The Pathogenicity and Biochemical Characteristics of Vibrio harveyi Isolated from the Pacific Abalone, Haliotis discus hannai (양식 전복(Haliotis discus hannai)으로부터 분리된 Vibrio harveyi의 생화학적 특성 및 병원성)

  • Recently, mass mortality of the young abalone Haliotis discus hannai has occurred in commercial seed production farms in Korea. The mortality rate was above 50% of the total cultured organisms in the farm, and the shell length of the moribund organisms was about 3cm. The mortal phenomenon was that the young abalones were weakly scattered on the bottom of the pond from the attachment matrix, or that they could not be moved back to their normal positions. The diseased farmed Pacific abalone had abdominal edema. From the edema in the moribund individuals, three bacterial strains were isolated and all the strains were identified as Vibrio harveyi. These strains were compared with thirty six strains isolated from the fish. The results was that the Vibrio harveyi from the fish were sorted into genogroup A or B; however, the three strains of the diseased farmed Pacific abalone were sorted into genogroup A and the new genogroup C. The identical mortality and pathological symptoms of the naturally infected organisms were reproduced by artificial infection with WA AG-1 and WA CS-5 strains. The $LD_{50}$ of WA AG-1 and WA CS-5 were each $1.0{\times}10^3cfu\;animal^{-1}$ and $1.7{\times}10^4cfu\;animal^{-1}$.

  • We experienced mortality of dark-banded rockfish (Sebastes inermis) maintained in the rearing facility located in Gangneung, after transportation. Moribund fish showed various symptoms such as exophthalmia, skin ulcers, tail rots, gill rots, discoloration of liver with petechiae, yellowish fluid in intestines and ascites. Two different colonies were dominantly appeared after spreading the lesions on the agar plates and incubation. One isolate (SI_1) showed swarming movement on TSA, and formed yellow colonies on TCBS agar. The other (SI_2) showed no swarming motility and green colonies on TCBS agar. Both of them were Gram-negative. All of these results are similar with those of Genus Vibrio. They were identified as V. harveyi and V. gigantis by PCR with subsequent sequencing of 3 different genes (16 rDNA, recA, rpoA). V. harveyi is well-known as a serious pathogen of marine fish and invertebrates, while V. gigantis is known to be often isolated from marine invertebrates, but the pathogenicity is still unknown. We suspect V. harveyi as the cause of the mortality of dark-banded rockfish, but challenge experiments with these 2 Vibrio species are thought to be necessary to make a clear conclusion.

Designing a novel mRNA vaccine against Vibrio harveyi infection in fish: an immunoinformatics approach

  • Vibrio harveyi belongs to the Vibrio genus that causes vibriosis in marine and aquatic fish species through double-stranded DNA virus replication. In humans, around 12 Vibrio species can cause gastroenteritis (gastrointestinal illness). A large amount of virus particles can be found in the cytoplasm of infected cells, which may cause death. Despite these devastating complications, there is still no cure or vaccine for the virus. As a result, we used an immunoinformatics approach to develop a multi-epitope vaccine against most pathogenic hemolysin gene of V. harveyi. The immunodominant T- and B-cell epitopes were identified using the hemolysin protein. We developed a vaccine employing three possible epitopes: cytotoxic T-lymphocytes, helper T-lymphocytes, and linear B-lymphocyte epitopes, after thorough testing. The vaccine was developed to be antigenic, immunogenic, and non-allergenic, as well as having a better solubility. Molecular dynamics simulation revealed significant structural stiffness and binding stability. In addition, the immunological simulation generated by computer revealed that the vaccination might elicit immune reactions in the actual life after injection. Finally, using Escherichia coli K12 as a model, codon optimization yielded ideal GC content and a higher codon adaptation index value, which was then included in the cloning vector pET2+ (a). Altogether, our experiment implies that the proposed peptide vaccine might be a good option for vibriosis prophylaxis.

  • 1999년 여름철 우리 나라의 몇몇 해수 어류 양식장과 종묘 배양장에서 Vibrio harveyi가 원인균으로 생각되는 질병이 발생하였다. 어종별 특징적인 증상으로는 넙치의 탈장, 복부팽만, 장충혈, 장액 삼출 및 간출혈과 조피볼락의 체표 궤양 및 간울혈을 들 수 있었으며 같은 시기에 감염증을 나타낸 turbot에서는 체표 출혈과 장출혈이 특징적인 증상이었다. 이 중에서도 특히 넙치의 병징은 에드와드병으로 추정되기 쉬운 특징이 있기 때문인지 각 양어장에서는 간단히 에드와드병으로 진단하고 그에 대한 치료가 실시되고 있었으나 치료 효과가 비교적 낮아서 피해가 증가되고 있는 실정이었다. Differentially Expressed Genes in Hemocytes of Vibrio harveyi-challenged Shrimp Penaeus monodon

  • Differential Display PCR technique (DD-PCR) was used for the analysis of altered gene expression in hemocytes of Vibrio harveyi-infected Penaeus monodon. Forty-four combinations of arbitrary and oligo(dT) primers were used to screen for differentially expressed genes. A total of 79 differentially expressed bands could be identified from 33 primer combinations. These included 48 bands (61%) whose expression level increased and 31 bands (39%) decreased after V. harveyi challenge. Subsequently, forty-eight differential display fragments were successfully reamplified and cloned. A total of 267 clones were randomly selected and sequenced. The sequence analysis showed that 85 (31%) out of 267 clones were matched with sequences in the GenBank database which represented 24 different genes with known functions. Among the known genes, glucose transporter 1, interferon-related developmental regulator 1, lysozyme, profilin, SERPINB3, were selected for further confirmation of their differentially expression patterns by real-time PCR. The results showed increasing in expression level of the selected genes in shrimp hemocytes after microbial challenge suggesting the involvement of such genes in bacterial response in shrimp. The anti-lipopolysaccharide factor type 3 (ALFPm3) gene, previously reported in P. monodon (Supungul et al., 2002) was found among the up-regulated genes but diversity due to amino acid changes was observed. Increase in ALFPm3 transcripts upon V. harveyi injection is in accordance with that found in the previous study.

Isolation and identification of Vibrio harveyi from chub mackerel (Scomber japonicus)

  • For several days, there was a series of mortalities of chub mackerel (Scomber japonicus) that were reared for public exhibition in a private aquarium in Seoul, Korea. As part of the diagnosis of the dead fish, a bacterial isolate from the kidney was cultured, identified, and confirmed to be Vibrio (V.) harveyi using Vitek System 2 and 16S rRNA gene sequencing. Phylogenetic analysis was also performed by the neighbor-joining method. As a result, the V. harveyi isolated from chub mackerels of a private aquarium in Korea, called as SNUVh-LW1, was clustered in the same group with V. harveyi ATCC33843.

  • A lot of cultured flounders died by ascitic disease blooming during the summer season in 2002, southern parts of Korea. This study was conducted to investigate the microbial flora for the ascitic cultivated olive flounder (Paralichthys olivaceus) which were collected in the Koe-je island in the southern of Korea from July 2002 to October 2003. Three Genera (Vibrio, Photobacterium and Edwardsiella), seven species of bacteria (V. harveyi, V. alginolyticus, V. parahaemolyticus, V. carcariae, V. metschenikovii, P. damsela and E. tarda) and a fish pathogenic birnavirus (marine birnavirus, MABV) were isolated from the liver, dropsy, spleen and identified by biochemical and molecular biological characterization.

  • The inhibitors against Vibrio harveyi quorum sensing (QS) signaling were developed by modifying the molecular structure of the major signal, N-3-hydroxybutanoyl-L-homoserine lactone (3-OH-$C_4$-HSL). A series of structural derivatives, N-(3-hydroxysulfonyl)-L-homoserine lactones (HSHLs) were synthesized by the solid-phase organic synthesis method. The in vivo QS inhibition by these compounds was measured by a bioassay system using the V. harveyi bioluminescence, and all showed significant inhibitory effects. To analyze the interaction between these compounds and LuxN, a 3-OH-$C_4$-HSL receptor protein of V. harveyi, we tentatively determined the putative signal binding domain of LuxN based on the sequence homology with other acyl-HSL binding proteins, and predicted the partial 3-D structure of the putative signal binding domain of LuxN by using ORCHESTRA program, and further estimated the binding poses and energies (docking scores) of 3-OH-$C_4$-HSL and HSHLs within the domain. In comparison of the result from this modeling study with that of in vivo bioassay, we suggest that the in silica interpretation of the interaction between ligands and their receptor proteins can be a valuable way to develop better competitive inhibitors, especially in the case that the structural information of the protein is limited.

Low-value Fish used as Feed is a Source of Disease in Farmed Fish

  • Low-value fish is the most commonly used feed in Asian fish farms despite the fact that its application is controversial in regard to the sustainability and biosecurity of aquaculture. In this study, the causal agent of a disease outbreak at a Korean rockfish Sebastes schlegeli farm was investigated to determine whether the low-value fish used at the farm was the source. Infected Korean rockfish and Pacific sand eel used as feed were sampled from the farm, and bacterial cultures recovered from the internal organs of all sampled rockfish were isolated as pure cultures and later identified as Vibrio harveyi. The causal agent of the disease was also isolated from the kidneys of some of the sampled Pacific sand eels. This study provides additional evidence that the low-value fish used as feed at fish farms can be a key source of infectious diseases.

  • 1999년 여름철 고수온기에 경북, 경남 및 부산 인근의 몇몇 넙치 양식장에서 세균성 감염증으로 보이는 질병에 의해 넙치의 폐사가 발생하여 그 원인을 조사하였다. 감염어는 복부팽만, 장충혈, 장액삼출 및 간출혈 둥의 특징적 증상을 보이면서 폐사하여, 에드워드병이나 연쇄구균증으로 의심되었다. 그러나 감염어의 환부 및 내부 장기에서 분리된 세균은 1.5% NaCl 첨가 TSA 평판배지에서 swarming을 하는 특성을 보였으며, TCBS에서 yellow 혹은 green colony를 형성하는 비브리오균인 것으로 나타나, 여러 가지 생화학 실험을 통해 V. harveyi로 추정되었다.
