The human immunodeficiency virus type 1 (HIV-1) Tat is one of the viral gene products essential for HIV replication. The exogenous Tat protein is transduced through the plasma membrane and then accumulated in a cell. The basic domain of the Tat protein, which is rich in arginine and lysine residues and called the protein transduction domain (PTD), has been identified to be responsible for this transduction activity. To better understand the nature of the transduction mediated by this highly basic domain of HIV-1 Tat, the Green Fluorescent Protein (GFP) was expressed and purified as a fusion protein with a peptide derived from the HIV-1 Tat basic domain in Escherichia coli. The transduction of Tat-GFP into mammalian cells was then determined by a Western blot analysis and fluorescence microscopy. The cells treated with Tat-GFP exhibited dose- and time-dependent increases in their intracellular level of the protein. the effective transduction of denatured Tat-GFP into both the nucleus and the cytoplasm of mammalian cells was also demonstrated, thereby indicating that the unfolding of the transduced protein is required for efficient transduction. Accordingly, the availability of recombinant Tat-GFP can facilitate the simple and specific identification of the protein transduction mediated by the HIV-1 Tat basic domain in living cells either by fluorescence microscopy or by a fluorescence-activated cell sorter analysis.
In order to find the cellular interaction factors of the Heliothis armigera nuclear polyhedrosis virus capsid protein VP39, a Heliothis armigera cell cDNA library was constructed. Then VP39 was used as bait. The host actin gene was isolated from the cDNA library with the yeast two-hybrid system. This demonstrated that VP39 could interact with its host actin in yeast. In order to corroborate this interaction in vivo, the vp39 gene was fused with the green fluorescent protein gene in plasmid pEGFP39. The fusion protein was expressed in the Hz-AM1 cells under the control of the Autographa californica multiple nucleopolyhedrovirus immediate early gene promoter. The host actin was labeled specifically by the red fluorescence substance, tetramethy rhodamine isothicyanete-phalloidin. Observation under a fluorescence microscopy showed that VP39, which was indicated by green fluorescence, began to appear in the cells 6 h after being transfected with pEGFP39. Red actin cables were also formed in the cytoplasm at the same time. Actin was aggregated in the nucleus 9 h after the transfection. The green and red fluorescence always appeared in the same location of the cells, which demonstrated that VP39 could combine with the host actin. Such a combination would result in the actin skeleton rearrangement.

국내에서 분리한 강독전염성후두기관염 바이러스 (ILTV)에 대한 세포융합방법에 의해 단크론성항채(MCA) 생산을 시도한 결과 총 8회의 세포융합을 통하여 총 1017개의 융합세포가 생산되었으며 그중 ILTV와 특이적으로 작용하늘 항체를 생산하는 3주의 Hybridoma를 작성하였다. 이 3주의 MCA는 모두 IgG형에 속하였으며 마우스 복강내접종하여 생산된 복수항체외 형광항체가는 $10^5$-$10^6$에 달하였고 약독 및 강독 ILTV에 차이가 없이 작용하였으며 중화능력은 인정되지 않았다. 이 MCA를 이용하여 간접형광항체법으로 인공감염계에서 ILTV 검출을 시도한 결과. 기관 및 안점막의 도말표본에서 감염후 10일 까지 진단이 가능하였으며 표준 양성혈청을 이용한 형광항체법이나 핵내봉입체 검출방법에 비해서 진단효율이 높았다.

CDC48 is a member of the AAA ATPase superfamily. Yeast CDC48 and its mammalian homolog p97 are implicated in diverse cellular processes, including mitosis, membrane fusion, and ubiquitin-dependent protein degradation. However, the cellular functions of plant CDC48 proteins are largely unknown. In the present study, we performed virus-induced gene silencing (VIGS) screening and found that silencing of a gene encoding a tobacco CDC48 homolog, NgCDC48, resulted in severe abnormalities in leaf and shoot development in tobacco. Furthermore, transgenic tobacco plants (35S:anti-NgCDC48), in which the NgCDC48 gene was suppressed using the antisense RNA method, exhibited severely aberrant development of both vegetative and reproductive organs, resulting in arrested shoot and leaf growth and sterile flowers. Approximately 57-83% of 35S:anti-NgCDC48 plants failed to develop mature organs and died at early stage of development. Scanning electron microscopy showed that both adaxial and abaxial epidermal pavement cells in antisense transgenic leaves were significantly smaller and more numerous than those in wild type leaves. These results indicate that NgCDC48 is critically involved in cell growth and development of tobacco plants. An in vivo targeting experiment revealed that NgCDC48 resides in the endoplasmic reticulum (ER) in tobacco protoplasts. We consider the tantalizing possibility that CDC48-mediated degradation of an as-yet unidentified protein(s) in the ER might be a critical step for cell growth and expansion in tobacco leaves.
본 연구는 WSSV의 주요 구조단백질인 VP19와 VP28을 모두 포함하는 VP19+28 fusion protein을 제조하여, Litopenaeus vannamei에서 WSSV에 대한 백신으로서의 효능을 평가하고자 수행하였다. VP19와 VP28 유전자를 fusion하여 제작한 VP19+28 유전자를 pET-28a(+) vector에 삽입하고 단일단백질로서 제작된 VP19+28 유전자를 E. coli BL21 (DE3)에서 발현시켰다. 백신실험을 위해 새우에게 2주 동안 실험용 사료를 급이하였으며, 그 후 바이러스액($1{\times}10^2$배로 희석한 WSSV)을 이용하여 새우에게 주사 감염에 의해 in vivo 공격실험(challenge test)을 수행하였다. 실험결과, vaccination을 하지 않은 새우들은 감염 후 11일째에 100%의 누적폐사율을 보였으며, host control로써 E. coli BL21을 사용하여 vaccination한 새우들은 감염 후 17일째에 100%의 누적폐사율을 보였다. rVP19, rVP28, rVP19+28을 이용하여 vaccination한 새우들의 경우 감염 후 21일째에 각각 66.7%, 41.7%, 41.7%의 누적폐사율을 보였다. 이상의 결과를 통해 rVP28과 rVP19+28이 WSSV에 대해 높은 백신효능을 가짐을 확인하였다. 또한 감염 후 21일째에 fusion protein rVP19+28과 rVP28의 누적폐사율은 동일하였지만 공격실험기간 동안 폐사율이 rVP19+28을 투여 한 실험군이 낮게 나타나는 것을 보아 WSSV에 대한 새우의 방어효능은 rVP19+28이 더 높음을 나타내는 것이다.
조류 독감바이러스(avian influenza virus, AIV)는 사람에게서 발생하는 인플루엔자 대유행에 중요한 역할을 한다. 특히 최근 AIV H9N2형에 의한 가금류 감염이 빈번히 나타나고 있어 인체 감염이 상당히 우려되는 실정이다. 본 연구에서는 최종적으로 AIV의 HA와 NA 단백질에 특이적으로 반응하는 항혈청을 생산하고자 하였다. 먼저 감염된 닭에서 분리된 AIV H9N2 한국분리주 A/Ck/Kr/MS96/96의 게놈RNA로부터 RT-PCR 방법으로 HA와 NA 단백질 N-말단부위에 해당하는 염기서열을 증폭하였다. 이렇게 증폭된 DNA단편은 E. coli 발현벡터 pGEX4T-1에 삽입한 후, BL21 세포에서 각각의 GST fusion protein (GST-HAln와 GST-NAn) 형태로 발현하였다. GST-HAln와 GST-NAn은 모두 glutathione sepharose column을 사용하여 분리 및 정제하였으며, 정제된 단배질을 항원으로 사용하여 토끼 항혈청을 생산하였다. 생산된 항혈청의 항원특이성은 AIV H9N2 한국분리주 A/Ck/Kr/MS96/96로 감염된 MDCK 세포의 cell extract를 사용하여 immunoblotting을 수행함으로써 확인하였다. 본 실험결과AIV H9N2의 HA와 NA단백질 N-말단부위에 해당하는 재조합GST fusion protein과, 이들 각각의 단백질에 특이적으로 반응하는 항혈청은 앞으로AIV 감염의 진단 뿐만 아니라, AIV에 대한 기초연구에 중요한 재료로 사용될 것으로 기대한다.

When endoplasmic reticulum (ER) functions are perturbed, the ER induces several signaling pathways called unfolded protein response to reestablish ER homeostasis through three ER transmembrane proteins: inositol-requiring enzyme 1 (IRE1), PKR-like ER kinase (PERK), and activating transcription factor 6 (ATF6). Although it is important to measure the activity of ATF6 that can indicate the status of the ER, no specific cell-based reporter assay is currently available. Here, we report a new cell-based method for monitoring ER stress based on the cleavage of $ATF6{\alpha}$ by sequential actions of proteases at the Golgi apparatus during ER stress. A new expressing vector was constructed by using fusion gene of GAL4 DNA binding domain (GAL4DBD) and activation domain derived from herpes simplex virus VP16 protein (VP16AD) followed by a human $ATF6{\alpha}$ N-terminal deletion variant. During ER stress, the GAL4DBD-VP16AD(GV)-$hATF6{\alpha}$ deletion variant was cleaved to liberate active transcription activator encompassing GV-$hATF6{\alpha}$ fragment which could translocate into the nucleus. The translocated GV-$hATF6{\alpha}$ fragment strongly induced the expression of firefly luciferase in HeLa Luciferase Reporter cell line containing a stably integrated 5X GAL4 site-luciferase gene. The established double stable reporter cell line HLR-GV-$hATF6{\alpha}$(333) represents an innovative tool to investigate regulated intramembrane proteolysis of $ATF6{\alpha}$. It can substitute active pATF6(N) binding motif-based reporter cell lines.
Tuberculosis (TB) remains a serious health issue around the word. Adenovirus (Ad)-based vaccine and modified vaccinia virus Ankara (MVA)-based vaccine have emerged as two of the most promising immunization candidates over the past few years. However, the performance of the homologous and heterologous prime-boost immunization regimens of these two viral vector-based vaccines remains unclear. In the present study, we constructed recombinant Ad and MVA expressing an Ag85B-TB10.4 fusion protein (AdH4 and MVAH4) and evaluated the impact of their different immunization regimens on the humoral and cellular immune responses. We found that the viral vector-based vaccines could generate significantly higher levels of antigen-specific antibodies, $IFN-{\gamma}$-producing splenocytes, $CD69^+CD8^+$ T cells, and $IFN-{\gamma}$ secretion when compared with bacillus Calmette-$Gu{\acute{e}}rin$ (BCG) in a mouse model. AdH4-containing immunization regimens (AdH4-AdH4, AdH4-MVAH4, and MVAH4-AdH4) induced significantly stronger antibody responses, much more $IFN-{\gamma}$-producing splenocytes and $CD69^+CD8^+$ T cells, and higher levels of $IFN-{\gamma}$ secretion when compared with the MVAH4-MVAH4 immunization regimen. The number of $IFN-{\gamma}$-producing splenocytes sensitive to $CD8^+$ T-cell restricted peptides of Ag85B (9-1p and 9-2p) and Th1-related cytokines ($IFN-{\gamma}$ and $TNF-{\alpha}$) in the AdH4-MVAH4 heterologous prime-boost regimen immunization group was significantly higher than that in the other viral vector-based vaccine- and BCG-immunized groups, respectively. These results indicate that an immunization regimen involving AdH4 may have a higher capacity to induce humoral and cellular immune responses against TB in mice than that by regimens containing BCG or MVAH4 alone, and the AdH4-MVAH4 prime-boost regimen may generate an ideal protective effect.
레트로바이러스는 바이러스 외막과 숙주세포막의 융합에 의해 세포내로 들어간다. Moloney 마우스레트로바이러스(murine leukemia virus)의 외막 단백질은 표면단백질(SU)과 막단백질(TM)을 포함하는 85 kDa의 전구체로 합성되는데 바이러스의 성숙과정에 막단백질의 카르복시 말단 16개의 아미노산(R-peptide)이 바이러스의 프로티아제에 의해 절단된다. R 펩타이드가 절단된 막단백질은 Moloney 마우스레트로바이러스에 대한 수용체를 가진 NIH3T3 세포주에서 합포체(syncytium)를 형성한다. R 펩타이드가 절단된 막단백질의 합포체 형성 기작 연구를 위해 R 펩타이드가 절단되어 있으며 표면단백질의 PRR (proline rich region) 부위가 EGFP로 삽입되어진 Moloney full length molecular clone을 만들었다. 이 clone은 NIH3T3 세포에서 합포체를 형성하였으며 형광이 세포질과 세포막에서 관찰되었으나 핵은 염색이 되지 않고 검게 보여 신속 정확하게 합포체 관찰이 가능하였다. 흥미롭게도 절단된 막단백질을 가진 비리온이 NIH3T3 세포에서 광학현미경으로 관찰하였을때는 합포체를 형성하였으나 형광현미경에서는 형광이 관찰되지 않아서 비리온이 세포감염 없이 바이러스-세포 융합 방식으로 합포체를 형성한 것으로 생각되었다. 본 연구에서는 형광의 발현 여부로 합포체 형성을 신속 정확하게 관찰할 수 있는 방법을 개발하였으며 R 펩타이드가 절단된 비리온이 세포 감염 없이 세포와 세포 사이의 융합을 매개할 수 있음을 밝혔다.

뉴켓슬병바이러스(NDV)인 LaSota 주를 SPF 발육난의 요막강내에 증식시켜 순수 정제한 것을 BALB/c 흰쥐에 면역시킨 후 추출한 비강세포와 흰쥐 골수암세포와의 융합방법에 의하여 NDV에 특이하게 작용하는 단크론성항체(MCA)를 생산하는 3주의 Hybridoma틀 작성하였다. 이 3주의 MCA는 모두 IgG형에 속하였으며 흰쥐 복강 내에 접종하여 생산된 복수항체의 항체가는 간접형광항체법으로 $10^3$-$10^6$에 달하였고 약독 및 강독 NDV에 모두 동일한 수준으로 작용하였다. 중화능은 인정되지 않았고 3주중 1주만이 별구응집 억제능을 약하게 나타냈다. 이 MCA를 이용하여 간접형광항체법으로 인공 감염시킨 닭에서 NDV항원 검출을 시도한 결과 기관점막을 비롯한 각종 장기의 도말표본에서 접종 3일 후부터 뚜렷한 검출이 가능하였다.
