A Proposal for Promotion of Research Activities by Analysis of KOSEF's Basic Research Supports in Animal Resources Science Field (한국과학재단의 동물자원과학 분야의 기초연구지원 추이분석을 통한 연구활동지원 활성화 제언)

  • This research was conducted to make suggestions for the promotion of research activities in the field of animal resources sciences, and we evaluated the research funding trend and the present status of research funding offered by KOSEF in this field. Comparative portion of the number of research projects and grants in this field to other fields has a tendency to be decreased year by year except recent few years. Researchers in this field have received more research funding from the group-based program than from the individual-based program. Also, they have received less money(per project) than did researchers in the fields of general agricultural sciences and other science and technology. Researchers in this field ranges from 43 to 51 years of age and showed 48 years of average age. It was found that researchers who has been funded in the field of animal resources sciences have showed tendency of publication of more articles to SCI journals in recent 5 years. The strong points of the animal resources of agricultural sciences field in South Korea include: lots of researchers, the establishment of research infra-structure, the excellence in research competitiveness and technology level. However, its weaknesses are: a lack of leadership in relevant societies and institutes, a predicted shortfall of researchers in the next generation and insufficient research productivity. The opportunities include: increasing the importance of the biotechnology industry, activating international cooperation researches and exploring the multitude of possible research areas to be studied. However, some concerns still exist, such as threats from developed countries for the government to open the agricultural market, the reduction of the number of full-time farms and intensification of needs for economic and social effects. The diverse actions and systems based upon the strongpoint, weakness, opportunity and threats above-mentioned are required to encourage research activities in the field of animal resources of agricultural sciences in Korea In addition, researchers in this fields would make an effort to keep pace with international society as well as domestic demands.

Effects of Addition of Mugwort Powder on the Quality Characteristics of Emulsion-type Sausage (쑥 분말 첨가가 유화형 소시지의 품질특성에 미치는 영향)

  • This study was carried out to investigate the effects of addition of mugwort powder 0.7%, 10/0, 2%) on the quality characteristics of emulsion-type sausages. The pH, color, TBARS, textural properties, minerals content and sensory evaluation were evaluated. The pH values of sausage containing mugwort powder were significantly lower as compared to control during 20 days of storage, but there were higher than those of control at 40 days of storage. The $L^*$ and $a^*$ values of sausage containing mugwort powder were significantly lower as compared to control, but the $b^*$ values were significantly higher in the sausage containing mugwort powder. The TBARS values of sausage containing mugwort powder were significantly lower than those of control at 20 and 45 days of storage. The hardness values of sausage containing mugwort powder were significantly lower than those of control. The Na content of sausage containing mugwort powder were significantly lower as compared to control, but Mg, Ca, Mn and Fe contents were significantly higher in the sausage containing 2%l mugwort powder. Sensory panels evaluated that sausage containing mugwort powder had the higher preference scores in mugwort flavor.

In vitro rumen fermentation kinetics, metabolite production, methane and substrate degradability of polyphenol rich plant leaves and their component complete feed blocks

  • Background: This experiment aimed at assessing polyphenol-rich plant biomass to use in complete feed making for the feeding of ruminants. Methods: An in vitro ruminal evaluation of complete blocks (CFB) with (Acacia nilotica, Ziziphus nummularia leaves) and without (Vigna sinensis hay) polyphenol rich plant leaves was conducted by applying Menke's in vitro gas production (IVGP) technique. A total of six substrates, viz. three forages and three CFBs were subjected to in vitro ruminal fermentation in glass syringes to assess gas and methane production, substrate degradability, and rumen fermentation metabolites. Results: Total polyphenol content (g/Kg) was 163 in A. nilotica compared to 52.5 in Z. nummularia with a contrasting difference in tannin fractions, higher hydrolysable tannins (HT) in the former (140.1 vs 2.8) and higher condensed (CT) tannins in the later (28.3 vs 7.9). The potential gas production was lower with a higher lag phase (L) in CT containing Z. nummularia and the component feed block. A. nilotica alone and as a constituent of CFB produced higher total gas but with lower methane while the partitioning factor (PF) was higher in Z. nummularia and its CFB. Substrate digestibility (both DM and OM) was lower (P < 0.001) in Z. nummularia compared to other forages and CFBs. The fermentation metabolites showed a different pattern for forages and their CFBs. The forages showed higher TCA precipitable N and lower acetate: propionate ratio in Z. nummularia while the related trend was found in CFB with V. sinensis. Total volatile fatty acid concentration was higher (P < 0.001) in A. nilotica leaves than V. sinensis hay and Z. nummularia leaves. It has implication on widening the forage resources and providing opportunity to use forage biomass rich in polyphenolic constituents in judicious proportion for reducing methane and enhancing green livestock production. Conclusion: Above all, higher substrate degradability, propionate production, lower methanogenesis in CFB with A. nilotica leaves may be considered useful. Nevertheless, CFB with Z. nummularia also proved its usefulness with higher TCA precipitable N and PF. It has implication on widening the forage resources and providing opportunity to use polyphenol-rich forage biomass for reducing methane and enhancing green livestock production.

Fuzzy reasoning for assessing bulk tank milk quality (Bulk tank milk의 품질평가를 위한 퍼지기반 추론)

  • Many dairy producers periodically receive information about their bulk tank milk with reference to bulk tank somatic cell counts, standard plate counts, and preliminary incubation counts. This information, when collected over a period of time, in combination with bulk tank mastitis culture reports can become a significant knowledge base. Several guidelines have been proposed to interpret farm bulk tank milk bacterial counts. However many of the suggested interpretive criteria lack validation, and provide little insight to the interrelationship between different groups of bacteria found in bulk tank milk. Also the linguistic terms describing bulk tank milk quality or herd management status are rather vague or fuzzy such as excellent, good or unsatisfactory. The objective of this paper was to develop a set of fuzzy descriptors to evaluate bulk tank milk quality and herd's milking practice based on bulk tank milk microbiology test results. Thus, fuzzy logic based reasoning methodologies were developed based on fuzzy inference engine. Input parameters were bulk tank somatic cell counts, standard plate counts, preliminary incubation counts, laboratory pasteurization counts, non agalactiae-Streptococci and Streptococci like organisms, and Staphylococcus aureus. Based on the input data, bulk tank milk quality was classified as excellent, good, milk cooling problem, cleaning problem, environmental mastitis, or mixed with mastitis and cleaning problems. The results from fuzzy reasoning would provide a reference regarding a good management practice for milk producers, dairy health consultants, and veterinarians.

Intratumoral distribution of 64Cu-ATSM and 18F-FDG in VX2tumor xenografted rabbit

  • $^{64}Cu$-labeled diacetyl-bis($N^4$-methylthiosemicarbazone) is a promising agent for internal radiation therapy and imaging of hypoxic tissues. In the study, we confirmed hypoxia regions in VX2 tumor implanted rabbits with injection $^{64}Cu$-ATSM and $^{18}F$-FDG using positron emission tomography (PET)/computed tomography (CT). PET images with $^{18}F$-FDG and $^{64}Cu$-ATSM were obtained for 40 min by dynamic scan and additional delayed PET images of $^{64}Cu$-ATSM the acquired up to 48 hours. Correlation between intratumoral $O_2$ level and $^{64}Cu$-ATSM PET image was analyzed. $^{64}Cu$-ATSM and $^{18}F$-FDG were intravenously co-injected and the tumor was dissected and cut into slices for a dual-tracer autoradiographic analysis. In the PET imaging, $^{64}Cu$-ATSM in VX2 tumors displayed a specific uptake in hypoxic region for48 h. The uptake pattern of $^{64}Cu$-ATSM in VX2 tumor at 24 and 48 h did not match to the $^{18}F$-FDG. Through ROI analysis, in the early phase (dynamic scan), $^{18}F$-FDG has positive correlation with $^{64}Cu$-ATSM but late phase (24 and 48 h) of the $^{64}Cu$-ATSM showed negative correlation with $^{18}F$-FDG. High uptake of $^{64}Cu$-ATSM in hypoxic region was responded with significant decrease of oxygen pressure, which confirmed by $^{64}Cu$-ATSM PET imaging and autoradiographic analysis. In conclusion, $^{64}Cu$-ATSM can utilize for specific targeting of hypoxic region in tumor, and discrimination between necrotic- and viable hypoxic tissue.

Variation of Nephrotoxicity Biomarkers by Urinary Storage Condition in Rats

  • Recently, there has been an increase in the use of several nephrotoxicity biomarkers in preclinical experiments. In addition, it has been indicated that the result may have been influenced by secondary factors, such as sample storage condition or storage period. In this study, we have assessed the variation in urinary nephrotoxicity biomarkers as a result of urine storage conditions and storage period of the urine. Urine was sampled from specific pathogen-free Sprague-Dawley rats (19 weeks old), which were housed individually in hanged stainless steel wire mesh cages. Urine was stored at $20^{\circ}C$, at $4^{\circ}C$, or at $-70^{\circ}C$ after sampling. The levels of the biomarkers such as beta-2 microglobulin (B2M), cystatin-C (Cys-C), N-acetyl-${\beta}$-D-glucosaminidase (NAG), micro albumin (MA), micro protein (MP) were measured at 6, 24, 48 and 144 hr after sampling. The B2M level was significantly decreased at 6, 24, 48, and 144 hr compared to 0 hr at $-70^{\circ}C$ (p < 0.05, p < 0.01, p < 0.05, and p < 0.05, respectively) and 24 and 144 hr at $20^{\circ}C$ (p < 0.01, p < 0.01, respectively). The Cys-C level was significantly decreased at 144 hr compared to 0 hr at $4^{\circ}C$ (p < 0.01), at $20^{\circ}C$ (p < 0.05) and at $70^{\circ}C$ (p < 0.01). MP and MA levels were not different for 144 hr in all storage conditions. Taken together, B2M and Cys-C levels were modulated by storage temperature and period. For the enhancement of test accuracy, it is suggested that strict protocols be established for samples to minimize the effects of the storage conditions on the detected levels of biomarkers.

Arsenite Acutely Decreases Nitric Oxide Production via the ROS-Protein Phosphatase 1-Endothelial Nitric Oxide Synthase-Thr497 Signaling Cascade

  • Chronic (>24 h) exposure of arsenite, an environmental toxicant, has shown the decreased nitric oxide (NO) production in endothelial cells (EC) by decreasing endothelial NO synthase (eNOS) expression and/or its phosphorylation at serine 1179 ($eNOS-Ser^{1179}$ in bovine sequence), which is associated with increased risk of vascular diseases. Here, we investigated the acute (<24 h) effect of arsenite on NO production using bovine aortic EC (BAEC). Arsenite acutely increased the phosphorylation of $eNOS-Thr^{497}$, but not of $eNOS-Ser^{116}$ or $eNOS-Ser^{1179}$, which was accompanied by decreased NO production. The level of eNOS expression was unaltered under this condition. Treatment with arsenite also induced reactive oxygen species (ROS) production, and pretreatment with a ROS scavenger N-acetyl-L-cysteine (NAC) completely reversed the observed effect of arsenite on $eNOS-Thr^{497}$ phosphorylation. Although protein kinase C (PKC) and protein phosphatase 1 (PP1) were reported to be involved in $eNOS-Thr^{497}$ phosphorylation, treatment with PKC inhibitor, Ro318425, and overexpression of various PKC isoforms did not affect the arsenite-stimulated $eNOS-Thr^{497}$ phosphorylation. In contrast, treatment with PP1 inhibitor, calyculin A, mimicked the observed effect of arsenite on $eNOS-Thr^{497}$ phosphorylation. Lastly, we found decreased cellular PP1 activity in arsenite-treated cells, which was reversed by NAC. Overall, our study demonstrates firstly that arsenite acutely decreases NO production at least in part by increasing $eNOS-Thr^{497}$ phosphorylation via ROS-PP1 signaling pathway, which provide the molecular mechanism underlying arsenite-induced increase in vascular disease.

Development of an Official Method for Measurement of Fluazinam Residues for Quarantine of Imported and Exported Horticultural Products (수출입 원예작물의 검역을 위한 살균제 Fluazinam의 공정 잔류분석법 개발)

  • This experiment was conducted to establish an official determination method to measure fluazinam residue in horticultural crops for import and export using GC-ECD/MS. Fluazinam residue was extracted with acetone from fresh samples of four representative horticultural products, the vegetable crops green pepper and kimchi cabbage, and the fruit crops mandarin and apple. The acetone extract was diluted with saline water and n -hexane partitioning was used to recover fluazinam from the aqueous phase. Florisil column chromatography was additionally employed for final purification of the extract. Fluazinam was separated and quantitated by GC with ECD using a DB-17 capillary column. The horticultural crops were fortified with three different concentrations of fluazinam. Mean recoveries ranged from 82.5% to 99.9% in the four crops. The coefficients of variation were less than 10.0%. The quantitative limit of fluazinam detection was $0.004mg{\cdot}kg^{-1}$ in the four crop samples. GC/MS with selected-ion monitoring was also used to confirm the suspected residue. This analytical method was reproducible and sensitive enough to measure the residue of fluazinam in horticultural commodities for import and export.

Studies on parasitic trematodes in bivalves -Variation of infection rate of Cercaria tapidis and Cercaria harengulae- (조개류에 기생하는 흡충류에 관한 연구 - Cercaria tapidis, Cercaria harengulae 의 기생율 변동)

  • We had reported the infection rates of trematode larvae, Cercaria tapidis and C. harengulae in the gonad tissues of Tapes philippinarum and Solen strictus in the estuary of Kum river, the west coast in Korea in 1980~1982. At the same sites from January 1994 to January 1995, we investigated the variation of the infection rates of these trematode larvae in the two clams. The results are follows. The infection rate of C. tapidis parasitized in T. philippinarum increased in 2.4 times (14.0%) higher than that (5.7%) in 1981. Monthly maxium infection rate was 32.0% in October 1994. Comparing the rate (23.3%) in December 1981, the rate in 1994 showed a higher trend than that in 1981. The mean infection rate of C. harengulae in T. philippinarum showed 2.3% in 1994. Comparing that (5%) in 1980, it showed a decreased rate than that in 1980. And monthly maximum infection infection rate of this clam was 6.7% in May 1994, compared with the rate (19.0%) in March 1980. It appeared furthermore decreased rate in 1994. The mean infection rate of the year of C. harengulae in S. strictus was 6.9% in 1994, while it was 10.2% between 1981 and 1982. It showed a decreased trend in 1994. Monthly maximum infection rate was 23.4% in August 1981, while it was 13.3% in April 1994. It showed a decreased trend in 1994 also. In case of C. tapidis the more large sizes of shell length the more infection rates were higher as same in the investigation in 1980, while in C. harengulae the smaller sizes, the more infection rates showed a higher trend.

Production and Characterization of vitellogenin monoclonal antibody on the Scorpion fish Sebastiscus marmoratus (쏨뱅이, Sebastiscus marmoratus의 vitellogenin 단클론 항체생산 및 특성에 관한 연구)

  • In order to establish bio-marker systems for the screening of endocrine-disrupting chemicals contaminated in various environment, Vitellogenin(Vtg) bio-marker have been developed to detect Scorpion fish's(Sebastiscus marmoratus) Vtg. Vtg has been induced by administration of estradiol into S. marmoratus, and purified by gel filtration and ion-exchange chromatography from serum of the fish. After immunization of the purified Vtg into BALB/c mouse, hybridomas secreting anti-Vtg antibodies have been produced. The size of induced Vtg in the serum was about 440 kDa by gel filtration using Sepharose CL-6B. By SDS-PAGE analysis, the main band of Vtg, however, was at 175 kDa, and several minor bands have been detected with the main band. Eight different monoclonal antibodies have been produced from established hybridomas and the antibodies did not cross-react with sera from different species of fishes tested in this study except with that of Sebastes hubbsi. These results suggested that the monoclonal antibody of S28 and S15 can used as capture and tracer antibodies for ELISA and ICG assays. The detection systems developed in this study can be used as Bio-marker assays to check endocrine disrupting activity of various chemicals as well as to detect known endocrine disrupting chemicals contaminated in environment.
