L-ascorbic acid가 포집된 BGsome의 특성 및 안정화 효과 (Characteristics of L-Ascorbic Acid Encapsulated BGsome and its Stabilization Effect)

  • Encapsulation of L-ascorbic acid(AA) into BGsome was attempted to improve its stability. BGsome is a bio-compatible vesicular system prepared by dispersion of hydrated liquid crystalline phase formed through hydration of 1,3-butylene glycol(BG)-dissolved lecithin with an aqueous solution containing hydrophilic component. The characteristics of AA encapsulated BGsome, such as droplet size, surface charge, and solution appearance, was investigated. The concentration of AA solution had considerable effect on droplet size and surface charge of BGsome. Several tens nanometer droplet made by sonication treatment did not showed any change of size with storage time. Stability of AA was improved by encapsulation into BGsome, which was verified through DPPH test and HPLC assay.

돼지난포의 배양에 의한 난자의 성숙 (Oocytes Maturation by Follicular Culture in Porcine)

  • The aim of this study was to investigate the effect of the follicular culture from which the oocytes originate on their subsequent in vitro maturation ability. Ovarian follicles were isolated and cultured according to size(1~2mm, 2~6mm and 6~8mm) for 42~44 h. The rates of germinal vesicle breakdown(GVBD) in each groups were 87%(65/75), 82%(80/97) and 89%(47/53), but the oocytes maturation were su, pp.essed at anaphase-I stage. In spite of the adding porcine follicular fluid and/or hormones in maturation medium, maturation ability of oocytes from follicle cultured for 21~22 h were inhibited. When oocytes from follicle cultured for 4 h at various temperature were incubated for 38~40 h, the rates of oocytes maturation from follicle cultured at 2$0^{\circ}C$(51%, 26/51) and 39$^{\circ}C$(54%, 26/48) were significant higher(P<0.05) than group cultured at 4$^{\circ}C$(33%, 19/58). On the other hand, the GVBD were stared 2 h after culture of follicle of oocytes. To summairze, oocytes maturation by follicular culture were inhibited at anaphase-I stage in porcine. When the follicle cultured for 4 h, maturation were completed to metaphase-II stage. However, rates of GVBD in oocytes from follicular culture were higher than oocytes cultured in medium.

마이크로플루다이저를 사용한 MLV liposome 형성에 관한 연구 (A study on manufacturing formation of the MLV liposomes by the microfludizer)

  • Liposome은 의약과 화장품 분야에서 많은 발전을 하였다. 본 연구에서는 세포 간지질의 주요 성분인 cholesterol, palmitic acid, cholesterol ester와 lecithin을 사용하여 swelling반응에 의해 MLV liposome을 제조 하였다. 지질베이스와 마이크로플루다이저를 사용하여 MLV liposome 형성에 대한 특성들을 연구 하였다. MLV liposome은 multilamellar vesicle로서 형성되었다. MLV liposome의 gelation효과는 고온(90$\pm$5도씨)에서 polyol과 water phase에 지질을 혼합한 것에 스웰링 반응을 하였다. MLV liposome에 alpha hydroxy acid 성분들을 투입 제조하였다. MLV liposome의 최적 조건은 마이크로플루다이저에 3회 통과하였으며 vesicle의 입자크기는 150-350nm범위이고 이것을 freeze-fracture electron microscopy로 확인 하였다.

불소화지방산염 첨가에 의한 디팔미토일포스파티딜콜린/콜레스테롤/불소화계면활성제 베지클의 제조와 물성 측정 연구 (The Preparation and Physicochemical Properties of Dipalmitoylphosphatidylcholine/Cholesterol/Fluorinated Surfactant Vesicle Incorporated Fatty Acid Salt)

  • 본 연구에서는 베지클의 응용성을 확장하기 위한 기초연구로서, DPPC(dipalmitoylphosphaticylcholine)/Chol(Cholesterol)계에 불소화계면활성제($C_8F_{17}(CH_2)_2OCO-CH_2CH(SO_3Na)COO(CH_2)_2C_8F_{17}$, FS)/불소화지방산염 (($C_7F_{15}COONH_4$, FFS)을 여러 가지 비율로 변화시켜 첨가함으로써 베지클을 변형 제조하고, 그 물리화학적 성질을 비교측정하였다. 제조된 베지클계에 대한 제타-전위 측정(Zetamaster Particle Electrophoresis Analyzer) 결과, 균일한 전하의 베지클이 형성되는 것으로 확인되었고, 이는 FFS 분자가 FS와 DPPC 사이에서 보조계면활성제로 기능하여 DPPC와 FS의 각 성분이 동일 베지클내에 공존하도록 작용했기 때문으로 풀이된다. 베지클의 입자크기 결정에서는 FFS의 농도가 증가할수록 그 크기가 감소하고, 제타-전위도 음의 값으로 더욱 작아지는 것으로 나타났다. 베지클의 분산도 변화에서도 DPPC/Chol/FS/FFS계의 경우가 DPPC/Chol/FS계에 비해 안정성이 훨씬 큰 것으로 조사되었다. 베지클의 서방유출 실험에서도 막내에 포집된 MB(Methylene Blue)의 유출속도가 DPPC/Chol/FS/FFS계에서 감소된다는 결과를 얻었고, 이는 FFS의 첨가로 인해 막의 유동성이 감소되기 때문으로 풀이된다. 이 결과는 형광방출 측정의 $I_{Excimer}/I_{Monomer}$값에서도 확인되는 것으로 막의 미세점도가 FFS 첨가 베지클에서 증가되는 것으로 나타났다. 단백질로서 알부민에 대한 친화도를 비교한 결과에서는 DPPC/Chol/FS/FFS계의 친화도가 다소 낮아지는 것으로 나타났다. 이들 실험결과에 바탕하여 FFS의 첨가에 의해 DPPC/Chol/FS 베지클계는 더욱 안정하고 균일한 막의 구조를 갖는 리포솜을 형성한다는 결론을 얻었다.

울릉도 부석 기질의 암석.광물학적 특성 (Petrological and Mineralogical Characteristics of Matrix of Pumice in Ulleung Island)

  • 울릉도에서 산출되는 치밀한 회색부석, 회색부석, 갈색 및 검은색부석의 기질에 대한 광물 암석학적 특성을 연구하기 위하여 X-선 회절분석, FT-IR, 열분석, XRF, SEM 분석을 실시하였다. 그 결과, 울릉도 부석의 기질은 전반적으로 비정질이며 미량의 새니딘과 아노르도클레이스의 결정구조적 특성을 보였다. FT-IR 흡수스펙트럼에서 흡착수분을 지시하는 O-H 피크가 관찰되었으나 열분석에서 나타나지 않는 것으로 보아 수분 함량은 미미한 수준인 것으로 판단된다. 이러한 낮은 수분 함량은 기질이 넓은 비표면적을 가지며 형성 이후 상당한 시간이 지났음에도 불구하고 수화변질 정도를 매우 낮게 만든 것으로 생각된다. SEM 이미지상에서 기질의 기공은 $2\sim2000{\mu}m$ 크기를 가지며 구형, 타원형, 실타래형 및 각상의 형태학적 특성을 보인다. 또한, 기공들은 융합하고 성장하면서 변형되는 연성특성을 보여준다. 다양한 크기와 형태를 보이는 기공의 특성은 마그마의 급격한 압력감소와 빠른 냉각에 기인하며 형성과정에서 마그마가 상당한 연성을 유지하였음을 시사한다. 초기에 형성된 치밀한 회색부석은 기공의 성장이 제한되고 구형 기공의 결핍과 각상의 매우 작은 기공($15{\mu}m$ 이하)을 포함하는 것으로 보아 수화프리니언 분출에 근접하였음을 시사한다. 기질표변에서 관찰되는 비정질 알루미나 규산염 덩어리인 극미립질 입자는 알칼리계열의 포놀라이트질 마그마가 급격히 상승하는 과정에서 미립자화된 마그마가 기질표면에 부착된 것으로 판단되며 부분적으로 결정화된 알칼리 장석입자와 공존할 가능성이 높은 것으로 추정된다.

The Ruddy Turnstone, Arenaria interpres interpres, a New Definitive Host for Gynaecotyla squatarolae (Digenea: Microphallidae)

  • The ruddy turnstone, Arenaria interpres interpres, a migratory Korean bird, was proved to be a natural definitive host for Gynaecotyla squatarolae (Digenea: Microphallidae). The ruddy turnstone was found dead at the seashore of Okgu-eup, Gunsan-si, Jeollabuk-do. The intestinal tract was examined, and 98 unknown flukes were recovered. The worms were $600{\times}284{\mu}m$ in size, and had 2 ventral suckers. The seminal vesicle was large, the genital atrium was prominent, and the average egg size was $20{\times}12.5{\mu}m$. Based on these results, the worms were identified as G. squatarolae. This is the first report on the ruddy turnstone as a natural definitive host of G. squatarolae in the Republic of Korea.

Porosome: the Universal Molecular Machinery for Cell Secretion

  • Porosomes are supramolecular, lipoprotein structures at the cell plasma membrane, where membrane-bound secretory vesicles transiently dock and fuse to release inravesicular contents to the outside during cell secretion. The mouth of the porosome opening to the outside, range in size from 150 nm in diameter in acinar cells of the exocrine pancreas, to 12 nm in neurons, which dilates during cell secretion, returning to its resting size following completion of the process. In the past decade, the composition of the porosome, its structure and dynamics at nm resolution and in real time, and its functional reconstitution into artificial lipid membrane, have all been elucidated. In this mini review, the discovery of the porosome, its structure, function, isolation, chemistry, and reconstitution into lipid membrane, the molecular mechanism of secretory vesicle swelling and fusion at the base of porosomes, and how this new information provides a paradigm shift in our understanding of cell secretion, is discussed.

In Vivo Evaluation of Multi Lamellar Vesicle Liposome’s Percutaneous Absorption and Stability

  • We had prepared MLV liposome with Hibiscus Esculentus Ext.(HEE) which have fluorescent light in order to evaluate its percutaneous absorption about hairless rat skin. Then we investigated particle size of MLV using confocal laser scanning microscope(CLSM) and transmission electron microscope(TEM), respectively. Stability of MLV liposome and penetration of MLV liposome to hairless rat skin was measured by CLSM. As a result of experiments, MLV was globular shape and the rage of particle size was 0.3-0.5$\mu\textrm{m}$ mostly. Cream-type MLV had high stability comparatively. When we treated with MLV to rat skin, skin penetration was enhanced, especially, the optimum concentration of MLV on penetration to rat skin was 10%. Optimum penetration time was 6hr-12hr. And MLV-type HEE was more effective on percutaneous absorption than HEE-cream or liposome-type HEE.

The Formation of Magnetite Nanoparticle in Ordered System of the Soybean Lecithin

  • A method of preparation of magnetite nanoparticles in ordered systems, as in vesicle and microemulsion, consisting of soybean lecithin and water has been introduced. The size of magnetite grain was controlled by the content of soybean lecithin and size of liposomes in the systems. It was found by experiment that magnetite nanoparticles were formed inside the double layer vesicles. The magnetite nanoparticles were separated by magnetic separation and centrifugation and the dispersion of the magnetite nanoparticles prepared at 10% (w/w) soybean lecithin was particularly stable. The formation of pure magnetite nanoparticles was confirmed by analyses of XRD and electron diffraction pattern.

한우에서 생식기질환의 치료 및 조기임신진단을 위한 초음파영상진단 (Ultrasonographic Diagnosis for the Treatment of Genital Disease and Early Pregnancy Diagnosis in Korean Native Cattle)

  • Ultrasonographic diagnosis of genital disease and early pregnancy diagnosis was performed in Korean native cattle. The size of ovarian follicle in preovulation, luteal stage and follicular cyst was 18.9, 9.2 and 27.6 mm, respectively, and the thickness of follicular wall was 2.3, 1.8 and 2.8 mm, respectively. The size of corpus luteums in formation stage, activity stage, regression stage, cystic corpora lutea and luteal cyst was 6.2, 11.3, 8.6, 26.7 and 25.9 mm, respectively. The thickness of luteal wall in cystic corpora lutea and luteal cyst was 8.4 and 4.9 mm, respectively. The size of embryo or fetus on day 25, 27, 30, 35, 40, 45 and 50 was 0.8, 0.9, 1.3, 1.5, 2.2, 2.8 and 3.8 cm, respectively. The size of amniotic vesicle on day 25, 27 and 30 was 1.2, 2.1 and 3,0 cm, respectively. The diameter of pregnant uterus on day 25 and 27 was 7.0 and 7.8 cm, respectively. It was concluded that the ultrasonographci values determined in this study can be used as references for the treatment of genital disease and early pregnancy diagnosis in Korean native cattle.
