Selective Sterilization of Vibro parahaemolyticus from a Bacterial Mixture by Low-Amperage Electric Current

  • The objective of this study was to investigate the possibility of using low-amperage electrical treatment (LAET) as a selective bacteriocide. Mixtures containing Escherichia coli, Staphylococcus aureus, and Vibrio parahaemolyticus were treated with different electric current intensities and for different times. The results showed that at 263 mA, treating bacteria for 100 ms eliminated all V. parahaemolyticus colonies. Although LAET reduced the populations of the three microorganisms, V. parahaemolyticus was more injured by LAET than S. aureus and E. coli when treated at the same processing conditions.

The Antibacterial Action of Garlic, Onion, Ginger and Red Pepper Juice (마늘, 양파, 생강, 고추즙의 항균작용)

  • This study was carried out to determine the inhibitory effect of juice of garlic, onion, ginger and red pepper against Staphylococcus aureus, Salmonella enteritidis, Vibrio parahaemolyticus and Enterobacter cloacae which are food born disease organisms. It was found that 1~2.5%(wt/vol.) garlic juice showed strong antimicrobial action against growth of all test bacteria at 1/20$\times$10-6 dilution. Especially 1% garlic juice completly inactivated Vibrio parahaemolyticus but approximately 71.9~88% of reduction at same concentration in other bacteria was occurred. 2.5% onion juice had 28.5% antimicrobial activity on Sal. enteritidis, but 50% reduction was occurred on St.aureus and V. parahaemolyticus at same concentration. Antibacterial activity of ginger juice was similar to that of onion juice. 2.5% ginger juice showed approximately 50% reduction on Sal. enteritidis and V.parahaemolyticus, but less antimicrobial activity was occured on St. aureus and E. cloacae. Red pepper juice showed the least antimicrobial activity on food born disease organism compared to that of other juices. 2.5% red pepper juice showed 11.3%, 18.7% and 8.1% reduction on St. aureus, Sal. enteritidis and V.parahaemolyticus, respectively.

Development of Predictive Growth Model of Vibrio parahaemolyticus Using Mathematical Quantitative Model (수학적 정량평가모델을 이용한 Vibrio parahaemolyticus의 성장 예측모델의 개발)

  • Predictive growth model of Vibrio parahaemolyticus in modified surimi-based imitation crab broth was investigated. Growth curves of V. parahaemolyticus were obtained by measuring cell concentration in culture broth under different conditions ($Initial\;cell\;level,\;1{\times}10^{2},\;1{\times}10^{3},\;and\;1{\times}10^{4}\;colony\;forming\;unit\;(CFU)/mL$; temperature, 15, 25 37, and $40^{\circ}C$; pH 6, 7, and 8) and applying them to Gompertz model. Microbial growth indicators, maximum specific growth rate (k), lag time (LT), and generation time (GT), were calculated from Gompertz model. Maximum specific growth rate (k) of V. parahaemolyticus increased with increasing temperature, reaching maximum rate at $37^{\circ}C$. LT and GT were also the shortest at $37^{\circ}C$. pH and initial cell number did not influence k, LT, and GT values significantly (p>0.05). Polynomial model, $k=a{\cdot}\exp(-0.5{\cdot}((T-T_{max}/b)^{2}+((pH-pH_{max)/c^{2}))$, and square root model, ${\sqrt{k}\;0.06(T-9.55)[1-\exp(0.07(T-49.98))]$, were developed to express combination effects of temperature and pH under each initial cell number using Gauss-Newton Algorism of Sigma plot 7.0 (SPSS Inc.). Relative coefficients between experimental k and k Predicted by polynomial model were 0.966, 0.979, and 0.965, respectively, at initial cell numbers of $1{\times}10^{2},\;1{\times}10^{3},\;and\;1{\times}10^{4}CFU/mL$, while that between experimental k and k Predicted by square root model was 0.977. Results revealed growth of V. parahaemolyticus was mainly affected by temperature, and square root model showing effect of temperature was more credible than polynomial model for prediction of V. parahaemolyticus growth.

Studies on the Isolation of Vibrio Species from Sea Water and Marine Products at Ulleung Island of Korea in 1985 (한국 울릉도 근해의 비브리오속의 분리연구)

  • Authors studies on the isolation of non-sucrose fermentation Vibrio species such as V. parahaemolyticus, V. vulnificus and V. damsela from sea water, shellfishes and various algae at Ulleung island in the east of Korea on summer in 1985. Authors carried out test for isolated strains to biochemical characteristics, halophilism, hemolysis and serological reaction. The results obtained were as follows: 1. Strains isolated from total specimens were 25 strains of V. parahaemolyticus, 2 strains of V. vulnificus and 1 strain of V. damsela, respectively. 2. Isolated frequency of Vibrio species from sea water and marine products was lower than on specimens in the southern sea of Korea from 1980 to 1985. 3. On hemolysis reaction on blood agar media using human and rabbit erythrocytes, 14 strains among 25 strains of V. parahaemolyticus isolated were positive, and V. vulnificus and V. damsela were positive, respectively. 4. The distributions of serotypes of V. parahaemolyticus isolated were from O1 group to O10 group, and from K 10 to K 69. The results of K serotypes were as follows ; serotype K 10 and K 33 were 2 strains, serotype K 59 and K 69 were 1 strain, respectively. And 2 strains of V. vulnificus isolated were not agglutinated by antiserum of V. vulnificus. Therefore, the probability is that 2 strains of V. vulnificus isolated were other serotypes distributed on sea weater and marine products of Ulleung island of Korea.

Virulence Factors and Antimicrobial Susceptibility of Vibrio parahaemolyticus Isolated from the Oyster Crassostrea gigas (양식 굴(Crassostrea gigas)에서 분리된 장염비브리오균의 독소 유전자 보유 및 항균제 감수성)

  • This study investigated the prevalence of Vibrio parahaemolyticus in the oyster Crassostrea gigas, which is commonly consumed raw. The presence of virulence factors and the antimicrobial susceptibility of isolates were also investigated. The overall prevalence rate of V. parahaemolyticus in oysters was 37.5% (36/96) and the range of concentrations was 30-11,000 MPN/100 g. PCR-based assays indicated that 9.6% (11/115) of the isolates were positive for the thermostable direct hemolysin-related hemolysin gene (trh), while none of the isolates were positive for the thermostable direct hemolysin gene (tdh). The Multiple Antibiotics Resistance (MAR) index was measured for 16 common antimicrobial agents and 46.1% (53/115) of the isolates had a MAR index > 0.2. The MAR index ranged from 0.07 to 0.73. The highest MAR index was observed with strain s150608, isolated in June 2015, which exhibited resistance to 11 antimicrobial agents. Our results demonstrate that oysters are high-risk sources of V. parahaemolyticus, although no antimicrobial agent was being used to promote growth or to treat bacterial infections in the sampled oyster-growing areas.

Distribution of Vibrio parahaemolyticus in Fishery products, Sold at Garak Wholesnle Market and Serological Characteristics of Isolated Strains (시판 수산물 중 장염비브리오균의 분포와 분리균주의 혈청학적 특성)

  • Totally, 742 fishery products were collected at Garak wholesale market in Seoul from Jan. to Dec. in 2001. Vibrio parahaemolyticus were 12.5%(93/742). The detection ratio of V. parahaemolyticus by month was reached the maximum in summer season (July to September). The highest ratio with 22.9% were abtained from molluscs, followed by shellfishes with 15.3%, and fishes and crustaceans with about 8%. The ratio of serotypable strains were 15.2%(10/66) and identified serotyps of V.parahaemolyticus were K24, K28, K34, K42, K47 and K53

  • Fifty four strains of pathogenic vibrios were isolated from the Gwangan Beach from May to October, 2002. The isolated vibrios were composed of 7 different species: Vibrio parahaemolyticus, V. cholerae non-O1, V. alginolyticus, V. vulnificus, V. hollisae, V. fluvialis, ane V. mimicus. In the detection rate, V. parahaemolyticus was most predominant as $46\%$(25/54). From the isolated strains, only 25 strains have hemolytic activity or 25 strains only proteolytic activity on agar plates. Eleven strains showed both hemolytic and proteolytic activity. No strains showed urease activity. All strains of V parahaemolyticus did not show hemolytic activity, while V. cholerae non-O1 strains showed $\beta$ hemolytic activity. Kanagawa phenomena of pathogenic vibrios did not accord with hemolytic activity of the culture supernatant at the late log phase. Some strains showed high hemolytic activity despite having proteolytic activity, but some weak hemolytic activities despite having no proteolytic activity.

Studies on Hemolysis of Vibrio Parahaemolyticus to Various Erythrocytes (각종 적혈구에 대한 장염비브리오의 용혈성에 관한 연구)

  • The authors isolated Vibrio parahaemolyticus from sea water, sea mud and various marine products in Busan shore area from 1981 to 1982, Among 100 isolated strains, 66 strains showed positive reaction in Kanagawa phenomenon. With the above 66 strains, the authors carred out test for detecting hemolysis activity of V.parahaemolyticus on human, rabbit, chicken, pig, goat, sheep and cow erythrocytes, in different media, such as modified Wagatsuma, nutrient, peptone and brain heart infusion agar plates media. The following results were obtained: 1. The media which can be used for Kanagawa phenomenon of V. parahoemolyticus were modified Wagatsuma, nutrient, peptone agar media, but not brain heart infusion agar medium. 2. The erythrocytes which showed positive Kanagawa phenomenon were those of human, rabbit, chicken and pig, but sheep, goat and cow erythrocytes showed no sensitivities.

Antimicrobial Activities of the Lignans from Abies koreana Wilson (구상나무 리그난류의 항균활성)

  • Abies koreana Wilson is a proper tree to Korea. We tested the antimicrobial activity of the lignans isolated from it. The lignans inhibited the growth of Klebsiella pneumoniae and Vibrio parahaemolyticus at the concentration of $10^3\;ppm\;and\;10^4\;ppm$. Especially, the compound V showed higher inhibition activity to Vibrio parahaemolyticus by 1.7 times compared with the control. The compound III showed activity of 36% of the control to Klebsiella pneumoniae. The compounds II and VI had the high growth inhibition activity of 48, 66% respectively to Vibrio parahaemolyticus.

Distribution and characteristic of growth of Vibrio spp. in Incheon coastal area (인천연안 해역의 Vibrio속 세균분포 및 증식특성)

  • This study was conducted to investigate the effects of environmental factor such as temperature, salinity, turbidity, pH and dissolved oxygen on the growth of Vibrio spp.. In this survey, total 56 seawater samples were obtained from 8 different sites of the Incheon coastal area during the periods from april 2008 to october 2008. Enumeration of Vibrio spp. was determined by using the most probable number(MPN) procedure. Isolation rates of V. parahaemolyticus, V. vulnificus, V. cholerae in all samples tested were 44.0%, 21.4% and 13.1%, respectively. The enumeration of Vibrio spp. was very low correlated with water temperature and pH and negatively correlated with salinity, dissolved oxygen and turbidity. We found salinity to be the parameter most highly correlated with the enumeration of Vibrio spp. The highest rate of antibiotic resistance of V.vulnificus and V.parahaemolyticus was Cefazolin(11.5%), Ampicillin(70.8%), respectively.
