Innibition of Cell Growth and Urease Activity of Helicobacter pylori by Medicinal plant Extracts (한약재 추출물에 의한 Helicobacter pylori의 생장 및 Urease 활성 억제)

  • Among 14 medicinal plants selected for the study ethanol (70%) extract of Coptis japonica Makino showed the highest anti-microbial activity against Helicobacter pylori followed by Perilla frutescens var. acuta KUDO, Caesalpinia sappan L. and Schizonepeta tenuifolia Briq. However, anti-urease activity of methanol (80%) extracts was best for Forsythiae Fructus followed by Caesaipinia sappan L. and Schizonepeta tenuifolia Briq. In the second fractionation using water, ethyl acetate and butanol more than 90% of the anti-urease activity was detected in the ethyl acetate fraction.

The Influence of Soil Physico-Chemical Properties on Urease Activity in Paddy Soils (답토양(畓土壤)의 이화학적(理化學的) 특성(特性)과 Urease의 활성(活性))

  • Paddy soil samples were collected from the plow layers of 19 soil series distributed in Yeongnam district to determine the relationship between soil urease activity and other soil characteristics. The results obtained were summarized as follows: 1. Simple correlation analyses showed that the urease activity was positively related with available $P_2O_5(r=.844^{**})$, potassium activity ratio($r=0.762^{**}$), available $SiO_2(r=.580^{**})$ and $SiO_2$/O.M ratio($r=0.591^{**}$). 2. Among soil chemical properties which had positive linear correlations with soil urease activity, the content of available $P_2O_5$ in soil had the highest contribution to the multiple regression equation of soil urease activity. 3. The activity of soil urease was especially lower in sandy texture than in clayey paddy soils, and a tendency was observed that the heavier soil textures the higher activity of soil urease. 4. Relatively well drained soils had the higher activity of soil urease while the soils in "poorly drained" had remarkably lower activity of soil urease. 5. The soils in higher classes of paddy soil equitability group had higher activities of soil urease.

  • A laboratory experiments were carried out to study the urea transformation and kinetics of soil urease in paddy soils with different properties. Urea transformation in paddy soils followed first-order kinetics, the rate of urea hydrolysis and the first-order constant was higher in the soil with high total urease activity (TUA) and accumulated urease activity (AUA) than those with low TUA and AUA. The values of Km and Vmax indicated that the Km values of accumulated urease in the soils were different in each soils and lower than that of microbial urease. However, the Km values of microbial urease were nearly same each other.

  • A incubation study was conducted to find out the effects of rice straw and gypsum as soil ameriolite on urease, nitrate and nitrite reductase activities in newly reclaimed saline sandy soil. The results obtained were summarized as follows: 1. Very low urease activities were observed in saline soil if contrast to high productive paddy soil. Urease activities were lower at 5 days than that of 25 and 50 days after incubation. Remarkably high urease activities were obtained by the application of rice straw and gypsum. 2. Comparing with NPK treatment, application of rice straw and gypsum were enhanced the activities of nitrate and nitraite reductase. 3. Positive correlation (r=0.5501 p=0.05) was obtained between urease activities and ammonium nitrogen concentration in soil. 4. Cyclic oxidation and reduction of nitrate and nitrite in soil were obtained in terms of first order microbial kinetics reaction in case of application of rice straw and gypsum, respectively. 5. Positive correlation (r=0.6296 p=0.05) was obtained between the activitie of nitrite reductase and nitrate reductase in soil.

  • To determine the urease inhibitory activity of various heavy metal ions, a photometric cuvette assay for measuring ammonia production was developed. In this assay, the absorbance values at 630 m were linearly increased according to the ammonia concentrations up to 3.0 mg/l (r : 0.998). The urease inhibitions upon addition of a single species of heavy metal ions were in the decreasing order of Hg(II) > Pb(II) > Cu(II) > Cd(II) > Zn(II) ions. As expected, the urease inhibitions at a fixed concentration of a single species and at varying concentrations of other species occurred in the additive way. The above results show the applicability of the current method to the selective detection on Hg(II) ions as well as the screening of heavy metal ions possibly present at various samples.

  • Changes of urease activity in plant leaves following foliar application of urea were investigated with soybean, rye, tomato, radish and cabbage which were actively growing in a field. In this experiment, the procedure of the enzyme assay included incubation of the reaction mixture at $60^{\circ}C$ for 3 hr in order to inactivate heat unstable enzmes which may utilize ammonia produced by urease. The leaves with urea application showed somewhat higher urease specific activities for 2-4 days immediately after the foliar spray as compared with controls. The most difference of the specific activies between urea treatment and control was usually observed 2 days after urea application regardless of the plants. The difference of the specific activities disappeared completely 4 or 5 days following urea treatment. Protein contents in the leaves of soybean and tomato were increased for about 5 days after urea treatment, while no significant difference was found with rye, radish and cabbage. Urea application showed slightly lower ammonia concentration in the leaves which had higher urease activities. These results suggest that foliar spray of urea is very effective when nitrogen supply is required for rapid growth.

Evaluation of Urease Inhibition Activity of Zerumbone in vitro (제럼본의 요소가수분해효소 활성 억제 평가)

  • A key virulence factor for urinary tract pathogens is the enzyme urease, which catalyzes the hydrolysis of urea into ammonium ions and carbonic acid. Urease activity plays an important role in the pathogenesis of urinary tract infection. In this study, the inhibitory effect of zerumbone against six urease-producing bacteria (Klebsiella oxytoca, K. pneumoniae, Morganella morganii, Proteus mirabilis, P. vulgaris, and Staphylococcus saprophyticus) and their urease activities were evaluated. The results of the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) tests showed that zerumbone had antibacterial effect against these six urease-producing bacteria. The MIC and MBC of zerumbone ranged from 0.5 to 2 mM and 1 to 4 mM, respectively. In the urease inhibitory assay, zerumbone showed better urease inhibition ($56.28{\pm}2.45-37.83{\pm}3.47%$) than the standard urease inhibitor, acetohydroxamic acid ($40.46{\pm}1.94-22.99{\pm}3.53%$). However, zerumbone did not affect the levels of the urease subunit. These results clearly indicated that zerumbone has antibacterial potential against urease-producing bacteria and possesses excellent bacterial urease inhibition properties.

Adsorption of Urease on Zeolite (Zeolite 에 의(依)한 Urease 의 흡착(吸着))

  • The urease adsorption on zeolite and the various properties of adsorbed urease were investigated to find out the influence of zeolite on activity and properies of urease. Free urease in solution was adsorbed on zeolite untill the max. adsorption, and the amount of max. adsorption was 11.3mg urease/100mg zeolite at pH 7.0. It is apparent that free urease was adsorbed on the outer surface of zeolite by cation exchange reaction, and more than 70% of urease adsorption was adsorbed within 30 min. The activity of adsorbed urease was decreased by 89.6%, whereas Km value was increased to 34.4mM, which is higher than that of free urease. The optimum pH of adsorbed urease was widened 6.5 to 7.0, compared to that of free urease 7.0. The resistance of urease to protease became weaker by adsorption, however substrate specificity and thermal stability were not affected.

  • A Helicobacter pylori urease inhibitor from Rubus coreanus Miquel has been isolated and partially characterized for aiming to Prevent H. pylori growth and decrease harmful accumulation of ammonia in human gastric mucosa. We screened urease inhibitory activities in 519 extracts library prepared by solvent extraction from 173 kinds of edible plants, medicinal herbs, herbs and seaweeds using a colorimetric urease assay system. As results of primary and secondary screening, 70% acetone extract of Rubus coreanus Miquel was selected as potent candidate, showing about 24% inhibitory activity. The acetone extract was sequentially partitioned into RCE/RCWI and RCB/RCW2 layers with ethyl acetate and butanol. The major active component in RCW2, water layer from butanol fractionation was revealed to be peptidic or proteinous substance by inhibitory activity determination after pronase digestion and periodate oxidation. RCW2-IIIc a was isolated by sequential column chromatography on DEAE-Toyopearl 650C, Butrl-Toyopearl 650M and Sephadex LH-20. The isolated urease inhibitor RCW2-IIIc $\alpha$, was highly pure proteinous substance with molecular weight of 13kDa by high-performance gel permeation liquid chromatography. RCW2-IIIc$\alpha$ has about 5 times higher inhibitory activity than 70% acetone extract, showing high stability against heat treatment and peptic digestion.

Effect of pyroligneous acids on urease inhibition (요소분해 저해에 미치는 목초액의 영향 평가)

  • This study was conducted to investigate the effect of pyroligneous acids on urea hydrolysis for the purpose of inhibiting ammonia volatilization during urea fertilizer application. Different types of synthetic urease inhibitors have been searched and developed, but their use is limited due to varying inhibition effects on soil urease, and environmental problems. In this study, the effect of pyroligneous acids, a natural substance, on urea hydrolysis in soil was evaluated by analyzing inhibition of urease activity. Pyroligneous acids inhibited plant urease and microbial urease activity, as well as soil urease with various urease complex. In addition, pyroligneous acids exhibited non-competitive urease inhibition effect through urease kinetics and inhibited urea hydrolysis in the soil. This study showed that pyroligneous acids treatment with urea fertilizer decreases the loss of urea fertilizer, improves the efficiency of nitrogen application on plant and reduces the amount of nitrogen fertilizers applied in soil.
