용혈독소를 생산하는 기수성 비브리오균의 생리.생태적 특성과 수산식품의 위생대책 2. 해수에서 분리된 vibrio mimicus SM-9의 생리적 특성 및 저온내성 (Physiological and Ecological Characteristis of Hemolytic Vibrios and Development of Sanitary Countermeasure of Raw Fisheries Foods. 2. Physiological and Psychrotrophic Characteristics of Vibrio mimicus SM-9 Isolated from sea Water)

  • Vibrio mimicus is a closely related species with V. cholerae, and has been reported to be associated with gastrointestinal infections. Although extraintestinal infections of these vibrios have also been reported in Japan and Southeast Asia. But little research papers on V. mimicus was reported in Korea. Therefore, we tried to isolate V. mimicus from the environmental sea water from April to July in Pusan, Korea. Among the isolated strains, we selected the strongest hemolytic strain and then named V. mimicus SM-9. In this paper, we checked the antibiotic susceptibility and psychrotrophic characteristics of the isolated strain. Hemolytic activity of the hemolysin produced by the isolated strain was also measured. V. mimicus was not detected from the sea water samples in April and May, but its detection rate was relatively high in June and July in Pusan, Korea. The bacteriological characteristics of V. mimicus SM-9 were Gram-negative rods, motile, oxidase positive, Voges-Proskauer negative and sucrose negative. In 23 kinds of antibiotics susceptibility test, V. mimicus SM-9 showed susceptibility to the most of antibiotics submitted while it was resistive against lincomycin, oxacillin, rifampin and vancomycin. Hemolytic activity of the hemolysin produced by V. mimicus SM-9 was highest in stationary phase of the growth curve in BHI broth at 37$^{\circ}C$ and its activity was reached 18 HU per $m\ell$ of culture supernatant. For checking the psychrotrophic property of V. mimicus SM-9, the decreasing rate of the strain in phosphate buffer solution and yellowtail flesh homogenate was examined during the storage at 4, 0, -4 and -2$0^{\circ}C$. The decreasing rates of the selected strain stored in phosphate buffer solution were greater than those in fish homogenate. Decreasing rates of V. mimicus SM-9 stored in phosphate buffer solution were not significantly different by the storage temperatures. The viable cell counts of the strain were decreased as 5 log cycles after 120 hours at all the tested temperatures. While decreasing numbers of the strain in fish homogenates were 2*4 log cycles after 120 hours. The decreasing pattern of the strain numbers were very slow after 200 hours at all the stored temperatures.

  • To investigate the epidemiological trait of intestinal diseases of animals caused by thermophilic Campyllobacter spp., isolation of etiological agent was carried out and the profiles of plasmids and the transfer of resistance plasmid in the isolated Campylobacter spp. were examined. The results were as follows. 1. A total of 110 isolates of C jejuni and C coli were subjected to the test for the presence of plasmid DNA. Of the isolates examined, 60% of the isolates were noted to harbor plasmid DNA. Plasmid occurrencer ate from pigs, chickens and cattle were 76.2%, 61.7% and 37.7%, respectively. The plasmids of a large molecular weight, ranging from 36 Md to 86Md, were identified with the strains of tetracycline resistant. 2. Transfer frequency of tetracycline resistant plasmids was higher in the case of the filter mating method than in the broth mating method by the factor of 10~1,000. 3. Tetracycline resistant plasmids of C jejuni were transferrable to C jejuni and C coli by conjugation. In a low frequency, the transfer of tetracycline plasmid was also possible to Vibrio parahemolyticus. However, it was impossible to transfer to Streptococcus fecalis, E coli and Vibrio cholerae. 4. Tetracycline resistant plasmids of C jejuni were impossible to transfer to Campylobacter spp. and related bacteria by transformation.

Role of TolC in Vibrio vulnificus Virulence in Mice

  • The role of a TolC homologue in the virulence of Vibrio vulnificus, a marine bacterium causing serious wound infection and fulminant septicemia in persons with underlying conditions, has been studied. TolC, an outer membrane protein, has been implicated in a variety of bacterial functions including export of diverse molecules ranging from large proteins to antibiotics. A homologue of the tolC gene of V. cholerae, which has been shown to be required for bile resistance, cytotoxicity and colonization of this organism, was identified in the partially determined genome sequence of V. vulnificus. To determine the role of TolC in the virulence of V. vulnificus, a TolC-deficient (TD) mutant was isolated by in vivo allelic exchange. Compared with the parent strain, the TD mutant was more sensitive to bile, and much less virulent in mice challenged subcutaneously. This mutant was noncytotoxic to the HEp-2 cells, but its metalloprotease and cytolysin activities in the culture supernatant were comparable to the parent strain. In addition, the resistance of the TD mutant to human serum bactericidal activity as well as its growth in either human or murine blood was not affected. Collectively, our data suggest that TolC may be involved in colonization and/or spread of V. vulnificus to the blood stream, probably by secreting a cytotoxin other than the cytolysin.

통영항의 해수 및 저질의 위생학적 특성 (Sanitary Characteristics of Seawater and Sediments in Tongyeong Harbor)

  • 이 연구는 통영항 내의 해수와 저질의 특성과 주변횟집의 수질의 품질 특성에 대하여 2008년 6월부터 2009년 5월까지 월 1회 조사를 실시하였으며 그 결과는 다음과 같다. 통영항의 해양수온은 평균 $17.4{\pm}5.77^{\circ}C$, 투명도 평균 $1.9{\pm}0.46$ m, 부유물질 $36.7{\pm}5.00$ mg/L, COD $2.36{\pm}0.80$ mg/L, DO $6.67{\pm}1.54$ mg/L, 염분농도 $32.3{\pm}0.64$‰를 각각 나타내었다. 아질산 질소는 $0.010{\pm}0.005$ mg/L, 질산 질소 $0.050{\pm}0.011$ mg/L, 암모니아 질소 $0.030{\pm}0.007$ mg/L, 인산 인 $0.050{\pm}0.006$ mg/L로 각각 나타났으며, TBT는 검출한계 이하로 검출되었다. 통영항 해수 중의 total coliform은 조사기간 중 < 1.8~22,000 MPN/100 mL 범위에 기하학적 평균값이 164.9 MPN/100 mL이었고 230 MPN/l00 mL을 초과하는 비율은 39.6%를 나타내었다. fecal coliform은 < 1.8~7,900 MPN/100 mL 범위에 기하학적평균값은 33.7 MPN/100 mL, 43 MPN/100 mL을 초과하는 비율은 47.9%로 조사되었다. total coliform에 대한 fecal coliform 검출 비율은 69.2%로 나타났다. 통영항 해수 중의 비브리오는 6월부터 11월에 240주가 검출되었으며, V. alginolyticus가 82주로 34.2%를 차지하여 가장 많이 검출되었고, 그 밖에 parahaemolyticus가 13.8%, V. culnificus 10.0%, V. cholerae non-O1이 0%, V. mimicus 12.5%가 검출되었다. 저질 특성은 함수율이 $56.63{\pm}5.51$ %, 강열감량 $9.20{\pm}1.36$%, COD $7.64{\pm}1.13$ mg/g, AVS $0.07{\pm}0.02$mg/g으로 다른 해역에 비하여 다소 높게 나타났다. 중금속의 함량은 카드뮴(Cd)은 $0.10{\pm}0.05$, 구리(Cu) $4.79{\pm}8.20$, 비소(As) $1.95{\pm}0.17$, 수은(Hg) $0.10{\pm}0.07$, 납은 불검출, 6가크롬은 ($Cr^{6+}$) $0.34{\pm}0.12$, 아연 (Zn) $125.33{\pm}16.40$, 니켈(Ni) $16.34{\pm}1.93$ mg/kg을 나타내었다. 통영항 주변 횟집의 total coliform 및 fecal coliform은 횟집에 따라서 다소 차이가 있었지만 여름철에 주로 검출되었으며, 여름철 집중관리가 요구되었다.

Antibacterial activity of methanol extract of roots of Heracleum nepalense D Don. on bacteria causing diarrhoea

  • Heracleum nepalense D Don. (Umbelliferae) is a small shrub having high glabrescent stem found in stream banks in Sikkim. Various medicinal properties which include antidiarrhoeal, antiseptic, anti-influenzal etc. have been attributed for this plant in the traditional system of medicine in Sikkim. In present investigation the methanol extract of roots of Heracleum nepalense was subjected for its effectiveness against both Gram-positive and Gram-negative bacteria causing diarrhoea. The roots extract was tested for its minimum inhibitory concentration (MIC) against both Gram-positive and Gram-negative organisms causing diarrhoea. Further, the zones of inhibition produced by the crude extract against few sensitive strains was measured and compared with those of standard antibiotic ciprofloxacin. It is evident that the methanol extract is very active against the bacteria causing diarrhoea at low concentrations. The antibacterial efficacy of the root extract was found to decrease in the following order against different tested bacterial strains like Shigella dysenteriae, Escherichia coli, Shigella boydii, Vibrio cholerae, Salmonella typhimurium.

Identification and Antibacterial Activity of Volatile Flavor Components of Cordyceps Militaris

  • Flavor characteristics of raw Cordyceps militaris significatntly different from those of dried one. In the case of raw Cordyceps militaris , major flavor components were composed of 5 alcohols, 3 ketones, 4 phenols, 9 alkanes , and 3 alkenes. The major alcohol was 1-octen-3-ol(22.56%, 1147.3% ng/ml), which contributed to the characteristic green flavor. Ketones (3-ocatone, inparticular )were present in the highest concentration in raw Cordyceps militaris . In contrast, major flavor components of dried Cordyceps militaris were composed of 4 alcohols, 4 ketones, 3 furans, 4 pyrizines, 2 dithiazines, 5 phenols , 8alkenes , 17 alkanes, and 8 fatty acids. Dried Cordyceps militaris had unique sweet aroma of sesame as wella s a milky flavor. Green or fruit flavor were rarely detected . In alkanes , 10 cosanes, component fo wax were present. Typical flavor components of alkanes such as $\beta$-caryophyllen and Δ-cadinene were also detected. Fatty acids of dried Cordyceps militaris ranged from myristic acid (14 :0) to linoleic acid (18 ; 2). The sweet aroma of dried Cordyceps militaris was mostly due to pryazines, dithaiazines, and furans. Two dithaizines were identified and characteristics of these flavor components was a roasted bacon flavor. Strong antibacterial acitivity was observed toward Vibrio spp. such as V. vulnificus, V.cholerae, V. parahaemlyticus. Relatively high antibacterial acitivity was shown toward Bacillus subtilis , B,cereus, Staphyllococcus aureus, and Corynebacterium xerosis.

Studies on Synonymous Codon and Amino Acid Usage Biases in the Broad-Host Range Bacteriophage KVP40

  • In this study, the relative synonymous codon and amino acid usage biases of the broad-host range phage, KVP40, were investigated in an attempt to understand the structure and function of its proteins/protein-coding genes, as well as the role of its tRNAs. Synonymous codons in KVP40 were determined to be AT-rich at the third codon positions, and their variations are dictated principally by both mutational bias and translational selection. Further analysis revealed that the RSCU of KVP40 is distinct from that of its Vibrio hosts, V. cholerae and V. parahaemolyticus. Interestingly, the expression of the putative highly expressed genes of KVP40 appear to be preferentially influenced by the abundant host tRNA species, whereas the tRNAs expressed by KVP40 may be required for the efficient synthesis of all its proteins in a diverse array of hosts. The data generated in this study also revealed that KVP40 proteins are rich in low molecular weight amino acid residues, and that these variations are influenced primarily by hydropathy, mean molecular weight, aromaticity, and cysteine content.

새로운 항균활성을 보이는 토양 분리 세균 Paenibacillus polymyxa DY1의 분류와 동정 (Identification and Characterization of Paenibacillus polymyxa DY1 Isolated from Korean Soil with New Antibacterial Activity)

  • 항생제 내성 세균의 출현으로 새로운 항생물질의 개발에 대한 필요성이 대두되고 있다. 본 연구에서는 새로운 항균활성물질을 개발하고자 강원도 대암산 용늪 토양으로부터 새로운 항균물질을 생산하는 균을 분리하였고, 이를 동정하였다. 생화학적인 시험과 16S ribosomal DNA 염기서열 분석결과 Paenibacillus polymyxa균과 가장 높은 상동성을 보여주었다. 지방산 조성의 분석에서도 이 균주는 Paenibacillus polymyxa와 가장 가까웠다. 이 균주가 생산하는 항균물질은 1군 법정 전염병을 일으키는 Samonella enterica serovar Typhi와 Shigella dysentery, enterohaemorrhagic Eschelichia coli, 그리고 Vibrio cholera등의 병원성 세균에 성장억제 효과를 나타냈으며, 다른 일반 식중독 장내세균에서도 성장억제 효과를 나타냈다. 이 균주가 생산하는 항균활성 물질은 과거에 보고된 것과 다른 새로운 것으로 보이며, 광범위한 항균활성으로 인하여 새로운 항생물질 개발 후보로 많은 잠재력을 가진 것으로 평가된다.

경남연안 기수지역에서 분리된 비브리오균의 항균제 내성 (Antimicrobial Resistance of Vibrio Strains from Brackish Water on the Coast of Gyeongsangnamdo)

  • Antimicrobial resistance patterns of Vibrio species isolated from brackish water in Geoje, Tongyeong and Goseong, Gyeongsangnamdo province into which streams, sewage and leachate all flowed. Only 19 strains (10.7%) of 177 V. parahaemolyticus were susceptible to 15 antimicrobials. 146 strains (69.5%) proved resistant against more than one antimicrobial and 12 strains (6.8%) were multi-drug resistant. The resistance rate of 152 strains were 85.9% against AM and 26.6% against RA, 16.4% against AN, 13.6% against Sand 13.0% against TMP. 86 strains of 129 V. cholerae non-O1 (66.7%) were susceptible to antimicrobials and 31 strains (24.0%) were resistant to more than one antimicrobial and 12 strains (9.3%) were multi-drug resistant. The antimicrobial resistance rate of 129 strains against 15 antimicrobials, with the exception of C, CIP, E and GM, i.e. 11 antimicrobials, was 0.7-16.2%, 16.2% of 129 strains proved resistant against RA and 13.9% against AM, 9.3% against TMP, 7.7% against SXT and 6.9% against TE. 19 of 49 strains of V. mimicus (38.8%) were susceptible to antimicrobials and 31 strains (61.2%) were resistant against more than one antimicrobial; none of the strains were multi-drug resistant. 15 strains of V. mimicus were resistant against only RA, AmC and TE. The resistance rate was 59.2% against RA (highest) 4.1% against AmC and 2.0% against TE.

ERIC-PCR genomic fingerprinting에 의한 주요 식중독 그람 음성 세균 4속의 구별 (Differentiation of Four Major Gram-negative Foodborne Pathogenic Bacterial Genera by Using ERIC-PCR Genomic Fingerprinting)

  • 본 연구에서는 높은 분리능을 가지고 있을 뿐만 아니라 실험의 재현성과 경제성의 측면에서도 많은 장점을 갖고 있는 ERIC DNA sequence를 응용한 ERIC-PCR을 이용하여 Salmonella, E. coli, Shigella, Vibrio 등 4속의 주요 그람 음성 식중독유발 세균들의 분리 동정 방법을 확립하고자 하였다. ERIC-PCR 결과, E. coli의 경우 0.3kb, 0.42kb 및 1.2kb의 band가 모든 균주에서 공통적으로 확인되었고, Salmonella속으로부터는 0.22kb, 0.4kb 및 0.7kb의 band가 증폭되었다. Shigella속은 모든 표준균주와 분리균주로부터 0.33kb와 1.25kb의 band가 증폭되었으며, S. sonnei의 경우 위의 주요 2개 band 이외에도 대부분의 균주에서 0.44kb, 2.0kb 및 3.05kb의 band가 증폭되어 다른 종의 Shigella와 구별되는 fingerprinting pattern을 나타내었다. 그리고 V. parahaemolyticus의 경우 표준균주와 분리균주 모두 0.51kb와 1.5kb의 band가 증폭되어 V. cholerae, V. mimicus 등과 같은 다른 종의 Vibrio와 구별되는 fingerprinting pattern을 나타내었다. 이와 같이 4속의 모든 식중독 균주마다 ERIC-PCR후 생성되는 fingerprinting pattern에서 3-5개의 공통적인 band가 증폭되는 것이 확인되어 이를 이용한 속 수준의 분리 동정과 이러한 주요 band들 이외의 부수적인 band들을 고려하여 종 수준까지의 분리도 가능함을 확인하였다. 따라서 본 연구의 결과는 ERIC 반복적 DNA 염기서열을 이용한 ERIC-PCR이 식중독균의 분리 동정 방법으로 사용될 수 있음을 확인하였으며, 나아가 더 많은 속(genus)의 식중독세균을 대상으로 한 새로운 분리 동정 방법을 확립하는데도 응용이 될 수 있을 것이다.
