Comparison of Relationship between Biochemical Indices and Bone Mineral Density of Pre- and Post- Menopausal Women in Gyeongnam Area (경남 일부지역 폐경 전·후 여성들의 골밀도와 생화학지수들과의 관련성에 관한 비교연구)

  • This study was carried out to compare the relationship between biochemical indices and bone mineral density (BMD) in 50 pre-menopausal and 50 post-menopausal women. The subjects were divided into normal and risk groups according to their bone status, as determined by T-scores of the lumbar spine and femur. The average T-score of the lumbar spine was higher (p<0.05) in pre-menopausal women ($0.42{\pm}0.18$) than post-menopausal women ($-0.08{\pm}0.21$). Serum levels of HDL-cholesterol, P, and Fe were significantly higher in the risk group than the normal group in pre-menopausal women (p<0.05). Serum levels of total protein, globulin, alkaline phosphatase (ALP), and osteocalcin were lower in the risk group than the normal group, whereas the level of estrogen was higher in the normal group than the risk group in post-menopausal women (p<0.05). In pre-menopausal women, P was positively correlated with Ca (p<0.01), and ALP was positively correlated with osteocalcin (p<0.01) and parathyroid hormone (PTH) (p<0.05). Further, insulin-like growth factor-I (IGF-I) was negatively correlated with the vitamin $25(OH)D_3$ and vitamin K (p<0.05). In post- menopausal women, the Ca was positively correlated with vitamin $25(OH)D_3$ (p<0.05) and vitamin K (p<0.01), and P was positively correlated with vitamin K (p<0.01), Ca (p<0.01), and IGF-I (p<0.05) and negatively correlated with PTH (p<0.05). IGF-I was negatively correlated with PTH (p<0.01) and estrogen (p<0.05), and ALP was positively correlated with osteocalcin (p<0.01) and negatively correlated with vitamin K and estrogen (p<0.05). In pre-menopausal women, the lumbar spine BMD was positively correlated with vitamin K level (p<0.01) and negatively correlated with P level (p<0.05). In post-menopausal women, the femur BMD was positively correlated with estrogen level and negatively correlated with PTH leves (p<0.05). These results suggest that vitamin K and P levels are associated with bone health in pre-menopausal women, and estrogen and PTH levels are associated with bone health in post-menopausal women.

Changes in drug metabolism during hypoxia/reoxygenation in isolated perfused rat

  • This study was done to investigate the effect of vitamin E on hypoxia/reoxygenation-induced hepatic injury in isolated perfused rat liver. Rats were pretreated with vitamin E or vehicle(soybean oil). Isolated livers from fasted 18 hours were subjected to 45min of low flow hypoxia or N$_2$ hypoxia followed by reoxygenation for 30min. The perfusion medium used was KHBB(pH 7.4) and 50${\mu}$㏖/$\ell$ of ethoxycoumarin was added to the perfusate to determine the ability of hepatic drug-metabolizing systems, In low flow hypoxia model, total glutathione and oxidised glutathione levels were significantly increased by hepoxia/reoxygenation with slight increase in LDH levels. These increases were prevented by vitamin E pretreatment. In N$_2$ hypoxia model, LDH, total glutathione and oxidized glutathione levels were increased significantly by hypoxia but restored to normal level by reoxygenation. Vitamin E had little effect on this hypoxic damage. There were no significant changes in the rate of hepatic oxidation of 7-EC to 7-HC in both hepoxic models. But, the subsequent conjugation of 7-HC by sulfate or glucuronic acid were significantly decreased by hypoxia, but restored by reoxygenation in both hypoxia models. As opposed to our expectation, treatment with vitamin E aggrevated the decrease of the rate of conjugation and even inhibited the restoration by reoxygenation. Our findings suggest that hypoxia/reoxygenation diminishes phase II drug metabolizing function and this is, in part, related to decreased energy level.

Effect of vitamin $B_6$ deficiency on antioxidative status in rats with exercise-induced oxidative stress

  • This study investigated the effect of vitamin $B_6$ deficiency on antioxidant enzyme activities and lipid profile in rats with exercise-induced oxidative stress. Forty eight rats were fed either a vitamin $B_6$ deficient diet (B6-) or a control diet (control) for 4 weeks and then subdivided into 3 groups: pre-exercise (PreE); post-exercise (PostE); recess after exercise (recessE). Compared to those of control group, plasma catalase and hepatic cytosol superoxide dismutase (SOD, EC 1.15.1.1) activities of B6- group were lower regardless of exercise. The ratio of reduced glutathione/oxidized glutathione (GSH/GSSG) of B6 - group was lower in PreE and there was no difference between PostE and recessE. The level of malondialdehyde (MDA) of B6- was significantly higher in PreE and PostE. High-density lipoprotein-cholesterol (HDL-C) level of B6- group was lower regardless of exercise. Atherosclerotic index of $B_6$- group was higher in PreE and there was no difference between PostE and recessE. It is suggested that a reduction in antioxidative status caused by vitamin $B_6$ deficiency may be aggravated under exercise-induced oxidative stress.

A Study on the Hypocholesterolemic Effect of Milk in Rats (랫트에 있어서 우유의 콜레스테롤 저하효과에 관한 연구)

  • The benefits of milk on the rats supplemented with cholesterol and vitamin D2 in diet was investigated. A total of 150 male rats of Sprague-Dawley strain, 5 weeks of age, were divided into 6 groups with the diet. Cholesterol control group was fed the diet containing 1.0% choleterol and 50,000 IU/100 g of vitamin D2 for 24 months, and M0.5, M1, M2 and M5 of milk groups were fed the diet containing cholesterol, vitamin D2 and 0.5, 1, 2 and 5 of reference daily intakes of whole milk, respectively. No remarkable differences of growth, diet intake, and food effciency ratio among groups were observed though rats in cholesterol control group showed abrupt decrease of diet intake and body weight showed slightly higher relative organ weights than did the rats in control group. The rats in cholesterol control group showed the highest serum total cholesterol level. The rats in milk groups showed lower total cholesterol level and higher high density lipoprotein (HDL)-cholesterol fraction than did the rats in cholesterol control group though no tendency was observed among milk groups. In milk groups, aorta, heart, kidney and liver of rats showed milder calcification and necrosis or fat degeneration compared with those in cholesterol control group. The above results suggest that whole milk could have beneficial effect on the cholesterol-and -vitamin D2-induced atherosclerosis in rats.

An Effect of Children's Food Alttitude on Nutritional Status and Personality (어린이의 식생활태도가 영양 상태 및 성격에 미치는 영향에 관한 연구)

  • This survey of 527 children, aged 4 to 6 years, focused on family and dietary environment, nutritional status and personality. It was conducted from June to October, 1986. The results showed that 68.0%(breakfast)and 55.4% (evening meal ) of the subjects indicated they had meals with the family everyday. Analysis of the nutritional status children showed all nutrient intake except protein an diron was insufficient compared with RDA. Carbohydrate provided 58.3% of total calorie intake ; protein accounted for 15.7% . fat provided 26.0%. There was a positive relation between income and nutrient intake. Children with employed mothers showed lower nutrient intake than children with unemployed mothers. Food attitude was positively related to nutrient intake, nutrient intake of children with good eating behavior was higher than that of children had poor eating behavior . Activity level had a significant correlation with intake of calories, fat, vitamin A, vitamin B$_1$, and Vitamin B$_2$. Mood was correlated with intake of protein, iron, vitamin A, vitamin B$_2$ and vitamin C.

The Effects of Vitamin C on Lipid Contents and Fatty Acid Compositions of Serum and Liver in Rats Treated with Radiation or Aflatoxin B1 (Vitamin C가 방사선과 Aflatoxin B1을 투여한 흰쥐의 혈청과 간장의 지질성분 및 지방산 조성에 미치는 영향)

  • Lipid peroxidation is one of the main manifestations of oxidative damage and has been found to play an important role in the toxicity and carcinogenesis of many carcinogens. This study was carried out to determine the effects of vitamin C on lipid contents and fatty acid compositions of serum and liver in male rats treated with radiation or aflatoxin $B_1\;(AFB_1)$. Six week-old male Sprague-Dawley rats were randomly divided into 7 groups; control group, radiation exposed group, $AFB_1$ treated group, X-ray and $AFB_1$ co-treated group. Three groups, except control group, were each further divided into vitamin C administered group and not administered groups. For this study, vitamin C was injected with 10 mg/kg of body weight by intraperitoneal injection and 1 hr later, 0.4 mg/kg of $AFB_1$ was injected by the same method. These administrations were repeated every 3 days over a period of 15 days. Only one time, X-ray was irradiated on whole liver with 1,500 cGy. Then vitamin C and AFB1 were administered by the same level and same method described above. On the 16th day of treatments, the animals were sacrificed. From the analysis of the serum lipid patterns, significant decrease (p<0.01) in triglyceride (TG) and total cholesterol levels were observed in X-ray and $AFB_1$ co treated group administered with vitamin C (group 7). In liver lipids, the levels of free cholesterol and total cholesterol were also decreased in X-ray and $AFB_1$ co treated group administered with vitamin C (group 7). The levels of serum free cholesterol and hepatic TG were not significantly different among all groups according to vitamin C administrations. The high density lipoprotein (HDL)-cholesterol level of serum was significantly (p<0.01) increased while the low density lipoprotein (LDL)-cholesterol level was decreased in X-ray and $AFB_1$ co treated group administered with vitamin C (group 7). In the phospholipid fatty-acid compositions of serum and liver tissue, group 3, 5 and 7 showed an increase in polyunsaturated fatty-acid (PUFA) but a decrease in saturated fatty acid (SFA) when compared to the control group. The composition ratio of fatty acid varied according to vitamin C administration. These results suggested that vitamin C has partly suppressive effects on lipid contents and fatty acid composition of serum and liver in rats treated by radiation and $AFB_1$.

Effects of Vitamin E and Selenium on the Antioxidative Defense System in Streptozotocin-induced Diabetic Rats (Vitamin E와 Selenium이 Streptozotocin 유발 당뇨쥐의 항산화계에 미치는 영향)

  • The purpose of this study was to investigate the effect of vitamin E and selenium on the antioxidative defense mechanism in the liver of streptozotocin(STZ)-induced diabetic rats. Sprague-Dawley male rats(120$\pm$10gm) were randomly assigned to one control and five STZ-diabetic groups. Diabetic groups were classified to STZ-0E (vitamin E free diet), STZ-40E(40mg vitamin E/kg of diet), STZ-400E(400mg vitamin E/kg of diet), STZ-S(0.5ppm Se/kg of diet) and STZ-400ES(400mg vitamin E and 0.5ppm Se/kg of diet) according to the level of vitamin E and selenium supplementation. Diabetes was experimentally induced by intravenous adminstration of 55mg/kg of STZ in citrate buffer(pH 4.3) after 4-weeks feedng of six experimental diets. Animals were sacrificed at the 4th day of diabetic states. Activities of the serum glutamic oxaloacetate transaminase(GOT) and the glutaminc pyruvate transaminase(GPT) in STZ-0E, STZ-40E and STZ-S rats were higher than those of control. Liver xanthine oxidase activities were similar to serum GOT and GPT. Liver superoxide dismutase(SOD) activities were higher in STZ-0E and STZ-40E groups by 33%, 22%, respectively than that of control. Glutathione S-transferase(GST) activities of liver were similar to GSH-Px activities. The contents of vitamin E in liver tissue were significantly lower STZ-0E, STZ-40E and STZ-S groups by 50%, 36%, 45% than that of control. Reduced glutathione(GSH) contents of liver were lower STZ-0E, STZ-40E, STZ-400E, STZ-S and STZ-400ES groups by 57%, 51%, 19%, 18%, 12% than that of control. Lipid peroxide values (LPO) in liver were higher 5.6, 2.3 and 2.3 times in STZ-0E, STZ-40E and STZ-S group than that of control. The present results indicate that STZ-induced diabetic rats are more sensitive to oxidative stress, leading to the acceleration of lipid peroxidation process, which can be more accelerated by feeding the low level of dietary vitamin E. In the coincident supplementation of high dietary vitamin E and selenium antioxidative enzymes activities and physiolosical antioxidants were increased more than those of the separate supplementation of vitamin E or selenium. Therefore, dietary vitamin E and selenium reduced peroxidative damage of tissue, promoting antioxidative defense mechanism against lipid peroxidation by diabetes.

The Effect of Vitamin B-Complex on Stress-induced Immune Alteration (비타민 B 복합제가 스트레스에 의해 유도된 면역변화에 미치는 영향)

  • The purpose of the study was to determine the effect of vitamin B complex on stress-induced immune alteration. 21 medical students participated in the study 4 weeks before an academic examination period(baseline), 2 weeks before the exam period and during the exam period. Among them, 10 subjects were given vitamin B complex for 4 weeks, and 11 were not given vitamin B during the whole period. Cell-mediated immune function was measured by lymphocyte proliferative response to phytohemagglutinin(PHA) and interleukin-2(IL-2) production. Global assessment of recent stress(GARS) scale and symptom checklist-90-revised(SCL-90-R) were used to measure the level of subjective stress and psychopathology. Vitamin group had significantly lower scores of anxiety scale on SCL-90-R than non-vitamin group. No significant differences were found in lymphocyte proliferative response to PHA and IL-2 production between vitamin and non-vitamin groups during each period. There were no significant differences in change of of each of the two immune parameters over time as well as between vitamin and non-vitamin groups. However, lymphocyte proliferative response to PHA was significantly increased over time. In conclusion, it was suggested that vitamin B complex is likely to decrease anxiety level, and that exam stress might enhance lymphocyte proliferation regardless of vitamin B.

Assessment of Vitamin A and E Status in Korean Rural Adult Population by Dietary Intake and Serum Levels (식이섭취조사와 혈청수준을 이용한 경기도 구리시거주 성인의 비타민 A와 E 영양상태평가)

  • Because the intake of vitamin A had been consistently found to be low among Koreans, there has been a concern that mild vitamin A deficiency might be prevalent in the population. However, the inaccuracy of vitamin A database has long been suspected database for the nutrient. This study was performed to assess the vitamin A and E status in 208 adults over 20 years of age living in a rural area of Korea. Newly modified database for vitamin A and E was used to estimate the dietary intake of the subjects in this study. A cross sectional survey of dietary intake with 24-hour recall method was conducted. Serum retinol and $\alpha$-tocopherol levels of the subjects were analyzed in fasting samples using HPLC. For vitamin A, mean levels of dietary intake and percent of RDA were 620.3$\pm$1087RE/day and 88.6$\pm$155.4%, respectively, which were considerably higher than the levels reported in 1995 Korean national Nutrition survey(470.1RE/day and 67.2%, respectively). For vitamin E, the levels were 9.74$\pm$6.30mg/day and 97.4$\pm$63.0%, respectively. Mean concentration of serum retinol was 83.1$\pm$30.1$\mu\textrm{g}$/dl and none of the subjects was below 10$\mu\textrm{g}$/dl, or deficient level. Mean serum retinol in men, 99.8$\pm$30.4$\mu$g/dl, was significantly higher than in women, 70.7$\pm$23.4$\mu\textrm{g}$/dl (p<0.01). Mean concentration of serum $\alpha$-tocopherol was 1.17$\pm$0.52mg/dl and that of one subject was below 0.5mg/dl, or deficient level. Serum $\alpha$-tocopherol levels of the old subjects were higher than the younger subjects in both sexes(p<0.05). These data suggest that contrary to the previous reports about low intake of vitamin a, intake and serum levels of vitamin A among the subjects are not low. EFFECTS OF DIETARY VITAMIN B6 LEVELS ON LIPID CONCENTRATION AND FATTY ACID COMPOSITION IN GROWING CHICKS

  • This experiment was designed to evaluate the effect of various dietary vitamin $B_6$ levels on conversion from linoleic acid to arachidonic acid in various tissues in growing chicks. Growing chicks were fed the purified diet containing 7% safflower oil with different levels of vitamin $B_6$ (0, 4, 8, 40, 80 mg per kg diet) for 14 days. Feed intake and weight gain in chicks fed the vitamin $B_6$-free diet were markedly depressed. Esterified and free cholesterol concentrations in serum were significantly higher, while the serum triglyceride concentration was significantly lover in chicks fed the vitamin $B_6$-free diet compared to that fed diets with vitamin $B_6$. The liver triglyceride content was also lower in chicks fed the vitamin $B_6$-free diet. The liver and serum cholesterol ester fractions in chicks fed the vitamin $B_6$-free diet showed higher rate of $C_{18:2n6}$ and lower rates of $C_{18:3n6}$, $C_{20:3n6}$ and $C_{20:4n6}$ as compared with vitamin $B_6$ fed groups. In serum phospholipid fraction of chicks fed the vitamin $B_6$-free diet, rates of $C_{20:3n6}$ and $C_{20:4n6}$ were markedly lower. As dietary vitamin $B_6$ level was increased, the rate of $C_{20:4n6}$ was slightly increased, although it was statistically not significant. The fatty acid compositions of adipose tissue showed almost the same pattern as those in liver and serum. This result suggests that the desaturation of $C_{18:2n6}$ to $C_{18:3n6}$, elongation to $C_{20:3n6}$ or both steps might be impaired by vitamin $B_6$ deficiency in growing chicks.
