양파껍질과 양파육질의 용매추출물에 따른 항산화 효과 (Antioxidant Effects on various solvent extracts from Onion Peel and Onion Flesh)

  • This study was designed to investigate the role of onion as a natural antioxidant. Onion was distinguished as yellow onion peel and onion flesh. Onion samples were extracted with 5 different kinds of solvents such as water, 70% ethanol, 99.9%ethanol, 99.9% methanol, and 96% butanol in order to select optimal extraction solvents, In this part of study linoleic acid was used s an model system for the purpose of determining the antioxidant activities. The optimal extraction rate of various solvents containing onion samples was determined by measuring extraction yield, electron donating ability(EDA), thiobarbituric acid(TBA), and thiocyanate, which are common methods for measuring activity. As a result 70% ethanol was shown as the most effective solvent.

접촉분해경유로부터 산화황화합물의 분리에 관한 추출용매의 영향 (Effect of Extraction Solvent on the Separation of Sulfur Components in Light Cycle Oil)

  • 접촉분해경유의 산화반응후 포함된 산화황화합물을 분리하기위해 다양한 용매를 사용하여 용매추출에 관한 연구를 수행하였다. 용매로는 극성을 가진 물, N-메틸피놀리논, 에틸아세테이트, 디메틸포름아마이드, 이소프로필알코올, 아세토니트릴, 메탄올등을 사용하였다. 실험결과, 접촉분해경유와 용매와의 층분리는 적절한 양의 물을 첨가한 경우에 이루어졌으며, 물과 N-메틸피놀리논을 혼합한 혼합용매가 접촉분해경유로부터 산화황화합물의 선택적인 분리에 가장 적절하였다. 또한 접촉분해경유로부터 황화합물을 99.5% 이상으로 제거하기 위해선, 4단 정도의 평형추출이 필요하였다.

  • Even if odorous compounds remain very low concentration in water, it cause strong odor. Because Geosmin and most of odorous compound had very low vaporization, those were difficult to analyze with GC/MSD and Purge & Trap. So, we needed pre-treatment method for decreasing amounts of extracting solvents, improving recovery efficiencies and increasing analytical efficiencies. This study developed efficient technology for analyzing odorous compounds, using various adsorbents and extracting solvents. The optimum adsorbent was XAD resins. Especially, XAD-2, XAD-7 and XAD-2010 were superior, but XAD-2 of these and the optimum extraction solvent is MTBE. Other extraction solvents' efficiency is in order of MTBE>Dichloromethane>n-Hexane>Diethylether. The optimum NaCl dosage for increasing efficiency is 5 g in liquid-liquid extraction method. The shaking time(0~24hr) has no concern with adsorption efficiency. The optimum adsorbent is XAD-2 resin and extraction solvent is MTBE. Dosing NaCl, adsorption efficiency is increased in liquid-liquid extraction method, but NaCl has no effect on liquid-solid extraction method. In this experimental results, this algae toxins(Mycrocystin, Anatoxin etc.).

대기 중 다환방향조탄화수소류의 분석 방법간 비교연구 (A Comparative Study on the Analytical Methods of Polycyclic Aromatic Hydrocarbons in the Atmosphere)

  • In order to compare analytical conditions for measuring PAHs(polycyclic aromatic hydrocarbons) in ambient air, a series of experiment were conducted using three different types of extraction methods that include:(1) accelerated solvent extraction (ASE), (2) soxhlet, and (3) sonification methods. Efficiency and relevant analytixal parameters for each of these methods were examined on five different types of solvents that include:(1) 10% diethyl ether in n-hexane(v/v), (2) dichloromethane, (3)cyclohexane.(4) toluene, and (5) Benzene: ethanol (4:1). Comparison of extraction efficiency for various combinations between methods and solvents was made through an application of both internal and external standard calibration techniques. The extraction efficiency tests, when checked using both internal and external standards, allowed us to derive meaningful comparison among different techniques and among solvents.

Development of Fast Screening Method for Crop Protection Agents in Tobacco by Stir Bar Sorptive Extraction and Thermal Desorption coupled to GC/MS

  • Simultaneous determination of crop protection agents(CPAs) in food are done with multi-residue methods, which are composed of sample clean-up, concentration, chromatographic separation and detection. Stir Bar Sorptive Extraction(SBSE) technique is used for sample preparation of various analytes in several fields. The aim of this study was to develop a sensitive and fast method based on SBSE followed by thermal desorption - gas chromatography - mass spectrometry(TD - GC/MS) to determine CPAs in tobacco sample. For the analysis of tobacco sample prior to the SBSE method, solvent extraction or ultrasound-assisted solvent extraction was performed. methanol was used as the extraction solvent. The extract was then diluted with water. Finally, the sample was subjected to SBSE. A method for fast screening of crop protection agents in tobacco using SBSE-TD - GC/MS has been developed. About 17 CPAs including organochlorine, organophosphorous and others were identified and quantified. This method showed good linearity and high sensitivity for most of the target CPAs. The method was applied to the determination of CPAs at ng/mL levels in tobacco sample. This method is simple, rapid and may be applied in detection of other components.

Characterization of Mulberry Root Bark Extracts (Morus alba L.) Based on the Extraction Temperature and Solvent

  • Mulberry root bark is one of potential plant sources for antioxidant materials which can be used for the relief of oxidative stress. To explore the effects of solvent type and temperature on the structural characteristics and antioxidant activity of the root bark extracts, we prepared various extracts of mulberry root bark (Morus alba L.) using 0 - 100 % ethanol (EtOH) at RT - 100℃. EtOH concentration and temperature critically affected the extraction yields, the content of bioactive components, and antioxidant activity of the extracts. Use of high content of EtOH solvent and low temperature resulted in the low extraction yield. Meanwhile, it was revealed that the extract prepared using absolute EtOH at room temperature contained polyphenols and flavonoids with the highest contents among other extracts. Interestingly, the temperature differently affected the polyphenol and flavonoid contents according to the solvent types. In the case of 30% EtOH solvent, polyphenol and flavonoid contents increased with an increase in temperature, whereas in the case of 70 and 100 % EtOH, these contents decreased. Using the radical scavenging assay, it was confirmed that the 100% EtOH extracts had higher antioxidant activity compared to distilled water (DW) extracts regardless of temperature. Also, heating might extract more antioxidant components from the root bark. Especially, the extract prepared using 30% EtOH solvent at 100℃ showed the highest antioxidant activity. Taken together, these experimental results imply that the extraction parameters should be designed carefully considering the productivity, the extracted bioactive components, and antioxidant activity.

  • The solvent extraction of platinum (IV) metal from hydrochloric acid media using 4-(4-ethoxybenzylideneamino)-5-methyl-4H-1,2,4-triazole-3-thiol (EBIMTT) in chloroform was studied as a function of several variables, such as reagent, acid and metal ion concentration, effect of various diluents, and diverse ions. The proposed method was further applied for the separation of platinum (IV) from binary mixtures, synthetic mixtures, alloys and commercially available samples.

  • This study was concentrated to investigate the dependency of recovery yield on the extraction conditions. From fruiting body, the extracted amount of polysaccharide was studied with various solvent. In order to maximize the yield, the optimum extraction conditions were elucidated with respect to solvent, temperature and time. In addition, the dialysis was applied to obtain the higher purity of polysaccharide with varying the exposure time.

  • Various bupivacaine-Ioaded microspheres were prepared with poly (d,l-Iactide) (PLA) by solvent extraction method. The internal solution of polymer(PLA R104) and drug in glacial acetic acid was introduced into the external phase of polyvinylpyrrolidone (PVP K-30) in polyethyleneglycol (PEG), and emulsified to be an oil-in-oil (o/o) emulsion. β-Glucan 추출에 미치는 온도, 용매 농도 및 pH의 영향 (Effect of Temperature, Solvent Concentration, and pH on the β-Glucan Extraction)

  • 추출조건에 따른 귀리겨 ${\beta}$-glucan의 추출조건을 확립하기 위하여 추출온도($40{\sim}60^{\circ}C$), 추출용매의 에탄올 농도(0~20%) 및 pH(5~9)를 변수로 중심합성계획법으로 설계하여 ${\beta}$-glucan 추출특성을 조사하였다. 추출시간의 증가에 따라 ${\beta}$-glucan의 추출속도는 추출초기에 급격히 증가하였으나, 이후에는 완만하였다. 총괄물질전달계수는 $3.363{\sim}8.552{\times}10^{-6}cm/min$ 범위로 낮은 추출온도에서는 추출용매의 농도가 낮을수록, 높은 추출온도에서는 추출용매의 농도가 높을수록 증가하였지만 pH에 의한 영향은 적은 것으로 나타났다. 추출용매의 pH는 중성일 때보다 산성이나 알칼리성일 때 ${\beta}$-glucan의 추출량이 증가하였다. 추출조건에 따라서 ${\beta}$-glucan 수용액의 점도는 유의적인 차이를 보였으며, 총괄물질전달계수가 높을수록 crude ${\beta}$-glucan 수용액의 점도가 높게 나타났으며, 점도, 추출율 및 총괄물질전달계수의 상관관계를 분석한 결과, 각각의 요인들이 높은 양의 상관관계(p<0.01)를 나타내었다.
