발효온도에 의한 효모의 에탄올 내성 요인 연구 (Study on the Factors Affecting the Ethanol Tolerance of Yeast Strains by fermentation Temperature)

  • Saccharomyces cerevosiae STV89와 Kluyveromyces fragilis CBS 397을 이용한 알콜발효에서 발효온도에 따른 세포내 에탄올 농도와 지방산 함량비율을 분석하여 효모의 에탄올 내성에 영향을 이치는 요인을 밝히고져 하였다. 실험결과 두 균주 모두 발효온도가 낮을수록 최대 세포내 에탄올 농도는 감소한 반면, 불포화지방산 중에서 oleic acid와 linoleic acid의 함량은 증가하였다. 따라서 세포내 에탄올 축적이 감소하면 세포막의 불포화지방산 함량이 증가하여 세포로 부터의 에탄올 확산이 촉진되기 때문에 효모의 에탄올에 대한 내성이 증가되는 것으로 추정된다.

Identification of the SNP (Single Nucleotide Polymorphism) for Fatty Acid Composition Associated with Beef Flavor-related FABP4 (Fatty Acid Binding Protein 4) in Korean Cattle

  • In this study, we investigated the relationship between unsaturated fatty acids influencing beef flavor and four types of SNPs (c.280A>G, c.388G>A, c.408G>C and c.456A>G) located at exon 2, 3 and 4 of the FABP4 gene, which is a fatty acid binding protein 4 in Korean cattle (n = 513). When analyzing the relationship between single genotype, fatty acids and carcass trait, individuals of GG, GG, CC and GG genotypes that are homozygotes, had a higher content of unsaturated fatty acids and marbling scores than other genotypes (p<0.05). Then, haplotype block showed strong significant relationships not only with unsaturated fatty acids (54.73%), but also with marbling scores (5.82) in $ht1{\times}ht1$ group (p<0.05). This $ht1{\times}ht1$ group showed significant differences with unsaturated fatty acids and marbling scores that affected beef flavor in Korean cattle. Therefore, it can be inferred that the $ht1{\times}ht1$ types might be valuable new markers for use in the improvement of Korean cattle.

  • The association of three candidate genes, fatty acid synthase (FASN), microsomal triglyceride transfer protein (MTTP) and fatty acid binding protein 3 (FABP3), with fatty acid (FA) composition in Duroc pigs was investigated. Identified single nucleotide polymorphisms (SNPs) were used for polymerase chain reaction-restriction fragment length polymorphism genotyping. The c.265C>T SNP of FASN gene was significantly associated with high levels of palmitoleic acid (C16:1) (p<0.05), oleic acid (C18:1) (p<0.01), and mono-unsaturated fatty acid (MUFA) (p<0.01), but low levels of linoleic acid (C18:2) (p<0.01), alpha linolenic acid (C18:3) (p<0.05), and poly-unsaturated fatty acid (PUFA) (p<0.01) in animals having the CT genotype. The c.2573T>C SNP in the MTTP gene had a significant effect only in elevating the level of palmitoleic acid (C16:1) (p<0.05) in heterozygote animals. The polymorphism in FABP3 showed no significant effects on any fatty acid composition traits. These results suggest that the identified SNPs in the FASN and MTTP genes can be useful markers for selecting Duroc pigs having desirable healthy fatty acid composition.

송순차 제조를 위한 송순 및 솔잎의 화학적 조성 (Chemical Composition of Pine Sprouts and Pine Needles for the Production of Pine Sprout Tea)

  • As a basic study for preparing pine sprout tea, chemical components in pine sprouts and pine needles were analyzed as follows: In proximate composition the contents of most components except for crude fat were different between in pine sprouts and pine needles. Moisture content in pine sprouts was higher than that in pine needles. Calcium and potassium were major minerals contained in pine sprouts and needles, and their contents in pine needles picked in December were higher than those picked in June. Soluble tannin and vitamin C contents in leaf part of pine sprouts were much higher than those in stem part and their contents in pine needles were increased according to their growth. Free sugars like fructose, glucose and sucrose were contained in both pine sprouts and needles, and their contents in pine sprouts were higher in stem part as compared that in leaf part. Although fourteen kinds of amino acids were detected in pine sprouts and pine needles, their contents were extremely low. Amino acid composition between pine sprouts and pine needles was different each other, but major amino acids contained in them were same, those are acidic amino acids such as aspartic acid and glutamic acid. Amino acid contents in pine needles were increased according to the growth. In fatty acid composition in leaf part of pine sprouts, saturated fatty acid contents were higher than unsaturated fatty acid contents, but in stem part unsaturated fatty acid contents were higher. In pine needles the amount of saturated fatty acid was increased with the growth, but the amount of unsaturated fatty acid was rather decreased.

The Change of Major Cellular Fatty Acids Composition and Morphology of Escherichia coli Affected by Toxic Substances

  • This study was performed to compare the growth rates, cellular fatty acid compositions and morphology by using electron microscope of Escherichia coli (E. coli) grown in various conditions including different concentrations of phenol, CdCl₂ and HgCl₂. Ninety eight E. coli strains were isolated from Naktong river and human feces. The content of unsaturated fatty acids, especially 16:1ω7c and 18:1ωc increased as the concentration of phenol and CdCl₂ increased. The content of unsaturated fatty acid increased up to 50 ppb of HgCl₂, but decreased at 75 ppb of HgCl₂. There were more unsaturated fatty acids than saturated fatty acid in the presence of toxic substances. However, the ration was reversed when the affected E. coli was transferred to toxic substance free fresh trypticase soy broth medium. Also, by using transmission electron microscope these cells were observed to various morphological deformation by heavy metals and their deposition on the surface. From these results, we suggested that the changes of major fatty acids composition and morphology of E. coli may be considered to indicate contaminated levels of heavy metals or organic solvents. The information presented here may be useful in predicting effects of heavy-metal and organic solvent contamination in streams and provides a basis for further studies of metal or organic solvent effects on microbial communities.

Effect of Oxygen and Unsaturated Fatty Acids on the Ethanol Tolerance of Yeast Strains

  • This study deals with investigation of the ethanol tolerance of yeast strains with respect to fatty acid composition and intracelluar ethanol concentration during alcohol fermentation. The cell viabilities and fermentation abilities of Saccharomyces cerevisiae and Kluyveromyces fragilis were improved by aeration and addition of unsaturated fatty acids into growth medium. Aeration decreases the accumulation of ethanol, while increases unsaturated fatty acid contents inside yeast cells. Thus it was found that oxygen and unsaturated fatty acids play decisive roles in the increase of ethanol tolerance of yeasts.

Identification of Exonic Nucleotide Variants of the Thyroid Hormone Responsive Protein Gene Associated with Carcass Traits and Fatty Acid Composition in Korean Cattle

  • The thyroid hormone responsive protein (THRSP) gene is a functional gene that can be used to indicate the fatty acid compositions. This study investigates the relationships of exonic single nucleotide polymorphisms (SNPs) in the THRSP gene and fatty acid composition of muscle fat and marbling score in the 612 Korean cattle. The relationships between fatty acid composition and eight SNPs in the THRSP gene (g.78 G>A, g.173 C>T, g.184 C>T, g.190 C>A, g.194 C>T, g.277 C>G, g.283 T>G and g.290 T>G) were investigated, and according to the results, two SNPs (g.78 G>A and g.184 C>T) in exon 1 were associated with fatty acid composition. The GG and CC genotypes of g.78 G>A and g.184 C>T had higher unsaturated fatty acid (UFA) and monounsaturated fatty acid (MUFA) content (p<0.05). In addition, the $ht1^*ht1$ group (Val/Ala haplotype) in a linkage disequilibrium increased MUFAs and marbling scores for carcass traits (p<0.05). As a result, g.78 G>A and g.184 C>T had significantly relationships with UFAs and MUFAs. Two SNPs in the THRSP gene affected fatty acid composition, suggesting that GG and CC genotypes and the $ht1^*ht1$ group (Val/Ala haplotype) can be markers to genetically improve the quality and flavor of beef.

참깨 품질 개량에 관한 연구 제1보 참깨 유질평가와 지방산조성의 품질간 차이 (Breeding of Sesame (Sesamum indicum L.) for Oil Quality Improvement 1. Study on the evaluation of oil quality and the differences of fatty acid composition between varieties in sesame)

  • 참깨의 유질을 개량하기 위한 기초연구로 참깨유구성지방산 종류별 함량의 품종간 차리와 조성의 특성을 밝히고자 시험을 하였는 바 기 결과를 요약하면 다음과 같다. 1. 불포화지방산인 Olieic acid는 평균 39% 함유 되어 있으며 최저함량은 32.6%에서 최고함량은47.3%까지 14.7%의 변이복을 가지적 주지방산인 Linoleic acid의 함량은 평균적으로 45.8%이며 최w저 31.1%에서 최고 54.9%까지 무려 23.8%의 품종간 차를 보였다. 2. 포화지방산인 Palmitic acid와 Stearic acid의 함량에서는 Palmitic acid의 평균함량은 9.5% 최저 4.6%, 최고 18%로서 13.4%의 품종간 차이가 있으며 Stearic acid 평균함량 4.8%, 최저 1%에서 최고 9%까지 8%의 품종간 변이를 보였다. 3. 품종 유래에 따른 지방산 조성 차이를 보면 한국품종과 미국품종들이 Palmitic, Stearic acid 함량이 가장 낮고 필수지방산인 Linoleic acid 함량이 가장 높았는데 특히 한국품종은 가장 양질유 품종들이었다. 대체로 재배기간의 온도가 높을수록 포화 지방산 함량이 높은 경향이었다. 4. 특히 한국재래종은 불포화양질유지방산 함량이 세계적으로 가장 높았으며 재래종인 만성종은 Linoleic acid 함량이 54.9%이며 Oleic + Linoleic acid 함량이 92%에 이르러 참깨 지방산 개량육종에 중요한 유전자원이라 할 수 있다. 5. 참깨 종피색별 지방산조성 차이에서는 양질지방산인 Linoleic acid 함량과 Oleic acid함량이 가장 높았던 것은 황색 종피종(47.5%)이었으며 그다음이 갈색 종피종이었다. 6. 지방산 조성의 숙기별 차를 보면 포화지방산에서는 숙기별 차이가 거의 없으나 불포화지방산중 Oleic acid는 조숙에서 만숙으로 갈수록 함량이 감소하는데 반해서 Linoleic acid는 만숙일수록 함량이 증가하는 경향이었다. 특히 조숙종에서는 Oleic acid 함량이 Linoleic acid 함량보다 약간 더 높아서 중ㆍ만숙종과 조성 패턴이 다름을 보여 주었다. 7 착과습성에 따른 지방산조성 차이에서는 1과성은 포화지방산이 높고 3과성은 불포화지방산중 Linoleic acid 함량이 1과성보다 월등히 높았으나 Oleic acid 함량은 1과성보다 약관 낮은 편이었다. 8. 실방수 차이에서는 3과성 2실4수이 불포화지방산이 가장 높았고 포화지방산에서는 1과성 2실4수이 가장 높았다. 4실8수에서는 일정한 영향이 보이지 않았다. 9. 지방산 간의 상관관계를 보면 불포화지방산과 포화지방산 간에는 r =-0.955**의 높은 부의 상관을 보여 지방산 개량육종의 전망을 밝게 해주며 O1eic과 Linoleic acid간에도 에우 높은 부상관을 나타내고 있어 필수지방산 함량을 높이는 개량 육종도 가능하다.

귀뚜라미 첨가 사료가 계육과 계란의 성분에 미치는 영향 (Effects of Cricket Supplements of the Chicken Meats and Its Eggs)

  • Chemical characteristics of the cricket, Gryllus bimaculatus, were investigated in total composition, amino acid composition, fatty acid composition and mineral components. After the treatment of 0.4% of G. bimaculatus in the chicken feed, the changes of fatty acid composition in the chicken meat and egg were also estimated. As the result, saturated fatty acid, especially palmitic acid, decreased 4% and unsaturated fatty acids, linoleic aced and arachidonic acid, increased 12 and 23%, respectively, compared with those of control. In addition, there was a small increase in Eicosapentaenoic acid(EPA). The sensory test of the chicken meat resulted in increased flavor, brightness and yellow color. The sensory scores of G. bimaculatus egg in the point of taste resulted in somewhat better estimate than control. These results are appear to be stemmed from fatty acids of cricket.

Effects of Rumen Protected Oleic Acid in the Diet on Animal Performances, Carcass Quality and Fatty Acid Composition of Hanwoo Steers

  • The effects of different rumen protected forms, oleamide, Ca oleate, of dietary oleic acid on the carcass quality and fatty acid composition in intramuscular and subcutaneous fat tissues of Hanwoo steer were examined. Sixty, 25 month old Hanwoo steers divided into three groups were fed no supplement (Control), 2% of oleamide (Oleamide) or Ca-oleate (Ca-Oleate) in their diet for 45 or 90 days. Disappearance rates of oleic acid supplements in digestive tracts (Rumen bypass, abomasal and intestinal disappearance rate) were 48.5, 68.4 for oleamide and Ca oleate, respectively. Both oleic acid supplements affected feed intake, growth rate, cold carcass weight and carcass fatness. Live weight gain, carcass weight, backfat thickness and marbling score were higher in the oleic acid supplemented steers compared with those from the control. Oleic acid supplements increased marbling score and ether extract in Hanwoo steer m. logissi thoracicmus. Rumen protected oleic acid increased not only the level of oleic acid but also polyunsaturated fatty acids in intramuscular and subcutaneous fat tissue. Total saturated fatty acid contents in both fat tissues were decreased whereas total unsaturated fatty acid content was increased compared with those from control. Linoleic acid, linolenic acid and polyunsaturated fatty acid contents were significantly higher in Ca oleate than any other steers. Lipid metabolites in blood were increased in rumen protected oleic acid treatments. HDL content in blood was increased in Ca-oleate supplemented steers whereas LDL was decreased compared with control. The changes of fatty acid compositions in the rumen protected oleic acid supplemented steers suggest that the oleic acid and unsaturated fatty acid were protected from rumen biohydrogenation and can be deposited in the fat tissues.
