The present study examined various environmental and cultural media conditions for in vitro "rescue" of cross-fertilized ovules formed through sexual crosses between Nicotiana rustica and N. tabacum cv. BY4. The response ovules to two cultural procedures was compared; ovules were cultured either separately or left attached to the placenta. Total yield of seedlings and percent of normal seedlings were increased by culturing individual ovules separately, rather than on excised placenta. Total yield of seedlings and number of normal seedlings were produced following in vitro culture of individual fertilized ovules of N. rustica X M tabacum cv. BY4 at four days post-pollination on NN medium containing 2% sucrose. In the in vitro culture of fertilized ovules, high sucrose concentration increased the frequency of seedlings of abnormal appearance. Therefore, sucrose should be supplied to developing ovules at gradually decreased concentrations. Culture of fertilized ovules from three to eight days after pollination gave increased number of seedlings, but with delayed cultral time the number of morphologically normal seedling were decreased. Hybrids were uniform in appearance and showed vegetative heterosis but flower characteristics were generally intermediate between those of the parents. All hvbrids evaluated were self-sterile.f-sterile.

Streptomyces coelicolor produces three kinds of catalases to cope with oxidative stress and to allow normal differentiation. Catalase A is the major vegetative catalase which functions in removing hydrogen peroxide generated during the process of aerobic metabolism. To understand the regulatory mechanism of response against oxidative stress, hydrogen peroxide-resistant mutant (HR4O) was isolated from S. coelicolor J1501 following UV mutagenesis. The mutant overproduced catalase A more than 50-fo1d compared with the wild type. The mutation locus catRI was mapped closed to the mthB2 locus by genetic crossings. An ordered cosmid library of S. coelicolor encompassing the mthB2 locus was used to isolate the regulator gene (catR) which represses catalase overproduction when introduced into HR4O. A candidate catR gene was found to encode a Fur-like protein of 138 amino acids (15319 Da).
Foliar sprays of three plant resistance inducers, including chitosan (CH), potassium sorbate (PS) ($C_6H_7kO_2$), and potassium bicarbonates (PB) ($KHCO_3$), were used for resistance inducing against Erysiphe cichoracearum DC (powdery mildew) infecting okra plants. Experiments under green house and field conditions showed that, the powdery mildew disease severity was significantly reduced with all tested treatments of CH, PS, and PB in comparison with untreated control. CH at 0.5% and 0.75% (w/v) plus PS at 1.0% and 2.0% and/or PB at 2.0% or 3.0% recorded as the most effective treatments. Moreover, the highest values of vegetative studies and yield were observed with such treatments. CH and potassium salts treatments reflected many compounds of defense singles which leading to the activation power defense system in okra plant. The highest records of reduction in powdery mildew were accompanied with increasing in total phenolic, protein content and increased the activity of polyphenol oxidase, peroxidase, chitinase, and ${\beta}$-1,3-glucanase in okra plants. Meanwhile, single treatments of CH, PS, and PB at high concentration (0.75%, 2.0%, and/or 3.0%) caused considerable effects. Therefore, application of CH and potassium salts as natural and chemical inducers by foliar methods can be used to control of powdery mildew disease at early stages of growth and led to a maximum fruit yield in okra plants.
The nucleolus is the largest, membrane-less organelle within the nucleus of eukaryotic cell that plays a critical role in rRNA transcription and assembly of ribosomes. Recently, the nucleolus has been shown to be implicated in an array of processes including the formation of signal recognition particles and response to cellular stress. Such diverse functions of nucleolus are mediated by nucleolar proteins. In this study, we characterized a gene coding a putative protein containing a nucleolar localization sequence (NoLS) in the rice blast fungus, Magnaporthe oryzae. Phylogenetic and domain analysis suggested that the protein is orthologous to Rrp8 in Saccharomyces cerevisiae. MoRRP8-GFP (translational fusion of MoRRP8 with green fluorescence protein) co-localizes with a nucleolar marker protein, MoNOP1 fused to red fluorescence protein (RFP), indicating that MoRRP8 is a nucleolar protein. Deletion of the MoRRP8 gene caused a reduction in vegetative growth and impinged largely on asexual sporulation. Although the asexual spores of DMorrp8 were morphologically indistinguishable from those of wild-type, they showed delay in germination and reduction in appressorium formation. Our pathogenicity assay revealed that the MoRRP8 is required for full virulence and growth within host plants. Taken together, these results suggest that nucleolar processes mediated by MoRRP8 is pivotal for fungal development and pathogenesis.
These researches have been conducted to obtain the basic information of the effects of paraquat on plant species and to screen the plant species showing specific responses to paraquat. Paraquat resistance related to ecotype and overwintering capacity. Perennial and biennial species showed higher resistance than annual species. In a family, most species showed higher resistance were overwintering species. Lamiaceae, Brassicaceae, and Caryophyllaceae were tolerant to paraquat, whereas Poaceae and Asteraceae were sensitive. Especially Mosla dianthera of Lamiaceae, Hemistepta lyrata and Aster pilosus of Asteraceae, and Paspalum thunbergii of Poaceae showed higher tolerance than others. The response patterns of plant species on germination stage were different to those on vegetative stage. Germination of Amarathus lividus, Arabis glabra, and Bidens frondosa was not inhibited by paraquat. But their seedling growth were highly inhibited.

Carbohydrate and soluble protein reserves and regrowth characteristics in response to cutting height were investigated over four regrowth cycles of turf-type perennial ryegrass(Lolium perenne L. cv. preludeII). When the plants were at the full-vegetative stage (twelve weeks-old), three sequential defoliations at 3, 6 and 9 cm above the root base were imposed at 2-week intervals. Shoot dry weight in all three treatments continuously decreased with progressing regrowth cycle and the decreasing rate was higher as cutting height was lowered. TNC (total non-structural carbohydrate) in stubble at the end of the fourth regrowth cycle in 3, 6 and 9 cm cutting height decreased by 98%, 82% and 27%, respectively, comparing the initial content. TNC in roots also largely decreased with similar pattern in response to cutting height, whereas the absolute amount was much less compared to stubble. Soluble protein in stubble in 3, 6 and 9 cm cutting height decreased by 98%, 82% and 57%, respectively, at the end of fourth regrowth. A significant correlations between TNC (r=0.906) or protein (r=0.879) at the fourth defoliation and dry weight of regrowing shoots at the end of fourth regrowth were observed. these results indicated that cutting height closely influences the levels of organic reserves available for new growth, and that the levels of reserves might provide a useful tool as a determinant for regrowth dynamics.
Brassinosteroids (BRs) are plant steroid hormones that play essential roles in growth and development. Mutations in BR-signaling pathways cause defective in growth and development like dwarfism, male sterility, abnormal vascular development and photomorphogenesis. Transition from vegetative to reproductive growth is a critical phase change in the development of a flowering plant. In a screen of activation-tagged Arabidopsis, we identified a mutant named abz126 that displayed longer hypocotyls when grown in the dark on MS media containing brassinazole (Brz), an inhibitor of BRs biosynthesis. We have cloned the mutant locus using adapter ligation PCR walking and identified that a single T-DNA had been integrated into the ninth exon of the GIGANTEA (GI) gene, involved in controling flowering time. This insertion resulted in loss-of-function of the GI gene and caused the following phenotypes: long petioles, tall plant height, many rosette leaves and late flowering. RT-PCR assays on abz126 mutant showed that the T-DNA insertion in GIGANTEA led to the loss of mRNA expression of the GI gene. In the hormone dose response assay, abz126 mutant showed: 1) an insensitivity to paclobutrazole (PAC), 2) an altered response with 6-benzylaminopurine (BAP) and 3) insensitive to Brassinolide (BL). Based on these results, we propose that the late flowering and tall phenotypes displayed by the abz126 mutant are caused by a loss-of-function of the GI gene associated with brassinosteroid hormone signaling.
Plants have evolved strategies to cope with drought stress by maximizing physiological capacity and adjusting developmental processes such as flowering time. The WOX13 orthologous group is the most conserved among the clade of WOX homeodomain-containing proteins and is found to function in both drought stress and flower development. In this study, we isolated and characterized OsWOX13 from rice. OsWOX13 was regulated spatially in vegetative organs but temporally in flowers and seeds. Overexpression of OsWOX13 (OsWOX13-ov) in rice under the rab21 promoter resulted in drought resistance and early flowering by 7-10 days. Screening of gene expression profiles in mature leaf and panicles of OsWOX13-ov showed a broad spectrum of effects on biological processes, such as abiotic and biotic stresses, exerting a cross-talk between responses. Protein binding microarray and electrophoretic mobility shift assay analyses supported ATTGATTG as the putative cis-element binding of OsWOX13. OsDREB1A and OsDREB1F, drought stress response transcription factors, contain ATTGATTG motif(s) in their promoters and are preferentially expressed in OsWOX13-ov. In addition, Heading date 3a and OsMADS14, regulators in the flowering pathway and development, were enhanced in OsWOX13-ov. These results suggest that OsWOX13 mediates the stress response and early flowering and, thus, may be a regulator of genes involved in drought escape.
Environmental factors play an important role in crop growth and development. In recent years, climate change has become a challenge that limits environmental factors. Light is an important environmental factor for photosynthesis in rice. In addition, temperature is one of the most important factors for rice production; thus, a 1℃ increase in temperature because of climate change can affect rice growth and development. Therefore, we investigated the effect of shading on the growth characteristics of rice under different temperature conditions from the vegetative stage to the flowering stage. Plants were grown at three different temperatures: 26℃/16℃ for 21℃, 29℃/19℃ for 24℃, and 22℃/32℃ for 27℃ in a phytotron. A 55% shade treatment was applied after 10 days of transplanting until the flowering stage. Plant height was not affected by the shading treatment. In the maximum tiller number response to shading, a lower tiller number and growth speed of tiller was found in the 27℃ condition. Among leaf characteristics, shading increased the flag leaf area, length, width, and effective leaf area; however, it decreased the leaf number on the main stem, especially at 27℃. In terms of stem characteristics, shading affected culm wall thickness in both varieties. Finally, regarding the panicle characteristics, lower panicle numbers, spikelet numbers per panicle, primary numbers, and secondary numbers per panicle were found under the shading treatment. Most of the desirable characteristics were affected by the shading treatment at 27℃. Overall, these results indicated that shading had a greater effect on rice plant growth at high temperature.
BACKGROUND: This experiment was conducted to investigate the effects of red light on inhibition of flowering and vegetative growth of perilla (Perilla Frutescens. L). METHODS AND RESULTS: To determine red light intensity for inhibiting floral induction of perilla 6h light plus daylength extension (17:00-23:00) with three different intensity of red lights 0.046, 0.114 and $0.177{\mu}mol/m^2/s$ were treated respectively, and control plants were grown under 11(06:00-17:00)/13(17:00-06:00)h light/dark environment. Red(660nm) and far-red(730nm) light were irradiated for night break treatment subsequently to investigate photoreversible flowering response of perilla 'Manchu'. The flowering was inhibited by night break with red light, but sequential far-red light induced floral induction of perilla. Perilla not flowered by red light intensity over $0.177{\mu}mol/m^2/s$. Red light of $0.2{\mu}mol/m^2/s$ was irradiated for 6 hours (20:00-02:00) with LEDs device in plastic house. Perilla not flowered and continued the vegetative growth by red light treatment and the plant length, number of leaves, fresh weight, and leaf area of perilla were increased by 3%, 7%, 21%, and 19%, respectively, compared to incandescent control. CONCLUSION: These results showed that red(660nm) light for daylength extension could be used to control flowering and to enhance production of perilla leaf.
