산란계의 Ceramics 급여 효과

  • This study was evaluated the dietary affect of ceramics on laying performance, intestinal microflora population, NH$_3$ and VFA gas emission from excreta and fatty acids composition of egg yolk in laying hens. A total of 180 layer at 34 weeks of age were fed the experimental diets containing 0.0 % (Control), 0.4 % and 0.8 % of ceramics powder for 6 weeks. It is concluded that 0.4 % ceramics powder supplementation in the 34 to 40 weeks laying hens diet, improves the laying performance and ratio of egg yolk n-6/n-3 fatty acids contents and decreasing emission of NH$_3$ and VFA gas from excreta.

Effect of Unsaturated Fatty Acids on Cellulose Degradation and Fermentation Characteristics by Mixed Ruminal Microbes

  • This experiment was conducted to evaluate the effects of supplemental unsaturated fatty acids (UFA) on fermentation characteristics, especially on gas production, cellulose degradation and volatile fatty acid (VFA) concentration by mixed ruminal microorganisms. In order to attain this objective, unsaturated fatty acids including oleic acid (C 18:1), linoleic acid (C18:2) and arachidonic acid (C22:4) were added at varying level. Mixed ruminal microbes used in this experiment were obtained from the rumen of a cannulated Holstein cow. Medium pH values after 7 d incubation were significantly affected by type and level of unsaturated fatty acids (p<0.01). All of UFA inhibited total gas production, and especially treatment of arachidonic acid at the levels of 0.01% gave the lowest gas. production after 7 d incubation (p<0.01). Comparison of the population of protozoa revealed that UFA did not have any significant effect on the total protozoa number. The addition of UFA did not effect dry matter degradation. Volatile fatty acid (VFA) composition of the culture was influenced little by UFA, although the considerable amount of iso-type VFA were detected in UFA supplemented incubations. The ratio of acetic acids to propionic acids, however, was lower than control in all the treatments after 7 d incubation (p<0.01).

  • This study's purpose is the investigating the created liquid post-acid fermentation of usability of denitrification as exterior carbon sources by loading capacity of Organic matters. The time of acid fermentation of food waste, the slower loading capacity of organic matter, the faster decomposition fate, but the density of generation $VFA_S$ was weak and, $SBOD_5$ : ST-N rate and $SBOD_5/SCOD_{Cr}$ rate was low. Between TS and VS, VS was decreased to 6th day rast, and then was decreased slowly. Two days after stating the experiments, $SCOD_{Cr}$, $SBOD_5$, STOC and $VFA_S$ was decreased or increased slowly, and then increased fast. And after showing the highest density, it was tended to decreased fast. About 5 days after, because about 5 days later $CH_4$ bacteria activity due to the accumulation of $VFA_S$ was limited, $CH_4$ density was weak. When highest density in the acquisition of VFA, $R_1$ was $10,120mg/{\ell}$, $R_2$ was $11,380mg/{\ell}$, $R_3$was $13,720mg/{\ell}$. So $R_3$ only cut was highest generation. At the time of $SBOD_5$ with the highest density, at $SBOD_5$ : ST-N ratio, $R_1$ was 243:1, $R_2$ was 278:1, $R_3$ was 293:1. All of these were high $SBOD_5$ : ST-N ratios. And $R_3$ was the highest. After studying $SBOD_5$ : ST-N ratio and $SBCD_5/SCOD_{Cr}$ ratio, as a carbon source of biological denitrification it was profitable composition ratio.

Effects of Branched-chain Amino Acids on In vitro Ruminal Fermentation of Wheat Straw

  • This study investigates the effects of three branched-chain amino acids (BCAA; valine, leucine, and isoleucine) on the in vitro ruminal fermentation of wheat straw using batch cultures of mixed ruminal microorganisms. BCAA were added to the buffered ruminal fluid at a concentration of 0, 2, 4, 7, or 10 mmol/L. After 72 h of anaerobic incubation, pH, volatile fatty acids (VFA), and ammonia nitrogen ($NH_3$-N) in the ruminal fluid were determined. Dry matter (DM) and neutral detergent fiber (NDF) degradability were calculated after determining the DM and NDF in the original material and in the residue after incubation. The addition of valine, leucine, or isoleucine increased the total VFA yields ($p{\leq}0.001$). However, the total VFA yields did not increase with the increase of BCAA supplement level. Total branched-chain VFA yields linearly increased as the supplemental amount of BCAA increased (p<0.001). The molar proportions of acetate and propionate decreased, whereas that of butyrate increased with the addition of valine and isoleucine (p<0.05). Moreover, the proportions of propionate and butyrate decreased (p<0.01) with the addition of leucine. Meanwhile, the molar proportions of isobutyrate were increased and linearly decreased (p<0.001) by valine and leucine, respectively. The addition of leucine or isoleucine resulted in a linear (p<0.001) increase in the molar proportions of isovalerate. The degradability of NDF achieved the maximum when valine or isoleucine was added at 2 mmol/L. The results suggest that low concentrations of BCAA (2 mmol/L) allow more efficient regulation of ruminal fermentation in vitro, as indicated by higher VFA yield and NDF degradability. Therefore, the optimum initial dose of BCAA for in vitro ruminal fermentation is 2 mmol/L.

Nutrient composition and in vitro fermentability of corn grain and stover harvested at different periods in Goesan, a mountainous area

  • With South Korea's limited capability of feed production because of its relatively small cultivable area, the country is pushed to depend on foreign feed imports despite the immensely fluctuating price of corn. Hence, intensive efforts to increase the total cultivable area in Korea like extending of farming to mountainous area is being practiced. Corn was planted in Goesan County, a mountainous area in the country. Grain and stover were harvested separately in three harvest periods: early-harvest (Aug 8), mid-harvest (Aug 18), and late-harvest (Aug 28). The nutrient composition such as dry matter (DM), crude protein (CP), crude fat (EE), organic matter (OM), neutral detergent fiber (NDF), acid detergent fiber (ADF), and non-fibrous carbohydrates (NFC) was determined after harvest. Effective degradability (ED) of the major nutrients (DM, NDF, ADF, and CP) were measured through in vitro fermentation of rumen fluid from Hanwoo (Korean cattle). pH, ammonia-N concentration, volatile fatty acid (VFA) concentration, and gas production were periodically measured at 0, 3, 6, 12, 24, 48, and 72 h. Corn grain showed higher nutrient content and ED than stover. It also had higher gas production but its pH, ammonia-N, and total VFA concentration were lower than corn stover. The best nutrient composition of corn grain was observed in early-harvest (high CP, EE, NDF, OM, NFC, and low ADF). Early-harvest of corn grain also had high effective degradability of dry matter (EDDM), effective degradability of neutral detergent fiber (EDNDF), effective degradability of acid detergent fiber (EDADF), and total VFA concentration. On the other hand, the best nutrient composition of stover was observed in mid-harvest (high DM, CP, NDF, and low ADF). EDDM, EDNDF, and EDADF were pronounced in early-harvest and mid-harvest of stover but the latter showed high total VFA concentration. Hence, early and mid-harvested corn stover and grain in a mountainous area preserved their nutrients, which led to the effective degradation of major nutrients and high VFA production.

Operation of High Performance Elutriation-Type Sludge Fermenter and Feasibility for Its Application (고성능 세정식 슬러지 산발효조의 운전 및 적용성 평가)

  • The performance of a novel fermentation process, adopting a sludge blanket type configuration for higher hydrolysis/acidogenesis of the municipal primary sludge, was investigated under batch and semi-continuous conditions with various pH and temperature conditions. This acid elutriation slurry reactor provided higher system performance with a short HRT (5 days) and higher acidogenic effluent quality under pH 9 and thermophilic ($55^{\circ}C$) conditions. The hydrolysis of the sludge was revealed to be significantly dependent on seasonal effects for sludge characteristics but with little impact on acidogenesis. Based on the rainy season at the optimum conditions, VFA production and recovery fraction ($VFA_{COD}/COD$) were $0.18\;g\;VFA_{COD}\;g^{-1}\;VSS_{COD}$ and 63%. As byproducts, nitrogen and phosphorus releasing were $0.006\;g\;N\;g^{-1}\;VSS_{COD}$ and $0.003\;g\;P\;g^{-1}\;VSS_{COD}$, respectively. For the mass balance in a full-scale plant($Q=158,880\;m^3\;day^{-1}$) based on the rainy season, the VFA and non-VFA(as COD) production were $3,110\;kg\;VFA_{COD}\;day^{-1}$ and $1,800\;kg\;COD\;day^{-1}$, resulting in an increase of organics of $31\;mg\;COD\;L^{-1}$ and $20\;mg\;VFA_{COD}\;L^{-1}$ and nutrients of $0.7\;mg\;N\;L^{-1}$ and $0.3\;mg\;P\;L^{-1}$ in the influent sewage. The economical benefit from this process application was estimated to be about $67 per $1,000m^3$ of sewage except for energy requirements and also, better benefits can be expected during the dry season. Also, the results revealed that the process has various additional advantages such as pathogen-free stabilized solids production, excellent solids control and economical benefits.

The Study on Ozone Treatment of Wasting Activated Sludge for VFA Production and Reuse as Carbon Source for Phosphorus Release (잉여슬러지의 오존분해에 따른 VFA의 생성 및 인 방출을 위한 탄소원으로의 재이용 가능성에 관한 연구)

  • Recently, the ozone treatment of wasting activated sludge has become one of the effective and feasible process for the sludge reduction. The objective of this study is to investigate the availability of ozonized wasting sludge on external carbon sources 13r phosphorus release. Experiment results showed that the ozone treatment of activated sludge could produce a large amount of VFA such as acetic acid and isobutyric acid. For example, 50.24 mg/L acetic acid was produced with the ozone dose of 0.05 g $O_3/g$ SS, and 123.56 mg/L acetic acid with 0.5 g $O_3/g$ SS. The higher ozone dose was applied, the more VFA was produced from sludge reduction into a limited point. Finally, using ozonated sludge as only carbon source, the batch experiment, to measure phosphorus release rate in anaerobic condition were performed. The specific phosphorus release rates were investigated as 0.94, 1.37, 1.48, 1.68 mg P/g VSS/hr with ozone dose of 0.05, 0.1, 0.2, 0.5 g $O_3/g$ SS, respectively. Considering the degree of mineralization, VFA production, phosphorus release rate, and economical aspect, the optimal ozone dose for sludge reduction and using carbon sources ranged from 0.05 to 0.1 g $O_3/g$ SS.

Effects of Bacterial Fraction and Proportion of Silage and Concentrate on Rumen Fermentation and Gas Production Profile

  • An in vitro experiment was carried out to investigate effects of solid associated (SAB) and liquid associated bacteria (LAB) and the type of incubation substrate on ruminal fermentation and gas production profiles. Bacterial fraction did not influence total numbers of bacteria. Gas production degradation parameters were significantly influenced by bacterial fraction and type of substrate (p<0.05). There was significant interaction between bacterial fraction and type of substrate in gas production (p<0.01). Total VFA concentration and acetic and propionic acid ratio were also influenced by bacterial fraction and type of substrate with little differences in individual VFA concentration.

  • The composition of rumen volatile fatty acids(VFA) and the blood chemistry were investigated in 5 clinically health dairy cows(Group I) reared on dairy farms and in 5 cows with post-parturition(POP) primary ketosis(Group II). The determinations were performed on days 5 to 7 pre-parturition(PRP), immediately POP, and on days 5, 10, 15, 20 and 25 POP. In both groups, the total VFA levels gradually increased starting from day 5 POP, but the levels were lower in Group II than in Group I. With regard to POP. changes in the composition of VFA, Group II occasionally showed lower levels of acetic acid and caproic acid than did Group I. Blood glucose levels decreased POP in both groups. In contrast, blood levels of ketone bodies and 3-hydroxybutyric acid were increased POP, but there was no statistically by significant difference between the groups. The aspartate aminotransferase level was transiently increased immediately POP in both groups, and the increase was more marked in Group II than in Group I. Both groups showed a tendency for total cholesterol, free cholesterol, ester cholesterol, phospholipid, and total bile acid to be increased POP, but there was no statistically significant difference between the groups. Clinically healthy dairy cows also showed POP changes in the composition of VFA and blood similar to those in dairy cows with ketosis, suggesting that even apparently healthy cows are at risk of subclinical ketosis POP.

Biosynthesis of polyhydroxyalkanoate by mixed microbial cultures from hydrolysate of waste activated sludge (혼합미생물배양체를 이용한 폐활성슬러지 가용화 산물로부터 polyhydroxyalkanoate 생합성)

  • A new approach to the solubilization of waste activated sludge (WAS) using alginate-quaternary ammonium complex beads was investigated under controlled mild alkaline conditions. The complex beads were prepared by the reaction of sodium alginate (SA) with 3-(trimethoxysilyl)propyl-octadecyldimethylammonium chloride (TSA) in acid solution, followed by crosslinking with $CaCl_2$. Treatment of WAS with SA-TSA complex beads was effective for enhancing the efficacy of WAS solubilization. The highest value of soluble chemical oxygen demand (SCOD) concentration (3,900 mg/L) was achieved after 10 days of treatment with 30% (v/v) SA-TSA complex beads. The WAS solubilization efficacy of the complex beads was also evaluated by estimating the concentrations of volatile fatty acids (VFAs). The maximum value of VFAs was 2,961 mg/L, and the overall proportions of VFAs were more than 75% of SCOD. The main components of VFAs were acetic, propionic, iso-butyric, and butyric acids. These results suggest that SA-TSA complex beads might be useful for enhancing the solubilization of WAS. The potential use of VFAs as the external carbon substrate for the production of polyhydroxyalkanoate (PHA) by a mixed microbial culture (MMC) was also examined. The enrichment of PHA-accumulating MMC could be achieved by periodic feeding of VFAs generated from WAS in a sequencing batch reactor. The composition of PHA synthesized from VFAs mainly consisted of 3-hydroxybutyrate. The maximum PHA content accounted for 25.9% of dry cell weight. PHA production by this process is considered to be promising since it has a doubly beneficial effect on the environment by reducing the amount of WAS and concomitantly producing an eco-friendly biopolymer.
