Bacillus cereus에 의한 대규모 집단식중독 원인 분석 (Analysis of the Causes of a Large Food Poisoning Outbreak Attributable to Bacillus cereus)

  • 본 연구는 2020년 10월 중 충청남도내 단체 급식소에서 발생한 대규모 집단 식중독 원인에 대하여 분석하였다. 전체 급식원 135명 중 21명(15.6%)에서 음식을 섭취한 후 1시간 이내에 주로 매스꺼움과 구토 증상을 보였다. 유증상자 21명 중 11명과, 조리종사자 1명, 조리기구 2건과 보존식 8건에서 B. cereus가 검출됨에 따라 B. cereus에 의한 집단 식중독으로 판단하였다. 분리된 21개의 균주를 PFGE 분석한 결과, 19개의 균주가 동일한 것으로 판단되었고, 이들 균주가 가지고 있는 독소 유전자는 CER, nheA, entFM이었다. 실험결과, CER을 포함하고, 증상 발현 시간이 1시간 이내로 매우 짧아 B. cereus의 구토형 식중독으로 판단하였다. 집단식중독 원인으로 안전하지 않은 급식환경과 제대로 관리되지 않은 음식에 의한 것이라 조사되었다. 이러한 결과는 단체급식에서의 급식환경과 제공되는 음식이 철저하게 관리되어야 한다는 것을 보여준다. 이와 더불어 보존식에서 원인 병원체를 찾아내는 것은 식중독의 원인을 추정하는데 매우 중요하므로, 집단급식소에서 규정에 맞는 보존식 용기를 이용하여 이를 적정온도에 잘 보관해야한다. 또한 정밀한 식중독 역학조사를 기반으로 사례를 분석하고 결과를 전파함으로써 유사한 식중독이 재발하지 않도록 해야 한다.

  • Background: Although respiratory tract infection is one of the most important factors triggering acute exacerbation of chronic obstructive pulmonary disease (AE-COPD), limited data are available to suggest an epidemiologic pattern of microbiology in South Korea. Methods: A multicenter observational study was conducted between January 2015 and December 2018 across 28 hospitals in South Korea. Adult patients with moderate-to-severe acute exacerbations of COPD were eligible to participate in the present study. The participants underwent all conventional tests to identify etiology of microbial pathogenesis. The primary outcome was the percentage of different microbiological pathogens causing AE-COPD. A comparative microbiological analysis of the patients with overlapping asthma-COPD (ACO) and pure COPD was performed. Results: We included 1,186 patients with AE-COPD. Patients with pure COPD constituted 87.9% and those with ACO accounted for 12.1%. Nearly half of the patients used an inhaled corticosteroid-containing regimen and one-fifth used systemic corticosteroids. Respiratory pathogens were found in 55.3% of all such patients. Bacteria and viruses were detected in 33% and 33.2%, respectively. Bacterial and viral coinfections were found in 10.9%. The most frequently detected bacteria were Pseudomonas aeruginosa (9.8%), and the most frequently detected virus was influenza A (10.4%). Multiple bacterial infections were more likely to appear in ACO than in pure COPD (8.3% vs. 3.6%, p=0.016). Conclusion: Distinct microbiological patterns were identified in patients with moderate-to-severe AE-COPD in South Korea. These findings may improve evidence-based management of patients with AE-COPD and represent the basis for further studies investigating infectious pathogens in patients with COPD.

  • Fish pathology is one of the main scientific bases upon which this expansion in aquaculture has been dependent and requires a wide knowledge of the environmental constraints, the physiology and characteristics of the various pathogens, the responses of the host and the methods by which they may be controlled. The primary disease and parasite problems in aquaculture animals relate to viral, bacterial, fungal and protozoan epizootics. Parasitic nematodes, trematodes and cestodes are commonly found in aquaculture animals, but seldom are they present in concentrations sufficinet to cause significant problems. When an epizootic does occur and chemical treatment is indicated, the appropriate chemical must be selected and properly applied. We have antibiotics, sulfa, nitrofuran and other chemicals for treatment of fish diseases. Some may be mixed with the fred during formulation, added to the pellets of feed as a surface coating, given in the dorm of an injection or used as a bath. Even though a drug or chemical has been officially approved for use in aquaculture, the substance should never be used unless there is a clear need. Some of the reasions for this view are as follows: (1) the constant use of antibiotics can lead to the development of resistant strains of bacteria, (2) biofilter efficiency may be impaired or destroyed by chemicals added to closed recirculating water systems, and (3) the injudicious use of chemicals can have a damaging effect on the environment as well as on human.

Comparative Analysis of the Complete Genome of Lactobacillus plantarum GB-LP2 and Potential Candidate Genes for Host Immune System Enhancement

  • Acute respiratory virus infectious diseases are a growing health problem, particularly among children and the elderly. Much effort has been made to develop probiotics that prevent influenza virus infections by enhancing innate immunity in the respiratory tract until vaccines are available. Lactobacillus plantarum GB-LP2, isolated from a traditional Korean fermented vegetable, has exhibited preventive effects on influenza virus infection in mice. To identify the molecular basis of this strain, we conducted a whole-genome assembly study. The single circular DNA chromosome of 3,284,304 bp was completely assembled and 3,250 protein-encoding genes were predicted. Evolutionarily accelerated genes related to the phenotypic trait of anti-infective activities for influenza virus were identified. These genes encode three integral membrane proteins, a teichoic acid export ATP-binding protein and a glucosamine - fructose-6-phosphate aminotransferase involved in host innate immunity, the nonspecific DNA-binding protein Dps, which protects bacteria from oxidative damage, and the response regulator of the three-component quorum-sensing regulatory system, which is related to the capacity of adhesion to the surface of the respiratory tract and competition with pathogens. This is the first study to identify the genetic backgrounds of the antiviral activity in L. plantarum strains. These findings provide insight into the anti-infective activities of L. plantarum and the development of preventive probiotics.

농촌지역 양돈 종사자의 인수공통감염병 (Zoonoses for Pig Farmers in Rural Communities in Korea)

  • The incidence of zoonoses in Korea has recently increased. But the study for high risk group such as pig farmers to zoonoses has not been conducted in Korea. Thus we reviewed the articles in order to obtain basic data for zoonoses among pig farmers, especially in rural communities. Pigs are one of the most important domestic livestock in Korea not only from economic standpoint but also from standpoint of food. Pigs also represent a potential reservoir for many novel pathogens, therefore may transmit these to humans via direct contact, vectors such as mosquitos, or contaminated meat. The zoonoses associated with pigs can be classified into bacterial pathogen, viruses and so on. Bacterial zoonoses include brucellosis, leptospirosis, listeriosis, enterohemorrhagic Escherichia coli infection, pasteurellosis, salmonellosis, yersiniosis, tuberculosis, anthrax, necrobacillosis, swine erysipelas, erysipeloid, melioidosis, Streptococcus suis infection, Clostrium difficile infection, and campylobactor infection. Viral zoonoses consist of Japanese encephalitis, swine influenza, Nipah virus, Reston ebolavirus, and hepatitis E virus infection. Other type of zoonoses include actinomycosis, toxoplasmosis and Taenia solium infection. These zoonoses were important in Korean health policy but lately they have been overlooked. For effective health policy, we need to study zoonoses associated with pigs, and clinicians and veterinarians must care deeply about these zoonoses.

Host-Induced gene silencing of fungal pathogenic genes confer resistance to fungal pathogen, Magnaporthe Oryzae in rice

  • Recently, host-induced gene silencing (HIGS) system has been successfully applied into development of resistant crops against insects, fungal and viral pathogens. To test HIGS-mediated resistance in rice against rice blast fungus, Magnaporthe oryzae, we first tested possibility of movement of small non-coding RNA from rice cells to rice blast fungus. The rice blast fungus expressing GFP transgene were inoculated to transgenic rice plants ectopically expressing dsRNAi construct targeting fungal GFP gene. Expression of dsRNAi construct for GFP gene in transgenic plants significantly suppressed GFP expression in infected fungal cells indicating that small RNAs generated in plant cells can move into infected fungal cells and efficiently suppress the expression of fungal GFP gene. Consistent with these results, expression of dsRNAi constructs against 3 fungal pathogenic genes of M. oryzae in transgenic rice specifically and efficiently suppressed not only the expression of fungal pathogenic genes, but also fungal infection. The conidia of M. oryzae applied on leaf sheath of transgenic rice expressing dsRNAs against 3 fungal pathogenic genes showed abnormal development of primary hyphae and malfunction of appressorium, which is consistent with the phenotypes of corresponding fungal knock-out mutants. Taken these results together, here, we suggest a novel strategy for development of antifungal crops by means of HIGS system.

Antiviral Activity of Seaweed Extracts against Feline Calicivirus

  • Norovirus, which causes gastroenteritis in humans, is an important food-borne pathogen worldwide. In an effort to discover an antiviral substance against norovirus, extracts from several seaweeds were evaluated for antiviral activity against feline calicivirus (FCV), which was used as a surrogate. The methanolic extract of Undaria pinnatifida exhibited the most significant antiviral activity and virucidal efficacy against FCV. The concentrations of the extract that reduced viral replication by 50% ($EC_{50}$) and resulted in the death of 50% of the host cells ($CC_{50}$) were 0.05 mg/mL and 1.02 mg/mL, respectively. The selectivity index, calculated from the ratio of the $CC_{50}$ and $EC_{50}$ was 20.4. No FCV infection of host cells occurred following a 1-h incubation in the presence of 12.50 mg/mL U. pinnatifida extract, indicating that the virus was completely inactivated by the extract treatment. The results obtained in this study will contribute to the development of a natural antiviral substance that will prevent food-borne disease caused by norovirus.

새로운 생물적 방제 전략: 미생물 인자 유래 식물면역 유도 (Augmenting Plant Immune Responses and Biological Control by Microbial Determinants)

  • 식물은 다양한 병원성 미생물에 대하여 효과적인 방어 기제를 발전시켜 왔다. 최근 유전체와 다중 오믹스 기술의 발전은 우리에게 미생물 인자에 의한 식물 면역을 폭넓게 이해할 수 있는 단초를 제공해 주었다. 하지만 아직까지는 이러한 기술을 병 방제 전략에 이용한 적은 많지 않다. 그래서 본 리뷰에서 식물 면역의 기본 개념을 소개하고 최근 얻어진 결과들을 소개하였다. 덧붙여 이미 논문에서 발표된 진균, 세균, 바이러스 유래 결정인자에 의한 생물적 방제 가능한 방법에 대해 기술하였다. 특히 미생물 결정인자인 chitin, glucan, LPS/EPS, 미생물분자패턴, 항생제, 식물유사호르몬, AHLs, harpin, 비타민, 휘발성물질에 대한 결과를 자세하게 기술하였다. 이 리뷰를 통하여 많은 과학자들과 농민들이 미생물 결정인자 기반의 생물적 방제에 대한 지식이 폭넓어지고, 다양한 미생물 결정 인자가 앞으로 농업현장의 종합적인 병방제 전략의 하나로 자리매김하기를 바란다.

제주도내 자연발생한 말유산태아에서 PCR을 이용한 Equine Herpesvirus Tyre-1 검출 (Detection of equine herpesvirus type-1 in naturally aborted equine fetuses in Jeiu by polymerase chain reaction)

  • It is impotent to identify the causative agents of abortion in equine for minimizing the loss of breeding costs in equine Industry. Recently, the abortion has often occurred in equine herds and thus the purpose of the study was aimed at the identification of equine herpesvirus-1, one of the frequent pathogens to abortion, using polymerase chain reaction. Six fetuses to be aborted at nine to ten months in pregnancy reared in six herds were used in the study. Two primers in the PCR were made from glycoprotein B gene of EHV-1. The primers specific for EHV-1 amplified 1880 bp of PCR products from DNA extracts from thorax fluids, livers, lungs, and spleens of four in six aborted fetuses. Consequently, PCR could be applied to diagnose the abortion of EHV-1 and also confirmed to play a major role of the viral pathogen associated with equine abortion in Jeju island.

무균성수막염과 뇌염환자 사이의 인구학과 검사소견의 비교: 나이와 C-반응단백질의 중요성 (Comparison of the Demographic and Laboratory Profiles of Patients with Aseptic Meningitis and Encephalitis: Significance of Age and C-reactive Protein)

  • Background: Viruses can cause either meningitis or encephalitis. It is unclear why some people suffer from aseptic meningitis, and others acquire aseptic encephalitis when infected with the same viral pathogens. The aim of this study was to compare demographic and laboratory factors between patients with aseptic meningitis and encephalitis. Methods: The demographic and laboratory differences were analyzed according to age, sex, diabetes, hypertension, C-reactive protein in the blood, white blood cell and protein in the cerebrospinal fluid, and glucose ratio (cerebrospinal fluid/blood). Additionally, we analyzed the nation-wide differencesin age between the patients with aseptic meningitis and those with encephalitis in Korea. Results: The patients with aseptic encephalitis were older, more likely to have hypertension, and had higher levels of C-reactive protein than did the patients with aseptic meningitis. However, the numbers of white blood cells in the cerebrospinal fluid were significantly higher in the patients with meningitis than in the patients with encephalitis. Multivariable analysis revealed that age >49 years, hypertension and a C-reactive protein level >5.81 mg/dL were independent and significant variables in the prediction of aseptic encephalitis. Additionally, the patients with aseptic encephalitis were older than those with aseptic meningitis in the nation-wide Korean database. Conclusions: Older age, hypertension, and higher levels of C-reactive protein are useful factors for the prediction of aseptic encephalitis.
