Gastrointestinal and Hepatic First-pass Effects of Triflusal in Rats (흰쥐에서 트리플루살의 위장관 및 간 초회통과효과)

  • In order to elucidate the influence of intestinal and hepatic first-pass effect on the pharmacokinetics of triflusal, the biotransformation of triflusal in the gastrointestinal tract and liver was designed. Moreover, we tried to establish an HPLC method applicable for bioassay and available to pharmacokinetics, not only with the simultaneous determination of triflusal and its active metabolite, 2-hydroxy-4-trifluoromethyl benzoic acid (HTB), but also with improving sensitivity. After the administration of triflusal (10 mg/kg) and HTB (10 mg/kg) into femoral vein, portal vein (only triflusal) and oral route (only triflusal), pharmacokinetic parameters were investigated from the plasma concentration-time profiles of triflusal and HTB in rats. An HPLC method was developed for the simultaneous determination of triflusal and HTB in rat plasma, urine and bile. The HPLC analysis was carried out using a C18 column and acetonitrile-methanol-water (25:10:65, v/v/v) as the mobile phase and UV detection at 234 nm. Furosemide was used as the internal standard. The calibration curves were linear over the concentration range $0.05-5.0\;{\mu}g/ml$ for triflusal and $0.2-200.0\;{\mu}g/ml$ for HTB with correlation coefficients greater than 0.999 and with intra-day or inter-day coefficients of variation not exceeding 10.0%. This assay procedure was applied to the study of metabolite pharmacokinetics of triflusal and HTB in rats. It was supposed that triflusal was almost metabolized in vivo because urinary and biliary excreted amounts of triflusal could be ignored as it was lower than 1.2% of the administered dose. According to the gastrointestinal and hepatic biotransformation pathways of triflusal, it was found that triflusal was hydrolyzed by about 5% in intestine and metabolized by about 53% in liver, and that the bioavailability of triflusal after oral administration of triflusal was 0.44, and also that the fraction of total elimination rate of triflusal which formed HTB in liver $(F_{mi},\;%)$ was about 98%. These results showed that triflusal was almost metabolized in liver, and the total elimination of triflusal in the body was dependent to the formation rate of HTB from triflusal in liver.

Gas Separations of Natural Zeolite by Chemical Treatments (화학처리에 의한 천연 Zeolite의 Gas 분리)

  • In the our country, especially in Yeongil and Wolsung area, abundant authigenic zeolites are found from the tuffaceous sediments and volcanic rocks of Miocene age showing wide variation in their mineralogy and abundance from horizon to horizon. The principal zeolite species identified are clinopti-lolite. mordenite. heulandite. ferrierite, and erionite. etc. Zeolite minerals are widely used in many countries in the following applications; (a) in air separation adsorption processes; (b)as desiccants; (c)in inorganic building materials; (d)in papermaking; (e)in fertilizers; (f)as soilconditioners-this application is based upon the ability of the zeolite to ion exchange with soil nutrients; (g)in the treatment of radioactive wastes; and (h)as adsorbents for toxic gases, etc. In the present paper, using natural zeolite mordenite treated with IN hydrochloric acid or IN sodium chloride solution as column packings, separation characteristics of argon, nitrogen, carbon monoxide, and methane gases have been studied by gas chromatography. By the use of mordenite treated with hydrochloric acid solution, the tailing peak of methane showed from untreated mordenite was satisfactorily reduced, although it was difficult to separate it from carbon monoxide with a column activated at $300^{\circ}C$. Using a column activated at $350^{\circ}C$, methane could be separated from carbon monoxide easily but only carbon monoxide eluted as a bad defined peak. Mordenite treated with sodium chloride solution was generally similar to chromatograms obtained by using the untreated mordenite. Both the above chemical treatments of mordenite had little effect on the separations of argon and nitrogen. The separations and the HETP values obtained from natural zeolite mordenite treated with continuously hydrochloric acid and sodium chloride solutions were almost identical with those obtained with synthetic molecular sieve 5A zeolite. On the other hand, the efficiency of column was good in the range 20~3Oml/min of the carrier helium gas rate.

Modeling the effects of excess water on soybean growth in converted paddy field in Japan 1. Predicting groundwater level and soil moisture condition - The case of Biwa lake reclamation area

  • In Japan, more than 80 % of soybean growing area is converted fields and excess water is one of the major problems in soybean production. For example, recent study (Yoshifuji et al., 2016) suggested that in the fields of shallow groundwater level (GWL) (< 1m depth), rising GWL even in a short period (e.g. 1 day) causes inhibition of soybean growth. Thus it becomes more and more important to predict GWL and soil moisture in detail. In addition to conventional surface drainage and underdrain, FOEAS (Farm Oriented Enhancing Aquatic System), which is expected to control GWL in fields adequately, has been developed recently. In this study we attempted to predict GWL and soil moisture condition at the converted field with FOEAS in Biwa lake reclamation area, Shiga prefecture, near the center of the main island of Japan. Two dimensional HYDRUS model (Simuinek et al., 1999) based on common Richards' equation, was used for the calculation of soil water movement. The calculation domain was considered to be 10 and 5 meter in horizontal and vertical direction, respectively, with two layers, i.e. 20cm-thick of plowed layer and underlying subsoil layer. The center of main underdrain (10 cm in diameter) was assumed to be 5 meter from the both ends of the domain and 10-60cm depth from the surface in accordance with the field experiment. The hydraulic parameters of the soil was estimated with the digital soil map in "Soil information web viewer" and Agricultural soil-profile physical properties database, Japan (SolphyJ) (Kato and Nishimura, 2016). Hourly rainfall depth and daily potential evapo-transpiration rate data were given as the upper boundary condition (B.C.). For the bottom B.C., constant upward flux, which meant the inflow flux to the field from outside, was given. Seepage face condition was employed for the surrounding of the underdrain. Initial condition was employed as GWL=60cm. Then we compared the simulated and observed results of volumetric water content at depth of 15cm and GWL. While the model described the variation of GWL well, it tended to overestimate the soil moisture through the growing period. Judging from the field condition, and observed data of soil moisture and GWL, consideration of soil structure (e.g. cracks and clods) in determination of soil hydraulic parameters at the plowed layer may improve the simulation results of soil moisture.

Mineralogical Composition and Physical Variation of Reproduced Potteries based on Chemical Composition and Firing Temperature (화학조성과 소성온도에 따른 재현 토기의 광물조성과 물성변화)

  • The purpose of this study aims to reveal characteristics of reproduced potteries which made with clays of various chemical compositions in different firing temperatures and thereafter to provide comparative samples for identifying the manufacturing techniques of earthenwares from archaeological excavation by reproducing and characterizing sample under controlled conditions. For this study, various samples of earthenware are reproduced using different types of raw clays at several different firing temperature, followed by physical and structural characterization. Chemical specification were varied from different types of clay, which were calculated by Seger formula, and four different types of clay were selected based on different mole ratio of acidic oxide. The temperatures of firing of 7 samples were varied between $600^{\circ}C$ to $1200^{\circ}C$ at the interval of $100^{\circ}C$ for each sample. The result of analysis revealed that each reproduced earthenware has different chemical compositions divided into two groups: 1. Sample Y(6.10) and Sample G(5.85) clay; 2. Sample H(3.41) and Sample S(2.85) clay. The former which has higher mole ratio of acidic oxide than the latter, shows higher level of rockwell hardness at the same firing temperature. In addition, all four samples presented that as the firing temperature was increased, absorption rates of Y and G were abruptly dropped at $1200^{\circ}C$. Furthermore the more mole ratio of acidic oxide increase, the more microtexture of samples were vitrificated. Such result reveals that mole ratio of acidic oxide influence physical and microtextural characteristics of earthenwares, and it can be used as the comparison data in the understanding of manufacture techniques for the earthenwares of similar chemical composition.

Determination and survey of tetracyclines residue in honey by high performance liquid chromatography (액체크로마토그래피를 이용한 벌꿀 중 테트라싸이클린계 항생물질의 정량분석 및 잔류조사)

  • Oxytetracycline, tetracycline, chlortetracycline and doxycycline in honey were separated by solid phase extraction (SPE) and determined with high performance liquid chromatography (HPLC) with UV/Visible detector. Analysis was carried out using following conditions: XTerra $C_8$ column $(3.9\times150mm\;i.d. 5{\mu}m)$, mobile phase composed of 0.01M oxalic acid : methanol : acetonitrile (820 : 80 : 100, v/v/v), isocratic pump at a flow rate of 0.9 ml/min. and $50{\mu}l$ of injection volume, UV/Visible detector with wavelength of 360nm. The calibration curves of four tetracyclines showed linearity $(\gamma^2>0.999)$ at concentration range of $100\~1,000 ng/ml$. The recoveries in fortified honey represented more than $70\%$ with low coefficient of variation $(<10\%)$ for concentration range of four tetracyclines. The detection limits for oxytetracycline, tetracycline, chlortetracycline and doxycycline were 13.8, 14.6, 26.2 and 24.9ng/g in acacia honey. respectively. We also monitored tetracyclines residue in domestic honey [n : 38, acacia (20), wild flower (18) ] and foreign honey [n=22, legally distributed (13), illegally distributed (9)] using modified Charm II screening and HPLC confirmation methods. Seven of the 60 samples $(11.7\%)$ were suspect positive using modified Charm II screening test. Chlortetracycline residue was found in one foreign honey (illegally distributed) tested at concentrations of 0.22 ppm. Conclusively, for more effective control of tetracyclines used in beekeeping should be further survey for residues in honey and also national guidelines (maximum residue limit : MRL) and methods should be obligatory.

Design of Roof Side Rail by Hot Blow Forming using High Strength Aluminum (핫블로우 포밍을 이용한 고강도 알루미늄 루프 사이드 레일 설계)

  • Recently, lightweight of automotive parts has been required to solve environmental problems caused by global warming. Accordingly, research and development are proceeded on manufacturing of parts using aluminum that can replace steel for lightweight of the automotive parts. In addition, high strength aluminum can be applied to body parts in order to meet both requirements of lightening and improving crash safety of vehicle. In this study, hot blow forming of roof side rail is employed to manufacturing of the automotive parts with high strength aluminum tube. In hot blow forming, longer forming times and excessive thinning can be occurred as compared with conventional manufacturing processes. So optimization of process conditions is required to prevent excessive thinning and to uniformize thickness distribution with fast forming time. Mechanical properties of high strength aluminum are obtained from tensile test at high temperature. These properties are used for finite element(FE) analysis to investigate the effect of strain rate on thinning and thickness distribution. Variation of thickness was firstly investigated from the result of FE analysis according to tube diameter, where the shapes at cross section of roof side rail are compared with allowable dimensional tolerance. Effective tube diameter is determined when fracture and wrinkle are not occurred during hot blow forming. Also FE analysis with various pressure-time profiles is performed to investigate the their effects on thinning and thickness distribution which is quantitatively verified with thinning factor. As a results, optimal process conditions can be determined for the manufacturing of roof side rail using high strength aluminum.

Corrosive Degradation of MgO/Al2O3-Added Si3N4 Ceramics under a Hydrothermal Condition (MgO/Al2O3가 소결조제로 첨가된 Si3N4 세라믹스의 수열 조건에서의 부식열화 거동)

  • Silicon nitride ($Si_3N_4$) ceramics have been considered for various components of nuclear power plants such as the mechanical seal of a reactor coolant pump (RCP), the guide roller for a control rod drive mechanism (CRDM), and a seal support, etc. Corrosion behavior of $Si_3N_4$ ceramics in a high-temperature and high-pressure water must be elucidated before they can be considered as components for nuclear power plants. In this study, the corrosion behaviors of $Si_3N_4$ ceramics containing MgO and $Al_2O_3$ as sintering aids were investigated at a hydrothermal condition ($300^{\circ}C$, 9.0 MPa) in pure water and 35 ppm LiOH solution. The corrosion reactions were controlled by a diffusion of the reactive species and/or products through the corroded layer. The grain-boundary phase was preferentially corroded in pure water whereas the $Si_3N_4$ grain seemed to be corroded at a similar rate to the grain-boundary phase in LiOH solution. Flexural strengths of the $Si_3N_4$ ceramics were significantly degraded due to the corrosion reaction. Results of this study imply that a variation of the sintering aids and/or a control (e.g., crystallization) of the grain-boundary phase are necessary to increase the corrosion resistance of $Si_3N_4$ ceramics in a high-temperature water.

Spatial and Temporal Variation of Phytoplankton Community in the Coastal Waters of Jeju Island (제주연안 식물플랑크톤 군집의 시공간적 변화)

  • Spatial and temporal characteristics of phytoplankton communities around the coastal waters of Jeju Island were investigated with environmental factors such as water temperature, salinity, and dissolved oxygen. Monthly samples were collected at 0 and 30 m depths of 10 stations from March 2015 to February 2016. During the survey period, water temperature ranged in 13.7~25.9 and $13.6{\sim}20.8^{\circ}C$ at 0 and 30 m, salinity in 31.51~34.47 and 33.03~34.47 psu at 0 and 30 m, and dissolved oxygen in 6.12~8.10 and $5.73{\sim}7.88mg\;L^{-1}$ at 0 and 30 m, respectively. Chlorophyll-a ranged in 0.28~2.48 and $0.44{\sim}1.01{\mu}g\;L^{-1}$ at 0 and 30 m, respectively. Phytoplankton abundance fluctuated in the range of $5,300{\sim}639,900cells\;L^{-1}$ during the year, showing the lowest in February at all stations, and the highest in July at the northern and western sea as well as in August at the southern and eastern sea of Jeju Island. A total of 362 species were occurred including 181 spp. of Bacillariophyta, 147 spp. of Dinophyta and 34 spp. of other phytoflagellates. Dominant species with occupancy rate over 20 percent of phytoplankton abundance showed apparent seasonal succession such as Paralia sulcata and Skeletonema costatum in spring and autumn, 6 spp. of genus Chaetoceros in summer, and 2 spp. of genus Chaetoceros and Thalassionema frauenfeldii in winter. Monthly abundance in the northern and western sea fluctuated with similar tendency, and the southern and eastern sea also showed similar pattern of monthly abundance variation. Species composition and dominant species succession mentioned above were quite different from previous studies, so some physical changes such as water temperature, salinity and current pattern might cause the changes of phytoplankton assemblages around Jeju Island.

Changes in the Seed Viability and DPPH-radical Scavenging of Helianthus annuus. Seeds According to Low Dose ${\gamma}$-ray irradiation (감마선 조사-해바라기 종자의 저장기간에 따른 종자활력과 추출물의 항산화활성)

  • This study was to elucidate the effect of low dose gamma (${\gamma}$)-ray irradiation on the germination, seedling growth and antioxidant activity in sunflower seeds. The seed germination was stimulated in the range of 2.7${\sim}$8.0% by low dose ${\gamma}$-ray irradiation compared with that of the control; however, the germination of seeds stored for 4 months after irradiated was decreased at all low dose ${\gamma}$-ray irradiation groups. Especially, the germination rate of 32 Gy-irradiated seed group was much lower than that of the other groups and 8 Gy was the optimal radiation dose for germination at room temperature. Though there was variation with storage time and temperature, seedling growth stored at 10$^{\circ}C$ for 4 months after irradiated was promoted at the 1 Gy ${\gamma}$-ray irradiation group compared to seed stored at 25$^{\circ}C$. For the antioxidant activity of ${\gamma}$-ray irradiated seeds extract, 4 Gy and 32 Gy groups were significantly increased by 68.83% and 95.44%, respectively, compared to control at the concentration of 100 ${\mu}g$/ml.

Yield of Green Manure and Nitrogen of Cornflower (Centaurea cyanus L.) in Different Upland Soil Textures (토성별 수레국화 (Centaurea cyanus L.)의 녹비수량 및 질소생산량)

  • Experiments were conducted to find out the landscape effects and green manure production at the same time in farmland. Cornflower was grown in different soil texture with sand, sandy loam, loam, clay loam, and was sowing with autumn and spring respectively. The overwintering rate of cornflower was at 58.7% in average, and the treatment at sand soil showed 62.1% that was highest among other soils, which cornflower is possible to winter landscape crop. After flowering of cornflower, the contents of total nitrogen (T-N) and total carbon (T-C) in plant were 15.0 and $409.2g\;kg^{-1}$, respectively, and the carbon-nitrogen ratio (C/N) was 28.6. The yield of cornflower biomass, which will be returned to soil as green manure, recorded $1,210{\sim}3,920kg\;ha^{-1}$ at the spring seeding higher than the autumn seeding as $1,540{\sim}3,170kg\;ha^{-1}$, and the biomass treated by soil texture were showed that the treatments at the clay loam had been the largest yields both spring and autumn seeding among at other treatment of soil. The heights of cornflower regardless of soil treatments were 52.8 to 73.6 cm at the autumn seeding and 35.5 to 79.2 cm at the spring seeding although it was more significant variation at the soil textures than the seeding periods. The flowering periods of cornflower ranged from $17^{th}$ to $20^{th}$ in May at the autumn seeding and from $19^{th}$ to $20^{th}$ in June at the spring seeding, which was faster 30 days approximately at the autumn seeding than the spring seeding. In a view of the cornflower application as green manure after flowering, the autumn seeding, when considered to combine with following crops, was more suitable and various than the spring seeding, even though the yield at spring seeding was higher than one at autumn seeding.
