Pregnancy-associated changes in morphological and histochemical characteristics of the uterine cervix of the native Korean cattle (한우 자궁경부의 임신경과에 따른 형태학적 및 조직화학적 연구)

  • This study was performed to investigate the effect of gestation on the histological and histochemical changes of the uterine cervix of the native Korean cattle. A total of 110 cows obtained from the Chonhuk abattoir were divided into six groups. The 1st group was of 9 non-pregnant cows and 101 singleton pregnant cows were grouped into 5 groups from pregnant I to pregnant V according to gestation periods by means of crown rump length measures. For light microscopy the tissues were fixed in 10% neutral formalin and processed routinely for paraffin sections The $6{\mu}m$ sections were taken and stained with H-E, Alcian blue pH 1.0, Alcian blue pH 2.5, Alcian blue pH 2.5/PAS, PAS reaction, toluidin blue, and trichrome. The results obtained were as follows: 1. The cervical lengths and widths were increased in relation to advancing gestation. 2. The cervical folds of the pregnant groups were increased and complicated with many branches, and the cervical muscosal epithelia were increased in according to advancing gestation. 3. As advancing gestation, the tunics muscularis of cervix was increased following moderate distribution of fibroblast and vascularity, meanwhile decreased mast cells were found. 4. The cervical mucosubstance was composed of mixed mucopolysaccharides; the acid mucus was increased from the early pregnant state but the neutral mucus was found after pregnant IV, and the mucus was stained deeply in each staining in the fold cavity in according to gestation state.

Benign Metastasizing Pulmonary Leiomyoma with Hemoptysis (객혈을 동반한 양성 전이성 폐 평활근종 1예)

  • A benign metastasizing pulmonary leiomyoma(BMPL) is a rare disease that is usually detected years after a hysterectomy or myomectomy. Despite the benign pathological appearance, these tumors can metastasize and become clinically malignant. Recently, we experienced case of BMPL with hemoptysis. A 43-year-old woman, who had undergone a hysterectomy due to uterine leiomyoma 8 years ago, visited our department complaining of intermittent hemoptysis. A chest CT showed bilateral multiple nodular lesions. Video-associated thoracoscopy was performed. The resected small nodular lesion revealed the proliferation of spindle cells without mitosis or nuclear atypism. The lesions tested positive to the smooth muscle marker (actin) by immunohistochemical staining. Therefore, the multiple nodules were considered as benign metastasizing pulmonary leiomyoma from a uterine leiomyoma. GnRH analogue therapy was initiated. A chest CT showed that the size and number of pulmonary lesions did not change after 3 months, and the patient was symptom free.

Gestational Exposure to Bisphenol A Causes DNA Hypomethylation and the Upregulation of Progesterone Receptor Expression in the Uterus in Adult Female Offspring Rats

  • Exposure to environmental chemicals, including endocrine-disrupting chemicals, during the gestational period can have profound adverse effects on several organs in offspring. Bisphenol A (BPA) can infiltrate the human body through food and drinks, and its metabolites can cross both the placental and the blood-brain barriers. In this study, we investigate the effect of gestational exposure to BPA on epigenetic, biochemical, and histological modifications in the uterine tissues of F1 adult offspring rats. Pregnant rats were exposed to BPA from gestational day 8-15, and changes in global DNA methylation in uterine tissues obtained from adult offspring born to the exposed mothers were analyzed. Global DNA methylation analysis revealed that gestational exposure to BPA resulted in DNA hypomethylation in the uterus. Progesterone receptor (PR) protein expression in uterine tissues was monitored using western blot analysis, which revealed that the PR protein content was considerably higher in all BPA-exposed groups than in the control. Immunohistochemical examination for the PR revealed that intense PR-positive cells were more frequently observed in the BPA-exposed group than in the control group. To date, the evidence that the upregulation of PRs observed in the present study was caused by the non-methylation of specific PR promoter regions is lacking. Conclusively, these results indicate that exposure to BPA during gestation induces epigenetic alterations in the uteri of adult female offspring. We speculate that the global DNA hypomethylation and upregulation of the PR observed simultaneously in this study might be associated with the uterus.

  • Adenomyosis is a nonneoplastic proliforation of uterine glands characterized by multicentric infiltration of endometrial tissues into the myometrium. A female domestic short hair cat with unknown age was referred to local animal hospital in Busan for ovariohysterectomy to prevent the unwanted offspring. At the time of surgery, the uterus was enlarged with multiple nodular protrusion on the serosa. On the cut surface of uterus, elevation of the endometrium and hypertrophied myometrium were observed. Microscopically, the uterus was characterized by severe proliforation of endometrial glands into the lumen and within the myometrium. The intra-myometrial endometrium is circumferentially surrounded by bundles of hypertrophic smooth muscle cells. These endometrial glands had tall columnar epithelium lacking nuclear atypia and mitoses. In our best knowledge, this is the first report of feline adenomyosis in Korea.

Mitochondrial tumor suppressor 1 is a target of AT-rich interactive domain 1A and progesterone receptor in the murine uterus

  • Objective: Progesterone receptor (PGR) and AT-rich interactive domain 1A (ARID1A) have important roles in the establishment and maintenance of pregnancy in the uterus. In present studies, we examined the expression of mitochondrial tumor suppressor 1 (MTUS1) in the murine uterus during early pregnancy as well as in response to ovarian steroid hormone treatment. Methods: We performed quantitative reverse transcription polymerase chain reaction and immunohistochemistry analysis to investigate the regulation of MTUS1 by ARID1A and determined expression patterns of MTUS1 in the uterus during early pregnancy. Results: The expression of MTUS1 was detected on day 0.5 of gestation (GD 0.5) and then gradually increased until GD 3.5 in the luminal and glandular epithelium. However, the expression of MTUS1 was significantly reduced in the uterine epithelial cells of $Pgr^{cre/+}Arid1a^{f/f}$ and Pgr knockout (PRKO) mice at GD 3.5. Furthermore, MTUS1 expression was remarkably induced after P4 treatment in the luminal and glandular epithelium of the wild-type mice. However, the induction of MTUS1 expression was not detected in uteri of $Pgr^{cre/+}Arid1a^{f/f}$ or PRKO mice treated with P4. Conclusion: These results suggest that MTUS1 is a novel target gene by ARID1A and PGR in the uterine epithelial cells.

Estrogen Receptor Is Activated by Korean Red Ginseng In Vitro but Not In Vivo

  • Ginseng has been used as a traditional medicine for treatment of many diseases and for general health maintenance in people of all ages. Ginseng is also used to ameliorate menopausal systems. We investigated the estrogenic activity of Korean red ginseng (KRG) in a transient transfection system, using estrogen receptor (ER) and estrogen-responsive luciferase plasmids in MCF-7 cells. The extract activated both ER${\alpha}$ and ER${\beta}$. KRG modulated the mRNA levels of estrogen-responsive genes such as pS2 and ESR1 and decreased the protein level of ER${\alpha}$. In order to examine in vivo estrogenic activity of KRG, sixteen female Sprague-Dawley rats separated into four groups were studied for nine weeks: non-ovariectomized (OVX) rats treated with olive oil, OVX rats treated with olive oil, OVX rats treated with 17-${\beta}$-estradiol (E2) in olive oil, and OVX rats treated with KRG extract in olive oil. The experiments were repeated for three times and the data of twelve rats were combined. Body weight of OVX rats was greater than that of sham-operated control rats and was decreased by E2 treatment. Uterine weight increased after E2 treatment compared to OVX rats. However, no difference in body or uterine weight was observed with KRG intake. KRG induced reductions in total cholesterol, low density lipoprotein cholesterol/total cholesterol, high density lipoprotein cholesterol/total cholesterol, and low density lipoprotein cholesterol/high density lipoprotein cholesterol, but not to the same degree as did E2 intake. These results show that KRG does contain estrogenic activity as manifested by in vitro study but the activity is not strong enough to elicit physiological responses.

Histopathologic Changes to Additive Effect of Endocrine Disruptors in Mice (마우스에서의 환경호르몬물질의 상가작용에 대한 조직학적인 변화)

  • To investigate additive effects of endocrine disruptors, we have histopathologically studied the exchanges of the reproductive organ to ovariectomized ICR mice. Female ICR mice were ovariectomized and then treated with endocrine disruptors after two weeks. Macroscopic exchanges, which were body weight, feed and water intake, of all groups were not seen during experiment period. Histopathological changes of uterine epithelial cells, vaginal epithelial cells, mammary glands and the diameter in uterine tubles were observed. In the results, the histopathological sensitivity to endocrine disruptors effect was more seen to the vaginal epithelial cell height than others. The additive estrogenic effects of endocrine disruptors, which were combinations of DEHP, DBP and BPA, were seen with E2 and BPA treatments. These results offers a sysmatic and mechanistically informative approach to assessing estrogenicity. It provides a useful profile of activity using a reasonable amount of resources and is compatible with the study of individual chemicals as well as the investigation of interactions among combinations of chemicals.

Localization (and profiles) of tyrosine-phosphorylated proteins in female reproductive organs of adult rats

  • Objective: Tyrosine phosphorylation is an essential process in many biological systems, including the male reproductive system. The presence of tyrosine-phosphorylated (TyrPho) proteins has been well documented in male reproductive organs, but research in fertile females is still limited. Methods: The ovary, oviduct, and uterus of adult female Sprague-Dawley rats in the estrus phase were used to localize TyrPho proteins using an immunohistochemical technique. These proteins were separated and their expression patterns were examined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Western blot analysis, respectively. Results: TyrPho proteins were localized in the cytoplasm of the oocyte except the antral fluid; in the granulosa cells, theca cells, and stromal cells of the ovary; at the apical surface of oviductal epithelial cells; and in the basal epithelium and submucosa of the uterine wall. Moreover, we found that 72-, 43-, and 28-kDa TyrPho proteins were localized in the ovary, while 170-, 55-, and 43-kDa proteins were localized in the oviduct. In the uterus, we detected four major bands, corresponding to 61-, 55-, 54-, and 43-kDa TyrPho proteins. Conclusion: Given that these TyrPho proteins were found in major reproductive organs in the estrus phase, these proteins may play important roles in female fertility.

Evaluation of "Atypical Squamous Cells of Undetermined Significance(ASCUS)" on Uterine Cervical Cancer

  • The recently proposed Bethesda system for cervical/vaginal cytology has made a standardization related to "atypia". In cellular change due to inflammation or repair, the word "benign cellular change" has been suggested as a substitute for atypia. Terminology related to atypia cells may become standardized, but the cytologic criteria has not been well defined yet Among 8113 cases screened, we evaluated 182 cases as atypia squamous cells undetermined significance (ASCUS) by the Bethesda system (TBS): 52 cases of ASCUS were cytologically diagnosed and 33 cases were histologically verified. ASCUS was diagnosed based on nuclear enlargement and nonclassical signs of condyloma. The results are as follows; Six cases (16.6%) turned out to have benign cellular change. Twenty five cases (69.1%) demonstrated squamous intraepthelial lesion, condyloma was 2 cases (6.0%) and mild dysplasia was not revealed. Moderate and severe dysplasia were 1 case (3.0%) and 16 cases (48.4%), squamous cell carcinoma and adenocarcinoma were 4 (12.1%) and 1 cases (3.0%), respectively. Our data suggested that patients with ASCUS are highly recommended to be examined colposcopic biosy to cornfim.

A Study on the Spermatogenesis and Spermatozoan Fate in the Female Reproductive Tract of the Korean Greater Horseshoe Bat (Rhinolophus ferrumequinum korai) (한국 관박쥐의 정자발생과 자성생식관내의 정자운명에 관한 연구)

  • The present study was designed in order to clarify ultrastructural characteristies of the spermatogenetic process and to examine survival state of the spermatozoa introduced into the female reproductive tract after autumnal coitus in the Korean greater horseshoe bats(Rhinolophus ferrumeguinum korai). The general morphological characteristics of spermatogenesis were principally similar to those of the other mammalian species; acrosomal formation, flagellar formation, middle piece formation and concentration of the spermatozoan nucleus. The spermatozoa introduced into the vagina were found to be dead forming a vaginal plug, the opaque central core of which consisted of trapped dead spermatozoa. Some spermatozoa introduced into the uterus were observed to be phagocytized by the polymorphonuclear leucocytes infiltrated into the uterine glandular lumen. The oviductal epithelium, consisted of ciliated and secretory cells; the luminal surface of secretory cells were covered by a number of microvilli with well developed glycocalyx, suggesting a close relationship to nutrient (e.g. glycogen) supply for the spermatozoa during hibernation.
