Do VDR Gene Polymorphisms Contribute to Breast Cancer?

  • Breast cancer is the first or second leading cancer among females across the globe. A large number of studies have been conducted to assess any relationship between vitamin D receptor (VDR) gene polymorphisms and breast cancer development. Epidemiological studies have indicated that ethnic traits exhibited by a group of people with a common ancestry and culture, alter the link between VDR gene and breast cancer. It has been hypothesized that VDR polymorphisms have the capacity to impact both on incidence of breast cancer occurrence and to predict its outcome. A survey was here conducted to assess and compare the impact of vitamin D receptor gene polymorphisms Fok1, Bsm1, Taq1, Apa1 and poly (A) on development of breast cancer. Information was obtained from electronic databases including PubMed and Google Scholar for articles published during the period from 1996 to 2015. This search was achieved by using the terms "genetics", "breast cancer", "VDR gene", "polymorphisms". However, due to inconsistent results, no conclusive statements could be presented about the significance of the VDR genotype as far as the development of breast carcinoma is concerned.

A Study on the Analysis System of Voyage Data Recorder

  • According to SOLAS Convention, a voyage data recorder(VDR) is to be fitted onboard ships to assist in the marine casualty investigation However, a review of Korean Maritime Safety tribunal(KMST), shows that the current VDR systems is inconvenient in practical because the storage format of VDR and the casualty reproduction method are different from one manufacturer to another making it economically and timely difficult for the maritime casualty investigation bodies to carry out investigation in a proper manner. To solve this problem, this paper proposed a newly designed VDR Analysis System(VDRAS), which decodes the voyage data of VDR producea by different manufacturers, reproduces the condition at the time of casualty in an accurate, and performs necessary checks.

  • VDR은 IMO의 협약에 따라 요구되는 모든 선박의 데이터를 기록하는 장비이다. 2002년 7월 1일 이후 건조되는여객선과 3,000톤 이상의 여객선을 제외한 모든 선박을 VDR를 탑재하여야 한다. 이것은 항공기의 블랙박스와 같이 사고조사를 가능하게 한다. 이 논문에서는 VDR을 이용한 선박의 거동 특성에 대하여 실선 M/S WS 선박을 대상으로 실시하기로 한다. 이 선박은 2014년 1월 여천에 있는 GS 칼텍스 터미널 부두와 충돌하여 상당량의 기름을 유출시킨 바 있다. 이 논문에서는 어느 쪽 타각을 더 많이 사용하였는지를 알기 위하기 위하여 타각면적을 도입하였다 그 외에 평균 선수방위, 평균속력, 평균 rpm 등을 사용하였다. 이 결과 이 선박은 우현타각을 많이 사용하는 것으로 나타났고 이것은 이 선박이 좌선회하는 경향이 있음을 확인할 수 있다.

Inhibition of Vitamin D Receptor Translocation by Cigarette Smoking Extracts

  • Background: Vitamin D can translocate a vitamin D receptor (VDR) from the nucleus to the cell membranes. The meaning of this translocation is not elucidated in terms of a role in pathogenesis of chronic obstructive pulmonary disease (COPD) till now. VDR deficient mice are prone to develop emphysema, suggesting that abnormal function of VDR might influence a generation of COPD. The blood levels of vitamin D have known to be well correlated with that of lung function in patients with COPD, and smoking is the most important risk factor in development of COPD. This study was performed to investigate whether cigarette smoke extracts (CSE) can inhibit the translocation of VDR and whether mitogen activated protein kinases (MAPKs) are involved in this inhibition. Methods: Human alveolar basal epithelial cell line (A549) was used in this study. 1,25-$(OH_2)D_3$ and/or MAPKs inhibitors and antioxidants were pre-incubated before stimulation with 10% CSE, and then nucleus and microsomal proteins were extracted for a Western blot of VDR. Results: Five minutes treatment of 1,25-(OH2)D3 induced translocation of VDR from nucleus to microsomes by a dose-dependent manner. CSE inhibited 1,25-$(OH_2)D_3$-induced translocation of VDR in both concentrations of 10% and 20%. All MAPKs inhibitors did not suppress the inhibitory effects of CSE on the 1,25-$(OH_2)D_3$-induced translocation of VDR. Quercetin suppressed the inhibitory effects of CSE on the 1,25-$(OH_2)D_3$-induced translocation of VDR, but not in n-acetylcysteine. Conclusion: CSE has an ability to inhibit vitamin D-induced VDR translocation, but MAPKs are not involved in this inhibition.

Vitamin D Receptor Gene TaqI, BsmI and FokI Polymorphisms in Korean Patients with Tuberculosis

  • Background: The active metabolite (1, 25- dihydroxycholecalciferol) of vitamin D (25-hydroxycholecalciferol) leads to activation of macrophages and deficiency of vitamin D seems to be involved in the risk of tuberculosis. The effects of vitamin D are exerted by interaction with the vitamin D receptor (VDR) and may be influenced by polymorphism in the VDR gene. In this study, variation in the VDR gene was investigated in Korean population with tuberculosis. Methods: We typed three VDR polymorphisms of restriction endonuclease sites for TaqI, BsmI and FokI in 155 patients with tuberculosis and 105 healthy volunteers. Results: The frequencies of FokI genotypes determined from TB patients were 29.13% for FF, 56.31% for Ff, and 14.56% for ff. We observed 1.4-fold increased prevalence of the Ff genotype in TB patients compared with normal healthy groups (p=0.0857). However, there was no significant association between the genotype groups, TB patient and normal control, for FokI polymorphism. There was also no significant association between VDR gene and tuberculosis in another polymorphism (BsmI and TaqI). Conclusion: Three polymorphisms (TaqI, BsmI and FokI) in the VDR gene do not appear to be responsible for host susceptibility to human tuberculosis in Korean population.

Association of Vitamin D Receptor Gene Polymorphisms with Prostate Cancer Risk in the Pakistani Population

  • Background: Vitamin D receptor (VDR) gene has been a subject of extensive pharmacogenetic research recently. Association studies between different types of cancers including prostate cancer (PCa) and VDR gene polymorphism have also been conducted. The objective of this study was to find possible associations between PCa and VDR gene polymorphisms in the Pakistani population. Materials and Methods: A total of 162 subjects, including prostate cancer patients and controls, were genotyped for Apa I, Taq I and Fok I polymorphisms in the VDR gene using allele specific PCR, PCR-RFLP and direct DNA sequencing. Allelic frequencies were tested for Hardy-Weinberg equilibrium and associations between the genetic markers and PCa were calculated using logistic regression. Results: Apa I CC genotype was found to have strongest association with PCa risk, and "A" genotype was found to have protective effect. Fok I and Taq I did not have appreciable levels of association with PCa, although Taq I "TC" heterozygotes seemed to have some protective effect. Similarly the "C" allele of Fok I also seemed to have protective effect. Conclusions: To our knowledge, this is the first report showing association between VDR gene polymorphisms and PCa in Pakistan. Our findings may be somewhat skewed because of small sample size and tendency of consanguineous marriages in Pakistani society; nevertheless, it shows the trend of association and protective effects of certain VDR gene polymorphisms against PCa.

사고 분석 사례 - Hyde Park, Cast Prosperity 충돌 사고 -

  • 2005년 9월 26일 컨테이너선 Cast Prosperity 호와 탱커선 Hyde Park은 Quebec 주 Trois-Riveres 로부터 남서쪽으로 12마일 떨어진 Lac Saint-Pierre 의 Channel 을 통과하던 중 충돌한다. Cast Prosperity 호는 VDR(Vessel Data Recorder)를 탑재하고 있었고 이 VDR의 기록과 양 선박의 AIS 기록을 토대로 사고를 재현하게 된다. 또한, 선주사로부터 제공된 자료를 바탕으로 사고 선박의 Database 및 사고 해역의 Database를 제작하여 ECS 의 항해 기록 및 각종 기상 Data 를 활용하여 사고 당시 선박의 항해과정을 3차원 영상으로 재현하여 사고의 원인을 규명한 사건이다.

  • The voyage data is very important for safety of ships, and duty being effectuated by the installation of voyage data record (VDR) on ships. VDR is a black-box, and it contains 14 kinds of voyage data as text. But it is not easy to understand when the accident happened because voyage data is saved as complicated texts. User interface (UI), analysis, visualization system, which works for assist to gather information about situation of accident, was developed. It will be possible to develop onboard ship monitoring and voyage prediction system by the VDR visualization system's development in near future.

VDL Mode-2 물리 계층 설계 및 성능 분석 (The Design and Performance Analysis of Physical Layer for VDL Mode-2)

  • 본 논문에서는 차세대 항공 데이터 통신에 사용되는 VDR(VHF Data Radio)의 물리계층을 국제규격에 만족하도록 설계하였다. VDR의 주파수 대역은 117.975~137MHz이고, CSMA(Carrier Sense Multiple Access) 다중 접속방식, D8PSK(Differential Eight Phase Shift Keyed), 25kHz의 채널 대역폭을 사용한다. VDR의 성능 요구 조건인 인접 채널 분리, 수신감도, 혼변조, 수신기의 동작영역, 송신기의 선형성, 불요 방사 전력을 선형 및 비선형 시뮬레이션을 통해 분석 하고, 제작을 위한 방향을 제시하였다. 설계 결과 송신부는 Po1dB보다 5dB 낮은 점에서 사용해야하고, 중간 주파수를 45MHz 선정하여 스퓨리어스 발생을 억제해야한다. 수신부는 잡음지수 4.5dB, Es/No 27.52dB로 설계 되었고, 혼변조 성분을 최소화하기위해 저 잡음 증폭기의 IIP3를 +10dBm 이상을 사용하고, 혼합기의 절연특성이 30dB이상이 되어야한다.
