Efficacy of ELISA for measurement of protective newcastle disease antibody level in broilers (육계의 뉴켓슬병 방어역가 측정에 있어서 ELISA 검사법의 효용성)

  • Newcastle disease (ND) is a highly contagious disease of poultry that can cause severe economic losses throughout the world. Vaccination has been used for a long time and proved as one of the most effective method to reduce the economic loss due to ND virus infection, The measurement of antibody titer such as hamagglutination-inhibition (Hl) test with sera has been used as a useful method to evaluate the immunity leve of host. However, Hl test is gradually being replaced by the enzyme linked-immunosorbent assay (ELISA), To evaluate the efficacy of ELISA in the chickens vaccinated with different procedure, present study has been performed. After SPF chicks and commercial broilers were vaccinated with different kinds of live vaccines such as V4, VG/GA and/or Bl at various time, the antibody level has been measured using both HI test and ELISA. Challenge test with velogenic viscerotropic NDV was also performed to measure the protective level of antibody. In the SPF chickens, the mean ELISA titer after vaccination and survival rate after challenge was increased and correlated with days post inoculation. More than 80% of chickens with higher than 1,000 ELISA titer after vaccination were survived after challenge with velogenic ND virus and had good correlation between survival rate and antibody titier. In commercial broiler chickens, most of them at market age had low level of ELISA titer regardless of the number of vaccination, and had a low correlation between survival rate and ELISA titer. However, the ELISA titer of remaining birds after challenge was increased. This result indicated that ELISA titer had good response against velogenic NOV infection compared to Hl titer.

  • This study has been designed to evaluate the efficacy of MD in-ovo vaccination in broiler and to find the contamination level of MD virus in broiler farms. A nested PCR test was used to find the level of filed contamination of pathogenic MD virus in the commercial broiler farms. The positive rate of farms contaminated with pathogenic MD virus was 26.09%. And to evaluate the efficacy of MD vaccination, performance factors such as the mortality, feed conversion rate and body weight at market age has been recorded and compared among farms. There was no difference on these factors between vaccinated and non-vaccinated farms except performance. We need further investigation of the film with low performance index. The B/B ratio, which is common index for the evaluation of bursal changes, was calculated and compared between vaccinated and non-vaccinated group There were significant decreases of B/B ratio in the both group without any statistical difference between groups. The reason of decreased bursal size may be due to other infection such as IBD. From this study, we can know that the contamination rate of fm with pathogenic MD virus was very low and not different from previous study. However, the difference of the contamination rate in different locations recommends periodical monitoring on these areas.

The Reactivity to 2TU PPD Tuberculin Skin Test after Percutaneous Multiple Puncture BCG Vaccination (건강한 영아에서 경피다자법 BCG 접종후 2TU 투베르쿨린 검사의 반응성)

  • Purpose : Tuberculosis, a major public health problem, is an important cause of childhood infectious diseases. To decrease the tuberculosis morbidity rate, BCG vaccination and chemoprophylaxis are performed. Recently 2TU PPD skin test was introduced as a diagnostic method for tuberculous infection. We studied the positive conversion rate of 2TU PPD test after percutaneous multiple puncture BCG vaccination. Methods : Four hundred seventy six infants from well baby clinic of Samsung Cheil Hospital were enrolled. They were immunized with percutaneous multiple puncture technique BCG(Japan BCG laboratory, Japan) at 1 month of age. Approximately 6 months later, tuberculin skin test using RT23 2TU PPD was performed. Induration size, family history of tuberculosis and number of BCG scars were evaluated. Induration greater than or equal to 5mm was defined as positive conversion. Results : Among 476 infants, 248(52.1%) were male and 228(47.9%) were female. PPD skin tests were performed $6.2{\pm}0.5$($mean{\pm}S.D.$) months after BCG vaccination. Mean induration size was $7.3{\pm}3.2mm$ and positive conversion rate was 85.5%. Total number of BCG scars was $15.5{\pm}3.2$. Conclusion : The seroconversion rate by 2TU PPD test after percutaneous multiple puncture BCG vaccination was high. But, more comparative studies with various age groups may be needed for 2TU PPD test used as diagnostic method of tuberculosis in the hospitals.

A Study on Health Belief of Convergency Relation of Preventive Behaviors and Seroprevalence Rate of Hepatitis A with Some Adults (예방행위와 항체 양성률과의 융합 관계를 통한 일부 성인의 A형 간염에 대한 건강신념 연구)

  • This study tries to look into the relationship between preventive behaviors and health belief for hepatitis A and seroprevanlence rate of hepatitis A. The cross ratio of seroprevanlence rate of hepatitis A significantly increased with an increase in age [1.210(95% CI, 1.158-1.264)], and those who had hepatitis A vaccination had a significantly higher cross ratio than those who didn't [19.591(95% CI 6.524-58.82)], and the more health motivation they had, the higher the cross ratio was [1.478(95% CI 1.053-2.075)]. To increase seroprevalence rate of hepatitis A, it is necessary to provide personal hygiene for health motivation and prevention education thoroughly and to give vaccination of hepatitis A.

A Study on the Validity of 'Hepa-S' Hepatitis B Antibody Detecting Reagent after Vaccination of 'Hepa-Vax' (B형 간염백신 'Hepa-Vax' 접종후 항체검사시약 'Hepa-S' kit의 정확도에 관한 연구)

  • To attempt to measure the effect of domestic product P.H.A. kit 'Hepa-S' after completion of 'Hepa-Vax' vaccination schedule, P.H.A. test and R.I.A. test on the 330 healthy adults were carried out. The results obtained were as follow ; 1. The positive anti HBs rate after completion of 'Hepa-Vax' vaccination were; in P.H.A. test with domestic product P.H.A. kit 81.2%, in P.H.A. test with foreign product P.H.A. kit 82.7%, and in R.I.A. test 95.8% 2. Using the result of R.I.A. test as the standard, sensitivity of P.H.A. test with domestic product P.H.A. kit was 84.8% and specificity was 100.0% 3. Using the result of R.I.A. test as standard, sensitivity of P.H.A. test with foreign P.H.A. kit was 86.4% and specificity was 100.0%. 4. The concordance rate of P.H.A. test with domestic product and foreign product kit was 98.5%. On the result of this study, there was no significant difference in the validity between the domestic product P.H.A. kit 'Hepa-S' and the foreign P.H.A. kit $'Hebsgencell^{TM}'$. So that it is recommendable to use domestic product P.H.A. kit instead of foreign product P.H.A. kit.

Factors influencing the Human Papillomavirus (HPV) vaccination of females in their twenties in some Busan areas (부산 일부지역 20대 여성들의 인유두종바이러스(HPV) 백신 접종에 영향을 미치는 요인)

  • Purpose: The goal of this study is to assess the factors influencing the HPV vaccination of females in their twenties. Methods: The survey was conducted during 2013.11.11-28 with a convenience sample of 488 participants. Results: The mean score of their knowledge about cervical cancer shows 2.54 out of 7. 96.7% and 31.5% of respondents have heard about cervical cancer and HPV, respectively. Only 2.7% are vaccinated by the recommendations from the medical teams. The higher the knowledge scores, the more the subjects are vaccinated. The factors influencing HPV vaccination appear to be prior knowledge about the HPV. Conclusion: The knowledge level is low compared to previous studies. Despite this, the trend of vaccination rate is higher. Medical teams need to promote vaccination. Accordingly, education and promotion are needed for the unvaccinated.

  • In this study, the accuracy duplication and register rate of the vaccination registration data in National Immunization Registry Information System were evaluated and analyzed. Through which undocumented vaccination status data, duplicate data, missing data, errors data into the vaccination registration data were analyzed. In addition, the quality control for the vaccination registration database quality improvement, were proposed for standard error checking. In this paper, we propose an efficient validation of a quality management system of the database.

Immunogenicity and Safety of Vi Capsular Polysaccharide Typhoid Vaccine in Healthy Persons in Korea

  • The purpose of this study was to evaluate the immunogenicity and safety of Salmonella Typhi Vi capsular polysaccharide vaccine (Vi vaccine) in Korea. The immunogenicity of a single dose of Vi vaccine was evaluated in 157 subjects (75 children and 82 adults) before and at 1, 6, and 12 months after vaccination. Immunogenicity was measured with a passive hemagglutination assay (PHA), quantified as geometric mean titers (GMTs) and seroconversion rates. The safety of the vaccine was investigated by determining adverse reactions occurring within 4h, 3 days, and 1 month after injection. The seroconversion rate for children and adults 1 month after vaccination was 96.92% and 89.02%, respectively. In the case of children, the GMTs of Vi antibodies before vaccination were $5.87{\pm}1.34\;and\;142.59{\pm}2.39$ at one month after vaccination. For adults, the GMTs before and one month after vaccination were $5.58{\pm}1.28\;and\;58.56{\pm}3.67$, respectively. Vi antibodies persisted for as long as 6 and 12 months after vaccination. All adverse reactions in adults and children were minor and did not require treatment. The Vi CPS vaccine was safe and immunogenic in adults and children older than 5 years.

  • In order to determine the knowledge of, attitudes to, and practice of housewives toward health care in a rural area, a survey with questionnaire was carried out with 87 housewives who were sampled randomly from 6 villages in Sudong Myun, from April 16th to 21st, 1979. The following results were obtained. 1. Of the housewives studied, 61.5% knew that B.C.G. is a vaccine for T.B prevention and 12.3% knew that D.P.T. is a vaccine for diphtheria, pertussis, and tetanus. 2. The vaccination rate of the children under six-year of the housewives studied was: polio 83.1%, B.C.G. 75.4%, D.P.T. 66.2%, and measles 55.4% respectively. 3. The vaccination rate was higher in children in the area near from the health subcenter than in there of the area further away. 4. Out of 87 respondants, 87.5% knew one or more methods of contraception for spacing children. These were: loop 69.0%, oral pill 66.7% and condom 14.9% respectively. 5. Out of 87 respondants, 82.2% knew the methods of contraception for sterilization. These were: laparascopy 87.5% and vasectomy 16.9%. 6. Out of 87 respondants those who had experience using contraceptive methods were 70.1% and present users were 47.1%. 7. Contraception practice rate was higher in the group of housewives having middle school education or above than those having primary school education or less. 8. Functions of the health subcenter listed by respondants were: patients care 72.4%, family planning 31.0%, vaccination 23.0%, T.B. control 3.4%, health education 3.4%, infant birth delivery assistance 1.1% respectively. 9. Housewives who knew that there is a village health voluntary worker in their own village were 63.2%(55), and 58.2% of those who knew appreciated her activities. 10. Purposes of expenditure of Myun community health development funds listed by respondants were: aid for patient care 34.5%, aid for health subcenter operation 16.1%, and aid for Myun health development 6.9% respectively. 11. It seems that both of the distance from the health subcenter and the utility rate level of the village health voluntary worker are co-related to the B.C.G. vaccination rate of children. 12. It seems that both of the distance from the health subcenter and the utility rate level of the village health voluntary worker are not co-related to the rate of contraception practice.
