상정 또는 태원(계) 등 원 조각가를 분류하는 작업을 시도하였다.

인터페론감마에 의한 A549 폐암세포주 세포독성의 기전 (The Mechanism of Interferon-$\gamma$ Induced Cytotoxicity on the Lung Cancer Cell Line, A549)

  • 연구배경: 인터페론감마-(interferon-$\gamma$)는 항바이러스 효과, 암세포의 형증식 효과, 대식세포 및 B 림프구의 활성화, 주면역복합체(MHC) 항원 발현의 증가 등의 생물학적 효과를 나타낸다. 특히, 인터페론감마의 항암 효과는 이미 생체 내외에서 입증되어 실제 폐암 환자에 대한 임상 연구가 시도되고 있다. 그러나, 인터페론감마의 항암효과 기전은 여러가지 가설이 제시되기는 하고 있지만 아직 확립된 것이 없다. 세포의 괴사(necrosis)는 심한 외부의 스트레스에 의해서 발생하는 세포 사망의 형태로 잘 알려져 있다. 생명현상 중 괴사와는 전혀 다른 세포 사망의 과정으로 아포프토시스(apoptosis)가 있다. 아포프토시스는 조직의 항상성(homeostasis of tissue volume), 개체의 발생과정, 장기의 퇴행(regression), tolerance 등의 여러 생명 활동 과정에서 발생하는 세포 사망의 과정으로서, 세포질 및 핵이 분절화(fragmentation)되어 죽어가는 능동적 사망과정으로 알려져 있다. 아포프토시스에서는 수동적으로 죽어가는 괴사에서 볼 수 없는 DNA 분절화(DNA ladder pattern)가 특징적으로 관찰된다. 인터페론감마의 암세포에 대한 세포독성 기전을 연구하기 위해서 인터페론감마를 폐암세포주인 A549세 처치한 후 현미경(inverted microscope)로 A549의 변화를 관찰하였는데 A549세포가 분절화되면서 죽어가는 것을 관찰할 수 있었다. 저자들은 인터페론감마의 항암기전으로서 아포프토시스의 가능성을 평가하고자 본 연구를 시행하였다. 방법: 폐암세포주인 A549세포를 대상으로 하였다. A549세포에 여러 농도의 인터페론감마를 투여하고 24시간, 72시간, 120시간 후에 MTT(dimethylthiazolyl diphenyltetrazolium bromide) bioassay법으로 세포독성을 정량화하였다. 그리고, 100 unit/ml의 인터페론감마를 A549 세포에 120시간 처치 후, 광학 현미경으로 세포 사망의 양상을 관찰하였다. 또한, 100 unit/ml의 인터페론감마를 투여하고 120시간이 경과한 후 사망 세포의 DNA를 추출하여 1.5% agarose gel에서 전기 영동을 시행하고 ethidium bromide로 염색 후 DNA ladder pattern 유무를 관찰하였다. 결과: 1) 인터페론감마에 의한 A549 폐암세포주의 세포독성 효과는 24시간에는 거의 없다가 72시간부터 120시간 사이에 나타나기 시작하여 120시간에는 더 증가하였다. 2) 인터페론감마에 의한 A549 세포의 사망 양상은 광학현미경상 A549 세포들이 작은 분절로 나뉘면서 사망하였다. 3) 인터페론감마를 A549 폐암세포주에 처치 후 죽어기는 세포의 DNA를 추출하여 전기영동시킨 결과 아포프토시스(apoptosis)에서 특징적으로 보이는 DNA ladder pattern을 관찰할 수 있었다. 결론: 인터페론감마(interferon-$\gamma$)의 A549 폐암세포주에 대한 세포독성의 기전은 아포프토시스 과정을 통해서 일어난다.
