Turnip crinkle vims (TCV) inoculation onto resistant Arabidopsis ecotype Dijon(Di-17) leads to a hypersensitive response (HR) on the inoculated leaves. A dominant gene, HRT, which confers an HR to TCV, has been cloned from Di-17 plants by map-based cloning. HRT is a LZ-NBS-LRR class resistance gene and it belongs to a small gene family that includes RPP8, which confers resistance to Peronospora parasitica Emco5. Outside of the LRR region, HRT and RPP8 proteins share 98% amino acid identity while their LRR regions are less conserved (87% identity). HRT-transformed Arabidopsis plants developed an HR but generally remained susceptible to TCV due to a dominant RRT allele, which is not compatible with resistance. However, several transgenic plants that over-expressed HRT much higher than Di-l7 showed micro-HR or no HR when inoculated with TCV and were resistant to infection. Both the HR and resistance are dependent on salicylic acid but independent of NPRI, ethylene, or jasmonic acid. Arabidopsis plants containing both TCV coat protein gene and HRT developed massive necrosis and death in seedlings, indicating that the TCV coat protein is an avirulence factor detected by the HRT.
Objectives: Hepatoma is a very serious disease in Korea and vvorldwiclc. Hepatitis B vims (HBV) has proved the most significant cause of hepatoma. We canied out this study to investigate the effect of Acanthopanax sessilifloms on inhibiting cell proliferation and DNA synthesis in HepG2.2.15 cell line and on inhibiting phosphorilation of oncogene (MAP kinase) in NIT/3T3-HBx ceIl. Methods: To investigate the anti-cancer effect of Acanthopanax sessiliflorus, we did the CellTiter 96 Aqueous Non-radioactive Cell Proliferation assay (Promega); MTS/PMS assay, [$^3H$]-thymicline incorporation assay, and we measured the gene expression through westem blotting. Results: Acanthopanax sessiliflorus showed an inhibiting effect on the increase of HepG2.2.15 in the NTS/PMS assay. It also showed an inhibiting effect on DNA synthesis of HepG2.2.15 in the [$^3H$]-thymidine incorporation assay. Acanthopanax sessiliflorus showed an inhibiting effect of phosphorilation of MAP kinase in HBV - X genes. too. Conclusions: The results suggested that this herb had an anti cancer effect. We may discover an effective anti-cancer herb medicine through further studies on this herb medicine.

An isolate of Odontoglossum ringspot virus (ORSV) was identified from Cymbidium var. 'Grace Kelly' showing ringspot symptom on the floral and leaf parts, and was denoted as cymbidium ringspot isolate (ORSV-CR). In ultrathin sections of leaf tissue from diseased Cymbidium plants, clusters of virus particles were observed in the vacuole and cytoplasm. In the Western blot hybridization, the virus strongly reacted with ORSV-specific antiserum indistinguishable from ORSV, suggesting that the vims is serologically identical with ORSV. ORSV-CR sap was inoculated onto 20 species belonging to 12 genera. Systemic infection occurred in Cymbidium sp., Nicotiana benthamiana and N. clevelandii, the host of which was found to be different from that of ORSV-Cy, the Korean strain of ORSV. The analysis of coat protein (CP) gene showed that ORSV-CR was highly homologous to the known isolates of ORSV, with over 95.6% identity in amino acid level. Phylogenetic tree analysis of CP showed that ORSV-CR was clustered with the known ORSV isolates, suggesting that ORSV is a very stable tobamovirus.

A mutant of Potato vims Y (PVY) was occurred in PVY resistance flue-cured tobacco breeding line KF0402 $(TC1146{\times}KF117)$ showing vein necrosis at Suwon in Korea. This isolate, PVY-SWM, was differentiated from other PVY based on biological properties and nucleotide sequence analyses of coat protein gene. PVY-SWM caused typical symptoms on 21 indicator plants as compared to the PVY-TOJC37. Remarkably, the PVY-SWM induced distinctly different symptom of systemic vein necrosis on tobacco cultivars V.SCR, PBD6, TN86, TN90, Virgin A Mutant (VAM), Wislica, NC744, KB108 and KB111, which were reported to have the recessive potyvirus resistance gene va. In RT-PCR assays with specific primers for detection of PVY, a single band of about 800bp in length was produced. The amplified DNA was cloned and the nucleotide sequence was determined. The coat protein gene of PVY-SWM showed 88.4%-99.0% and 92.5%-98.5% identities to the 12 different PVY isolates of Genbank Database at the nucleotide and amino acidi respectively. Multiple alignments as well as cluster dendrograms of PVY-SWM isolate revealed close phylogenetic relationship to the $PVY^{NTN}$ subgroup.
Hemagglutination-inhibition titers (log2) to Newcastle disease (ND) vims were chronologically observed in chicks, which were orally inoculated with 5 × 105 oocysts of Cwptospori,drum bcileyi at 2 days of age and subsequently vaccinated with inactivated ND virus at 4 and 21 days postinoculation. In general, the titers were considerably lower in the infected chicks than those in the uninfected control throughout the experimental period (p < 0.01), and rapid negative seroronversions were observed in the infected chicks. The titers reached a peak on weeks 2 and 4 post-booster-vaccination in the control and infected chicks, respectively. Thus, C. bciLeWi infection was shown to have an immunosuppressive effect on ND vaccination when the agent was given to 2-day-old chicks. It is suggested that C. bniLeWi infection in chicks may increase the host susceptibility to ND virus.
Systemic virus symptoms caused by a Potexvirus were observed on leaves of infected hosta (Hasta spp.) plants cultivated in Seoul, Korea. Symptoms on diseased hosta plants include mosaic, mottle, irregular blotchy patches, and chlorotic spots on or distortion of the leaves. No other viruses, such as Cucumber mosaic virus, Lily symptomless virus, or Potyvirus, were detected from the same plants by electron microscopy and by Western blot and RT-PCR analyses, indicating that they were singly infected by the potexvirus. The symptoms differed among cultivars and species of hosta, and affected the quality of plants for commercialization, as well as, plant growth and flowering of susceptible cultivars. Most of the cultivars and species investigated were susceptible to the virus, while some were not infected by the virus at all. Purified virus particles were of filamentous type with unaggregated forms 540 nm in length, which is a typical potexviral morphology. The virus consisted of a single-stranded RNA molecule of 6 kb long for genome and single component of coat protein (CP) about 27 kDa. The CP strongly reacted with the antiserum against Hosta vims X (HVX), suggesting that the virus is an isolate of HVX. This is the first report of the occurrence and identification of HVX from hosta plants in Korea.
The occurrence of potato(Sotanum tuberosum) viral diseases caused by Potato virus X(PVX), Potato virus Y (PVY), Potato leafroll virus(PLRV), Potato vims S(PVS), Potato virus M(PVM), Potato virus A(PVA), Potato virus T(PVT), Alfalfa mosic virus(AIMV), Tobacco mosic virus(TMV), Potato mop top virus(PMTV) Tobacco rattle virus(TRV) and Potato spindle tuber viroid(PSTVd), potato witches' broom phytoplasma, have been identified so far in Korea. Major viral diseases such as PVX, PVY and PLRV had been studied more deeply, however, the others are just identified and only partially characterized since the first study on the relation between PVX nucleic acid and virus protein by Kim in 1961. The most studies on potato viral diseases are mainly focused on the problems of seed potato production. The National Alpine Agricultural Experiment Station(NAAES), since it began its activities in 1961, has given special attention to this problem by doing studies to identify, characterize and control potato virus diseases. This effort resulted in the development of new potato virus detection methods as a basis for elaborating new method of control, such as the production of seed potato free of virus and the selection of new virus-resistant transgenic potatoes. The further studies of potato viral diseases required would be fallowings: the continuous monitoring for the occurrence of identified or not identified potato viruses in Korea, the isolation of resistant viral genes, the development of control method for the non-persistently transmitted viruses like PVY, special vectors such as nematode and fungus transmitted viruses, TRV and PMTV and the development of control methods against potato viral diseases by viral cross protection, therapy, transgenic plant, and the use of the agents or molecules, such as virus inhibitors and antiviral proteins, etc., blocking viral replication.
This paper focuses on the rigorous and holistic fatigue analysis of mooring chains for a deep draft semi-submersible platform in the challenging environment of the central Gulf of Mexico (GoM). Known for severe hurricanes and strong loop/eddy currents, this region significantly impacts offshore structures and their mooring systems, necessitating robust designs capable of withstanding extreme wind, wave and current conditions. Wave scatter and current bin diagrams are utilized to assess the probabilistic distribution of waves and currents, crucial for calculating mooring chain fatigue. The study evaluates the effects of Vortex Induced Motion (VIM), Out-of-Plane-Bending (OPB), and In-Plane-Bending (IPB) on mooring fatigue, alongside extreme single events such as 100-year hurricanes and loop/eddy currents including ramp-up and ramp-down phases, to ensure resilient mooring design. A detailed case study of a deep draft semi-submersible platform with 16 semi-taut moorings in 2,500 meters of water depth in the central GoM provides insights into the relative contributions of wave scatter diagram, VIMs from current bin diagram, the combined stresses of OPB/IPB/TT and extreme single events. By comparing these factors, the study aims to enhance understanding and optimize mooring system design for safety, reliability, and cost-effectiveness in offshore operations within the central GoM. The paper addresses a research gap by proposing a holistic approach that integrates findings from various contributions to advance current practices in mooring design. It presents a comprehensive framework for fatigue analysis and design optimization of mooring systems in the central GoM, emphasizing the critical importance of considering environmental conditions, OPB/IPB moments, and extreme single events to ensure the safety and reliability of mooring systems for offshore platforms.
Rice black-streaked dwarf virus(RBSDV) was purified from infected maize leaves. Antiserum against RBSDV was prepared for virus detection by enzyme-linked immunosorbent assay(ELISA). In detection of RBSDV by ELISA, effective dilution range of antiserum extracted in RBSDV-containing host plants and insect vectors was from 320 to 2,560 times in rice plant, 320 to 5,120 in maize plant, and 160 to 2,560 times in insect vector, Laodelphax striatellus F, respectively. The percentage of viruliferous vector in overwintered nymphs of Laodelphax striatellus determined by ELISA were 3.0 in Milyang, 2.3 in Chilgok, and 3.7 in Sunsan area. Dead insect vector which could not be tested for vims infection by conventional rice seedling inoculation test could be tested by ELISA. One hundred plants of rice and maize were randomly sampled in the field and tested whether or not they were infected with RBSDV. In rice plants, 4 out of 98 plants turned out to be infected with RBSDV by ELISA. In maize plant, 3 out of 92 plants which were excepted 8 plants to be appeared symptom already were infected. As a result, ELISA could be detected even in case of symptomless plants at early stage of viral infection.

Turnip mosaic Potyvirus (TuMV) and Ribgrass mosaic Tobamovirus (RMV) are major viruses infecting crucifer crops in Korea. RMV-FG22 was isolated from oriental cabbage. TuMV isolates were TuMV-CA7 from oriental cabbage, TuMV-TU and TuMV-TU2 from turnip, TuMV-RA from rape, TuMV-ST from stock, and TuMV-R9 from radish. The six isolates of TuMV were classified by symptom expression in inbred lines of crucifers. TuMV-CA7 and TuMV-TU isolates infected mostly oriental cabbages; TuMV-ST, TuMV-TU2, and TuMV-R9 infected radishes; and TuMV-RA infected both oriental cabbages and radishes. Crops used in six combinations of mixed infections were 'Tambok' cultivar resistant to TuMV,'SSD63' susceptible inbred line of oriental cabbage, pure line of leaf mustard, and‘Daeburyungyeorum’cultivar of radish. External symptoms in 'Tambok' and radish by each of the six single infections of TuMV showed similar results by bioassay. Synergistic response of necrotic death occurred within 1 week after inoculation in all combinations mixed with TuMV and RMV-FG22 on leaf mustard. In oriental cabbage 'SSD63' , synergism of necrosis occurred in four TuMV isolates, but not in TuMV-ST and TuMV-R9. In oriental cabbage 'Tambok' , synergism was expressed only in two combinations of RMV-FG22+TuMV-CA7 and RMV-FG22+TuV-TU, but other combinations had the same symptoms produced by RM-FG22. In radish‘Daeburyungyeorum’, only mild mosaic symptoms were induced by combinations of RMV-FG22+TuMV-CA7, RMV-FG22+TuMV-TU, RMV-FG22+TuMV-RA, and RMV-FG22+TuMV-R9. Mosaic and severe mosaic were induced in combinations of RMV-FG22 +TuMV-TU2 and RMV-FG22+TuMV-ST, respectively.
