도롱뇽(Hynobius leechii) 정충의 미세구조(Amphibia, Urodela) (Ultrastructure of Spermatozoa in Urodeles, Hynobius leechii (Amphibia: Urodela))

  • The ultrastructure of spermatozoa in Hynobius leechii was examined with transmission electron microscope and compared with those of other urodeles and anurans. The perforatorium and the tail show most of the common feature of urodeles. However, there were several ultrastructural characteristics in an acrosome, nucleus, neck, axial rod and mitochondria related to the nucleus. The acrosome was trifoliate in transverse sections and the perforatorium consisted of two different concentric parts with a fine sharp point in a subacrosomal lumen. The nucleus consisted of two different regions of chromatin area and nuclear ridge. The nuclear ridge was composed of several bundles of five to ten minute tubular subunits of 19 nm diameter in this species, while in higher urodeles it was well developed in multi layers. The protoplasmic bead was separated by a cytoplasmic canal except the connection with only the distal portion of the nucleus. The neck was a short cylinder and contained pericentriolar material with transverse striations. In Hynobius the ring was not elongated to the tail and the mitochondria were distributed only in the protoplasmic bead around the nucleus as in Cryptobranchus, while in higher groups of urodeles it elongates to the length of the middle piece and the mitochondria follow the ring. The ring elongation may be related to the distribution of mitochondria. Hynobiidae and Cryptobranchidae are closely related based on structure of neck, the axial rod and location of mitochondria, although they are different in the composition of the axial rod.

유미류와 하등 무미류 정충의 미세구조 비교와 계통적 고찰 (Ultrastructure of spermatozoa in Urodela and Primitve Anura(Amphilbia) with Phylogenetic Considerations)

  • 유미류와 하등 무미류 정충의 미세구조를 비교하고 미세구조적 형질에 의한 계통적 관계를 고찰하였다. 대부분의 유미류 정충은 subacrosomal rod, endonuclear canal, perforatorium, ring, marginal filament, 꼬리의 구성과 꼬리의 주축이 axial rod인7가지 양서 류의 원시 공유형질(symplesiomorphies)을 나타내었다. 하등 무미류 정충은 marginalfilament가 없으며 subacrosomal rod와 ring의 구조는 단지 Ascaphus와 Discoglossus의 두 속에서만 각각 보고되었다. 이러한 미세구조적 형질은 유미류와 하등 무 미류의 원시 공유형질로 간주되며 두 분류군 사이의 계통적 연속성을 보여준다. 또한 정자 꼬리의 구성, endonuclear canal과 perforatorium의 형성에서도 매우 밀접한 계통적 관계를 보여주었다.

Phylogenetic Relationships among Groupers (Genus Epinephelus) Based on Mitochondrial Cytochrome b DNA Sequences

  • To infer phylogenetic relationships among Epinephelus species inhabiting coastal regions of Korean peninsula, mitochondrial cytochrome b genes from 9 species belonging to the subfamily Epinephelinae were PCR-amplified, cloned and sequenced. Aligned cytochrome b sequences of 10 species containing one additional sequence from GenBank were 1,140 base pairs in length, including 439 variable and 330 parsimony informative sites. The cytochrome b genes of 10 species, as other vertebrates studied to date, exhibit unequal base compositions: an entirely low G content ($15.2{\pm}0.3{\%}$on average) and almost equal T, C and A contents ($29.3{\pm}0.8{\%},\;30.7{\pm}1.0{\%},\;and\;24.8{\pm}0.5{\%}$ on average, respectively).In third codon positions, transitional substitutions especially between Epinephelus species and outgroup species are almost certainly saturated or near saturation. Phylogenetic analyses were performed with sequence data from 8 Epinephelus species and 2 outgroup species (Cephalopholis urodela and Vaviola louti) by using distance-based (neighbor-joining and minimum evolution) and parsimony-based (maximum parsimony) methods. The results showed that the monophyly of the genus Epinephelus was supported by relatively high bootstrap values. However, phylogenetic relationships among E. areolatus, E. moara, E. septemfasciatus, and Epinephelus sp were poorly resolved. Within the genus Epinephelus, three resolved monophyletic groups were found: clade 1 included E. akaara and E. awoara;, clade 2 included E. fasciatus and E. merra; and clade 3 included E. akaara, E. awoara, E. fasciatus, E. merra, E. areolatus, E. moara, E. septemfasciatus and Epinephelus Sp.

Reproductive Function of the Body and Tail Undulations of Hynobius leechii (Amphibia: Hynobiidae): A Quantitative Approach

  • To clarify the reproductive function of vibration signals in Hynobius, which has externally fertilized eggs, we quantitatively analyzed the body and tail undulations of male Korean salamanders (Hynobius leechii) in sixteen mating events. One large and one small male, and one female were used in each mating event. We analyzed behaviors recorded over a total of 3 hrs for each mating event; 2 hrs before and 1 hr after female's oviposition. Males touched females using their snouts or body trunk throughout the entire mating periods, but females touched males increasingly more after approximately 1 hr before oviposition. Males conducted body undulations more than 50 times per 10 minutes at a mean frequency of 0.64 Hz. Large males conducted more body undulations than small males, particularly on the tree twig where females attached their egg sacs. Males responded to other males' body undulation throughout the mating period by orienting their head towards, approaching, and touching the undulating male. Females only responded for about 10-20 min before ovipositing, and most responses were directed to the large male's body undulation. Males conducted tail undulations 3.0 times per 10 min at a mean frequency of 1.7 Hz and most tail undulations occurred after one male bit the other male. These results suggest that body undulations function in both male-female and male-male interactions, while tail undulations mainly functions in male-male competition. Also, male H. leechii appear to actively attract females, while females respond to the males only at times close to oviposition.

Age structure and growth rates of two Korean salamander species (Hynobius yangi and Hynobius quelpaertensis) from field populations

  • We studied and compared the age structure, body size, and growth rates of field populations of two Korean salamander species (Hynobius yangi and Hynobius quelpaertensis) to elucidate important aspects of basic population dynamics of these two endemic Hynobius species. In both populations, females were sexually mature at three years of age, while H. yangi and H. quelpaertensis males matured at two and three years of age, respectively. Both males and females of H. yangi and H. quelpaertensis attained a maximum age of 11 years and 10 years, respectively. In both species, the snout-vent length (SVL) and body weight (BW) of the females were greater than those of the males. The SVL, BW, and asymptotic SVL of both male and female H. yangi were smaller than those of H. quelpaertensis. The adult growth rates after sexual maturation of male and female H. yangi were lower than those of H. quelpaertensis, possibly resulting in the smaller body size of the former, although overall growth coefficients were not significantly different between the two species. We also compared the age structure and growth rates of three Korean and three Japanese species of Hynobius.

기후변화에 따른 한국꼬리치레도롱뇽(Onychodactylus koreanus)의 분포 예측에 대한 연구 (Distribution Prediction of Korean Clawed Salamander (Onychodactylus koreanus) according to the Climate Change)

  • 기후변화는 동·식물의 서식지와 개체군을 감소, 소멸시키며, 생물다양성 보존에 위협이 되고 있다. 특히, 도롱뇽과 (Hynobiidae)에 속한 종들은 다른 분류군들에 비해 행동권이 작고, 분산 능력이 극히 제한되기 때문에 기후변화에 매우 취약한 분류군이다. 본 연구에서는 한국꼬리치레도롱뇽(Onychodactylus koreanus)의 관찰지점과 종 분포 모델링 기법을 바탕으로 국내 서식하고 있는 한국꼬리치레도롱뇽의 주요 분포지역과 서식특성을 파악하고 기후변화에 따른 분포변화를 예측하였다. 그 결과 고도가 그들의 분포에 가장 주요한 영향을 끼친 환경변수로 확인되었으며, 강원도와 경상북도와 같은 고도가 높은 산림 지역에 밀집된 분포 형태를 보였다. 이처럼 종 분포 모델에서 예측된 공간적 분포 범위와 서식특성은 선행 조사 결과를 충분히 포함하고 있었다. 기후변화에 따른 분포변화를 확인한 결과, 한국꼬리치레도롱뇽은 현재 분포 범위에 비해 RCP4.5 시나리오에서 62.96% 가 감소할 것으로, RCP8.5 시나리오에서는 98.52% 감소할 것으로 예측되어 기후변화로 인해 서식 적합 공간들이 급격하게 감소하는 것으로 확인되었다. 모델의 AUC값은 현재에서 0.837, RCP4.5에서 0.832, RCP8.5에서 0.807로 높게 측정되었다. 이러한 결과들은 기후변화로 인해 영향을 받는 양서류의 보전 대책 수립에 중요한 기초자료가 될 수 있을 것이다. 추후, 한국꼬리치레도롱뇽의 생활사에 따른 서식지 특성과 미세한 서식 요인들이 반영된 다양한 분석기법을 통한 추가적인 연구가 수행된다면 종 감소에 영향을 끼치는 주요환경 요인들을 밝혀낼수 있을 것으로 판단된다.
