Purpose: This study was conducted to investigate the effects of Ginseng Radix Alba extract solution on the cell cycle regulation, apoptosis of human uterine myoma cells. Methods: Primary cultured human uterine myoma cells were treated with extract solution of Ginseng Radix Alba concentration of 1, 10 and $100mg/m{\ell}$ for 48 hours. We underwent flow cytometry and western blotting for cell cycle and apoptosis related factors. Results: In flow cytometry, a slight promotion of G1 phase in $1mg/m{\ell}$ group had been observed but, simultaneously a delay of G1 phase in 10 and $100mg/m{\ell}$ groups were also observed. The manifestation of cyclin D1 in Ginseng Radix Alba extract solution medicated group increased compared to the control group. The manifestation of Bax controlling apoptosis increased in terms of concentration but Bcl-2 showed no change. Also VEGF increased in terms of concentration. Conclusion: The present study suggests that Ginseng Radix Alba extract solution treatment in human uterine myoma cells induce the delay of cell cycle and apoptosis. These results suggest that Ginseng Radix Alba will be a promising agent for use in therapeutics agents against human uterine myoma.
Objectives: The purpose of this study is to examine the effects of Sagunjatang-Gami(SGJT) on uterine and ovarian function in the ovariectomized rat postmenopause model. Methods: SGJT was administered in ovariectomized Wister albino female rats for three month. After that, uterine weight, uterine index, serum estradiol-$17{\beta}$ levels and phosphorylation of ERK or AKT, and histological analysis of uterus were measured to assess the impact on uterine and ovarian function in ovariectomized rats. In addition, phosphorylation of $ER{\alpha}$, ERK, AKT by SGJT in MDA-MB-231 cells were measured. To identify safety of SGJT, the cell cytoxicity in MDA-MB-231 cells and serum GOT, GPT levels were measured in ovariectomized rats. Results: The results were as follows. 1. SGJT decreased the viability of MDA-MB-231 cells in a dose-dependent manner. 2. The level of serum GOT, GPT in SGJT-treated group showed significant decrease in comparison with control group. 3. Phosphorylation of $ER{\alpha}$, ERK, AKT by SGJT in MDA-MB-231 cells were increased. 4. Uterus index in SGJT-treated group showed significant increase in comparison with control group. The level of serum estradiol-$17{\beta}$ in SGJT-treated group showed significant increase in comparison with control group. Phosphorylation of ERK or AKT by SGJT in the uterus of ovariectomized rats was increased significantly. 5. Uterus index and the level of serum estradiol-$17{\beta}$ in SGJT-treated group increased at higher rates in comparison with estrogen-treated group. Conclusions: Taken together, we suggest that SGJT has been shown to be effective in preventing postmenopausal uterine and ovarian degeneration and curing postmenopausal low estrogen related symptoms.
The significance of endometrial cells on cervicovaginal smears is underestimated. The aim of this study is to evaluate the detection rate of endometrial cells on cervicovaginal smears. The materials consisted of two groups. Group I was 701 cervicovaginal smears from patients with no gynecological problems. Group II was 208 cervicovaginal smears from patients with abnormal uterine bleeding followed by endometrial curettage; 31 cases of endometrial adenocarclnoma(CA), 19 cases of endometrial hyperplasia(HP), 83 cases of dysfunctional uterine bleeding(DUB), and 75 cases of normal endometrium. Cervicovaginal smears were reviewed according to the criteria of The Bethesda System. Endometrial cells were identified in 15 of 701 cases(2.1%) in group I and 64 of 208 cases(30.8%) in group II. Among group II, detection rate of endometrial cells was the highest in CA (51.6%) compared to HP(26.3%), DUB(41.0%), and normal endometrium(12.0%) (p<0.05). Cytologic atypia of endometrial cells was not found In group I, but was more frequently identified in CA(87.5%) than in HP(10.5%) or DUB(14.7%) (p<0.05). Exfollatlon of endometrial cells might be related to abnormal endometrial lesion, and reporting of endometrial cells in the cervicovaginal smear may increase a chance to detect endometrial lesions especially in patients with abnormal uterine bleeding.
Small cell neuroendocrine carcinoma of the uterine cervix is a distinct subtype of cervical cancer that appears analogous to oat cell carcinoma and carcinoid tumors of the lung. It has been assumed to be derived from the neural crest via argyrophilic cells in the normal endocervix. We have recently encountered a case of small cell neuroendocrine carcinoma of the uterine cervix coexisting with adenocarcinoma which was argyrophil negative. A 66-year-old multiparous woman was admitted because of vaginal bleeding for 2 months. Cervicovaginal smear revealed several scattered clusters and sheets of monotonous small cells with some peripheral palisading in the background of hemorrhage and necrosis. Radical hysterectomy specimen revealed an ulcerofungating tumor on endocervical canal which was composed of two components. Major component of the tumor was made up of monomorphic population of small oval-shaped tumor cells arranged in sheets and partly in acinar structures or trabecular fashion. Other component was adenocarcinoma, endocervical well-differentiated type. Argyrophilia was present on the Grimelius stain and immunohistochemical studies revealed diffuse positivity to neuron-specific enolase and carcinoembryonic antigen. Electron microscopic examination showed clusters of small round to oval cells, which had a few well-formed desmosomes and several membrane-bound, dense-core neurosectetory granules.
Purpose : Polygonum cuspidatum extract is an oriental herb which has been used for uterine diseases. In this study, the effects of Polygonum cuspidatum extract were investigated on inducing growth inhibition and apoptosis of human uterine cervical carcinoma cells. Methods : Viability of Polygonum cuspidatum extract-induced ME-180 cells was measured by MTT assay. Apoptotic cells were visualized by EtBr/AcOr staining under fluorescent microscope. Nucleosomal DNA fragmentation was assayed by agarose gel electrophoresis. Cell cycle distribution and changes in mitochondrial membrane potential were observed by flow cytometry. Results : Polygonum cuspidatum extract induced ME-180 cell death in a dose- and time-dependent manner. In the cells treated with Pc, the population of cells at sub-G1 phase significantly increased, and the condensed nuclei, apoptotic bodies and nucleosome-sized DNA were detected. Moreover, reduction in mitochondrial membrane potential was detected. Conclusion : Polygonum cuspidatum extract inhibits the growth and proliferation of ME-180 cells by apoptotic induction and facilitates its activity initiated by depolarization of mitochondria.
The present study was conducted to investigate the regulatory mechanism of plasminogen activators (PAs) activation by $17{\beta}$-estradiol ($E_2$) and progesterone ($P_4$) in porcine uterine epithelial cells (pUECs). pUECs were collected from porcine uterine horn and cultured at 80% confluence. Then, 0.1% (v/v) DMSO, 20 ng/mL $E_2$, and $P_4$ with or without fetal bovine serum (FBS) treated to cultured cells for 24 hours. The supernatants were used for measurement of PAs activity and expression of urokinase-type PA (uPA), tissue-type PA (tPA), uPA specific receptor (uPAR), and type-1 PA inhibitor (PAI-1) mRNA were analyzed by real-time PCR. The expression of PAs-related genes was not affect by steroid hormones in both of serum treatment groups. However, PAs activity was increased by treatment of $E_2$ compared to 0.1% DMSO treatment in serum-free group (p<0.05). Then, $E_2$ and $P_4$ were diluted with 0.002% (v/v) DMSO for reduction of its effect and treated to cultured cells without FBS. Only tPA mRNA was significantly increased by $E_2$ treatment (p<0.05). PAs activity was enhanced in $E_2$ treated group compared to control groups (p<0.05). These results indicate that serum-free condition is more proper to evaluate effect of steroid hormones and activation of PAs in pUECs was mainly regulated by estrogen. These regulation of PAs activation may be associated with uterine remodeling during pre-ovulatory phase in pigs, however, further studies are needed to investigate precise regulatory mechanism.
The present investigation has been undertaken to understand the mechanism of implantation process, by demonstrating the role of ovarian steroids in connection with phosphatase activity in the differentiation of uterine endometrium for implantation. The results obtained are as followings: The differentiation of the uterine endometrial tissue was closely influenced by the ovarian steroid hormones; at first, 17${\beta}$-estradiol initiated the differentiation of the uterine luminal and glandular epithelial cells, and then progesterone induced differentiation of stromal cells, and thereby two steroids maintain decidualization of the uterine tissues. We observed that the phosphatase activities seem to be dependent upon the ovarian steroids; that is the activity showed higher level in progesterone treated group than in estradiol treated one, and the highest activity was found in the group treated with both estradiol and progesterone. Acid phosphatase showed the highest activity whereas alkaline phosphatase showed the lowest in the rat uterine endometrium during early pregnancy.
A 2-year-old female Maltese dog was admitted with a history of pyometra and resulting peritonitis and septicemia. Uterine specimen sampled by ovariohysterectomy was processed routinely for histopathological observation. Grossly, the uterine mucosa was covered with necrotic debris and on the cut surface, lesion extended into the uterine wall. Microscopically, severe necrosis was observed throughout thickened mucosa, submucosa, and wall of uterus. Tumorous lesions composed of anaplastic cells with bizarre nuclei or tubular structures of cuboidal to short columnar cells were infrequently observed around the necrotic lesions and muscular layer far from necrotic areas. Immunohistochemically, central necrotic area with ambiguous cell and tissue structures, peri-necrotic tumor lesions, and muscular layer were strongly positive for cytokeratin. Since huge necrosis of adenocarcinoma lesions in this case made it difficult to diagnose, immunohistochemical results enable to diagnose as a severe necrotizing adenocarcinoma. Thus, histopathological and immunohistochemical findings in this case may serve as an important knowledge to diagnose uterine adenocarcinoma with huge necrosis in the veterinary field.

Uterine leiomyoma is the most common tumor in the female genital tract. Although the tumor is benign, it is a matter of paramount importance since it often causes profuse menstrual bleeding, pressure symptoms and infertility. Nevertheless, the etiology and pathophysiology of this abnormality remain poorly understood. The traditional definitive treatment for uterine leiomyomas is hysterectomy and, even today, symptomatic leiomyomas are the leading cause of hysterectomy in Korea. Clearly, the development of a safe, effective, and nonsurgical method of treatment for leiomyoma would be of great benefit to many women. This study demonstrated growth inhibition of uterine leiomyoma cells using Haeohyultang (HT). When human leiomyoma cells were treated with Haeohyultang, cells showed dose-dependent growth inhibitory effect. Cell growth was inhibited by over 40% as determined by both cell counts and MTS assay. Reduction of cellular viability as a consequence of exposure to Haeohyultang resulted from induction of apoptosis, as assessed by DNA fragmentation, PARP cleavage, caspase 9 and caspase 3 assay. Flow cytometry analysis with uterine leiomyoma cells demonstrated sub G1 cell cycle arrest after treatment with drug Haeohyultang. But, the expression levels of p27 and p21 were not changed in Haeohyultang treated cells compared with control. However, the expression levels of clAP1 were reduced by Haeohyultang compared with control. This reduction of clAP1 data means activation of the caspase family, and then induction of PARP cleavage and apoptosis. These results suggest that Haeohyultang may be potential therapeutic approach in the clinical management of uterine leiomyoma.
To investigate the role of interleukin-l$\beta$ (IL-1$\beta$) in the embryonic development, in vivo and in vitro expression patterns of IL-1$\beta$ gene in the preimplantation mouse embryos were examined by RT-PCR, and the effects of explanted mouse ovi-duct and uterine epithelial cells on the expression of IL-1$\beta$ gene in the pleimplantation mouse embryos were examined by co-culture. IL-1$\beta$ mRNA was detected in the embryos from 4-cell stage to blastocyst stage in vivo and from morula stage to hatching blastocyst stage in vitro. This transcript was not detected from the GV stage to late 2-cell stage in vivo, and not at the 4-cell and 8-cell stages in vitro. For the co-culture of late 2-cell embryos with the explanted mouse oviduct and uterine epithelial cells, oviducts and uterine epithelial cells were isolated at 48 hour alter the hCG injection. The explanted oviduct and uterine epithelial cells in co-culture groups facilitated the IL-1$\beta$ gene expression of the mouse embryos in comparison with the control. Taken together these results suggest that the presence of IL-1$\beta$ plays an important role in preimplantation embryonic development. In addition, the up-regulation of IL-1$\beta$ gene expression by the explanted oviduct and uterine epithelial cells demonstrates that embryonic expression of IL-l$\beta$ gene may be regulated by the interaction with oviductal and uterine factor (s).
