Improvement of Virus Safety of an Antihemophilc Factor IX by Virus Filtration Process

  • Viral safety is an important prerequisite for clinical preparations of plasma-derived pharmaceuticals. One potential way to increase the safety of therapeutic biological products is the use of a virus-retentive filter. In order to increase the viral safety of human antihemophilic factor IX, particularly in regard to non-enveloped viruses, a virus removal process using a polyvinylidene fluoride membrane filter (Viresolve NFP) has been optimized. The most critical factor affecting the filtration efficiency was operating pH and the optimum pH was 6 or 7. Flow rate increased with increasing operating pressure and temperature. Recovery yield in the optimized production-scale process was 96%. No substantial changes were observed in the physical and biochemical characteristics of the filtered factor IX in comparison with those before filtration. A 47-mm disk membrane filter was used to simulate the process performance of the production-scale cartridges and to test if it could remove several experimental model viruses for human pathogenic viruses, including human hepatitis A virus (HAV), porcine parvovirus (PPV), murine encephalomyocarditis virus (EMCV), human immunodeficiency virus type 1 (HIV), bovine viral diarrhea virus (BVDV), and bovine herpes virus (BHV). Non-enveloped viruses (HAV, PPV, and EMCV) as well as enveloped viruses (HIV, BVDV, and BHV) were completely removed during filtration. The log reduction factors achieved were $\geq$6.12 for HAV, $\geq$4.28 for PPV, $\geq$5.33 for EMCV, $\geq$5.51 for HIV, $\geq$5.17 for BVDV, and $\geq$5.75 for BHV. These results indicate that the virus filtration process successfully improved the viral safety of factor IX.

토양전염성 맥류바이러스 발생지역 및 맥류품종의 저항성 조사 (Survey of Barley (Hordeum vulgare L.) Cultivars Resistance to Mosaic Viruses and Areas Infected with Soil-borne Barley Mosaic Viruses)

  • 1999년부터 2001년까지 전국42개 지역에서 토양전염성 맥류바이러스를 조사한 결과 강원도 영월, 전라북도 무주, 충청남도 공주, 보령, 서천 포장을 제외한 37개 포장에서 BaYMV, BaMMV 및 SbWMV가 발생하고 있었다. 또한 토양전염성 맥류바이러스 상습 발병지 10개 장소에 겉보리 10품종, 쌀보리 16품종, 맥주보리 6품종, 밀 2품종을 파종하고 저항성 조사를 실시한 결과 겉보리는 조강보리, 쌀보리는 내한쌀보리 및 논산과1-6품종이 비교적 여러지역에서 저항성 높은 품종으로 나타났다. 그리고 일본에서 분양 받은 15품종도 대부분이병성으로 나타났으나 특이하게 익산, 영광 및 예산지역에서 저항성 품종이 많았다. 지역간 품종에 따른 바이러스 혼합 감염을 조사한 결과 조사 10개 지역 모두 BaYMV, 및 BaMMV가 혼합 감염되어 있었고, BbWMV는 예산, 진주, 나주 및 익산지역에 혼합감염되어 있었다. 그러나 영광지역에서는 사천6호 품종을 제외하곤 BaYMV만이 발생하였다. 일본 분양 품종은 예산에서만 BaYMV가 감염되었고 나머지 지역에서 모두 BaMMV 및 BaYMV에 혼합 감염되 어 있었다.

  • 호흡기 바이러스는 감염된 방문자와의 직접적인 접촉을 통해 병원 환경에서 감염된다. 감염관리 분야에서 호흡기 바이러스에 의한 병원내 획득 감염의 주요한 문제를 유발한다. 임상 검사실에서 파생된 감시 데이터는 또한 의료 서비스를 제공하는 기관과 공중 보건 분야에서 치료, 소모품 및 진단 제품 구매를 위해 병원과 지역 사회에 의료자원을 적절하게 배분하기 위해 종종 사용된다. 호흡기바이러스의 감염에서 조기 진단은 필수적이며 호흡기 검체를 사용하는 진단법에 활용될 수 있는 방법에는 바이러스 배양, 분자 진단 및 분석 등이 포함된다. 랩온어칩(LoC)/마이크로칩은 보다 다양하고 강력한 기술인 차세대 현장검사 시험법을 개발할 수 있는 새로운 전략으로 제공한다. 호흡기계의 중요성은 의료관련 종사자의 직업적 건강과 안전을 보장하기 위해 감염 관리 지침에 엄격히 적용되어야 한다. 이 연구를 포함하여 점점 더 많은 임상적 효능 증거가 감염 전파에 대한 오랜 패러다임에 도전하고 있다. 바이러스의 감염 가능성을 의심하는 새로운 호흡기 증상이 시작된 입원 환자로부터 호흡기 바이러스를 탐지하기 위한 빈번한 검사에 대해 추가 지원이 요구되고 감염통제의 노력에 집중적으로 도움이 이루어져야 할 것이다.

Development of an Automatic PCR System Combined with Magnetic Bead-based Viral RNA Concentration and Extraction

  • Human respiratory viral infections such as COVID-19 are highly contagious, so continuous management of airborne viruses is essential. In particular, indoor air monitoring is necessary because the risk of infection increases in poorly ventilated indoors. However, the current method of detecting airborne viruses requires a lot of time from sample collection to confirmation of results. In this study, we proposed a system that can monitor airborne viruses in real time to solve the deficiency of the present method. Air samples were collected in liquid form through a bio sampler, in which case the virus is present in low concentrations. To detect viruses from low-concentration samples, viral RNA was concentrated and extracted using silica-magnetic beads. RNA binds to silica under certain conditions, and by repeating this binding reaction, bulk samples collected from the air can be concentrated. After concentration and extraction, viral RNA is specifically detected through real-time qPCR (quantitative polymerase chain reaction). In addition, based on liquid handling technology, we have developed an automatic machine that automatically performs the entire testing process and can be easily used even by non-experts. To evaluate the system, we performed air sample collection and automated testing using bacteriophage MS2 as a model virus. As a result, the air-collected samples concentrated by 45 times then initial volume, and the detection sensitivity of PCR also confirmed a corresponding improvement.

Twindemic Threats of Weeds Coinfected with Tomato Yellow Leaf Curl Virus and Tomato Spotted Wilt Virus as Viral Reservoirs in Tomato Greenhouses

  • Tomato yellow leaf curl virus (TYLCV) and tomato spotted wilt virus (TSWV) are well-known examples of the begomovirus and orthotospovirus genera, respectively. These viruses cause significant economic damage to tomato crops worldwide. Weeds play an important role in the ongoing presence and spread of several plant viruses, such as TYLCV and TSWV, and are recognized as reservoirs for these infections. This work applies a comprehensive approach, encompassing field surveys and molecular techniques, to acquire an in-depth understanding of the interactions between viruses and their weed hosts. A total of 60 tomato samples exhibiting typical symptoms of TYLCV and TSWV were collected from a tomato greenhouse farm in Nonsan, South Korea. In addition, 130 samples of 16 different weed species in the immediate surroundings of the greenhouse were collected for viral detection. PCR and reverse transcription-PCR methodologies and specific primers for TYLCV and TSWV were used, which showed that 15 tomato samples were coinfected by both viruses. Interestingly, both viruses were also detected in perennial weeds, such as Rumex crispus, which highlights their function as viral reservoirs. Our study provides significant insights into the co-occurrence of TYLCV and TSWV in weed reservoirs, and their subsequent transmission under tomato greenhouse conditions. This project builds long-term strategies for integrated pest management to prevent and manage simultaneous virus outbreaks, known as twindemics, in agricultural systems.

  • A single-step multiplex reverse transcription polymerase chain reaction (RT-PCR) assay was developed for the simultaneous detection of five cucurbit-infecting viruses: cucumber mosaic virus (CMV), watermelon mosaic virus 2 (WMV2), zucchini yellow mosaic virus (ZYMV), cucumber green mottle mosaic virus (CGMMV), and kyuri green mottle mosaic virus (KGMMV). The multiplex RT-PCR provides a simple and rapid method for detecting various viruses in cucurbit plants, which will help diagnose many cucurbit plants at a time.

Conformation and Linkage Studies of Specific Oligosaccharides Related to H1N1, H5N1, and Human Flu for Developing the Second Tamiflu

  • The interaction between viral HA (hemagglutinin) and oligosaccharide of the host plays an important role in the infection and transmission of avian and human flu viruses. Until now, this interaction has been classified by sialyl(${\alpha}2-3$) or sialyl(${\alpha}2-6$) linkage specificity of oligosaccharide moieties for avian or human virus, respectively. In the case of H5N1 and newly mutated flu viruses, classification based on the linkage type does not correlate with human infection and human-to-human transmission of these viruses. It is newly suggested that flu infection and transmission to humans require high affinity binding to the extended conformation with long length sialyl(${\alpha}2-6$)galactose containing oligosaccharides. On the other hand, the avian flu virus requires folded conformation with sialyl(${\alpha}2-3$) or short length sialyl(${\alpha}2-6$) containing trisaccharides. This suggests a potential future direction for the development of new species-specific antiviral drugs to prevent and treat pandemic flu.

  • A survey was conducted to investigate viral infections in 184 whiteleg shrimp (Litopenaeus vannamei) collected from nine farms and one wholesale fish vendor during 2018 and 2019. Gill and abdominal muscle of shrimp were tested for the presence of five viruses, viz. infectious hypodermal and haematopoietic necrosis virus, taura syndrome virus, infectious myonecrosis virus, yellow head virus genotype 1, and covert mortality nodavirus by reverse transcription-polymerase chain reaction (RT-PCR) and PCR. These viruses were not detected in any of 184 samples, screened under the study.

라우터기반 바이러스 감염파일에 대한 블록킹 시스템 설계 (The design of the blocking system against file infected with virus on router)

  • 인터넷 기술은 사용자의 수만큼이나 빠르게 발전하고 있다. 그러나, 이 기술이 사용자들에게 항상 좋은 면만을 보여주지는 않는다. 해커나 어린이조차도 컴퓨터 바이러스를 만들어 네트웍상에 배포한다. 그래서 바이러스의 위협은 점점 심각해져가고 있다. 본 논문에서는 이전에 개발되었던 바이러스 블록킹 시스템을 분석한 후 라우터 상에서 바이러스에 감염된 파일을 차단하는 시스템을 개선하여 설계하였다. 다른 시스템과 비교하여, 라우터와 블록킹 시스템간의 전송시간을 단축하여 통신의 복잡성에서도 더 효율적이다.

  • The genetic algorithm that simulates the virus evolutionary theory has been developed applying to combinatorial optimization problems. The algorithm in this study uses only one individual and a population of viruses. The individual is attacked, inflected and improved by the viruses. The viruses are composed of flour genes (a pair of top gene and a pair of tail gene). If the individual is improved by the attacking, the inflection occurs. After the infection, the tail genes are mutated. If the same virus attacks several times and fails to inflect, the top genes of the virus are mutated. By this mutation, the individual can be improved effectively. In addition, the influence of the immunologic mechanism on evolution is simulated.
