여수 연안 및 동중국해의 세균상 2. 분리균의 수평분포 (Bacterial Flora of East China Sea and Yosu Coastal Sea Areas 2. Horizontal Distributions of Bacteria Isolated from The Sea Area)

  • 1992년 8월 6일 부터 8월 14일 까지에 걸쳐 조사한 동중국해와 여수 연안에서 분리한 세균상은 174균주, vibrio spp. 132균주, 대장균군 183균주에 대한 분리 동정 결과는 다음과 같다. 1. 동중국해 세균상은 분리균 74균주중 40균주가 Pseudomonas spp. $(54\%)$였고, 여수 연안에서는 분리균 100균주중 60균주가 Enterobacteriaceae $(60\%)$였다. 2. Vibrio spp.는 동중국해에서 분리된 5균주중 V. alginolyticus가 4균주, V. parahaemolyticus가 1균주 밖에 분리되지 않았고, 여수 연안에서는 127균주중 V. alinolyticus가 54균주로 $42.5\%$를 차지 하였으며, V. parahaemolyticus는 21균주로 $16.5\%$를 차지하여 이들 두 균종이 Vibrio의 우점종이였다. 3. 대장균군은 동중국해에서 분리한 27균주중 E. coli I이 3균주, C. freundii I이 1균주였고 여수 연안에서는 분리균 156균주중 E. coli I이 79균주 $(50.6\%)$였고 C. freundii I, II가 각각 1균주였다. 4. 대장균군의 IMViC system, $44^{\circ}C$의 발육, gelatin 액화 실험에 의해 16균형으로 분별 되었고 비동정균은 $8\~16$균형으로 분리 균 183균주중 99균주로 $54\%$를 차지하였으며 5. $5^{\circ}C$ 가능한 저온성 대장균군은 15균주였다.

  • this study was performed for the purpose of surveying the distribution and characteristics of the Vibrio species in the diseased farm flounders (Paralichthys olivaceus) during the winter season in Korea. Samples were collected along the southern coast in the Sinam, Keoje Island, and Teosu areas from November '90 to Feburary '91. Nine species of Vibrio were identified as V. tubiashii (85 strains), V. damsela (78 strains), V. anguillarum (21 strains), V. compbelli (9 strains), V. fluvialis (9 strain), V. costicola (1 strain), V. alginolyticus (1 strain), V. gazogenes (1 strain), V. marinus (1 strain), and unidentified 3 groups of bacteria (154 strains) which may have related with V. tubiashii were isolated. Eleven kinds of discs (BBL Co.) were used for antibiotic susceptibility test and 116 strains were selected for the experiment. Most bacteria were powerfully inhibited their growth by 4 antibiotics such as cephalothin, chloramphenicol, colistin, and tetracycline, but they were poorly inhibited by 4 antibiotics such as carbenicillin, kanamycin, penicillin G, and streptomycin.

  • The author was carried out bacteriological identification, and in order to evaluate the sensitivity of the different chemotherapeutic agents including chloramphenicol to Vibrio parahaemolyticus isolated from the stool of the patient's diarrhea. The results obtained were as follows: 1) Biochemical properties of Vibrio parahaemolyticus strains isolated from patients with diarrheal food poisoning was showed Table 1. 2) The sensitivity pattern of the isolated strains of Vibrio parahaemolyticus were sensitive to chloramphenicol, sulfonamide, kanamycin and colistin. But tetracycline, penicillin and leucomycin were resistant.

Detection and Identification of Vibrio Species Using Whole-Cell Protein Pattern Analysis

  • Outbreaks of foodborne diseases associated with Vibrio species such as V. parahaemolyticus, V. vulnificus, and V. cholerae frequently occur in countries having a dietary habit of raw seafood consumption. For rapid identification of different Vibrio species involved in foodborne diseases, whole-cell protein pattern analysis for 13 type strains of 12 Vibrio species was performed using SDS-PAGE analysis. Pathogenic Vibrio species such as V. parahaemolyticus, V. vulnificus, V. cholerae, V. alginolyticus, V. fluvialis, and V. mimicus were included in the 12 Vibrio species used in this study. Each of the 12 Vibrio species showed clearly specific band patterns of its own. Two different strains of V. parahaemolyticus showed two different SDS-PAGE whole-cell protein patterns, giving the possibility of categorizing isolated strains in the same V. parahaemolyticus species into two subgroups. The 36 Vibrio isolates collected from sushi restaurants in Busan were all identified as V. parahaemolyticus by comparing their protein patterns with those of Vibrio type strains. The identified isolates were categorized into two different subgroups of V. parahaemolyticus. The whole-cell protein pattern analysis by SDS-PAGE can be used as a specific, rapid, and simple identification method for Vibrio spp. involved in foodborne diseases at the subspecies level.

Vibrio parahaemolyticus에 대한 황금추출물의 항균활성 (Antimicrobial Activities of Scutellariae Radix Extract against Vibrio parahaemolyticus)

  • 식품 가물로부터 분리.동정 된 V. parahaemolyticus에 대하여 황금추출물의 항균력을 검토하였다. 바지락 72건 가검물 중에서 66건(91.7%)이 양성으로서 높은 검출율을 보였다. 슬라이드 응집법으로 혈청형을 확인한 결과 분리 된 V. parahaemolyticus 66균주 중에서 혈청형을 결정할 수 없는 UT(untypeable) K-group이 46균주로서 69.7%였으며, 확인된 혈청형 중에서는 serotype K-I group이 7균주로서 10.6%의 검출율을 나타내었고, K-IV group 5균주(7.6%), K-II, V, VII group이 각각 2균주(3.0%)이 었으며, K-VI, VIII group 1균주(1.5%)의 순으로 분리되었고, K-III와 IX group 혈청형은 검출되지 않았다. 식품에 있어서 생육관계를 조사하기 위하여 시판되는 바지락을 시료로 분리균주들을 각각 접종한 다음35$^{\circ}C$에서 120시간동안 배양하여 얻은 생육곡선에서 V. parahemolyticus균주들 모두가 유도기, 대수기, 정지기, 사멸기를 거치는 전형적인 S자 모양의 생육곡선을 보였다. 식품에 대한 황금추출물의 항균작용을 알아보기 위하여 시판되는 바지락을 시료로 하여 증기 멸균한 후, 동량의 멸균생리식염수를 넣고 황금추출물의 농도가 0, 500, 1000, 5000 ppm (mg/kg)이 되도록 균질화시킨 시료에 분리균주들을 각각 접종한 다음, 35$^{\circ}C$에서 48시간 동안 배양하여 얻은 생육저해곡선에서 V. parahaemolyticus 균주들 모두가 유도기를 거친 다음인 6시간이 경과된 후 황금추출물의 농도에 따라 시험균주의 균수 차이를 보였고, 식품중의 Vibrio parahaemolyticus에 있어서 생육이 억제되는 황금추출물 유효저해농도는 1000ppm이었다. 세포형태변화를 관찰하기 위한 투과형전자현미경(TEM) 상에서 황금추출물 처리로 인한 V. parahaemolyticus균주들의 세포막기능의 파괴로 세포내용물들이 용출된 것을 볼 수 있었다.

The Viable But Nonculturable State of Kanagawa Positive and Negative Strains of Vibrio parahaemolyticus

  • Ingestion of shellfish-associated Vibrio parahaemolyticus is the primary cause of potentially severe gas-troenteritis in many countries. However, only Kanagawa phenomenon (hemolysin) positive (KP$\^$+/) strains of V. parahaemolyticus are isolated from patients, whereas >99% of strains isolated from the environment do not produce this hemolysin (i.e. are KP$\^$-/). The reasons for these differences are not known. Following a temperature downshift, Vibrio parahaemolyticus enters the viable but noncultur-able (VBNC) state wherein cells maintain viability but cannot be cultured on routine microbiological media. We speculated that KP$\^$+/ and KP$\^$-/ strains may respond differently to the temperature and salinity conditions of seawater by entering into this state which might account for the low numbers of cul-turable KP$\^$+/ strains isolated from estuarine waters. The response of eleven KP$\^$+/ and KP$\^$-/ strains of V. parahaemolyticus following exposure to a nutrient and temperature downshift in different salinities, similar to conditions encountered in their environment, was examined. The strains included those from which the KP$\^$+/ genes had been selectively removed or added. Our results indicated that the ability to produce hemolysin did not affect entrance into the VBNC state. Further, VBNC cells of both biotypes could be restored to the culturable state following an overnight temperature upshift.

A Profile of Naturally Occurring Plasmids from Selected Strains of Vibrios

  • The naturally occurring plasmids of Vibrio species have been isolated in part to investigate their genetic traits. Among six different Vibrio species tested, Vibrio anguillarum, Vibrio fluvialis, Vibrio vulnificus, Vibrio mimicus and Vibrio furnissi did not show any presence of plasmid. One environmental isolate of Vibrio pnrahemolyticus harboring plasmid was observed. The isolated plasmid was 8.7 kb by analysis with restriction endonuclease digestion. No common feature was shown relationships between the presence of plasmid and resistance against commonly used antibiotic compounds from the tested Vibrios. Activities of Essential Oils from Perilla frutescens var. acuta against Antibiotic-Susceptible and -Resistant Vibrio and Salmonella Species

  • We determined the inhibitory activity of the essential oil fraction obtained by steam distillation from the fresh and dried leaves of Perilla frutescens var. acuta against some pathogenic Salmonella and Vibrio spp. The activities of compounds isolated from the essential oils, apiol and myristicin, were also tested and the results were compared with those of the essential oil fraction. The Perilla essential oil fraction and its main components showed significant inhibition against antibiotic-susceptive and antibiotic-resistant strains of the tested Salmonella and Vibrio strains. Synergistic or additive effects were identified by combing the oils with ampicillin by checkerboard-titer tests. We conclude that essential oils from P. frutescens can be useful in the treatment of Salmonella and Vibrio infections and as safe additives to food materials for the prevention of contamination of food by these bacteria. This is especially important because of the rapid increase in antibiotic-resistant strains, which could cause severe symptoms in humans.

한국연안의 호염성 비브리오균에 관한 연구 3. 부산시 해수욕장의 호염성 비브리오균의 분포 (Vibrio parahaemolyticus IN KOREAN COASTAL WATERS 3. The Ditribution of Vibrio Flora in the Public Beach of Busan City)

  • 호염성 비브리오균의 계절별 분포를 알고 식중독 구명자료를 얻고자 해운대, 수영, 광안리, 용호, 송도, 다대포에서 해수, 이토, 어패류 198 시료를 수집하여 이들 시료에서 분리된 세균에 관하여 몇 가지 견해를 얻었기에 보고한다. 1. 계절별 분포율을 보면 V. parahaemolyticus는 5월부터 11월까지 분포하고 8월부터 9월까지 높은 분포를 모였으며, V. alginelyticus는 4 계절 분포를 나타내었다. 2. 시료별 분포율은 해수 40 시료에서 40균주, 이토 40 시료에서 25균주, 맛조개 48 시료에서 10 균주, 진주담치 40 시료에서 2 균주, 전어 10 시료에서 1 균주, 숭어 10 시료에서 1 균주가 분리되었다. 3. 해역별 분리율은 해운대 20 시료 중 4 균주, 수영에서 25 시료 중 19 균주, 광안리 26 시료중 3균주, 용호만 35 시료 중 29균주, 송도 29 시료 중 6균주, 다대포 63시료중 19균주로서, 용호만과 수영만에서 높은 분포를 보였다. 4. 분리된 총균주수는 80 균주였는데 V. parahaemolyticus가 20 균주, V. alginelyticus 60 균주였다.
