Quality Changes of the Extracts of Salt-Fermented Ammodytes personatus during the Storage (까나리 염장품 추출물 저장 중의 품질 변화)

  • The quality changes of the extracts of salt-fermented Ammodytes personatus during the storage were examined at 1 month interval during 3 months fermentation period. To prepare the extracts of Ammodytes personatus, 17%, 20% and 25% salt were added respectively and then stored for 3 months at room temperature and then extracted with hot water. During the storage, moisture and lipid contents of the samples decreased, but the crude protein contents increased gradually. On the other hand, ash content and pH showed little changes during the fermentation storage. After 6 months fermentation, the extracts of salt-fermented Ammodytes personatus were rich in free amino acids, such as glutamic acid, alanine, lysine, leucine, isoleucine, valine and aspartic acid in the order. As the fermentation progressed, the fish odor, turbidity, salty taste, savory flavor and overall preferences were similar to those of Ammotlytes personatus sauce in the market. The total cell numbers were 450∼570 cfu/mL at 0 day, but there were none revealed during the fermentation.

  • Several hydroxylated cinnamic acid derivatives were prepared from the corresponding acids and amino acid residues, and their hypocholesterolemic activities were evaluated in high cholesterol-fed mice. The presence of the double bond in hydroxylated cinnamide derivatives decreases cholesterol-lowering activities and the number of free phenolic hydroxy groups affect greatly the activities. 3,4-Dihydroxy hydrocinnamides obtained from amino acid derivatives containing a hydrophobic side chain such as alanine, valine, phenylalanine, and isoleucine exhibited potent cholesterol-lowering activities.

  • The resolution of five amino acids derivatives by gas chromatography on phenylthiohydantoin derivatives as chiral stastionary phase was investigated. (S)-5-isopropyl, isobutyl, (sec)-butyl and benzoyl derivatives of $N^3-phenyl-2-thiohydantoin$ were synthesized from L-valine, L-leucine, L-isoleucine and L-phenylalanine, respectively. The phases were employed at several column temperatures ($140^{\circ}C{\sim}210^{\circ}C$). The separation factors were compared with each stationary phase with varying R groups. The possible mechanism of chiral recognition was investigated by NMR technique.

  • In order to find 2-(5-nitro)furylacrylic acid derivatives possessing antimicrobial activity, nine new 2-(5-nitro)furylacrylamino acids were synthesized which were obtained by the action of 2-(5-nitro)furylacryla chloride on amino acids, such as $_{L}$-phenylalanine, glycine, $_{L}$-isoleucine, $_{L}$-glutamic acid, $_{DL}$-methionine, $_{L}$-threonine, $_{L}$-valine, $_{L}$-tryptophan and $_{DL}$-alanine, according to Schotten-Baumann method. These compounds generally showed a good bactericidal and bacteriostatic activity against Bacillus subtilis but were less effective against staphylococcus aureus, Proteus vulgaris and Escherichia coli. Of the above nine compounds, 2-(5-nitro)furylacryl glycine exhibited a good bactericidal activity.

  • Quantitative analysis was achieved by gas-liquid chromatographic method (GLC) with a single column system of OV-17 for 16 of protein amino acids in mushroom (agaricus bisporus). 방선균이 생산하는 아미노산 대사길항물질, YS-460의 분리 정제 및 특성

  • An amino acid antimetabolite named YS-460 was isolated from the culture filtrate of a newly isolated Actinomycetes identified as Streptomyces sp. Fermentation was carried out in the synthetic medium at 30$\circ$C for 5 days. Purification was done by ion exchange resin, active carbon, silica gel column chromatography and obtained 38 mg of pure active substance per liter of the broth. YS-460, an amino acid like substance, has the molecular formula of C$_{7}$H$_{11}$NO$_{3}$- Its structure determined to be furanomycin by spectral analysis. It is active against some bacteria on a chemically defined medium and reversed competitively by L-isoleucine and non-competitively by L-leucine and L-valine.

  • A gas chromatography/electron impact mass spectrometric assay method was developed for the determination of Hb-adduct, 2-(hydroxyethyl)valine (HEVal), of ethylene oxide(EO). Globin was separated from hemoglobin by acid iso-propanol and ethyl acetate, then HEVal was isolated as PFPITH-HEVal by Edman degradation. PFPITH-HEVal was silylated with N-methyl-N-(tert-butyl-dimethylsilyl)trifluoroacetamide(MTBDMSTFA)-NH4I (1000:4) under catalysis of dithioerythritol. The detection limit of the assay was 5.8 pmol/g based upon assayed hemoglobin of 0.1g. Two groups of mice were exposed to EO for 0.5 and 1.0 hr/day, respectively at 400ppm during 4 weeks. As the result, the adduct levels increased according to the exposure time with the linearity of 0.7011 and 0.8914, respectively, HEVal was very valuable as biomarker for the exposure of EO. In human, HEVal was analyzed until 8.33 pmol/mg.

  • For the investigation of major taste components in loquat(Eiobotrya japonica) flesh its conte-nts and compositions of free sugars organic aicds and free amino acids were analyzed Major free sugars of the fully ripened loquat were fructose glucose and sucrose and their contents were 3,71, 3.42 and 0.46%(w/w) respectively. The content of total sugar 13.7% was 2 times higher than that of the unripe fruit. The content of total organic acid was about 0.2% (w/w) and major organic acids were malic acid -89mg% formic acid -32mg% and oxalic acid -26%mg% Thirteen kinds of free amino acids from the fully ripened loquat were confirmed. Major free amino acids were aspartic acid valine glutamic acid serine alanine and histidine and their contents were in the range of 18-30mg%.

  • Properties of a protease purified from Neungee[Sarcodon aspratus(Berk, ) S. Ito] have been investigated. The enzyme displays a glycosylated serine protease. The enzyme is able to hydrolyze alanine glycine methionine glutamine and cysteine of N-CBZ and N-t-BOC-L-amino acid derivatibes relatively strongly but splits valine proline and isoleucine derivatives with low affinity which means the enzyme has the broad substrate spectrum toward the amino acids. Interestingly the enzyme was inhibited by bromelain inhibitor. That is the active site environ-ment of the enzyme is believed to be similar to that of bromelain However peptide mapping studies show that the two enzymes have distinct different cleavage sites.

Functional Amino Acid Residues of Recombinant Tobacco Acetolactate Synthase

  • Acetolactate synthase (ALS) is the common enzyme in the biosynthetic pathways leading to leucine, valine, and isoleucine. Tobacco ALS was expressed in E. coli and purified to homogeneity. The recombinant tobacco ALS was inactivated by thiol-specific reagents, N-ethylmaleimide (NEM) and 5,5'-dithio-bis-(2-nitrobenzoic acid) (DTNB). Inactivation of the ALS by NEM followed pseudo-first order kinetics and was first order with respect to the modifier. The substrate pyruvate protected the enzyme against the inactivation by NEM and DTNB. Extrapolation to complete inactivation of the enzyme by DTNB showed modification of approximately 2 out of 4 total cysteinyl residues (or 2 cysteinyl and 1 cysteinyl residues), with approximately 1 residue protected by pyruvate. The tobacco ALS was also inactivated by the tryptophanspecific reagent, N-bromosuccinimide (NBS), and was similarly protected by pyruvate. The kinetics of the inactivation was first-order with respect to NBS. The present data suggest that cysteinyl and tryptophanyl residues play a key role in the catalytic function of the enzyme.
