Vibrio parahaemolyticus에 대한 Immunoglobulin yolk (IgY)의 생산과 특이성 (Production and Specificity of Imunoglobulin yolk (IgY) on Vibrio parahaemolyticus)

  • 본 연구는, Vibrio parahaemolyticus에 대한 면역 글로블린 Y(IgY)를 생산하기 위하여 시행되었다. Vibrio parahaemolyticus는 중요한 병원성 균이며, 위장염을 유발시키는 원인 균이다. 따라서, 난황에서 Vibrio parahaemolyticus에 대한 항체를 생산하기 위하여 항원 LPS(lipopolysaccharide)를 암탉에 주사하였다. 면역은 2주 간격으로 4차까지 실시하였으며, 추가면역으로 2주 간격으로 3차까지 실시한 후 ELISA로 그 항체 역가를 측정하였다. 난황에서의 항체생성은 1차 면역 후 1주일부터 나타나기 시작하였으며, 7주에 최고 역가에 도달하였고, 이러한 역가는 13주 이후까지 지속되는 것으로 나타났다. 한편 혈청에서의 항체 역가도 난황에서의 경향과 유사하였다. 정제시킨 IgY를 본 연구의 water-dilution방법과 상업용 IgY분리 kit로 그 정제 정도를 비교해본 격과 큰 차이를 나타내지 않았으며, Vibrio parahaemolyticus외의 다른 Vibrio 속 및 세균들과의 교차반응성 조사에서 특별한 교차반응이 없었다. 본 연구에 나타난 결과로, 아주 적은 양의 LPS 항원으로 역가가 높은 항체의 생산이 가능하였으며, 이러한 결과들로 보아 난황은 유행성 위장염을 유발하는 병원성 균의 특이 항체 개발을 위한 좋은 공급원이 될 것으로 생각된다.

제주 양식장 유입수과 방출수에서 분리한 다제내성 Vibrio균 플라스미드 프로파일링 (Plasmid profiling of multi-drug resistant Vibrio sp. isolated from influent and effluent water samples of fish farms in Jeju, South Korea)

  • 이 실험은 제주도내 양식장으로부터 유입수와 방출수의 다제내성(MDR)을 가진 Vibrio 균의 plasmid profiling을 위해 진행하였다. Plasmid profiling을 위해 사용한 다제내성을 가진 균주는 디스크 확산법을 통해 확인하였고, 유입수와 방출수으로부터 각각 150개의 Vibrio 균주를 분리하였다. 모든 다제내성 균주를 대상으로 plasmid profiling을 실시하였으며, 유입수와 비교하여 방출수에서 많은Vibrio 균이 열거되었다(유입수 39%, 방출수 61%). 방출수에서는 neomycin, sulfamethoxazole, amoxicillin 및 oxytetracycline의 내성을 가진 균주가 유의적으로 많은 것으로 확인되었고, 대조적으로 Vibrio 균주는 florfenicol, chloramphenicol, ciprofloxacin 및 nalidixic acid에 더 민감한 것으로 확인이 되었다. 99종의 다제내성 균주(유입수 39종과 방출수 60종) 중에서 총 58종(유입수 38종과 방출수 20종)이 1.7 kb에서 10 kb 이상의 플라스미드를 가지고 있는 것을 확인하였으며 플라스미드 크기마다 19가지의 다른 다제내성 패턴을 보였다. 6종의 유입수와 4종의 방출수에서 다제내성 균주는 특이적인 plasmid profile이 확인되었다. 방출수 샘플은 보다 많은 플라스미드를 가진 다제내성 Vibrio 균주와 다양한 plasmid profile들과 다제내성 패턴을 가지고 있었고 이는 양식장의 저장탱크가 항생제내성 유전자의 저장소 역할을 할 수 있다는 점을 시사한다. 양식장의 방출수에서 plasmid를 가진 다제내성 Vibrio 균주의 존재는 항생제 내성 유전자의 전파에 기여할 수 있으며 이로 인해 인간의 건강을 위협할 수 있다.

복어로부터 복어독(Tetrodotoxin) 생성능이 있는 해양 미생물의 분리 및 동정 (Isolation and Identification of Tetrodotoxin-producing Marine Microorganism from Pufferfish)

  • A novel marine microorganism, Vibrio sp. YE-101, was isolated from pufferfish and investigated for its ability to synthesize tetrodotoxin (TTX). Various strains isolated from the intestine of pufferfish were grown on TCBS agar plate, and then cultured on Ocean Research Institute (ORI) medium supplemented with 3% NaCl at 23$^{\circ}C$ for 3days. The cells were harvested, disrupted, fractionated by Bio-Gel P-2 column chromatography and then TTX-producing strain, Vibrio sp. YE-101, was identified using mouse bioassay. The isolated TTX from Vibrio sp. YE-101 was also analyzed and identified by HPLC and gas chromatography-mass spectrometer (GC-MS). The mass fragmentation of trimethylsilyl derivatives of C9-base of TTX from Vibrio sp. YE-101 was interpreted and the pattern of fragmentation was same with that of authentic standard. The purfied TTX was also positive to the mouse bioassay, which clearly represents that Vibrio sp. YE-101 can synthesize TTX.

꼬막에서 분리된 장염 Vibrio의 성상 (The Characteristics of Vibrio parahaemolyticus Cultures Isolated in Tegillaria)

  • Distribution of Vibrio parahaemolyticus in the Tegillaria granosa Linne in Costal area Inchun. The survey has been done chiefly from the Inchun Sea ports during 5 months from Oct. 1974 to May 1975. 1. The number of Vibrio parahaemolyticus isolated 17(13.8%) of 117 specimens from Anadara Subcrenata. 2. The number of Vibrio alginolyticus isolated 92 strains of 119 specimens from Tegillaria granosa Linne. 3. Bacteriological characterigation such as morphological, ecological, biochemical results of 17 isolated strain has been identified the category of typical Vibrio parahaemolyticus differentiation. 4. Most of Kanagawa positive strains was positive in Greig test, and considerable number of Kanagawa negative strains was also positive. The result of Greig test was almost the same between cultures in brain heart infusion broth and brain heart infusion agar. 5. Kanagawa positive strains showed same lethal activity in inoculated mice into peritoreal cavity as compared with Kanagawa negative strain.

  • In this study, the ${\beta}$-lactamase (VPA0477) gene was used as a new target for the PCR-based detection of Vibrio parahaemolyticus. Primers specific for the ${\beta}$-lactamase (VPA0477) gene of V. parahaemolyticus, were designed and incorporated into a PCR-based assay. The assay was able to specifically detect all of the 191 V. parahaemolyticus strains tested, but did not result in amplification of 39 other Vibrio spp. and non-Vibrio spp. strains tested. The detection limit of the assay was 10 CFU of V. parahaemolyticus RIMD2210633 from pure culture broth. The ${\beta}$-lactamase (VPA0477) gene-based assay developed in this study was sensitive and specific, and has great potential for the accurate detection and identification of V. parahaemolyticus in seawater or seafood samples.

Vibrio vulnificus cytolysin의 흰쥐 혈소판 응집 기전 (Mechanism of Vibrio vulnificus Cytolysin on Rat Platelet Aggregation)

  • Vibrio vulnificus cytolysin has been incriminated as one of the important virulence determinants in V. vulnificus infection. In the present study, the effects of Vibrio vulnificus cytolysin on platelets were examined. Vibrio vulnificus cytolysin induced platelet aggregation and increased intracellular calcium concentration ($[Ca^{2+}]_i$) of rat platelets. These effects were abolished in $Ca^{2+}-free$ buffer (2 mM EGTA). Cytolysin also potentiated ADP-and collagen-induced platelet aggregation. Lanthanum (2 mM) inhibited cytolysin-diduced platelet aggregation. However, another $Ca^{2+}$ channel blockers, verapamil ($20{\;}{\mu}M$) or mefenamic acid ($20{\;}{\mu}M$) did not block cytolysin-induced platelet aggregation. Osmotic protectants, sucrose (50 mM) and raffinose (50 nM) suppressed platelet aggregation by 35.9% and 63.4%, respectively. V. vulnificus cytolysin increased membrane conductances of platelet membranes. These results suggest that cytolysin-induced platelet aggregation is mediated via lanthanum sensitive-calcium influx which resulted from the pore formation by V. vulnificus cytolysin.

서해안에서 분리한 비브리오균의 항생제 감수성 특성 (Antibiotic Susceptibility of Vibrio spp. Isolated from West Sea)

  • 비브리오균은 해양 및 하구 환경에서 상재하는 균으로 그람 음성 간균, 호염성, 비포자형성의 특징을 가지고 있다. 질병과 관련된 균주로는 Vibrio parahaemolyticus, Vibrio vulnificus, Vibrio cholera 등이 있다. 매년 어패류의 섭취 등을 통해 식중독 사고가 빈번하게 발생하고 있으며, 이에 본 연구에서는 서해안 6곳에서 패류를 채취하여 비브리오균의 분포도를 모니터링하고, 이들의 항생제 내성을 분석하였다. 총 120건의 패류를 TCBS 평판배지와 API 20E kit 기법으로 동정하였을 때 23.3%의 시료에서 비브리오균이 검출되었다. 이들 분리균주들의 항생제 내성을 16종의 항생제에 대해 분석한 결과 82.1%가 각각 vancomycin과 ampicillin에 대해 내성을 보였고, rifampin (71.4%)과 cephalothin (53.6%)에 대한 내성비율도 다른 항생제들에 비해 상대적으로 높은 것으로 나타났다. 분리균주들의 항생제 감수성은 chloramphenicol (92.9%), sulfamethoxazole/trimethoprim (92.9%)과 tetracycline (96.4%)에 대해 가장 높았다. 분리균주들의 약 71.4%가 vancomycin과 ampicillin을 포함하여 3개 이상의 항생제에 대해 다중내성을 보였다.

Vibrio vulnificus의 인혈청살균력에 대한 감수성과 Vibrio 감염이 마우스의 Hematocrit치에 미치는 영향 (Susceptibility of Vibrio vulnificus to Human Serum Bactericidal Activity and Effect of Vibrio Infections on Hematocrit Value in Mice)

  • Vibrio vulnificus, a halophilic Vibrio has gained worldwide attention as a cause of severe and frequently fatal wound infections and life-threatening septicemia. For this reason V. vulnificus is thought to produce extreme hemoconcentration and rapid death after infection, and because V. vulnificus is thought to be less susceptible to bactericidal activity of normal human serum, the present study was undertaken to assess the susceptibility of V. vulnificus to human serum bactericidal activity with that of V. parahemolyticus and V. cholerae and to assess the effect of Vibrio species, Salmonella typhimurium and E. coli on hematocrit values in experimentally infected mice. Serum bactericidal activity against both V. vulnificus and V. cholerae was higher than against V. parahemoltyicus. Survival of the test strains in heat-inactivated human serum was greater than that in heat-uninactiveted serum. Both V. parahemolyticus and V. cholerae produced slight hemoconcentration within 2 to 6 hr after intraperitoneal injection of $10^7$ viable bacteria into mice. In contrast, V. vulnificus, S. typhimurium and E. coli produced hemodilution rather than hemoconcentration after 4 or 6 hr after infection. With these results the author can conclude that V. vulnificus is more susceptible to serum bactericidal activity than other Vibrio species, and V. vulnificus did not produce hemoconcentration.

남해안 어류양식장에서 분리된 Vibrio parahaemolyticus와 Vibrio aiginolyticus의 항균제 감수성 (Antimicrobial Susceptibility of Vibrio parahaemolyticus and Vibrio alginolyticus from Fish Farms on the Southern Coast of Korea)

  • The antimicrobial resistance patterns to the 10 antimicrobial agents of potential pathogenic vibrios isolated from seawater and a variety of farmed fishes, including oliver flounder (Payalichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagyus major) and sea bass (Lateolabyax japonicus), were investigated from May to October, 2004. A total of 314 strains of the genus vibrios were isolated from 126 collected samples, and the number of isolated strains of Vibrio payahaemolyticus and Vibrio alginolyticus were 194 and 120, respectively. Apparently $98.5\%$ of V. parahaemolyticus and $100\%$ of V. alginolyticus isolates demonstrated antimicrobial resistance against at least one antimicrobial agent. The resistance of V. parahaernolyticus isolates to ampicillin ($97.9\%$) was highest, followed by oxolinic acid ($26.8\%$), amikacin ($19.1\%$) and tetracycline and amoxicillin/clavulanic acid ($6.7\%$). V. alginolyticus isolates were resistant to ampicillin ($100\%$), solfamethoxazoleit,imethopenem ($25\%$), amikacln ($21.7\%$), amoxicillin/clavulanic acid ($15.8\%$), ciprofloxacin ($13.3\%$), and tetracycline and doxycycline ($11.7\%$). The rate of multiple antimicrobial resistance to at least four antimicrobials was higher in the V. alginolyticus isolates ($20.8\%$) than in the V. parahaemolyticus ($6.7\%$).

Vibrio parahemolyticus에 대한 한방처방(금궤요략) 및 그 구성약재의 항균활성에 관한 연구 (Antimicrobial Activity of Korean Traditional Prescription (Kumkue-Yoryak) and Herb Simplexes Extracts to Vibrio Parahemolyticus)

  • Vibrio become prevailing if superficial temperature of ocean is raised and their activities of area are expanded and most of ocean creatures(fishes, oysters etc) are polluted with vibrio. The one who has taken these polluted fishes and oysters uncooked caused foodpoisoning and diarrhea from Vibrio. Frequently these diseases breakout is disposed in westsea shore of Korea. According to ancient and traditional Korean medical book - Kumkue Yorak - and their single, we carried out experiment check the activities of natural medicinal effects on Vibrio parahemolyticus. The prescriptions of trial materials are processed from extraction boiling water and 80% methanol and followed by freeze dried and adsorbed to every discs in dosage of 10mg. Gentamycin of 10mg were used for control. The result of compound prescription displayed special diseases in antimicrobial activities of boiling water and MeOH extraction compared with control. In single prescription, extraction of boiling water and MeOH of Schisandra sphenanthera(clear zone : 15mm) presented extraordinal antimicrobial activity. The MBC of Schisandra sphenanthera extracts was expressed 2.56mg/ml in boiling water and MeOH.
