장염(腸炎) 비브리오의 역학(疫學) 및 병원성(病原性)에 관(關)한 연구(硏究) - III. 마산시(馬山市) 연안(沿岸)에서의 분리상황(分離狀況) (A Study of Epidemiology and Pathogenicity of Vibrio parahaemolyticus - III. Isolation of Vibrio parahaemolyticus around seashore of Masan)

  • This study was undertaken to know the distribution of V. parahaemolyticus in sea fishes feeding at an aquarium and from kitchen environments of restaurants located in Masan, Kyungnam, during May and June, 1979. Results obtained are as follows: The detection rate of V. parahaemolyticus was calculated as 21.1% from 1103 specimens; as 38.1% from 336 sea fishes and as 10.4% from 675 kitchen environments. The detection rate of V. parahaemolyticus were 49.6% from 127 sea fishes feeded at an aquarium contaminated by V. parahaemolyticus and 31.1% from 209 sea fishes feeded at an aquarium not contaminated by V. parahaemolyticus. The detection rate of V. parahaemolyticus were shown as 14.6% from 261 kitchen environments having an V. parahaemolyticus contaminated aquarium but as 7.7% from 414 kitchen environments of restaurants having an aquarium not contaminated by V. parahaemolyticus.

  • Acute hepatopancreatic necrosis disease (AHPND), previously known as early mortality syndrome (EMS), is an emerging disease in shrimp caused by Vibrio parahaemolyticus. Some V. parahaemolyticus strains are associated with foodborne diseases in humans. To date, studies on the relationship between AHPND and pathogenic V. parahaemolyticus are very limited. In this study, we monitored the thermostable direct hemolysin-related hemolysin (trh) gene and AHPND-related genes, such as Photorhabdus insect-related (pir) genes, in 892 strains of V. parahaemolyticus isolated and identified in 24 areas of the West Coast of Korea from May to October 2019. The trh gene was detected in 9.6% of the isolates from short neck clam samples. However, the pirA and pirB genes related to AHPND were not found in any of the isolates despite using both duplex and nested PCR assays, suggesting that AHPND-related genes were nonexistent in the V. parahaemolyticus strains isolated. This study contributes to the current understanding of the relationship between AHPND and V. parahaemolyticus in Korea, as well as provides data on spatial and seasonal distributions of V. parahaemolyticus.

Quantitative microbial risk assessment of Vibrio parahaemolyticus foodborne illness of sea squirt (Halocynthia roretzi) in South Korea

  • The annual consumption of fishery products, particularly sea squirt (Halocynthia roretzi), per person has steadily increased in South Korea. However, the quantitative risk of Vibrio parahaemolyticus following intake of sea squirt has not been analyzed. This study focuses on quantitative predictions of the probability of consuming sea squirt and getting of V. parahaemolyticus foodborne illness. The prevalence of V. parahaemolyticus in sea squirt was evaluated, and the time spent by sea squirt in transportation vehicles, market displays, and home refrigerators, in addition to the temperature of each of these, were recorded. The data were fitted to the @RISK program to obtain a probability distribution. Predictive models were developed to determine the fate of V. parahaemolyticus under distribution conditions. A simulation model was prepared based on experimental data, and a dose-response model for V. parahaemolyticus was prepared using data from literature to estimate infection risk. V. parahaemolyticus contamination was detected in 6 of 35 (17.1%) sea squirt samples. The daily consumption quantity of sea squirt was 62.14 g per person, and the consumption frequency was 0.28%. The average probability of V. parahaemolyticus foodborne illness following sea squirt consumption per person per day was 4.03 × 10-9. The objective of this study was to evaluate the risk of foodborne illness caused by Vibrio parahaemolyticus following sea squirt consumption in South Korea.

장염비브리오의 biofilm 형성에 영향을 미치는 인자 (Factors that Influence Biofilm Formation in Vibrio parahaemolyticus)

  • Biofilm is a structured community of microorganism encapsulated within a self-developed polymeric matrix and adherent to a living or a solid surface. In this study, we investigated the effects of various substrates on the formation of biofilm in Vibrio parahaemolyticus. We found that biofilm formation profoundly increased in a substrate, that consisted of calcium chloride, calcium nitrate, and calcium sulfate in 1% peptone water. On the other hand, a dramatic reduction in biofilm formation was observed in a substrate, that consisted of glucose and ferric chloride in LB broth. These results suggest that V. parahaemolyticus prefer to form a biofilm on the surface of a crustacean or a clam, where calcium ion is rich, and also where seawater temperature is relatively lower. In contrast, high levels of glucose in a crustacean or a clam body resulting from increased seawater temperature, can make V. parahaemolyticus detach from it and lead to free floating.

해수에서 분리한 장염비브리오의 항생제 내성 및 암피실린 내성 유전자의 동정 (Antibiotic-Resistance Profiles and the Identification of the Ampicillin-Resistance Gene of Vibrio parahaemolyticus Isolated from Seawater)

  • The antibiotics-resistance profiles of 28 strains of Vibrio parahaemolyticus isolated from seawater were investigated. All of the strains studied were resistant to ampicillin (100%), but susceptible to 12 other antibiotics. The minimum inhibitory concentration (MIC) of V. parahaemolyticus to ampicillin was as high as $1,024-2,048\;{\mu}g{\cdot}mL^{-1}$. The phenotype of strain 8 changed from ampicillin-resistant to susceptible with an in-frame deletion mutant of VPA0477, a putative ${\beta}$-lactamase gene, and the MIC for ampicillin of the mutant strain was $1{\mu}g{\cdot}mL^{-1}$. In conclusion, our findings suggest that the VPA0477 gene acts as a ${\beta}$-lactamase in ampicillin-resistant V. parahaemolyticus strains.

오배자 발효추출물의 항세균활성 (Antibacterial activity of Bio-fermented Galla Rhios Extract)

  • Objectives : This experimental study was performed in order to investigate the antibacterial effect of bio-fermented Galla Rhois extract. Methods : The Galla Rhois extract was fermented by Streptococcus thermophilus, Saccharomyces cerevisiae and Lactobacillus delbrueckii, and their products was tested for antibacterial activity against six pathogenic microorganisms namely, Bacillus cereus, Bacillus subtilis, Staphylococcus aureus, Vibrio parahaemolyticus, Escherichia coli and Salmonella typhimurium by paper disc diffusion method. Results : The Galla Rhois fermented extract by Lactobacillus delbrueckii and Saccharomyces cerevisiae showed more effective antibacterial activity than not fermented extract against Bacillus subtilis and Vibrio parahaemolyticus. Antibacterial activity of fermented extract using especially Lactobacillus delbrueckii and Saccharomyces cerevisiae was proved that it was good with even 2 percents concentration. Antibacterial activity of Galla Rhois extract within pH 3 to pH 7 had been safe regardless of pH but low over pH 9. The growth of Bacillus cereus, Staphylococcus aureus, and Vibrio parahaemolyticus had a tendency to decrease depend on the increasing concentration of the extract. EtOEt, EtOAc and n-BuOH fractions of the Galla Rhois extract had a high level of antibacterial activity against Bacillus cereus, Bacillus subtilis, Staphylococcus aureus and Vibrio parahaemolyticus, respectively. Surprisingly, EtOAc fractions of the Galla Rhois extract showed higher antibacterial activity against Vibrio parahaemolyticus alone. And antibacterial activity against six pathogenic microorganisms had a tendency to increase depend on the increasing concentration of the fractions of the Galla Rhois extract. Conclusions : Bio-fermented Galla Rhois extract, efficiently inhibited the growth of Bacillus cereus and Vibrio parahaemolyticus.

Vibrio 속 16S rRNA 유전자 염기서열의 이질성 분석 (Heterogeneity Analysis of the 16S rRNA Gene Sequences of the Genus Vibrio)

  • 세균 16S rRNA 유전자 염기서열은 분자계통분류, 진화역사 규명, 미생물 검출 등 다양한 목적으로 이용되어 왔다. 세균 제놈(genome)은 multiple rRNA 오페론을 갖고 있으며, 이들 유전자 염기서열은 일부 변이가 있는 것으로 알려져 있다. 본 연구에서는 Vibrio 속의 16S rRNA 유전자 염기서열을 이용하여 세포 내 16S rRNA의 이질성을 규명하였다. 분석은 GenBank 자료 중에서 제놈 염기서열 annotation이 완료된 V. cholerae, V. harveyi, V. parahaemolyticus, V. splendidus, V. vulnificus를 이용하여 실시하였다. Vibrio 속은 1번 염색체에 7~10개의 16S rRNA 유전자 copy를 갖고 있으며, 이들의 세포 내 유전자 변이는 0.9% 이하 상이성(99.1%이상 DNA 상동성)을 보였다. 2번 염색체에서는 16S rRNA 유전자가 1개 이하로 존재하였다. 유전체내 16S rRNA 유전형은 최소 5개(V. vulnificus #CMCP6)에서 최대 8개(V. parahaemolyticus #RIMD 2210633, V. harveyi #ATCC BAA-1116)로 조사되었다. 본 결과는 Vibrio 속의 16S rRNA 유전자 염기서열이 높은 이질성을 갖는 것을 제시해 준다.

Pathogenic Vibrio spp. Isolated from the Gwangan Beach of Busan in 2003

  • A total of 52 pathogenic Vibrio strains was isolated from the Gwangan Beach during summer in 2003. The isolated vibrios were composed of 6 different species: V. parahaemolyticus, V. cholerae non O1, V. fluvialis, V. vulnificus, V. alginolyticus, and V. mimicus. V. parahaemolyticus was most predominant as $46\%$ (24/52), V. cholerae non O1 was the second with $23\%$ (12/52), and V. fluvialis was the third with $17\%$ (9/52). Among the isolated strains, 22 strains showed hemolytic, proteolytic or ureolytic activity. Eight strains showed both hemolysin and protease activities, and either 6 strains showed only hemolysin activities and 7 strains only protease activities. Only one strain of V. parahaemolyticus isolates showed urease activity. The urease-positive V. parahaemolyticus strain (V. parahaemolyticus S25) showed the same biochemical characteristics as the reference strain, V. parahaemolyticus KCTC 2471 (urease­negative) except for urease production. To compare the degree of virulence of Vibrio strains having different pathogenic factors, hemolysin, protease, or urease-positive strains were injected into groups of 10 each of ICR mice (7- to l0-week-old male). The lethal rate of urease-positive V. parahaemolyticus S25 was significantly high, being $70\%$. Protease-positive strains showed $40-60\%$ of lethal rate. Hemolysin-positive strains showed no mortality, similar to non-pathogenic V. parahaemolyticus KCTC 2471 and V. parahaemolyticus FM12.

생선회에 오염원 장염비브리오균에 미치는 저장온도의 영향 (The Storage Temperature Effect on Vibrio parahaemolyticus in Fish Homogenates)

  • 생선회에 사용되는 선어육중에 오염된 장염비브리오균이 저장온도와 시간에 따른 균의 증감정도를 파악하여 식중독 예방대책에 필요한 기초자료를 얻고자 가다랭이와 방어의 마쇄육에 Kanagawa현상 양성인 장염비브리오균을 접종하여 $30^{\circ}C,\;18^{\circ}C,\;4^{\circ}C$$-20^{\circ}C$에서 24시간 저장하면서 매 2시간마다 균수의 변화를 실험한 결과는 다음과 같다. 1. 가다랭이나 방어의 마쇄육에 장염비브리오균을 접종하여 상온에 방치하여 두었을때 처음 2시간까지는 균수감소 경향을 나타내었으며 그 이후부터 급격한 균의 증식현상이 나타났다. 2. 생선 마쇄육에 장염비브리오균을 $10^3/g$ 정도 오염시켜 $18^{\circ}C$에 10시간정도 보관하여 두었을때 $10^4/g$ 정도로 증가하는데 반하여 $30^{\circ}C$에 같은 시간 보관하였을 경우에는 $10^6/g$인으로 증가하였다. 3. 장염비브리오균을 생선마쇄육에 오염시켜 $-20^{\circ}C$에서 24시간 저장했을 때에 약 $20\%$의 생잔율을 나타내었다. 4. 생선회를 $4^{\circ}C$ 냉장고에 보관했을에 4시간후에는 약 $10\%$ 감소하였고, 24시간 후에는 약 $50\%$ 감소하였다. 5. 용선회를 $4^{\circ}C$에 냉장보관할때 신선한 방어육에 오염된 장염비브리균이 동결해동한 가다랭이육에 오염된 경우 보다 균 감소율이 $10{\sim}15\%$정도 높았다.

Vibrio parahaemolyticus에 대한 황금추출물의 항균활성 (Antimicrobial Activities of Scutellariae Radix Extract against Vibrio parahaemolyticus)

  • 식품 가물로부터 분리.동정 된 V. parahaemolyticus에 대하여 황금추출물의 항균력을 검토하였다. 바지락 72건 가검물 중에서 66건(91.7%)이 양성으로서 높은 검출율을 보였다. 슬라이드 응집법으로 혈청형을 확인한 결과 분리 된 V. parahaemolyticus 66균주 중에서 혈청형을 결정할 수 없는 UT(untypeable) K-group이 46균주로서 69.7%였으며, 확인된 혈청형 중에서는 serotype K-I group이 7균주로서 10.6%의 검출율을 나타내었고, K-IV group 5균주(7.6%), K-II, V, VII group이 각각 2균주(3.0%)이 었으며, K-VI, VIII group 1균주(1.5%)의 순으로 분리되었고, K-III와 IX group 혈청형은 검출되지 않았다. 식품에 있어서 생육관계를 조사하기 위하여 시판되는 바지락을 시료로 분리균주들을 각각 접종한 다음35$^{\circ}C$에서 120시간동안 배양하여 얻은 생육곡선에서 V. parahemolyticus균주들 모두가 유도기, 대수기, 정지기, 사멸기를 거치는 전형적인 S자 모양의 생육곡선을 보였다. 식품에 대한 황금추출물의 항균작용을 알아보기 위하여 시판되는 바지락을 시료로 하여 증기 멸균한 후, 동량의 멸균생리식염수를 넣고 황금추출물의 농도가 0, 500, 1000, 5000 ppm (mg/kg)이 되도록 균질화시킨 시료에 분리균주들을 각각 접종한 다음, 35$^{\circ}C$에서 48시간 동안 배양하여 얻은 생육저해곡선에서 V. parahaemolyticus 균주들 모두가 유도기를 거친 다음인 6시간이 경과된 후 황금추출물의 농도에 따라 시험균주의 균수 차이를 보였고, 식품중의 Vibrio parahaemolyticus에 있어서 생육이 억제되는 황금추출물 유효저해농도는 1000ppm이었다. 세포형태변화를 관찰하기 위한 투과형전자현미경(TEM) 상에서 황금추출물 처리로 인한 V. parahaemolyticus균주들의 세포막기능의 파괴로 세포내용물들이 용출된 것을 볼 수 있었다.
