Plant regeneration through multiple-shoot induction and ex vitro rooting in Vaccinium oldhamii Miq. (정금나무(Vaccinium oldhamii Miq.)의 다신초 유도 및 기외발근을 통한 식물체 재분화)

  • In vitro techniques were developed for propagating Vaccinium oldhamii using shoots with apical buds. Explants having an apical bud were cultured on Murashige and Skoog (MS) medium supplemented with 1.0, 2.0, and 5.0 mg/L of each zeatin, thidiazuron, 6-benzylaminopurine (BA), and 6-(γ,γ-dimethylallylamino)purine (2-iP) in order to induce multiple shoots. Among the tested treatments, the 2.0 mg/L of 2-iP proved to be most suited for the multiplication and growth of shoots; the multiple shoot induction rate was 100.0%, the average number of shoots was 7.4 per explant, and the average shoot length was 51.7 mm. The in vitro elongated shoots were rooted on half-strength MS medium containing various concentrations of indole-3-butyric acid (IBA) and 1-naphthaleneacetic acid (NAA). However, overall callus overgrowth was observed in all treatments and resulted in necrosis and abnormal shoot growth in root formation. A low concentration (0.5 mg/L) of IBA was appropriate for normal root development and the in vitro rooting rate was 30%. Ex vitro treatments on root formation using various concentrations of IBA with Talc powder and two types of rooting substrates (Flexi-Plugs or Horticultural soil) were examined. The ex vitro rooting rate (80%) and length of roots (32.9 mm) were obtained when the cut ends of the shoots were treated with 1.0 mg/L IBA and cultivated in Horticultural soil for 2 months. These findings suggest that ex vitro rooting is the more effective method for improving root formation in Vaccinium oldhamii than in vitro rooting.

Functional Bioactive Compounds and Biological Activities of Vaccinium oldhamii (정금나무의 기능성 생리활성 물질과 생리활성)

  • In modern society, interest in antioxidants is increasing as the stress caused by oxidants increases. However, the demand for synthetic antioxidants is decreasing because some studies have confirmed that they are harmful when consumed in large quantities; thus, studies on antioxidants derived from natural substances are actively being conducted to replace synthetic antioxidants. Blueberry, known as one of the world's top ten long-lived foods, is a plant of the Vaccinium (Ericaceae) family, and various pharmacological activities of blueberry including antioxidant activity have been studied. Vaccinium oldhamii (VO) is a deciduous broad-leaved shrub in the same genus as blueberries, and in this paper, we summarize the studies on the efficacy analysis of VO extracts and purified products. The content of phenolic compounds in VO fruits was proportional to antioxidant and anti-influenza activity such as the inhibition of NO production, and the total content of polyphenols and anthocyanin was higher than that in blueberries. VO fruit extracts showed anti-inflammatory activity and anti-cancer activity against human acute leukemia; in contrast, VO branch extracts showed anti-inflammatory activity, activity to inhibit osteoclast differentiation and bone resorption due to inflammatory response, and anti-cancer activity against several human cancer cell lines. Compared to blueberries, VO showed higher phenolic compound content, antioxidant activity, and various physiological activities. In addition, VO is considered to have sufficient value as an alternative crop to blueberries, such as it can be grown natively in Korea, with simple mass cultivation and no need to pay royalties for commercialization.

Compositions and Contents Anthocyanins in Blueberry (Vaccinium corymbosum L.) Varieties (블루베리(Vaccinium corymbosum L.) 품종별 안토시아닌 조성 및 함량)

  • BACKGROUND: Anthocyanins, potential health-promoting compounds, were major natural pigment in the blueberry (Vaccinium corymbosum L.). The objectives of this study was to investigate anthocyanin glycosides in the blueberry varieties.METHODS AND RESULTS: A total of seventeen anthocyanins were identified from highbush blueberry using HPLC (representatives, 530 nm) and ESI-MS in positive ion mode. The individual anthocyanins are containing cyanidin, delphinidin, malvidin, peonidin, and petunidin moieties which are acylated with aliphatic acid (acetic acid) and conjugated with sugar moieties (arabinose, galactose, and glucose). Among them, delphinidin 3-O-galactoside (D3Ga), peonidin 3-O-glucoside (Pn3G) + malvidin 3-O-galactoside (M3Ga) were major compounds in varieties. Total anthocyanins were found the highest level in 'Elizabeth' (1,406.3 mg/100 g dry weight) which was 3-fold higher than 'Darrow' (465.7). Especially, D3Ga was presented the 32% of total anthocyanins followed by Pn3G + M3Ga (20%) in 'Elizabeth'.CONCLUSION: This result was showed as valuable information regarding nutritional properties of the different varieties of the highbush blueberry.

Anthracnose of Blueberry (Vaccinium spp.) of Caused by Colletotrichum gloeosporioides in Korea (Colletotrichum gloeosporioides에 의한 블루베리 탄저병 발생)

  • Anthracnose of blueberry (Vaccinium spp.) caused by Colletotrichum gloeosporioides was observed in the exhibition orchard of Gyeongsangnam-do Agricultural Research and Extension Services in Korea, 2007. The typical symptoms were initially water-soaked lesions and then rotten on fruits, the infected fruits were dropped but some of them were hunged and mummified. The pathogenic fungus grown on potato dextrose agar showed greyish to dark gray color. Conidia were straight, cylindrical apex obtuse in shape and $10{\sim}20{\times}3{\sim}5\;{\mu}m$ in size. Appressoria were brown in color, clavate or ovate in shape and $6{\sim}18{\times}4{\sim}12\;{\mu}m$ in size. Optimum temperature for mycelial growth was $25{\sim}30^{\circ}C$. On the basis of mycological characteristics and pathogenicity test on host plants, the fungus was identified as Colletotrichum gloeosporioides (Penz.) Penz. & Sacc.. This is the first report on the anthracnose of blueberry (Vaccinium spp.) caused by C. gloeosporioides in Korea.

Quality Characteristics of Pudding Using Fruit of Corni Fructus and Vaccinium Oldhamii Miq. (산수유와 정금나무 열매로 제조한 푸딩의 품질특성)

  • Purpose: This study chose Vaccinium oldhamii as a material to complement the redness of Sansuyu, which is unstable to heat, to Sansuyu pudding. Methods: The level of browning and hunter color of Corni fructus juice supernatant (CFS) and Vaccinium oldhamii extract (VOE) were examined and dependent on heating time and storage period. Results: A value (redness) of VOE was high, browning was moderate during the storage period, and though the storage period was extended, the value stayed stable, meaning overall redness was maintained. With the increase in the amount of added VOD, the color preference of Sansuyu pudding, and D (VOE 10.0%) was marked the highest at 5.80. As for the preference of texture, B (VOE not added) was the highest at 5.35, but C (VOE 5.0%) was marked the highest at 5.10 for overall preference. The DPPH radical scavenging ability of Sansuyu pudding was 15.86-21.39% at a concentration of 1,000 ppm, and the total polyphenol content was 136.76-139.62 mg/100 g. Since the redness of Sansuyu is unstable to heat and then is heated, its degree of browning and b value (degree of yellowness) increases. Conclusion: So if a material with a red color that is stable to heat is added to Sansuyu, the preference of Sansuyu products will improve.

  • Two lignan glycoside compounds were isolated from the twigs of Vaccinium oldhami (Ericaceae) through repeated column chromatography. Their chemical structures were elucidated as lyoniside and ssioriside, respectively, by spectroscopic analysis.

The Branch Extracts of Vaccinium oldhamii Stimulate Melanin Synthesis Through Activation of Tyrosinase Activity in B16F10 Melanoma Cells

  • In this study, we investigated the effect of branch extracts from Vaccinium oldhamii (VOB) on melanin synthesis in B16F10 cells. VOB promoted melanin production in absence or presence of ${\alpha}$-melanocyte-stimulating hormone (${\alpha}-MSH$) in B16F10 cells. However, VOB did not affect the expression of tyrosinase and TRP-1 associated with melanin synthesis at the mRNA and protein level in B16F10. But, VOB decreased TRP-2 protein level and induced tyrosinase activation in B16F10 cells. Inhibition of tyrosinase activity and tyrosinase knockdown attenuated VOB-mediated melanin synthesis. In conclusion, it is thought that VOB may stimulate melanin synthesis through activating tyrosinase activity.

Antioxidant Potential of Enzymatic Extracts from Blueberry (Vaccinium corymbosum L.) (블루베리 (Vaccinium corymbosum L.) 유래 효소 추출물의 항산화성)

  • Enzymatic extracts were prepared from the blueberry (Vaccinium corymbosum L.) collected in Jeju, Korea. Five carbohydrases namely AMG, Celluclast, Termamyl, Ultraflo and Viscozyme, and five proteases namely Alcalase, Flavourzyme, Kojizyme, Neutrase and Protamex were used to prepare the enzymatic extracts. Antioxidant properties of each extracts were studied using stable 1,1-diphenyl 2-picrylhydrazyl (DPPH), reactive oxygen species (ROS), nitric oxide (NO) scavenging, metal chelating assays and lipid peroxidation inhibitory activity in hemoglobin-induced linoleic acid system. The phenolic content of all enzymatic extracts was in the range of 517.85-597.96 mg/100 g dried sample. DPPH and NO${\cdot}$scavenging, and metal chelating assays exhibited prominent activities. Viscozyme showed the highest DPPH activity $(0.046{\pm}0.002\;mg/mL)$ while AMG Showed the highest activity in NO${\cdot}$scavenging $(0.339{\pm}0.011\;mg/mL)$. All the extracts exhibited strong metal chelating activities. Blueberry enzymatic extracts also showed relatively good activity in hydrogen peroxide scavenging. AMG showed the highest lipid peroxidation inhibitory activity $(0.28{\pm}0.01\;mg/mL)$ in hemoglobin-induced linoleic acid system. In this results, the blueberry, which has potential antioxidant components, may be a good candidate as a natural antioxidant source.

Anticancer Activity of the Branch Extracts from Vaccinium oldhamii through Cyclin D1 Proteasomal Degradation in Human Cancer Cells

  • In this study, we investigated the effect of the extracts from Vaccinium oldhamii on cell proliferation and the regulatory mechanisms of cyclin D1 protein level in human cancer cells. The branch extracts from Vaccinium oldhamii (VOB) showed higher inhibitor effect against the cell growth than leave extracts (VOL) and fruit extracts (VOF) in human colorectal cancer, breast cancer, prostate cancer, non-small lung cancer, pancreatic cancer and liver cancer cells. In addition, VOB decreased cyclin D1 level at both protein and mRNA level. MG132 treatment attenuated VOB-mediated cyclin D1 downregulation. A point mutation of threonine-286 to alanine attenuated cyclin D1 degradation by VOB. In addition, the inhibition of nuclear export by leptomycin B (LMB) attenuated cyclin D1 degradation by VOB. But, the treatment of PD98059 (ERK1/2 inhibitor), SB203580 (p38 inhibitor), SP600125 (JNK inhibitor), LiCl ($GSK3{\beta}$ inhibitor), LY294002 (PI3K inhibitor) or BAY 11-7082 ($I{\kappa}K$ inhibitor) did not affect VOB-induced cyclin D1 degradation. In conclusion, VOB induced cyclin D1 degradation through redistribution of cyclin D1 from the nucleus to cytoplasm via T286 phosphorylation of cyclin D1, which resulted in the inhibition of cancer cell proliferation.

Analysis of Total Phenolic Content and Antioxidant Activity from Fruits of Vaccinium oldhamii Miq. (정금나무 열매의 총 페놀 함량 및 항산화활성 분석)

  • Vaccinium oldhamii Miq. is a Korean native tree, which is deciduous and shrub tree with broad leaf. To develop superior cultivars for edible or medicinal purposes, we evaluated total phenolic content and antioxidant activity. Total phenolic content was measured by folin-denis method and antioxidant activity was measured by DPPH free radical scavenging activity (DPPH method) and reducing power (potassium ferricyanide method). Total phenolic content, DPPH free radical scavenging activity (at 400 ppm) and reducing power (at 700 ppm) were 15.8~19.9 mg/g, 69.1~91.4% and 0.56~0.96, respectively. A linear correlation was shown between DPPH-radical scavenging activity and reducing power (r=0969).
