Metastatic lipid-rich mammary carcinoma in a dog

  • An adult female dog was presented for evaluation of mammary gland masses. Complete blood count and serum chemistry data were within normal limits. Fine-needle aspiration cytology of the mammary masses revealed clusters of malignant epithelial cells with clear cytoplasmic vacuoles. Based on histopathological findings, a diagnosis of lipid-rich mammary carcinoma was made. Approximately 5 weeks after surgical removal, the tumor recurred at the surgery site and metastasis to the tibia was detected. Due to the poor prognosis and deterioration of the condition, the dog was euthanized.

  • With electron microscope, author studied on the pulp structure of human primary tooth in shedding stage. Non-carious human primary molar teeth were selected for this study. Using standard methods, specimens were sectioned and examined by light and electron microscope, The results were as follows; 1. In coronal pulp, odontoblasts were replaced by multinucleated odontoclasts, which contained a large number of mitochondria of varying shape and vacuoles in cytoplasm. Where odontoclasts were in contact with tooth surface, the characteristic ruffled border and clear zone were observed. 2. Fibrous tissue with plentiful collagen fibers and fibroblasts was observed adjacent to the dentin in the pulp. Fibroblast contained a number of mitochondria and well-developed rough-surfaced endoplasmic reticulum. 3. Inflammatory cells were observed in the pulp and active fibroblasts could be seen between inflammatory cells. In many cases, cervical epithelium proliferated toward absorbed area. 4. Inflammatory cells consisted of a number of lymphocytes, polymorphonuclear leukocytes, plasma cells and macrophages. Macrophage containing lysosomes in digestive state or phagocyting PMN could be seen. 5. In the primary molar of delayed root resorption, odontoblast layer, zone of Weil and cell-rich zone could be seen at roof of pulp chamber and odontoblast in this area cont과ained some lipid droplets.

EFFECT OF SEVERAL RESTORATIVE MATERIALS ON ULRASTRUCTURAL CHANGES OF L929 CELLS USING TRANSMISSION ELECTRON MICROSCOPE (투과전자현미경을 이용한 수종의 수복재가 L292세포의 미세구조 변화에 미치는 영향)

  • Cytotoxicity of dental restorative materials using cell culture technique has been extensively studied by various quantitative assays. The aim of this study was to investigate the microstructural change of damaged L292 cells which could not observed with light microscope. Cytotoxic effect of ZOE, Prisma APH (Densply International Inc., U.S.A.), Clearfil FII(Kuraray Co., Japan), Fuji II(GC Co., Japan) and Fuji II LC(GC Co., Japan) on cultured L292 cells were observed. Irreversible cell damage and cytolysis were found in ZOE and Fuji II groups. In Clearfil FII, mild to moderate cell damage was observed. APH group showed variable cytotoxicity. Moderate cell damage was found in Fuji II LC group. Cytotoxic effect were as follows : A condensation of the chromatin occureds along or adjacent to the inner membrane of the nuclear envelops. The nuclear envelope remained resonably intact but the contents were partially or completely lost. The cell nucleus contains clusters of markedly electron-dense interchromatin granules. The rough endoplasmic reticulum were dilated. In some mitochondira, matrix was disoriented and fused cristae were discernible. Mitochondiral swelling and woolly appearance were recognized. Large vacuoles and autolysosmes were found in cytoplasm. Some breaks of the cytoplasmic membrane and even cytolysis could be seen in dying cells.

  • The gametogenesis of the surf clam, Tresus keenae, were investigated by SEM and TEM. Both the testis and the ovary had follicle tubes surrounded by inter-tubal tissue composed of adipogranular cells that provided storage function. In the vitellogenic oocyte, lipid droplets and lipid yolk granules were found in the vacuoles formed by the Golgi apparatus. Proteid yolk granules were formed by the endoplasmic reticulum and cortical granules in the cytoplasm during vitellogenesis. The mature sperm was primitive and resembled a jar with a cover. The sperm heads were approximately $2.00-2.30 {\cal}um$. The acrosomal rod projected in front of the acrosome. In addition, four large mitochondria were in the midpiece.

  • Ewing's sarcoma (ES)/PNET is common in both axial and appendicular skeletons, but is extremely rare in the talus. Here, we report a case of ES/PNET of the left talus in a 29-year-old male patient diagnosed by fine needle aspiration cytology, and review the literature on similar cases. The cytological smears were composed of individually dispersed small round cells and occasional clusters of loosely cohesive cells. The tumor cells were fragile, frequently exhibiting naked nuclei. Two distinct types of cells were observed. The light (chief) cells displayed round or slightly oval nuclei with frequent indentations, generally inconspicuous nucleoli, and a thin rim of cytoplasm, which sometimes harbored small vacuoles. The dark cells were smaller, displaying scanty cytoplasm with dense hyperchromatic nuclei, intermixed with chief cells, and often manifesting as small molded groups. However, no significant nuclear pleomorphisms or mitoses were noted. Tumor cells in the ceil block revealed positive cytoplasmic glycogen, as determined by a PAS stain with diastase control, and also exhibited positive immunoreactivity for CD99.

The Cytotoxicity of Rat Hepatocytes on Viscum album Lectin (겨우살이 lectin의 흰쥐 간세포 독성에 미치는 영향)

  • In the present study, the author investigated to the cytotoxocity in cultured rat hepatocytes of Viscum album lectin. The cytotoxcity effect in Viscum album lectin on the activity of LDH was also investigated. Viscum album lectin significantly increased LDH leakage into medium of hepatocytes treated or untreated with $CCl_4$ (p<0.001). However, Viscum album lectin significantly increased LDH leakage from $CCl_4$-induced hepatocyte (p<0.001). There was a significant increase in LDH levels relative to the control group. Histological observation basically supported the result obtained from LDH assay. The livers of rats challenged with $CCl_4$ produced a marked increased cytoplasmic vacuoles and inflammatory cells in number, while the number of necrotic cells and swollen hepatocytes did not change significnatly. Rats administered DMSO alone did not alter the normal hepatic architecture. Histological observation of liver section in rat treated 72 hrs with either Viscum album lectin $CCl_4$-induced liver damage showed number of cytoplasmic vaculoe and necrotic cell. The number of inflammatory cell increased markedly. This results suggest to the conclusion that Viscum album lectin has a effect of hepatotoxicity activator.

Histopathological Effects of Diazinon in Rat Liver (Diazinon을 투여한 rats에 있어서 간의 병리조직학적 변화에 관한 연구)

  • The toxicity and histopathologic changes of diazinon (O, O-diethy-O-(2-isopropyl-6-methyl-4-pyrimidinyl)phosphorothioate) was investigated in rat. Rat was treated with diazinon (100 mg/kg/day) by oral administration for 12 days. The experimental results were summarized as follows. Biochemical parameters such as ALT, AST, LDH and glucose in serum were significantly increased and hematological parameters such as Hb, Hct and PLT in blood were slightly increased in treated groups. Also the activities of serum cholinesterase were very significantly decreased in treated groups. In the histopathological changes, the normal lobular architectural pattern of the liver was well preserved in all treated groups. However, vacuolation or fatty change were represent in hepatocytes. Sections of liver from rats treated with diazinon for 3 and 6 days contained slight lipid infiltration in the form of small droplets randomly distributed that were graded minimal (+) or moderate (++) compared with the respective control condition. After 9 days, there were numerous small and large vacuoles in the terminal hepatic venule and perilobular areas of many serial sections of these rats indicative of fatty infiltration which were graded moderate (++) and severe (+++). After 12 days, fatty infiltration progressed periportal tract areas and graded severe (+++) and very severe (++++) in experimental groups. Note absence of cellular necrosis or inflammation.

ULTRASTRUCTURAL STUDY ON THE EFFECT OF RADIATION IN THE RAT FETUS TONGUE. (방사선조사가 태내백서의 설조직에 미치는 영향에 관한 전자현미경적 연구)

  • The author observed the effects of /sup 60/Co irradiation on the development and subcellular structure of tongue tissue of the fetal rats. The lower left abdomen of mothers were exposed to radiation on 15½th day of gestation with 300R. The fetuses were removed on the 6hr, 14hr, 24hr, 48hr and 72hr after irradiation and the light microscopic and electron microscopic observations of the lingual epithelium, lamina propria and muscle layer were carried out. The results were as follows: 1. The irradiated fetuses showed the retardation of filiform papillae formation. 2. Epithelial cells revealed fusion and myelination of mitochondria, large autolysosomes, increased lipid droplets, retardation of tonofilaments and desmosome formation. 3. In the lamina propria, undifferentiated cells showed bleb formation of nuclear membrane, pyknosis and fragmentation of nucleus, edema of cytoplasm I and nucleus, increased auto-lysosomes, dilatation of cell membrane and cell necrosis. Also, collagenous fibril formation was inhibited by irradiation. 4. In the muscle layer, growth of myotubes was inhibited. Myotubes showed swelling of mitochondria, loss of mitochondrial cristae, autolysosomes, retardation of myofibril formation, and large vacuoles. Undifferentiated cells adjacent myotube contained pyknotic nucleus and autolysosomes. 5. Among the various tissues of tongue, it seems that mesenchymal cells were most radiosensitive.

Mechanisms and Physiological Roles of Mitophagy in Yeast

  • Mitochondria are responsible for supplying of most of the cell's energy via oxidative phosphorylation. However, mitochondria also can be deleterious for a cell because they are the primary source of reactive oxygen species, which are generated as a byproduct of respiration. Accumulation of mitochondrial and cellular oxidative damage leads to diverse pathologies. Thus, it is important to maintain a population of healthy and functional mitochondria for normal cellular metabolism. Eukaryotes have developed defense mechanisms to cope with aberrant mitochondria. Mitochondria autophagy (known as mitophagy) is thought to be one such process that selectively sequesters dysfunctional or excess mitochondria within double-membrane autophagosomes and carries them into lysosomes/vacuoles for degradation. The power of genetics and conservation of fundamental cellular processes among eukaryotes make yeast an excellent model for understanding the general mechanisms, regulation, and function of mitophagy. In budding yeast, a mitochondrial surface protein, Atg32, serves as a mitochondrial receptor for selective autophagy that interacts with Atg11, an adaptor protein for selective types of autophagy, and Atg8, a ubiquitin-like protein localized to the isolation membrane. Atg32 is regulated transcriptionally and post-translationally to control mitophagy. Moreover, because Atg32 is a mitophagy-specific protein, analysis of its deficient mutant enables investigation of the physiological roles of mitophagy. Here, we review recent progress in the understanding of the molecular mechanisms and functional importance of mitophagy in yeast at multiple levels.

Study on Antihyperlipidemic Effect of Kamiondamtang (가미온담탕(加味溫膽湯)의 항고지혈 작용에 대한 연구)

  • Kamiondamtang(KODT) has been used in oriental medicine as stress by many medical practitioner for the study of KODT, we had fed mice divided to three groups(basal diet, hyperlipidemic diet, hyperlipidemic diet + KODT), and observed the change of weight, total cholesterol, triglyceride, SGOT, SGPT, HDL-cholesterol, LDL-cholesterol on the serum per every 7days for 6weeks. To help comparison with the results above, we had tested endothelial cells and liver. The results of this Study were obtained as fallows ; 1. Total cholesterol, triglyceride and LDL-cholesterol were decreased significantly by KODT. 2. HDL-cholesterol was increasd significantly by KODT. 3. SGOT was intended to decrease by KODT, 4. %W was decreased significantly by KODT. 5. In case of supplying KODT, WBC attatched on endothelial cells and vacuoles in muscular layer were not observed. 6. In case of supplying KODT, fatty degeneration was not observed in liver portal area.
