유기산처리 갑오징어갑을 이용한 두부의 저장성 개선 (Improvement on Storage Stability of Soybean Curd using Cuttle Bone Powder Treated with Acetic Acid)

  • 수산가공 부산물인 갑오징어갑의 효율적 이용을 목적으로 고 용해성 아세트산 처리 갑오징어갑 분말을 응고제로 이용하여 보통 두부를 제조한 다음, 이의 저장성에 대하여 살펴보았다. 염화칼슘을 응고제로 하여 제조한 두부의 pH, 담금액의 탁도, 적정산도 및 생균수는 저장 6일째까지 거의 변화가 없었으나, 이후 pH 및 담금액의 탁도는 급격히 감소를, 적정산도 및 생균수는 급격히 증가하였다. 그러나 갑오징어갑 분말을 응고제로 하여 제조한 두부의 pH 담금액의 탁도, 적정산도 및 생균수는 저장 9일까지 거의 변화가 없었고, 이후 PH 및 담금액의 탁도는 미미한 감소를, 적정산도 및 생균수는 미미한 증가를 하여 염화칼슘을 응고제로 한 두부와 차이가 있었다. 한편, 백색도는 응고제의 종류 및 저장기간에 관계없이 저장 중 변화가 없었다. 조직감은 경도 및 부스러짐성의 경우 응고제의 종류에 관계없이 저장 3일째까지 감소하였고, 이후 염화칼슘으로 제조한 두부의 경우 저장 6일째까지, 갑오징어갑 분말로 제조한 두부의 경우 저장 9일째까지는 변화가 적었으나, 이후 염화칼슘으로 제조한 두부의 경우 크게 감소하였고, 갑오징어갑 분말로 제조한 두부의 경우 미미하게 감소하였다. 관능검사의 경우 응고제의 종류에 관계없이 물리화학적 분석의 결과와 유사하였다. 이상의 결과로 미루어 보아 두부의 유통기한은 갑오징어갑 분말을 응고제로 한 두부의 경우 9일, 염화칼슘을 응고제로 한 두부의 경우 6일로 판단되었다.

사군자탕(四君子湯) 재료를 첨가한 스폰지 케이크의 품질특성 (The Quality characteristics of sponge cake added with the ingredients of Sagoonja-Tang)

  • 본 연구에서는 사군자탕 구성 재료의 기능성을 연구하고자 사군자탕가루를 스폰지 케이크에 첨가하여 제조하였다. 케이크에 대한 한약재의 첨가량은 6%로 5개(S1(사군자탕가루), S2(인삼가루), S3(백복령가루), S4(백출가루), S5(감초가루))의 첨가군으로 구성하였다. 한약 재료의 첨가에 따라 완제품의 부피감소가 SC> S1>S3>S2>S4>S5 순으로 낮게 나타났다. 제품의 저장 중 미생물학적 품질 평가에서 생균수는 SC에서 가장 높게 나타났으며 SC는 저장 7일째에 $9{\times}10^7CFU/g$로 가장 높다가 저장 10일째에는 $2.5{\times}10^6CFU/g$으로 감소하였다. 한약재가 들어간 다른 시료에서는 낮게 나타나 특히 S5의 경우 같은 7일째에 $1.2{\times}10^2CFU/g$으로 적게 나타났다. 제품의 조직특성은 견고성, 점착성, 씹힘성이 SC에 비해 다 높은 값을 가졌다. 항산화성을 측정한 결과 과산화기질 생성억제효과의 크기는 스폰지 케이크에서 S5>S4>S3>S2>S1>SC의 순으로 나타나 한약 재료의 첨가군의 항산화효과가 나타났다. 관능검사에서는 전체적인 기호도는 SC에 비해 S3가 가장 유의적으로 높은 점수를 나타냈고, S3>SC,S2>S1>S5>S4순으로 전체적인 기호도를 나타내었다.

Production of Hydrolyzed Red Ginseng Residue and Its Application to Lactic Acid Bacteria Cultivation

  • Enzymatic treatment conditions for red ginseng residue (RGR) were investigated to apply RGR as a microbial medium. Polysaccharide hydrolyase and protease were screened to obtain high solid and carbohydrate yields, and a good degree of carbohydrate hydrolysis. The optimal dosage and reaction time for Viscozyme, the chosen polysaccharide hydrolyase, were found to be 1.0% (w/w) and 3 h, respectively. Of the tested proteases, Flavourzyme, whose optimal dosage was 0.5% (w/w), was selected. Co-treatment with the optimal dosages of Flavourzyme and Viscozyme increased solid yield, carbohydrate yield, and degree of carbohydrate hydrolysis by 76%, 65%, and 1,865%, respectively, over levels in non-treated RGR. The culture characteristics of Leuconostoc mesenteroides strain KACC 91459P grown in enzymatically hydrolyzed red ginseng residue (ERGR) and RGR suspensions were compared. After cultivation for 6 h, the viable cell counts of both cell suspensions rapidly increased to $1.3{\times}10^9$ colony-forming units (CFU)/g. Moreover, while the viable cell population drastically decreased to $2.4{\times}10^6\;CFU/g$ for cells grown in RGR medium, it was maintained in cells fermented in ERGR medium for 24 h.

Effect of Sedative Dose of Propofol on Neuronal Damage after Transient Forebrain Ischemia in Mongolian Gerbils

  • This study investigated whether propofol, an intravenous, non-barbiturate anesthetic, could reduce brain damage following global forebrain ischemia. Transient global ischemia was induced in gerbils by occlusion of bilateral carotid arteries for 3 min. Propofol (50 mg/kg) was administered intraperitoneally 30 min before, immediately after, and at 1 h, 2 h, 6 h after occlusion. Thereafter, propofol was administered twice daily for three days. Treated animals were processed in parallel with ischemic animals receiving 10% intralipid as a vehicle or with sham-operated controls. In histologic findings, counts of viable neurons were made in the pyramidal cell layer of the hippocampal CA1 area 4 days after ischemia. The number of viable neurons in the pyramidal cell layer of CA1 area was similar in animals treated with a vehicle or a subanesthetic dose of propofol. In terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick end-labeling (TUNEL) assay, semiquantitative analysis of dark-brown neuronal cells was made in the hippocampal CA1 area. There was no significant difference in the degree of TUNEL staining in the hippocampal CA1 area between vehicle-treated and propofol-treated animals. These results show that subanesthetic dose of propofol does not reduce delayed neuronal cell death following transient global ischemia in Mongolian gerbils.

마 첨가에 의한 유산균 생육에 미치는 효과 (Effect of Yam on the Growth of Lactic Acid Bacteria)

  • The growth of Lactobacillus casei in milk was enhanced by adding yams. Addition of 1% yam(raw or dry) promoted the cell growth and acid production in fermented milk. The milk containing 1% yams formed the complete curd by lactic acid fermentation at 37oC for 19hr while the milk without yams showed the incomplete curd formation. The crude mucilage extracted from a raw yam also enhanced the cell growth as well as the acid production. Addition of mucilage(0.08%) showed the similar effects with adding heat treated yam(1%). The milk fermented by adding various yams showed the high scores for sensory evaluation comparing with the milk fermented without yams. The fermenting ability of Lactobacillus acidophilus, Lactobacillus kefir and Leuconostoc mesenteroides was evaluated by adding 1% of a dry yam in milk. A dry yam also enhanced the cell growth of L. acidophilus resulting in the high acid production. The viable cell counts of L. casei, L. acidophilus and Leuc. mesenteroides except L. kefir were increased by adding 1% of a dry yam.

도계처리 단계별 도체와 처리수의 세균오염 및 염소처리 효과 (Efficacy of Chlorine for Reducing Bacterial Populations and Bacteriological Contamination on Carcass and Treatment Water at Different Stage of Poultry Processing)

  • This study was carried out to assess the effect of the chlorine treatment into water for processing chicken products in each stage of slaughtering, with a special viewpoint related with reducing the viable number of microorganisms by which the water and the chicken body were contaminated. The mean bacterial number on chicken samples after picking process was log5.37$\pm$0.20~5.84$\pm$0.160CFU/$\textrm{cm}^2$. When assessed by standard plate count method, it was the higher one than any other processing stage in which eviscerating, pinning, packaging, and chilling was followed in order of the mean bacterial number. The coliform bacterial numbers on carcasses after sampling from different processing stages were log2.11$\pm$0.63~2.88$\pm$0.25MPN/$\textrm{cm}^2, which show almost similar numbers in each processing stage. But, after chilling process the number was decreased slightly. The bacterial counts in the water for scalding and chilling showed log3.43 $\pm$ 0.59~5.06$\pm$0.21 and log4.30$\pm$0.21~6.62$\pm$0.33CFU/$m\ell$, respectively. In the coliform counts for the water taken out from the 2nd chilling tank, the number was log1.97$\pm$0.35~2.91$\pm$0.22MPN/$m\ell$ which showed higher than those of the 1st and the 3rd chilling tank water. The effect of chlorination in reducing the bacterial numbers was accepted at the residual chlorine concentration of 1$m\ell$/$\ell$by showing the reduction from $10^8$ to $10^4$CFU level and the numbers were decreased less than 10CFU at the concentration of 5mg/$\ell$, when assessed by viable cell counts. In conclusion, these results suggested that chlorination In chilling water with final concentration of 5mg/$\ell$was strongly recommended to reduce the bacterial numbers on final chicken products.

Production of Carrot Pomace Fortified with Mucilage, Fibrinolytic Enzyme and Probiotics by Solid-state Fermentation Using the Mixed Culture of Bacillus subtilis and Leuconostoc mesenteroides

  • Bioactive compounds were produced from carrot pomace by solid-state fermentation using Bacillus subtilis HA and Leuconostoc mesenteroides. The carrot pomace (CP) fermented by B. subtilis HA with 3% monosodium glutamate (MSG) showed higher production of various bioactive compounds, with 1.64 Pa·sn of consistency, 2.31% of mucilage content, 16.95 unit/g of fibrinolytic enzyme activity, 35.3 unit/g of proteolytic activity and 37.5 mg% of tyrosine content. The mucilage production was greatly dependent upon the concentration of MSG added. Most MSG added in CP was converted into mucilage (2.3%) including 0.83% poly-$gamma$-glutamic acid (PGA) with 1,505 kDa of molecular weight. The CP fermented secondly by Leuc. mesenteroides showed acidic pH and lower consistency. However, the fibrinolytic and proteolytic activities were increased. The secondly fermented CP showed the viable cell counts with $2.5{\time}108$ CFU/g of B. subtilis HA and $3.7{\time}109$ CFU/g of Leuc. mesenteroides, respectively. The freeze-dried fermented CP showed 2.88 Pa·sn of consistency, 24% of mucilage content and 104.9 unit/g of fibrinolytic enzyme activity, respectively. Also, the powder of fermented CP indicated viable cell counts of $8.0{\time}107$ CFU/g of B. subtilis and $4.0{\time}108$ CFU/g of Leuc. mesenteroides. Therefore, the fermented CP that was fortified with dietary fibers, fibrinolytic enzyme and probiotics could be utilized as valuable ingredients of functional foods in food or cosmetic industries.

Physicochemical Properties of Roasted Soybean Flour Bioconverted by Solid-State Fermentation Using Bacillus subtilis and Lactobacillus plantarum

  • To produce novel cheese-like fermented soybean, the solid-state fermentation of roasted soybean flour (RSF) was performed using 1.0% inoculum Bacillus subtilis HA and Lactobacillus plantarum, with the initial 60% substrate moisture for 10 hr at $42^{\circ}$, resulting in pH 6.5, 0.82% acidity, 3.5% mucilage, 14.3 unit/g protease activity, 7.6 unit/g fibrinolytic activity, 216 mg% tyrosine content and $1.7{\times}10^{10}$ CFU/g of viable cell counts. After the second lactic acid fermentation with 10~30% skim milk powder, the fermented RSF resulted in an increase in acidity with 1.64~1.99%, tyrosine content with 246~308 mg% and protease activity in the range of 5.2~17.5 unit/g and 0.966 water activity. Viable cell counts as probiotics indicated $1.6{\times}10^8$ CFU/g of B. subtilis and $7.3{\times}10^{10}$ CFU/g of L. plantarum. The firmness of the first fermented RSF with 2,491 $g{\cdot}{\o}mm^{-1}$ greatly decreased to 1,533 $g{\cdot}{\o}mm^{-1}$ in the second fermented RSF, although firmness was slightly increased by adding a higher content of skim milk. The consistency of the second fermented RSF also decreased greatly from 55,640 to 3,264~ 3,998 in the presence of 10~30% skim milk. The effective hydrolysis of soy protein and skim milk protein in the fermented RSF was confirmed. Thus, the second fermented RSF with a sour taste and flavor showed similar textural properties to commercial soft cheese.

Quality and storage characteristics of yogurt containing Lacobacillus sakei ALI033 and cinnamon ethanol extract

  • Background: This study was conducted to examine the quality and storage characteristics of yogurt containing antifungal-active lactic acid bacteria (ALH, Lacobacillus sakei ALI033) isolated from kimchi and cinnamon ethanol extract. The starter was used for culture inoculation (1.0 % commercial starter culture YF-L812 and ALH). Results: The antifungal activity of cinnamon extracts was observed in treatments with either cinnamon ethanol extracts or cinnamon methanol extracts. Changes in fermented milk made with ALH and cinnamon extract during fermentation at $40^{\circ}C$ were as follows. The pH was 4.6 after only 6 h of fermentation. Titratable acidity values were maintained at 0.8 % in all treatment groups. Viable cell counts were maintained at $4{\times}10^9CFU/mL$ in all groups except for 1.00 % cinnamon treatment. Sensory evaluations of fermented milk sample made with ALH and 0.05 % cinnamon ethanol extract were the highest. Changes in fermented milk made with ALH and cinnamon ethanol extract during storage at $4^{\circ}C$ for 28 days were as follows. In fermented milk containing ALH and cinnamon ethanol extracts, the changes in pH and titratable acidity were moderate and smaller compared with those of the control. Viable cell counts were maintained within a proper range of $10^8CFU/mL$. Conclusions: The results of this study suggest that the overgrowth of fermentation strains or post acidification during storage can be effectively delayed, thereby maintaining the storage quality of yogurt products in a stable way, using cinnamon ethanol extract, which exhibits excellent antifungal and antibacterial activity, in combination with lactic acid bacteria isolated from kimchi.

Microbial Contamination by Bacillus cereus, Clostridium perfringens, and Enterobacter sakazakii in Sunsik

  • The powdered cereal sunsik is a partially thermal-processed product that required safety evaluations for food-borne pathogens. Thirty-six sunsik products from Korean markets were collected and analyzed for contamination by total viable cell counts, coliforms, Escherichia coli, and the spore-forming Clostridium perfringens and Bacillus cereus. Enterobacter sakazakii, as a newly emerging pathogen, was also analyzed. Approximately 28% of sunsik were contaminated at 5 log CFU/g for total viable counts. Coliforms and E. coli were detected in 33 and 4% of the samples, respectively. The spore-forming B. cereus was found in 42% of the samples at a maximal level of 3 log CFU/g on average. About 6% the samples were contaminated with Cl. perfringens at an average level of 15 CFU/g. Forty-five % of sunsik contained E. sakazakii, at levels from 0.007 to over 1.1 cell/g by MPN method. In addition, one sunsik product for infants and children showed over 3 log CFU/g for both B. cereus and E. sakazaki. Therefore, concern should be placed on controlling for microbial hazards such as B. cereus and E. sakazakii in sunsik, particularly for the products fed to infants under 6 months of age.
