NUTRITIONAL EVALUATION OF MAIZE FODDER AT TWO DIFFERENT VEGETATIVE STAGES

  • Different fractions of maize plant and whole mixed fodder were analysed for their chemical composition and dry matter digestibility (DMD). Highest crude protein (CP) values were found in leaves as compared to the other portions. Younger plants contained more CP as compared to the matured ones. The crude fiber (CF) content of various fractions of the plant ranged between 19.12 to 35.60% with maximum values in the bottom portion of the stem. Matured plants contained more CF. The analysis of cell wall constituents indicated that the maximum values for neutral detergent fiber (NDF) were found in the bottom portion and in the whole mixed plant. The highest levels of acid detergent fiber (ADF) were observed in bottom fraction followed by whole mixed plant, whereas the other plant fractions did not show any differences. Variation in acid detergent lignin (ADL) values existed in different fractions of the plant and the lowest were in the top portion of the stem. Although there existed a variation in the mineral composition of different fractions of the plants, the results were non significant. Maximum DMD was found in leaves followed by the whole mixed plant, middle and bottom portion of the stem. The values of DMD were higher in younger plants as compared to the matured ones. It may be concluded that younger plants and the upper portion of the plants have a higher nutritive value as compared to the matured plants and lower portion of the plants.

Extreme pH Reduced Vegetative Growth and Biomass Accumulation in Alfalfa

  • Soil acidity or alkalinity are serious limitations for crop production. The purpose of this study was to clarify the negative effects of extreme pH stress (low and high) on alfalfa vegetative growth (VG) and biomass accumulation (BA). Two-week-old alfalfa seedlings were exposed to different pH (4.0, 4.5, 7.0, 8.0 and 8.5, respectively) levels for 72 hours. Alfalfa grown at pH 4.0 and 8.5 significantly reduced VG and BA, wherein as neutral pH (7.0) comparably exhibited better growth and biomass yield. These results indicate that extreme acidic or alkaline level are critical limiting factors for growth and biomass yield in alfalfa.

한국잔디(Zoysia spp.)의 종자 및 영양체를 이용한 carpet 잔디 생산 (Production of Carpet Sod Using Seed and Runner of Zoysiagrass (Zoysia spp.))

  • 한국잔디의 종자 및 영양체를 이용한 carpet 잔디 생산체계 확립은 한국잔디의 고성 효율과 이용 증대를 높일 수 있는 유용한 방법이다. 또한 덧파종 기법을 이용해 한국잔디류의 carpet 잔디를 생산하는 것도 효율적인 carpet 잔디 생산 및 푸른 기간 연장의 가능성을 제시할 수 있다고 생각한다. Z. japonica cv. Zenith 종자를 이용해 carpet 잔디를 생산시 6g/$m^2$ 파종시 가장 효율적으로 확인되었으며, 영양체를 이용할 경우는 초종에 따라 피복률에 차이를 보였다. 사용한 초종 중 중엽 Z. japonica가 우수한 피복률을 보였으며, 영양체 사용량에 있어서는 2.5L/$m^2$을 사용하고, 영양체의 길이는 3~4마디씩 잘라서 이용할 때 효과적으로 나타났다. 덧파종용 한지형 잔디로는 발아속도가 빠른 퍼레니알라이그라스가 효과적이었으며, 적절한 덧파종 시기는 9월 중순이 효과적이라고 생각된다. 덧파종을 통해 carpet 잔디의 생산기간을 단축할 수 있으며, 푸른 기간을 연장할 수 있을 것으로 생각되나, 조성 후 고관리 등의 문제점을 해결해야 할 것으로 생각된다.

Black Locust (Robinia pseudoacacia L.) in Bulgaria

  • Robinia pseudoacacia is one of most widely cultivated exotic species in Bulgaria. The total area of black locust plantations amounts to 2.9% of the total forest area of the country. 15.34% of the plantations are of management afforestations category, where the priority is given to timber production. They have been created on rich and moisture soils, which are the most appropriate for the species in order to achieve its biological potentials of high productivity. The rest of the available plantations in the country are planted on poorer and drier soils up to 600~800 m altitude. The high adaptive ability of the species to unsuitable environmental conditions as well as the high sprout potential was used for their creation. These stands are mainly done with the aim to protect and ameliorate damaged environments and production of small-size timber and fire woods. They are cultivated until 15-20 years and are revived by sprouts. Therefore the management goals searched, 45.69% of those forests are low productive and 38.97% with average productiveness. The present report deals with growth and productivity capacity of black locust plantations; production of sowing materials; production of reproductive and vegetative saplings for afforestation; the technological aspects of afforestation works; the management and use of black locust plantations as well as some more important diseases and pests affecting the species. The report is entirely based on Bulgarian scientific research works and experience with the Robinia preudoacacia. Recommendations for optimizing the use of species are given.

Optimization of Medium Composition and Cultivation Parameters for Fructosyltransferase Production by Penicillium aurantiogriseum AUMC 5605

  • Fructooligosaccharides have been mainly produced by microbial fructosyltransferases (FTase) enzymes. The present work focuses on the optimization of medium composition and cultivation parameters affecting FTase produced by Penicillium aurantiogriseum AUMC 5605 in shake flask cultivation. FTase production was optimized in two steps using DeMeo's fractional factorial design. A 1.46-fold increase in FTase production (105.4 U/mL) was achieved using the optimized culture medium consisting of (g/L): sucrose, 600; yeast extract, 10; $K_2HPO_4$, 5; $MgSO_4{\cdot}7H_2O$, 0.5; $(NH_4)_2SO_4$, 1.0 and KCl, 0.5. The obtained results showed that the maximum FTase enzyme activity was produced at initial cultivation pH values ranging from 6.0-6.5, at agitation speed of 200 rpm and using vegetative fungal cells as inoculum. Moreover, results showed that optimization of medium composition and some cultivation parameters resulted in an increase of about 93.7% in the enzyme activity than the nonoptimized cultivation conditions after 96 h of cultivation. Additionally, maximum production and specific production rates recorded 2340 U/L/h and 102 U/L/h/g cells, respectively.

Reproduction rate and stolon production rate after transplantation of grass germplasm

  • Grass breeding starts with the process of selecting grass with good traits, and this operation consumes a lot of resources. Therefore, there is a need for an efficient screening method. Stolon is a unit for the storage of carbohydrates and vegetative propagation, which enhances grass growth and grass sod. Grass varieties with active production of stolons have excellent traits because they reproduce quickly and have a high density. To select grass with such a trait, the survival rate and the production rate of stolon after transplantation of 72 grass germplasms were investigated. After transplantation, the survival rate ranged from 75% to 100%. The majority of the grass cultivars showed a 100% survival rate. Therefore, the group was divided into two groups: A grass variety showing 100% survival and a grass variety that did not show 100% survival. The grass cultivar group, which showed a 100% survival rate after transplantation, included 61 turf varieties, and the rates of stolon production in these grass varieties ranged from 0 to 100%. In contrast, 10 varieties were included in the grass cultivar group that did not show 100% survival after transplantation. These cultivars had a stolon production rate of 0 to 33%. The results suggest that grass germplasms with a 100% survival rate should be selected.

The Mitogen-Activated Protein Kinase Signal Transduction Pathways in Alternaria Species

  • Mitogen-activated protein kinase (MAPK) cascades are conserved signaling modules in the eukaryotic cells. They are involved in many major cell processes in fungi such as stress responses, vegetative growth, pathogenicity, secondary metabolism and cell wall integrity. In this review, we summarized the advances of research on the MAPK signaling pathways in Alternaria species. As major phytopathogenic fungi, Alternaria species reduce crop production. In contrast to the five MAPK pathways known in yeast, only three MAPK pathways as Fus3/Kss1-type, Hog1-type, and Slt2-type have been characterized in Alternaria. The Fus3/Kss1-type MAPK pathway participates in regulation of vegetative growth, conidiation, production of some cell-wall-degrading enzymes and pathogenicity. The Hog1-type pathway is involved in osmotic and oxidative stress, fungicides susceptibility and pathogenicity. The Slt2-type MAP kinases play an important role on maintaining cell wall integrity, pathogenicity and conidiation. Although recent advances on the MAPK pathways in Alternaria spp. reveal many important features on the pathogenicity, there are many unsolved problems regarding to the unknown MAP kinase cascade components and network among other major signal transduction. Considering the economic loss induced by Alternaria spp., more researches on the MAPK pathways will need to control the Alternaria diseases.

Above- and below-ground vegetative responses to prescribed fire regimes in a Chesapeake Bay tidal brackish marsh

  • The primary purposes for using fire are to enhance marsh vegetation to support waterfowl, and to manage invasive plant species. The study was conducted for two consecutive years in 2004 and 2005, investigating the effects of prescribed fire regimes on vegetation biomass in tidal brackish marsh areas of the Blackwater National Wildlife Refuge located on the eastern shore of Maryland, USA, that are under relatively similar environmental conditions. Four different burn regimes (i.e., annual burn, 3-5 year burn, 7-10 year burn, and no burn) were applied in the study. Above- and below-ground vegetation biomass samples as affected by the different burn regimes were harvested in each year for five plant species native to the marsh; Distichlis spicata, Spartina alterniflora, Schoenoplectus americanus, Spartina cynosuroides and Spartina patens. No significant difference was found either in total above-ground biomass or in above-ground biomass by species between burn regimes in 2004. However, more total above-ground biomass was produced in annual burn regime in 2005 than in the other burn regimes. There were no consistent effects of burning on vegetative biomass production by species, but it seemed D. spicata was somewhat benefited by prescribed burning for its biomass production. Moreover, the stem density for D. spicata under annual burn regime was significantly higher than that in the other burn regimes, showing some positive effects of burning on vegetation. The below-ground biomass was significantly greater in 2004 than in 2005, yet with no significant difference between burn regimes in either year. A longer-term monitoring is strongly recommended.

Dry Matter Accumulation, Harvest Index, and Yield of Soybean in Response to Planting Time

  • Planting date of soybeans [Glycine max (L.) Merr.] is one of production components in cultural systems. The objective of the current study was to identify the components of soybean production and cultural practices encompassing planting dates and cultivars that respond to dry matter accumulation, harvest index and yield components. Three determinate soybean cultivars were planted on May 13 (early), June 3 (mid), and June 24 (late). Planting density was 60$\times$15cm with 2 seeds (222,000 plants per ha). Soybean plants were sampled every 10 days interval from the growth stages of V5 to R8 and separated into leaves including petioles, stems, pods, and seeds. Dry matter accumulations, harvest indices, and yield components were measured. Early planting had taken 55 days from VE to R2 and late planting taken 39 days indicating reduced vegetative growth. Early planting showed higher leaf, stem, pod and seed dry weights than late planting. However, late planting appeared to be higher harvest index and harvesting rate. Vegetative mass including leaf and stem increased to a maximum around R4/R5 and total dry weight increased to a maximum around R5/R6 and then declined slightly at R8. The highest seed yield was obtained with mid planting and no difference was found between early and late plantings. Cultivar differences were found among planting dates on growth characteristics and yield components. The results of this experiment indicated that soybean yield in relation to planting dates examined was mainly associated with harvest index and harvesting rate, and planting date of cultivars would be considered soybean plants to reach the growth stage of R4/R5 after mid August for adequate seed yield.

A Putative Transcription Factor pcs1 Positively Regulates Both Conidiation and Sexual Reproduction in the Cereal Pathogen Fusarium graminearum

  • The plant pathogen Fusarium graminearum causes Fusarium head blight in cereal crops and produces mycotoxins that are harmful to animals and humans. For the initiation and spread of disease, asexual and sexual reproduction is required. Therefore, studies on fungal reproduction contribute to the development of new methods to control and maintain the fungal population. Screening a previously generated transcription factor mutant collection, we identified one putative $C_2H_2$ zincfinger transcription factor, pcs1, which is required for both sexual and asexual reproduction. Deleting pcs1 in F. graminearum resulted in a dramatic reduction in conidial production and a complete loss of sexual reproduction. The pathways and gene ontology of pcs1-dependent genes from microarray experiments showed that several G-protein related pathways, oxidase activity, ribosome biogenesis, and RNA binding and processing were highly enriched, suggesting that pcs1 is involved in several different biological processes. Further, overexpression of pcs1 increased conidial production and resulted in earlier maturation of ascospores compared to the wild-type strain. Additionally, the vegetative growth of the overexpression mutants was decreased in nutrient-rich conditions but was not different from the wild-type strain in nutrient-poor conditions. Overall, we discovered that the pcs1 transcription factor positively regulates both conidiation and sexual reproduction and confers nutrient condition-dependent vegetative growth.
