Electron Microscopical Observations of Hepatic Cells in Korean Native Goat Injected with Clostridium perfringens Toxin (Clostridium perfringens독소(毒素)를 주사(注射)한 재래산양(在來山羊) 간장(肝臟)의 전자현미경적(電子顯微鏡的) 관찰(觀察))

  • This paper dealt with the light microscopical and electron microscopical findings on the morphological changes of the liver of Korean native goat injected with toxin (culture filtrate) of Clostridium perfringens which was isolated from Korean native cattle died of acute Clostridium perfringens enterotoxemia. The results observed are summarized as follows. In the microscopical findings, hyperemia and minute hemorrhage of the liver parenchyma, dilatation of hepatic central vein and centrilobular necrosis of liver, cloudy swelling and hydropic degeneration of hepatic cells, and appearance of light eosinophilic granular bodies in the vacuoles were recognized. In the electron microscopical findings, appearance of pinocytotic vesicle (coated vesicle), fusion of these vesicles, formation of vacuole and accumulation of minute granular proteinous materials in the vacuole were observed in the hepatic cells. Decreased number of glycogen granules, swelling and destruction of mitochondria, proliferation of smooth-surfaced endoplasmic reticulum, enlargement of rough-surfaced endoplasmic reticulum, dispersal of thready agranular membranous structure and appearance of secondary lysosome were recognized in the hepatic cell cytoplasm.

Electron Microscopec Observations on the HeLa Cells treated with culture Filtrates of Mycotoxin-Producing Fungi (Mycotoxin분비 수종균주들의 배양여과액으로 처리된 HeLa세포의 전자현미경적 관찰)

  • The fine structure of HeLa cells treated with several mycotoxin-producing fungi (Aspergillus flavus ATCC 15517, Aspergillus parastiticus RIB 1037, Penicillium toxicarium RIB 4002, Penicillium cirinum SWU)238, Penicillium islandicum IFO 5235, Penicillium tadum IFO 5787 and Pencillium brunneum RIB 1172) has been examined and some details have been descried. The normal HeLa cell have numerous microvilli, large ovoid nucleus, pleomorphic mitochondria, electron-dense body, Golgi complex, mid-body and endoplasmic reticulum etc. Certain specific structural changes induced by culture filtrates of several mycotoxin-producing fungi have been noted. These alterations induced disappearance of Golgi complex, rER vacuolization, nucleolus attachment to the nuclear envelope nad appearance of certain vacuoles. There were not any changes by the treatment of culture filtrates of non-toxic fungi and only cell debris of some specimens can be observed by the injury of culture filtrates. The experimental animals treated with mycotoxin-producing fungi (Aspergillus flavus ATCC 15517, Aspergillus parasilicus RIB 1037, Penicillum citrinum SWU 238, Penicillium toxicarium RIB 4002, and Penicillium islandicum IFO 5235) were mal cells treated with culture filtrates.

Effect of autophagy in human tongue squamous cell carcinoma SCC 25 cells from Scutellariae Radix by ethanol extract (에탄올에 의해 추출한 황금이 구강암 세포에서 나타나는 자가포식작용)

  • Objectives : The purpose of the study is to examine the cell growth effect and autophagy effect of Scutellariae Radix by ethanol extract in SCC 25 cells. Methods : Cell growth inhibitory effect and autophagy induced by Scutellariae Radix were confirmed by WST-1 assay, monodansylcadaverine(MDC) stain, and flow cytometry by acridine orange(AO) stain. Results : The Scutellariae Radix treatment decreased the cell proliferation in a dose and time dependent manner. Scutellariae Radix has anticancer effects that autophagic vacuoles were apparent by MDC and AO staining in SCC 25 cells. Conclusions : Scutellariae Radix showed anticancer activity against SCC 25 cells via autophagy. The data provided the possibility that Scutellariae Radix may potentially contribute to oral cancer treatment.

  • Tubulolobular carcinoma is a tumor of the breast that shares histoiogic features of both tubular and lobular carcinoma of the breast. The cytologic features of this neoplasm have not been reported in Korea. We described a case of tubulolobular carcinoma diagnosed by fine needle aspiration cytology in a 46-year-old female. The tumor contained several cytologic features of both tubular and lobuar carcinoma, including tubular structures, single filing of cells, intracytoplasmic vacuoles, and low nuclear grade.

  • The site of the synthesis of Japanese encephalitis virus(JEV) in the actinomycin-treated and infecter PS Y15 cells(a porcine kidney stable cell line) was observed by the immunofluorescent antibody technique, acridine orange staining, and the autoradiographic analysis. In the parallel studies by immunofluorescent technique and acridine orange staining it the infected cells, Viral protein(as an antigen) and viral RNA were detected at the same site of cytoplasm. In the autoradiographic analysis, the cytoplasmic labeling of $^3H$-uridine was due to the synthesis of JEV-RNA, while the nucleolus and nucleus were not involved. In the autoradiographic studies on the secton of infected cells, the $^3H$-uridine was frequently incorporated around the cytoplasmic vacuoles. This localization of labeling agreed with the site of acridine orange positive granules. The results suggest that the syntheses of the viral RNA and viral protein occurred in the similar site of cytoplasm of the infected cells, and also the virus particles seem to be assembled in the sites of the viral RNA and protein syntheses.

  • Attempts were made in order to make clear the meiotic irregularities, male gametophyte formation and spermatogenesis for the purpose of applying these embryological facts to taxonomic works. Followings are the results obtained : 1. Meiosis is extremely irregular, giving rise to giant, micro, and empty, pollens. Meiotic division in each PMC is observed synchronizing. 2. In the microspore immediately before the vegetative-generative nuclear division, the nucleus locates appressed to the inner wall, most of the other part of the cell being occupied by large vacuoles. 3. Spindle symmetry in the microspore nuclear division is of intermediate type. 4. Nuclear divisions in each micropore are not synchronized as in meiosis. 5. Generative nucleus is located in close contact with the inner wall, while the vegetative nucleus occupies the central part of pollen grain. The pollen in this stage looks somewhat like broad bean. Afterward the generative nucleus loses contact with the wall, the nuclear shape changing from lenticular to roundsh. 6. The generative nucleus in fully matured pollen grain usually takes the shape of crescence, those in abortive pollen, lenticular or ellipsoidal, etc.

Studies on the Haustorium of Cuscuta japonica Choisy (새삼(Cuscuta japonica Choisy)의 흡기에 관한 연구 I. 숙주조직 침투전의 흡기)

  • The portion of Cuscuta japonica haustorium which lies external to the host tissues, the upper haustorium, was investigated at the light- and electron-microscopic levels. The haustorium lightly contacted with the host was formed by the expansion of the epidermis and cortex of the stem at the contact side, and to have a group of meristematic cells within the haustorial cortex. When such a haustorium was closely contacted with the host, the meristematic region transformed into a primordial structure of the endophyte (endophyte primordium, EP) which may penetrate into the host tissues. EP consisted of the three kinds of cell group: dividing cells at the adaxial or proximal side; large, elongate cells (idioblasts) at the middle portion,; compressed cells at the abaxial or basal side. the idioblasts were characterized by the presence of large nucleus, dense cytoplasm, several small vacuoles, and abundant cell organelles including the multilamellar structures and cytosegresomes, and thus suggested to have a high metabolic activity. The features of the EP were discussed in relation to the possibility of the penetrating into host tissues.

Infiltrative Lipoma in a Dog (개에서의 침윤성 지방종)

  • A 7 year-old male Maltese dog was referred to Seoul National University Veterinary Medical Teaching Hospital with a recurrent mass in the right scapular region. The patient had no pains associated with the mass. On aspiration cytology, the neoplastic mass is composed of clusters of mature adipocytes with large lipid vacuoles, intermixed with frequent microcapillary vessels which is characteristic of lipoma. At surgery, the mass was infiltrated into the surrounding muscles and therefore the complete resection of the mass was difficult. Histologically, the mass was diagnosed as infiltrative lipoma. The patient has no recurrent signs 8 months after surgery.

  • Phenolic compounds in food and plant materials belong to the simple phenols, phenolic acids, coumarins, flavonoids, stilbenes, tannins, lignans and lignins, all of which are considered as secondary plant metabolites. These compounds may be synthesized by plants during normal development or in response to stress conditions. Phenolics are not distributed uniformly in plants. Insoluble phenolics are components of cell walls while soluble ones are present in vacuoles. A cursory account of phenolics of cereals, beans, pulses, fruits, vegetables and oilseeds is provided in this overview. The information on the bioavailability and absorption of plant phenolics remains fragmentary and diverse. Pharmacological potentials of food phenolics ave extensively evaluated. However, there are many challenges that must be overcome in order to fully understand both the function of phenolics in plant as well as their health effects.

Ultrastructural Changes of the Hermaphrodite Duct Epithelium by Season in a Korean Slug Incilaria fruhstorieri (한국산 산민달팽이(Incilaria fruhstorferi) 장웅동체관 상피의 계절에 따른 미세구조의 변화)

  • A studv on the ultrastructural changes of the hermaprodite duct epithelium in spring and summer specimens of a Korean slug Incirovia fruhstorferi was conducted. In spring specimen, the hermaphrodite duct was 0.3 H 0.2 mm in diameter and seemed to be a little bit abnormal feature, due to shrinkage. The epithelium of the duct was composed of cells containing various-sized vacuoles and their cytoplasm showed high electron density. In summer specimen, the hermaphrodite duct was 0.4 $\times$ 0.23 mm in diameter and seemed to be more voluminous than that of the spring specimen. The lumen of the duct in the summer specimen was lined by a epithelium composed of several cell types such as ciliated cuboidal, pseudostratified cuboidal, ciliated columnar and irregular cells. Phasocvtotic activities of the duct epithelium are observed only in the summer specimen. Both of the duct epithelia in the spring and summer specimens were surrounded by connective tissues at their bases. The results obtained suggest that the duct epithelia of the slugs experience seasonal variation in relation to their reproductive cycles.
