  • This study was to investigate water quality and biotic community characteristics in Juam Lake. In water quality, water temperatures was $3.8{\sim}21.2^{\circ}C$, 6.7~8.6 in pH, $64{\sim}76{\mu}s\;cm^{-1}$ in Conductivity, $5.3{\sim}13.2mg\;L^{-1}$ in DO, $2.5{\sim}3.3mg\;L^{-1}$ in COD, $1.0{\sim}5.1mg\;L^{-1}$ in SS, $0.622{\sim}0.841mg\;L^{-1}$ in T-N, $0.007{\sim}0.019mg\;L^{-1}$ in T-P and $2.8{\sim}8.8mg\;m^{-3}$ in Chl-a. Revised Carlson's Index (TSIm) assessment using total phosphorus (TP) and chlorophyll-a domonstrated that the trophic states of Juam Lake were rated as mesotrophic. A total of 53 species of phytoplankton were identified. They were 28 Bacillariophyceae, 13 Chlorophyceae, 3 Cyanophyceae, and 9 Other algal taxa. The standing crops of phytoplankton was ranged from $113cells\;mL^{-1}$ to $2,909cells\;mL^{-1}$. A total of 16 species of zooplankton were identified (10 rotifers, 4 cladocerans and 2 copepods). Total zooplankton abundance was $309ind.\;L^{-1}$ to $435ind.\;L^{-1}$. The collected benthic macroinvertebrates from the surveyed sites in Juam Lake were 1,038 individuals, 33 species, 21 families and 12 orders. A dominant species was Uracanthella rufa and a subdominant species was Ecdyonurus kibunensis. Hydrophytes recorded from Juam Lake were identified 9 taxa. Emerged plants, floating plants among the hydrophytes was classified 8, 1 taxa, respectively. Ecosystem disturbance wildplant by Environment Ministry found were Paspalum distichum var. distichum and Ambrosia artemisiaefolia. A total of 30 species (6 families) were collected fishs from Juam Lake. There were 10 Korean endemic species (33.3% of collected species number) and 3 exotic species (10.0%).

  • To investigate the oviposition preference and development of azuki bean weevil (Callosobruchus chinensis L.), the following six different leguminous seeds were used in this study: red bean (Vigna angularis (Willd.) Ohwi & Ohashi), black soybean (Glycine max (L.) Merr.), soybean (Glycine max (L.) Merr.), seoritae (Glycine max (L.) Merr.), small black bean (Rhynchosia nulubilis) and kidney bean (Phaseolus vulgaris var. humilis Alef.). In the study of oviposition preference, the numbers of eggs per leguminous seed on red bean, black soybean, soybean, seoritae and small black bean were 1.23, 0.61, 0.69, 1.05 and 1.13, respectively. The maximum daily number of eggs was observed at 48 hours and the minimum was at 96 hours. According to each host leguminous seed, developmental time for each host seed was different. The shortest adult emergence time was on red bean (25.27 days). The other five leguminous seeds increased or doubled the adult emergence time. Adult emergence rates feeding on red bean, seoritae, black soybean, soybean, small black bean were 83.33%, 28.23%, 27.87%, 20.44%, and 11.59%, respectively. Emergence rate on red bean was four times higher than the rate on other seeds. The longevity of emerged female adults was almost all longer than that of males. The male adults weighed the lowest of feeding on small black bean. Female adults weighed the lowest of feeding on soybean. Adult weights were the heaviest for both males and females feeding on red bean. As a result, hosts of azuki bean weevil could decrease oviposition rate, emergence rate, adult longevity, and adult weight but increase emergence time. Especially in kidney bean, adult was not completely emerged. No eggs were laid. These results suggest that there might be emergence inhibitors in kidney bean. These imformation might be used to control damages caused by azuki bean weevils.

  • In order to clarify the ecophysiological characteristics of Chenopodiaceae which widely distribute on saline and arid habitats, we collected 10 chenopodiaceous plant species, examined their inorganic and organic solute patterns, and confirmed several common physiological characteristics. In spite of high soil Ca/sup 2+/ contents, chenopodiaceous plants had a little water-soluble Ca within cells, but contained high contents of acid-soluble Ca particularly as a result of Ca-oxalate formation. These plant species also showed accumulation of inorganic ions such as K/sup +/, NO₃/sup -/ and Cl/sup -/, and Na/sup +/especially in saline habitats instead of K/sup +/ Meanwhile, with respect to nitrogen metabolism they retained high N contents in leaves, but showed very low amino acid contents. Additionally, they contained very little proline known to act as a cytoplasmic osmolyte. To ascertain whether this physiological characteristics in the field also can be found under controlled conditions, 7 chenopodiaceous plants (Atriplex gmelini, Corispermum stauntonii, Salicornia herbacea, Suaeda aspayagoides, Suaeda japonica, Chenopodium album var. centrorubrum, C. serotinum) were selected and cultivated under salt treatments. As well as field-grown plants, selected plant species showed similar solute pattern in growth experiment. In summary, the family of Chenopodiaceae represents the following physiological properties; high storage capacity for inorganic ions (especially alkali cations, nitrate and chloride), oxalate synthesis to maintain lower soluble Ca contents within cytoplasm, and low contents of amino acids. In addition to some characteristics mentioned above, the physiological plasticities of Chenopodiaceae which can properly regulate their ion and solute pattern according to soil conditions may enable its representative to grow in dry sand dune and salt marsh habitats.

  • The over-expression in E. coli of the pHLN1-SO(+) and pHLN2-80(-) plasmids cloned an insecticidal crystal protein (ICP) gene (crylAal type) from Bacillus thuringiensis var. kurstaki HD 1 was investigated through in part, the deletion of -80 bp promoter and an alternative change of cloning vector system. Two recombinant plasmids were constructed in an attempt to analyze the over-expression of the ICP in relations to its gene structure possessing only -14 bp [Shine-Dalgarno (SD) sequence of -80 bp promoter]. Also, anther two recombinant plasmids similarly cloned the icp gene in a different vector system. The amounts of ICP produced from the recombinants were measured by SDS-PAGE and confirmed by Western blot analysis. One clone, pHLRBS1-14 clone in which only the SD sequence in the inverted orientation icp gene appeared, was more evident than the pHLRBS2-14 clone in which only the -14 bp SD sequence of the right orientated icp gene was shown to exist. The pHLN2-80(-) clone produced more ICP proteins than the pHLRBS1-14 clone. In the two clones, pHLNUC1-80 right-oriented icp gene and the pHLNUC2-80 clone inverted-orientation icp gene in a new different vector, the pHLNUC2-80 produced more ICP proteins in E. coli system. These results indicate that the P/ac promoter, the inverted icp gene insertion and -80 bp promoter (-66 bp part of the icp gene promoters), were concerned with the expression of the icp gene in the recombinant plasmids. In addition, the expression mechanism might result from the disruption of the transcription-suppressing regions in the promoter regions.

  • This study was conducted to compare the regeneration of the seedlings under different vegetation types and to identify the presence of allelopathy in Abies koreana(Ak) natural forest in Banyabong Peak(elevation, 1715m), of Mt. Chiri. Twenty quadrats($10m{\times}10m$) were placed in May, 1996 to classify vegetation structure using TWINSPAN. Water-soluble extracts from leaves and soil humus of different vegetation types were collected to test their effects on both seed germination of Ak and mycelial growth of ectomycorrhizal fungi. Phenolic compounds from soil humus were quantified using HPLC. Among the four vegetation types, Sasa borealis(Sb) was found in both Ak-Quercus mongolica(Qm) and Ak-Rhododendron schlippenbachii(Rs) communities. Natural seeding of Ak was $230,000{\pm}90,000seeds/ha$ in 1995 and their germination rate was 25% in an ideal laboratory condition. Density of Ak seedlings less than 5cm in height was 52,000/ha in 1996, while that of seedlings taller than 5cm in height was only 4,000/ha. In the case of Ak-Qm community, density of Ak seedlings with Sb understory was only 7% of the density of seedlings with Rs understary, suggesting the inhibitory effect of Sb. The germination rate of Ak seeds was significantly reduced by leaf extracts of Sb, and Rhododendron mucronulatum var. ciliatum(Rm) and Ak. Soil humus extract of Ak-Qm-Sb subcommunity reduced germination of Ak seeds by 81% and also reduced by 19% the respiration of mycelia of ectomycorrhizal fumgus, Lactarius sp. Among the seven phenolic compounds identified from the soil humus, extract, Ak-Qm-Sb subcommunity contained significantly high content of p-hydroxybenzoic acid, vanillic acid, and syringic acid. Particularly, p-hydroxybenzoic acid was present at 4.2ppm in the Sb roots and at 16.5ppm in the Sb humus, suggesting that it could be the primary allelopathic compound in Abies koreana forests with Sasa borealis understory.

  • This study traces the development of physic gardens in Korea and explores their significance in the history of landscape architecture. For this purpose, records related to physic gardens from medical sources from the period of the Three States to the Joseon dynasty, when herbal medicine was systematized as a field, were searched. Physic gardens had been developed by the time of the late Goryeo and early Joseon dynasties, in the 13th and the 15th centuries. Yakpo(kitchen gardens for medicinal herbs) were cultivated by a group of new high-level officials in the late Goryeo dynasty, when an increasing interest in hyangyak(native herbs) emerged under the influence of the Neo-Confucian perspective on nature, which emphasized locality. The sources analyzed in this study confirm that physic gardens called jong-yakjeon(royal medicinal herb gardens) were in operation in the early Joseon dynasty when policies to investigate, discover, cultivate, and research native herbs were put into place. It is likely that the jong-yakjeon were established at the beginning of the Joseon dynasty as subsidiary facilities under its central medical institutions, the Naeuiwon and Hyeminseo, and then declined in the late Joseon dynasty. Jong-yakjeon can be confirmed to have existed in the mid-15th century. Physic gardens were located in several places outside the Fortress Wall of Hanyang, such as Yakhyeon, Yuldo, Yeoudo, and Saari. The total area encompassed by physic gardens was about 160,000 square meters in the early 18th century. In jong-yakjeon, dozens of medicinal herbs were cultivated, including Schizonepeta tenuifolia var. japonica, Rehmannia glutinosa, and Glycyrrhiza uralensis Fischer, and these gardens were operated by physicians dispatched from the Naeuiwon and dozens of provincial slaves. In conclusion, the jong-yakjeon were similar to the physic gardens of Renaissance medical universities in that they reflected the interest in and development of theories about new herbs, and were similar to the physic gardens of medieval castles and monasteries in terms of species types, location, and function. This paper has limitations in that it does not present the specific spatial forms of the yakpo or the jong-yakjeon. Nevertheless, this paper is significant for the field of garden history because it shows that physic gardens in Korea appeared in the late Goryeo and early Joseon dynasties concomitantly with the development of medicine towards native herbs and functioned as utilitarian gardens to cultivate community remedies.

  • This study was conducted to investigate the effect of climate conditions during cultivation and harvesting on the quality and storability of fresh bulb cabbage (Brassica oleracea L. var. capitata). Plug seedlings of six cabbage cultivars were transplanted to Gangneung-Wonju University high elevation research station in Gangwon province (780 m above sea level, lat. $37.5^{\circ}N$.) and harvested with four different harvest times like August 3 ($1^{st}$), August 13 ($2^{nd}$), August 23 ($3^{rd}$), and September 10 ($4^{th}$), respectively from 50 days after transplanting. Weight loss, Hunter color factors, firmness, and soluble solids content (SSC) of the cabbage bulbs were investigated during storage at $3^{\circ}C$ (85% RH) and $25^{\circ}C$ (60% RH). Decreased bulb weight and poor quality cabbages were apparent at the late transplanting (July 14) and harvest (September 10) respectively. Quality index such as firmness and SSC at August 23 ($3^{rd}$) harvested cabbage was better than August 3 ($1^{st}$) and August 13 ($2^{nd}$) cabbages due to the good weather condition just before harvesting. The cv. 'Speed king' and 'Minix 40' showed good qualities among the cultivars, especially when the bulbs were harvested during sunny day conditions from one week before harvesting. Also SSC was influenced by weather condition before harvesting rather than transplanting date, while firmness was influenced by transplanting and harvest date. However, the differences among the cultivars were not significant. The potential of storage as maintaining the quality was different, depending on weather conditions at harvest time. Generally the storage periods of six cultivars were around 3~5 days and 9~10 days at room and low temperature, respectively. However, the August 3 ($1^{st}$) harvested cabbage lost their marketable quality very fast because of rainy and cloudy weather condition before harvesting and also storability of bulbs was 2 days and 4 days at room temperature and $3^{\circ}C$, respectively. Quality index was also not significant difference among cultivars.

  • The contents of chemical components such as total nitrogen, total amino acid and vitamin C were somewhat higher in $N_2$ gas treatment at $10^{\circ}C$ for 3 hours than those of other treatments. However, the contents of tannin and chlorophyll were slightly lower than that in the other treatments. The content of ${\gamma}-Aminobutyric$ acid with $N_2$ gas treatment was higher $1.5{\sim}6$ times with values of $51{\sim}205mg/100g$ than in control (35mg/100g). The scores of sensory test was not different between $N_2$ gas treatment for 3 hours and control. The contents of chemical components such as total nitrogen, total amino acid including theanine and caffeine were slightly higher in $N_2$ gas treatment at $20^{\circ}C$ for 3 hours than those of other treatments. However, the contents of tannin and vitamin C were slightly lower than those of other treatment. The content of GABA in tea leaves treated with $N_2$ gas was higher $2.5{\sim}7$ times with values of $85{\sim}225mg/100g$ than in control (35mg/100g). The sensory test was lower in $N_2$ gas treatment($76.3{\sim}78.1$ point) than in control(80.4 point). The contents of chemical components were not different between $N_2$ gas treatment at $30^{\circ}C$ for 3 hours and control. Whereas the contents of chemical components were somewhat lower in $N_2$ gas treatment for 1 hour and 5 hours than in control. The content of GABA in tea leaves treated with $N_2$ gas was higher $3{\sim}7$ times with values of $115{\sim}217mg/100g $than in control(35mg/100g). The sensory test was lower in $N_2$ gas treatment ($74.3{\sim}78.4$ point) than in control(80.4 point). Consequently, tea mading within 5 time $N_2$ gas treatment at $10^{\circ}C$ or 3 time $N_2$ gas treatment at 20, $30^{\circ}C$ after plucking was considered to be the best green tea in terms of functional nature as well as taste nature.

  • The flora of the vascular hydrophytes and hygrophytes and the change of flora according to the fluctuation of the water level before and after flooding were investigated from September 2002 to October 2003 in Woopo Wetland and Topyeong Stream, Changnyeong-gun, Gyeongsangnam-do, Korea. The flora of the Woopo, Mokpo, Sajipo, Jokjibyeol, Topyeongcheon Upstream, and Topyeongcheon Downstream were composed of 256, 242, 265, 177, 201, and 180 taxa, respectively. The flora of total study area was 86 families, 232 genera, 302 species, 42 varieties, 6 form, or total 350 taxa. Among them, hydrophytes, hygrophytes, and others were 36, 96, and 218 taxa, respectively. The life form of the vascular hydrophytes was classified as 19 taxa of emergent plants, 6 taxa of floating-leaved plants, 4 taxa of free-floating plants, and 7 taxa of submersed plants. The specified wild plants designated by the Korean Association for Conservation of Nature, Ministry of Forest, and Ministry of Environment were Euryale ferox, Hydrocharis dubia, Persicaria amphibia, Acorus calamus var. angustatus. And Aristolochia contorta was distributed in the study area. The floras of the naturalized plants of the Woopo, Mokpo, Sajipo, Jokjibyeol, Topyeongcheon Upstream, and Topyeongcheon Downstream were composed of 27, 25, 35, 21, 26 and 24 taxa, respectively. The flora of the naturalized plants of total study area was 43 taxa, this equalled 12.3% of total vascular plants in the study area. And 165~244 taxa were surveyed in the each study area before flooding, total 299 taxa, and 86~212 taxa after flooding, total 299 taxa. Among them, hydrophytes, hygrophytes, and others were 33 and 29, 93 and 83, 183 and 187 taxa before and after flooding, respectively. Before and after flooding, the species number of emergent plants, floating-leaved plants, free-floating plants, and submersed plants was nearly the same. It was guessed that vascular hydrophytes and hygrophytes adapted to fluctuation of the water level in the study area.
