Fed-Batch Fermentation of High-Content RNA Yeast by Using Molasses Medium. (당밀 배지를 이용한 고함량 RNA효모의 유가배양)

  • In order to maximize the RNA accumulation and biomass production is Saccharomyces cerevisiae MTY62, a high-content RNA yeast strain, batch and fed-batch cultures were performed. Among the feeding modes of fed-batch cultures examined, the intermittent feeding mode R\`(IFB-lV), in which 50 ml of 40% molasses and 20% com steep liquor (CSL) solution was intermittently fed for 5 times, resulted in the cell concentration of 33.8 g- dry cell weight/1 and the RNA concentration of 5221 mg-/l, and RNA content of 153 mg-RNA/g-dry cell weight. The constant fed-batch with feeding mode III (CFB-III), in which the feeding rate of 40% molasses and 20% CSL solution was stepwisely decreased from 48 mph (9-13 h), to 24 mph (13-21 h), and to 18 ml/h (21∼ 48 h), gave the highest cell concentration of 42.7 g-dry ceil weigh71 and R7IA concentration of 5536 mg-RNA/1, which were about 2.4-fold and 1.9-fold increased levels, respectively, compared to the results of batch culture. However, the RNA con- tent of 130 mg-RNA/g-dry cell weight of the fed-batch was lower than that of the batch culture (171 mg-RNA/g-dry cell weight) and other fed-batch cultures. When the specific growth rates in the fed-batch cultures were increased, the RNA contents increased. This result indicates that the RNA content is adversely proportional to the cell concen- tration. However, at the same specific growth rate, the RNA content was maintained at higher level in the intermit- tent fed-batch than in the constant fed-batch culture.

Change of Somatosensory Evoked Field Potential according to the Severity of Hydrocephalus in Kaolin-induced Hydrocephalus of Rats (수두증 흰쥐 모델에서 수두증 정도에 따른 체성 감각 유발 장전위의 변화)

  • Objective : Somatosensory evoked potential(SSEP) has been known to be a good method for evaluating brain stem function, but it is not sufficient to check the fine changes of cortical functions. A fine change of cortical function can be expressed with somatosensory evoked cortical field potential(SSEFP) rather than general SSEP. To confirm the usefulness of SSEFP for evaluating the cortical function, the authors simultaneously measured SSEFP and the intracranial pressure-volume index(PVI) in kaolin-induced hydrocephalic rats. Method : Hydrocephalus was induced with injection of 0.1ml kaolin-suspended solution into the cisterna magna in 60 Sprague-Dawley rats. The authors measured PVI and SSEFP 1 week after injection of kaolin-suspended solution. To evaluate the severity of induced hydrocephalus, we measured the transverse diameter of the lateral ventricle on the coronal slice of the rat brain 0.40mm posterior to the bregma. Result : The typical wave form of SSEFP in control rats showed a negative-positive complex wave at early latency. In SSEFP of normal rats, N0 is 10.0 msec, N1 15.3 msec, P1 31.2 msec and N1-P1 amplitude $15.4{\mu}V$. As hydrocephalus progressed, the peak latency of N1 and P1 were delayed. In mild hydrocephalus, negative peak waves were split. The N1-P1 amplitude was decreased only in severe hydrocephalus. The changes of the characteristics of SSEFP according to the severity of hydrocephalus were well correlated with the changes of PVI. Shunting normalized the characteristics of SSEFP in relation to ventricular sizes and PVI in hydrocephalic rats. Conclusion : SSEFP may be useful for evaluating the impairment of cortical function in hydrocephalus.

  • The conventional pattern-methods for discrimination of acupoint, meridian line which is the basic object of diagnosis and medical treatment in oriental medicine is discriminated the conduction point by the stimulation in body skin with DC. But, it is not sufficient to truth in discrimination ratio, coincident ratio, body effect, reproductivity. Therefore, this paper is extracting the optimal parameter of frequency and waveform in order to improve the conventional pattern, and proposing the SPAC(Single Power Alternative Current) stimulus pattern applying that. Also, this algorithm proposes to be able to discriminate with low pressure of the electrode by displaying in the level meter both the absolution and relation value of the skin current. It is able to decrease pain and body effect by electrode pressure and discriminate acupoint regardless of skin current in difficult discrimination spot. It is compared the performance of system applying the extracted optimal parameter and algorithm, and it is confirmed that there is difference in discrimination parameter of acupoint reacted to the individual and the meridian. It is compared that discrimination, coincident ratio of the traditional acupoints as the acupoint stimulation pattern. It is confirmed truth of optimal parameter and discrimination algorithm.   • Methyl alcohol is one of the basic intermediates in the chemical industry and is also being used as a fuel additive and as a clean burning fuel. In this study, conversion of carbon dioxide to methyl alcohol was investigated using catalytic chemical methods. Ceramic monoliths (M) with $400cell/in^2$ were used as catalyst supports. Monolith-supported CuO-ZnO catalysts were prepared by wash-coat method. The prepared catalysts were characterized by using ICP analysis, TEM images and XRD patterns. The catalytic activity for carbon dioxide hydrogenation to methyl alcohol was investigated using a flow-type reactor under various reaction temperature, pressure and contact time. In the preparation of monolith-supported CuO-ZnO catalysts by wash-coat method, proper concentration of precursors solution was 25.7% (w/v). The mixed crystal of CuO and ZnO was well supported on monolith. And it was known that more CuO component may be supported than ZnO component. Conversion of carbon dioxide was increased with increasing reaction temperature, but methyl alcohol selectivity was decreased. Optimum reaction temperature was about $250^{\circ}C$ under 20 atm because of the reverse water gas shift reaction. Maximum yield of methyl alcohol over CuO-ZnO/M catalyst was 5.1 mol% at $250^{\circ}C$ and 20 atm.

  • Effects of methionine on cephalosporin C(CPC) production in a fluidized-bed bioreactor were investigated using bioparticles of Cephalosporium acremonium. Since methionine was found to be an important metabolic regulator on the synthesis of cephalosporin C, the effects of its concentration in the cuture broth and feeding mode to the bioreactor were studied. It was observed that the presence of initial methionine was essential for higher cephalosporin C production and there existed an optimal content of methionine. Carbon consumption rate also increased significantly under the presence of methionine. Production of cephalosporin C was most active when methionine was exhausted in the broth; however its additional feeding did not enhance the antibiotic production in the fluidized-bed bioreactor as much as expected. It was therfore considered important to feed an optimal content of methionine at the early operating stage for a higher cephalosporin C production in a fluidized-bed bioreactor. An interesting thing to note was that titre of the antibiotic with reused bioparticles was about 2 times higher in the methionine containing medium than that without methionine. Therefore repeated use of bioparticles, with an optimal content of methionine, was believed to be very useful to enhance to process productivity.

  • Modeling urban climate caused by land use conversion is critical for human welfare and sustainable development, but has hampered because detailed information on urban characteristics is hard to obtain. With the advantage of satellite observations and the new statistical boundary system, this paper measures the economic and environmental effects of green area loss due to land use conversion in urban areas. To perform this purpose, data were collected from the various sources basic statistical unit data from the National Statistical Office, digital maps from the National Geographic Information Institute, satellite images, and field surveys when necessary. All data (maps and attributes) are built into the geographic information system (GIS). This paper also utilizes Landsat TM 5 imagery of Daegu city to derive vegetation index and to measure average surface temperature. The satellite data were examined using standard image processing software, ERDAS IMAGINE, and the results of the digital processing were presented with ARCVIEW(v.3.3). SAS package was used to perform statistical analyses. This study presents that there exists a strong relationship between land use change and climatic change as well as land price change. Based on results of the analysis, this paper suggests that planners should implement effective tools and policies of urban growth management to detect environmental quality and to make right decisions on policies concerning smart urban growth.

  • Inferior alveolar nerve dysfunction may be the result of trauma, disease, or iatrogenic injury. Inferior alveolar nerve injury is inherent risk in endodontic therapy, orthognathic surgery of the mandible, and extraction of mandibular teeth, particularly the third molars. The sensory disturbances of inferior alveolar nerve associated with such injury have been well documented clinical problem that is commonly evaluated by several clinical sensory test including Tinels sign, Von Frey test(static light touch detection), directional discrimination, two-point discrimination, pin pressure nociceptive discrimination, and thermal test. These methods used to detect and assess inferior alveolar nerve injury have been subjective in nature, relying on the cooperation of the patients. In addition, many of these techniques are sensitive to differences in the examiners experience and skill with the particular technique. Data obtained at different times or by different examiners are therefore difficult to compare. Prior experimental studies have used electro diagnostic methods(sensory evoked potential) to objectively evaluate inferior alveolar nerve after nerve injury. This study was designed with inferior alveolar nerve of rabbit. Several types of injury including mind, moderate, severe compression and perforation with 19 gauze, 21 gauze needle and 6mm, 10mm traction were applied for taking the sesory evoked ppterntial. Latency and amplitude of injury rabbit inferior alveolar nerve were investigated with sensory evoked potential using unpaired t-test. The results were as follows : 1. Intensity of threshold (T1) was $128{\pm}16{\mu}A$ : latency, $0.87{\pm}0.07$ microsecond : amplitude, $0.4{\pm}0.1{\mu}V$ : conduction velocity, 23.3 m/s in sensory evoked potential of uninjured rabbit inferior alveolar nerve. 2. Rabbit inferior alveolar nerve consists of type II and III sensory nerve fiber. 3. Latency was increased and amplitude was decreased in compression injury. The more injured, the more changed in latency and amplitude. 4. Findings in perforation injury was similar to compression injury. Waveform for sensory evoked potential improved by increasing postinjured time. 5. Increasing latency was prominent in traction injury rabbit inferior alveolar nerve. 6. In microscopic histopathological findings, significant degeneration and disorganization of the internal architecture were seen in nerve facicle of severe compression and 10mm traction group. From the above findings, electrophysiological assessment(sensory evoked potential) of rabbit injured inferior alveolar nerve is reliable technique in diagnosis and prognosis of nerve injury.

A Study on the Quantitative Analysis for the Forest Landscape (삼림경관에 관한 계량적 분석에 관한 연구)

  • The purpose of this thesis is to suggest objective basic data for the environmental design through the quantitative analysis of the visual quality included in the physical environment of forest landscape. For this, landscape values of forest landscape have been evaluated by using the Iverson method, the images structure of forest landscape's main utilizing space have been analysed by the factor analysis algorithm, degree of visual preferences have been pleasured mainly by questionnaries and SBE method, and finally these thesis can be summarized as fallow LCP with high values of Iverson factors I and IV yield high landscape value. Specifically, Iverson factor IV has been found to play the dominant. For all experimental points, significant seasonal variations in S.D. scale values have been observed. In natural parks, where artificial structures are complementary to the natural landscape, main factors of image are S.D. scales such as the visual sequence, the formal simplicity of structures, the emphasis, the unification of heterogeneous factors and the assimilation. Factors covering the spatial image of natural parks have been found to be the overall evaluation, the individual characteristics, the tidiness, the potentiality, the dignity, the intimacy and the space volume. For all seasons, factors such as the individual characteristics, the dignity, the tidiness, the potentiality, yield high factor scores. As for factors determining the degree of visual preference, variables such as the summit, the skyline, rocks, the water and the degree of natural destruction by artificial structures yield high values for all seasons.

Economic Cooperation between Russia and Republic of Korea;Problems and Prospects (러-한 경제협력;과제와 전망)

  • 러시아와 한국간의 공식적인 접촉은 1990년 6월 샌프란시스코에서 개최된 정상회담이다. 동년 9월 뉴욕에서 개최된 양국의 외무장관 회담에서 러시아와 한국의 외교관계가 수립되었다. 그간 양국간에는 협정을 통한 협력의 법적기반이 조성되었는데 주요 분야를 보면 무역, 투자보장, 어업, 이중과세방지, 군사기술분야, 원자력의 평화적 이용, 문화교류에 관한 협정들이 있다. 역시 양국간의 주요 관심사항은 무역과 경제협력 분야이다. 2007년도 러시아와 한국의 무역액은 150억 달러를 초과함으로써 전년도에 비해 55.5% 증가하였다. 러시아의 한국으로의 수출은 약 70억 달러로서 전년도에 비해 52.6% 증가했고 한국으로부터의 수입은 80억 달러를 초과함으로써 전년도에 비해 56.1% 증가하였다. 그리하여 러시아는 한국의 10대 교역국으로 성장하였다. 가스와 오일 프로젝트 추진과 관련하여 사할린지역에서 사용하기 위해 한국으로부터 수입하는 드릴기계장비, 탱커, 자동차, 휴대전화는 한국과 러시아 양국간 경제성장의 주요요인이 되고 있다. 2008년 1/4 분기 양국간 무역액은 42억 달러를 초과했는데 이는 전년도 동기와 비교해서 72.1% 증가한 것이다. 이 중 한국에의 수출은 17억 달러를 초과했는데 이는 전년도 1/4분기에 비해 91.7% 성장한 것이다. 한국으로부터의 수입은 약 25억 달러로서 전년도 1/4 분기와 대비해서 60.6% 성장했다. 러시아와 한국간의 경제교류가 크게 증대하게 된 배경에는 러-한경제과학기술협력공동위원회가 큰 역할을 담당했다. 이 논문에서 필자는 무역과 투자측면에서 본 양국간 경제협력의 특징을 고찰하는 한편 러시아의 동부지역(러시아 극동 및 바이칼횡단지역)과 한국과의 협력과 관련된 역동성과 과제를 제시하는데 연구의 목적을 두었다. 한국은 외국기업과 함께 러시아 극동지역의 무역과 경제협력 증진에 상당한 공헌을 함으로써 러시아 극동경제권에서 주요 무역파트너가 되고 있다. 또한 본 연구에서는 한국과 러시아간의 주요 협력프로젝트에 관하여 고찰하는 한편 앞으로의 전망을 제시하였다. 이 문제는 무엇보다도 한-러간의 공동 에너지프로젝트 및 에너지자원의 무역과 관련되어 있다. 한국과 러시아는 이미 많은 분야에서 상당한 정도로 상호 협력관계를 발전시켜왔다. 여기에는 정책, 에너지, 경제, 문화, 과학기술 분야가 포함되어 있다. 현재 러시아와 한국간에는 에너지 분야에서의 전략적 대화채널 구축에 관한 문제와 함께 연료 및 에너지공단 건설과 관련하여 협력문제가 논의되고 있다. 에너지 분야에서의 전략적 대화개념 구축을 위해서는 전략, 예측 및 투자환경의 문제가 양국간에 집중논의 되어야 하고 법제상의 조화문제도 논의되어야 한다.

  • Death and dying of human being is a comprehensive system, and death orientation, the subjective meaning related to every component of the death system is developed throughout life. This study was designed and carried out to identify, describe and classify the orientations of Korean adult towards the death system. In an attempt to measure the subjective meaning of death and dying, unstructured Q-methodology was used. The 65 Q-statements developed by Kim(1994), used by Kim(1994) and Park(1996) were adopted as Q-population and 39 Q-statements were selected by the three researchers for Q-items for this study. Thirty-three P-samples were sampled from P-population of literate Korean men and women, 35 and 55 years of age, lived in urban Korea for the last 10 years. Sortings of the 39 Q-items according to the level of personal agreement, and a forced normal distribution into the 9 levels were carried out by the P-samples. The Z-scores of the Q-sort data were computed, and the principal components factor analysis by PC-QUANL Program were carried out. The demographic, socio-cultural and health-related attributes of the P-samples were descriptively analysed. Eight types of death orientation were identified ; Type I ; 'naturalist'. Six P-samples. Death is a natural phenomena, to be accepted as it is and to follow its natural course. Prefer to be informed of all facts and possibilities concernig the course of dying and death to occur to self. Type II ; 'life-after-life negator'. Three P-samples. Time and process of death is the destiny of each person. Death means 'darkness' and 'end to every thing, the absolute end'. Yet, wish physical integrity at the dying and after death. Type III ; 'life-after-life believer'. Six P-samples. Men are travellers passing by this life bound to the life-after-life. Priority concerns are on the activities to prepare self for the eternal life ahead. Disregard premature and sudden death. Type IV ; 'here-now believer' Five P-samples. Positive regard to the cremation of the body and donation of the organs on death. Regard religious and customary post-motem rituals meaningless. Negate life-after life. Type V; 'believer of rituals'. Five P-samples. Death being accepted as a part of, a natural end to, and destiny of human life. Concerned to ensure a dignified end to personal life and dignified post-mortem rituals. Type VI ; 'Realist'(derived from Type I). Two P-samples. Life and death as universal reality. The abrupt death at golden age at the peak of happiness is favored to avoid inevitable physical and mental distress of self and the family. Agreed to the cremation of the body. Disregard rituals. Type VII : 'Fatalist' (derived from Type II). Five P-samples. Not favored, yet, all man are destined to death, the inevitable end of all living beings. To ensure dignified end by personal consummation, information on one's dying and imminent death are to be shared. Type VIII ; 'reality avoider'(derived from Type III). One P-sample. Negative to longevity, artificial prolongation of, meaningless and distressful life. Highly positive to postmortem organ donation.
