Isolation of Wild Yeasts from Freshwaters and Soils in Nonsan Stream and Sapgyoho in Chungcheongnam-do, Korea, and Microbiological Characteristics of the Unrecorded Wild Yeasts (충남 논산천과 삽교호로부터 야생효모의 분리 및 국내 미기록 효모들의 균학적 특성)

  • This study aimed to isolate wild yeasts from water and soil sample of the Nonsan stream and Sapgyoho (lake) in Chungcheonnam-do, Korea, and to further characterize previously unrecorded wild yeast strains. In total, 102 strains, representing 55 different species of wild yeast were isolated from 95 samples collected from the Jangseoncheon and Ipchoncheon of Nonsan stream in Jellabuk-do and Chungcheonnam-do. Among these, 33 strains were isolated from alkalophilic yeast extract-peptone-dextrose (YPD) medium (pH 9.0), and 9 strains were isolated concurrently on general YPD medium (pH 6.5) and alkalophilic medium. Seventeen strains of Cryptococcus laurentii were predominantly isolated. Additionally, 65 strains, representing 27 different species of wild yeast were isolated from 58 samples obtained from Sapgyoho (lake). Among the 82 isolated wild yeast strains, 8 strains, including Candida fructus JSC 72-1(JSL-GGU 015), had not previously been recorded. All 8 previously unrecorded yeasts were oval in shape except C. fructus JSC72-1(JSL-GGU-015), and only the Filobasidium chernovii JSC39-1(JSL-GGU-013) strain formed spores. All strains except Pseudosydowia eucalypti JSC23-6(JSL-GGU-012) grew well in yeast extract-peptone-dextrose (YPD) and yeast extract-malt extract media and grew in vitamin-free medium. Four strains, including P.eucalypti JSC23-6(JSL-GGU-012) grew well in 15% NaCl-containing YPD medium. F.chernovii JSC39-1(JSL-GGU-013) and Sirobasidium intermedium JSC7-3(JSL-GGU-014) assimilated lactose, and five strains, including F. chernovii JSC39-1(JSL-GGU-013) also assimilated starch. All strains were resistant to 800 ppm of Ca, Cu, Li, and Mg ions.

  • This study focused on the isolation of wild yeasts from the soils of an herb park in Seoul, Korea, and characterization of unrecorded yeasts. Fifteen yeast strains of nine species were isolated and identified. Among them, strains of the Cryptococcus genus, such as Cryptococcus laurentii (2 strains), Cryptococcus saitoi (two strains), Cryptococcus podzolicus (2 strains), and Cryptococcus sp. (2 strains) were dominant. Candida boleticola SU14-2, Candida novakii SU6-2 and Cryptococcus podzolicus SU18-3 were also screened as newly recorded yeasts in Korea. Candida novakii SU6-2 formed ascospores and a pseudomycelium, and Candida boleticola SU14-2 grew well in the 5% NaCl-containing yeast extract-peptone-dextrose medium.

Microbiological Characteristics and Physiological Functionalities of Unrecorded Wild Yeast Strains in the Soils of Hajodae and Gyungpodae Beaches in Korea (동해안 하조대와 경포대 해수욕장 주변 토양으로부터 분리한 국내 미기록 야생효모들의 균학적 특성과 생리활성)

  • The goal of this study was to isolate and characterize the wild yeast strains in the soils of Hajodae and Gyungpodae Beaches, Gyungpo Lake in Kangwon-do, Korea and evaluate their anti-demential and tyrosinase inhibitory activities. Among the 27 yeast strains isolated from 30 soil samples from Hajodae Beach, Cyberlindnera culbertsonii HJ31-1, Dothichiza pithyophila HJ22-4, Metschnikowia bicuspidata HJ16-1, and Sakaguchia cladiensis HJ14-1 were recorded for the first time in Korea. Additionally, six of the 55 wild yeast strains isolated from 45 soil samples from Gyungpodae Beach and Gyungpo Lake had not previously been detected in Korea, such as Aureobasidium melanogenum JR3-1. The microbiological characteristics and anti-demential and tyrosinase inhibitory activities of these previously unrecorded yeasts were investigated. Almost all of them were oval-shaped and had ascospores except for Dothichiza pithyophila HJ22-4, Diutina siamensis JR37-5, and Canadida gelsemii DC 35-1. Furthermore, they were all sugar-tolerant and able to grow in 20% glucose-containing YPD broth, except for Cyberlindnera culbertsonii HJ31-1. Diutina siamensis JR37-5 had the highest tyrosinase inhibitory activity (30.9%).

Characterization of Unrecorded Yeasts Isolated from Leaves of Trees of Oknyeobong Peak and Yeonjasan Mountain in Daejeon, Korea (대전광역시 옥녀봉과 연자산 나뭇잎으로부터 야생효모의 분리 및 국내 미기록 효모의 특성)

  • Twenty-two yeast strains of 15 species were isolated from the leaves of 20 trees on Oknyeobong Peak, and 24 yeast strains of 12 species were isolated from the leaves of 20 trees on Yeonjasan Mountain in Daejeon city, Korea. Cryptococcus bestiolae (5 strains) and Aureobasidium pullulans (8 strains) were the predominant isolates from Oknyeobong Peak and Yeonjasan Mountain, respectively. Of a total of 46 yeast strains, Cryptococcus kuetzingii JSL508, Cryptococcus vishniacii JSL509, and Dioszegia takashimae JSL510 from Okyeobong Peak, and Plowrightia periclymeni JSL514, Erythrobasidium hasegawanium JSL0193, and Rhodotorula nothofagi JSL0196 from Yeonjasan Mountain were determined to be yeast strains that were yet unrecorded in Korea. Morphological and cultural characteristics of these unrecorded yeasts were investigated. Erythrobasidium hasegawanium JSL0193 and Rhodotorula nothofagi JSL0196 did not form ascospores and pseudomycelia. All the strains, except Dioszegia takashimae JSL510, were halotolerant or halophilic, and Cryptococcus kuetzingii JSL508 and Dioszegia takashimae JSL510 were thermophilic, growing at $37^{\circ}C$.

Isolation of Wild Yeasts from Humus-rich Soil in City Park of Daejeon Metropolitan City, Korea, and Characterization of the Unrecorded Wild Yeasts (대전광역시 도시 자연공원 부엽토와 주변 산림토양들로부터 야생효모의 분리, 동정 및 국내 미기록 야생효모들의 특성)

  • Totally 91 strains of wild yeasts were isolated from the humus rich soil in the Bogyong city park of Daejeon city, Korea. Majority of the strains belonged to Cryptococcus spp., which included 11 strains of Cryptococcus aureus. Among them, Kwoniella mangroviensis JSS0500, Candida corydalis JSS0501, Candida bombi JSS0503, Candida multigemmis JSS0504, Cryptococcus dimennae JSS0506, Cryptococcus saitoi JSS0507, Cryptococcus victoriae JSS0508, Metschnikowia pulcherrima JSS0502, Papiliotrema aurea JSS0505, Debaryomyces vanrijia JSS0509, Occultifur externus JSS0510, Filobasidium floriforme JSS0511 and Yamadazyma scolyti JSS0512 represented newly recorded yeast strains in Korea, and their microbiological characteristics were investigated. All of these unrecorded yeasts showed oval shape and also formed ascospores and pseudomycelia, except for Kwoniella mangroviensis JSS0500, Candida bombi JSS0503, and Metschnikowia pulcherrima JSS0502. Seven strains including Candida corydalis JSS0501 grew in vitamin-free medium, and 4 of the wild yeasts including Cryptococcus victoriae JSS0508 were halotolerant, i.e., capable of growing in 10% NaCl-containing yeast extract-peptone-dextrose (YPD) broth. Debaryomyces vanrijia JSS0509 was found to be a thermophilic yeast that grew at $37^{\circ}C$.

  • The goal of this study was to isolate wild yeasts from reed fields in the Seocheon-gun county in Chungcheongnam-do, South Korea. Molecular analysis of the D1/D2 domain of the 26S rDNA of the yeasts isolated from soil samples was performed using Basic Local Alignment Search Tool (BLAST). In total, 20 yeast strains from 11 species were isolated from 20 different soil samples. Candida species were observed to be the most common yeasts. Of these 20 strains, Candida subhashii (6 strains), Candida tropicalis (3 strains), Candida boleticola MS008, and Lachancea thermotolerans (2 strains) were identified as alcohol-fermenting yeasts. Further, Bullera japonica YJ10-1, Candida subhashii J7-1, Kluyveromyces yarrowii YJ11-1, and Ustilago shanxiensis Y10-1 were newly recorded yeast strains in Korea, and therefore, their microbiological characteristics were investigated further. All of these unrecorded yeast strains had oval cells; only Candida subhashii J7-1 formed ascospores and pseudomycelia. Kluyveromyces yarrowii YJ11-1 grew in vitamin-free medium, and all of these strains, except Candida subhashii J7-1, grew in 5% NaCl-containing YPD broth.

Isolation of Wild Yeasts and Characterization of Physiological Functionalities of Unrecorded Wild Yeasts Obtained from Flowers and Soils of the Wolpyung Park, Daejeon City and Gykpo Beach, Buan, Jeollabuk-do in Korea (대전광역시 월평공원과 전북 격포해수욕장 주변 야생화와 토양들로부터 야생효모의 분리 및 국내 미기록 효모들의 특성과 생리 활성)

  • This study aimed to isolate wild yeasts obtained from flowers and soil of the Wolpyung park, Daejeon city and Gykpo beach, Buan, Jeollabuk-do in Korea, and to further characterize previously unrecorded wild yeast strains. In total, 88 strains of 62 different species of wild yeasts were isolated from 75 samples obtained from the Wolpyung park. Among these, six strains of Trichosporon moniliiforme and four strains each of Papiliotrema flavescens and Candida melibiosica were isolated. Additionally, 39 strains of 30 different species of wild yeasts were isolated from 35 samples collected from the Gykpo beach. Among the 127 isolated wild yeast strains, 10 strains, including Apiotrichum porosum ASCM32-1, were previously unrecorded. All the 10 previously unrecorded yeasts were oval or global in shape, and three strains, including Candida athensensis WP4-90-3, formed spores. Three strains, including Vishniacozyma taibaiensis WP13-2, were halophilic yeasts which grew in 15% NaCl-containing YPD(yeast extract-peptone-dextrose) medium. Five strains, including C. athensensis WP4-90-3, showed 15% ethanol resistance. Cell-free extracts from Candida oleophila WP5-19-1 and Wickerhamomyces anomalus HO9-2 showed the highest β-glucuronidase inhibitory activity (49.0%) and neutrophil elastase inhibitory activity (38.4%), respectively.

Microbiological Characteristics, Enzyme Activity, and Physiological Functionality of Unrecorded Wild Yeasts from the Wolsung Valley of South Deogyu Mountain and Jinjamcheon in Daejeon, Korea (남덕유산 월성계곡과 대전광역시 진잠천주변 등으로부터 선발된 국내 미기록 야생효모들의 균학적 특성과 주요 효소활성 및 생리활성)

  • This study aimed to screen and investigate the microbiological characteristics, enzyme activity, and physiological functionality of unrecorded wild yeasts from the Wolsung valley of South Deogyu mountain and Jinjamcheon in Daejeon city, Korea. Candida sorboxylosa WSC25-4 (NNIBRFG47336), Arxula adeninivorans WSC6-3 (NNIBRFG47335), Farysizyma taiwaniana JRC6-2 (NNIBRFG47339), Saturnispora dispora JTS19-1 (NNIBRFG47334), Vanderwaltozyma polyspora JJCH5-3 (NNIBRFG47337), and Vanrija fragicola SW-9 (NNIBRFG47338) have not been previously recorded. None of these formed spores or pseudo-mycelium, and four strains, including C. sorboxylosa WSC25-4, were global in shaped. Except for V. polyspora JJCH5-3, all strains grew well in the yeast extract-peptone-dextrose (YPD), potato-dextrose and yeast malt extract media. A. adeninivorans WSC6-3 and S. dispora JTS19-1 grew well in 40% glucose-containing YPD medium and V. fragicola SW-9 in 5% NaCl-containing YPD medium. The supernatant of A. adeninivorans WSC6-3, F. taiwaniana JRC6-2, and V. fragicola SW-9 exhibited 58.0, 59.0, and 59.0% of glucoamylase activity, respectively. C. sorboxylosa WSC25-4 also produced extracellular alkaline protease. Antioxidant activity of the supernatant from F. taiwanian JRC6-2 was high (79.8%), while the other five unrecorded yeasts demonstrated elevated antiaging superoxide dismutase (SOD)-like activity (99%). Anti-gout xanthin oxidase inhibitory activity of S. dispora JTS19-1 and V. fragicola SW-9 was also elevated at 79 and 80%, respectively.

Isolation and Characterization of Wild Yeasts from Water and Riverside Soils of Geumgang Midstream in Gongju City, Korea (금강 중류 공주시 주변 물과 토양으로부터 야생효모의 분리 및 국내 미기록 효모 특성)

  • The goal of this study was to elucidate wild yeast diversity of the midstream region of the Geumgang river, Gongju, Chungnam province, Korea. Fifty strains of 24 species of wild yeasts were isolated from 45 water and soil samples among which Cryptococcus spp. (11 strains), Cryptococcus magnus (7 strains), Rhodotorula spp. (9 strains), and Hanseniaspora spp. (6 strains) were dominant. Four species, Candida chauliodes WJSF 0201, Candida oleophila WJSF 0202, Candida catenulata WJSF 0203, and Candida jaroonii WJSF 0204, represented newly recorded yeasts in Korea. All of these unrecorded yeasts were oval in shape, formed ascospores and pseudomycelium, and grew in vitamin-free medium. Candida oleophila WJSF 0202 was thermophilic which can grow at $37^{\circ}C$.

  • We isolated 120 strains of wild yeasts from the water and riverside soils of Daejeoncheon and Gapcheon in Daejeon, Korea. We identified Debaryomyces udenii JSF601, Kazachstania telluris JSF602, Trichosporon faecale JSF614, Candida infanticola WJSL0039, Candida palmioleophila WJSL0048, Pichia spartinae WJSL0087, and Trichosporon coprophilum WJSL0093 from Daejeoncheon, and Leucosporidium golubevii WJSL0108 and Ustilentyloma graminis WJSL0118 from Gapcheon, as newly recorded yeast strains in Korea and investigated their microbiological characteristics. All of these previously unrecorded yeasts were oval- or ellipsoidal-shaped with ascospores, except C. infanticola WJSL0039, C. palmioleophila WJSL0048, and D. udenii JSF601. L. golubevii WJSL0108 and U. graminis WJSL0118 grew in vitamin-free medium, and C. infanticola WJSL0039 was halotolerant and grew in 10% NaCl-containing YPD broth. K. telluris JSF602 and P. spartinae WJSL0087 were thermophilic yeasts, which grew at $37^{\circ}C$.
