Two Unrecorded Macrofungal Species from Sohwangbyeongsan in Korea

  • Indigenous fungi from Mount Sohwangbyeongsan, Odaesan National Park, Pyeongchanggun, Gangwon-do, South Korea, were investigated between 2013 and 2016. Our survey identified a total of 137 macrofungi species. These species were systematically categorized into 2 phyla, 4 classes, 15 orders, 41 families, and 73 genera. We identified collected specimens to the species level based on their morphology and rDNA sequences. Two species-Lentinellus flabelliformis and Tricholoma stans were newly recorded as macromycota in Korea.

  • 1995년 4월부터 1997년 10월까지 주로 한라산 일대를 중심으로 버섯을 채집한 결과 총 127속 292종이 동정되었다. 이들 중 담자균문의 41속 103종과 자낭균문의 13속 88종이 새롭게 제주도에 기록되게 되었으며 Typhula phacorriza(제주노랑부들국수버섯), Coryceps crinalis(깊은주름동충하초), Cordyceps cochlidiicola(쐐기나방번데기동충하초), Coryceps longissima(제주긴뿌리동충하초), Cordyceps prolifica(나무가지동충하초) 등 5종이 새롭게 한국기록종으로 등록되었다.

Description of 39 unrecorded bacterial species in Korea, belonging to the class Alphaproteobacteria

  • During an investigation of the biodiversity of bacterial species in Korea, we discovered many indigenous prokaryotic species. A total of 39 bacterial strains in the class Alphaproteobacteria were isolated from various environmental samples collected from marine organisms, sea water, fresh water, tap water, mud flats, activated sludge, mineral water, tidal flats, soil and decayed plants. From the high 16S rRNA gene sequence similarity (>98.7%) and formation of robust phylogenetic clades with the most closely related species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that any of these 39 Alphaproteobacteria species have been described in Korea. Specifically, 18 species in 11 genera in the order Sphingomonadales, 11 species in 10 genera in the order Rhizobiales, two species in two genera in the order Caulobacterales, six species in six genera in the order Rhodobacterales and two species in two genera in the order Rhodospirillales were found in Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs are described in the species description section.

  • During a comprehensive investigation of indigenous prokaryotic species in Korea, a total 31 bacterial strains assigned to the class Alphaproteobacteria were isolated from diverse environmental habitats including freshwater, seawater, brackish water, ginseng soil, plant roots, natural caves, and tidal flats. Based on their high 16S rRNA gene sequence similarities(>99.1%) and formation of robust phylogenetic clades with the closest type species, each strain was assigned to an independent and predefined bacterial species. Because there were no published or official reports regarding the isolation of these 31 species in Korea, this study identified three species in two genera in the order Caulobacterales, 12 species in 10 genera in the order Rhodobacterales, three species in two genera in the order Rhizobiales, two species in two genera in the order Rhodospirillales and 11 species in seven genera, all in the order Sphingomonadaceae within the Alphaproteobacteria are reported as new alphaproteobacterial species in Korea.   • During a series of extensive surveys of prokaryotic species diversity in Korea, bacterial strains belonging to Gammaproteobacteria were isolated from various sources of aquatic and terrestrial environments. A total of 39 isolates were obtained, which represented 39 unrecorded species in Korea belonging to 20 genera in 12 families. Enterobacteriaceae was the largest family, as eight species were assigned, which was followed by Moraxellaceae (6 species) and Pseudomonadaceae (5 species). At the genus level, Marinobacter (6 species), and Pseudomonas (5 species) were the main genera, and at least two species were obtained for Acinetobacter (3 species), Psychrobacter (3 species), Shewanella (2 species), Dickeya (2 species), Salinivibrio (2 species), Vibrio (2 species) and Rhodanobacter(2 species). A report of sixteen unrecorded haloarchaea species in Korea, isolated from a solar saltern

  • In July 2018, solar saltern samples were collected from Siheung, Gyeonggi-do Province to obtain unrecorded haloarchaea in Korea. The samples were suspended in a 20% NaCl (w/v) solution, and serial dilution was performed in fresh DB Characterization media No. 2. The strains isolated in this study showed at least 98.7% sequence similarity or more compared to the previously reported. Finally, 16 haloarchaeal strains, which were not reported in Korea but validly published under the International Code of Nomenclature of Prokaryotes (ICNP), were obtained from a solar saltern in Siheung. These 16 isolates were allocated to the orders Halobacteriales and Haloferacales. The 10 Halobacteriales strains were classified into the family Halobacteriaceae and Haloarculaceae. Each family belonged to three genera, respectively. The other six Haloferacales belonged to the families Haloferacaceae and Halorubraceae. Each family belonged to genus genus, respectively. Collectively, the unrecorded haloarchaeal strains belonged to two orders, four families, and eight genera. During the research, the possibility of discovering previously unknown species in domestic solar saltern was established. Gram-staining, cell morphology, physiological and basic biochemical parameters, and phylogenetic analysis were all performed in this study and are described in detail for each strain.

A report of four unrecorded Proteobacteria species isolated from soil in Korea

  • In 2015 and 2017, the National Institute of Biological Resources has isolated four unrecorded prokaryotic species designated as R-1-5, R-2-13, R-2-1, and R-1-8 from the peatland soil of Yongneup. Phylogenetic analysis based on 16S rRNA gene sequence similarity determined the four strains (R-1-5, R-2-13, R-2-1, R-1-8) were most closely related to Curvibacter lanceolatus (99.93%), Massilia brevitalea (98.7%), Pseudomonas lini (99.54%), and Pseudomonas vancouverensis (99.93%), respectively. The four unrecorded strains belong to the phylum Proteobacteria, in which the genera Curvibacter and Massilia are assigned to the class Betaproteobacteria, and the genus Pseudomonas to the class Gammaproteobacteria. Since there are no publications or official reports on these four strains, these four species are new records to Korea. The strains were further characterized by Gram reaction, colony and cell morphology, basic biochemical properties, and phylogenetic position. Descriptive information of the four unrecorded species is provided.

A report of 30 unrecorded bacterial species in Korea, isolated from marine ecosystems in 2021

  • To obtain unrecorded bacterial species in Korea, various marine samples were collected from Jeollanam-do Province, Korea in 2021. After plating the samples on marine agar and marine R2A agar, and incubating aerobically and anaerobically, approximately 1200 bacterial strains were isolated and identified using 16S rRNA gene sequences. A total of 30 strains showed ≥98.7% 16S rRNA gene sequence similarity with validly published bacterial species but not reported in Korea, indicating that they are unrecorded bacterial species in Korea. The unrecorded bacterial strains belonged to 4 phyla, 7 classes, 13 orders, 19 families, and 22 genera, which were assigned to Azospirllium, Loktanella, and Pseudovibrio of the class Alphaproteobacteria; Grimontia, Halomonas, Marinobacter, Microbulbifer, Photobacterium, Pseudoalteromonas, Pseudidiomarina, Ferrimonas, Shewanella, Simiduia, Thalassotalea, and Vibrio of the class Gammaproteobacteria; Priestia and Enterococcus of the class Bacilli; Persicobacter of the class Cytophagia; Aureivirga of the class Flavobacteriia; Propionigenium and Psychrilyobacter of the class Fusobacteriia; and Tepidibacter of the class Clostridia. The details of the unreported species including Gram reaction, colony and cell morphology, biochemical characteristics, and phylogenetic position are also provided in the description of the strains.

A report of 12 unrecorded bacterial species isolated from Suncheon Bay in Korea

  • Suncheon Bay Ecological Park, possessing abundant fisheries and biological diversity, was registered as a Ramsar wetland in Korea. Approximately 300 bacterial strains were isolated from the Suncheon Bay in a comprehensive study of indigenous prokaryotic species conducted during 2019-2020 in South Korea. A total of 12 bacterial strains were identified using 16S rRNA gene sequencing, demonstrating >98.7% sequence similarity with validly published species. These species were determined to be unrecorded bacterial species in Korea. A total of six strains were isolated from brackish water and Phragmites communis Trin (reed) species. These unrecorded species were phylogenetically diverse and belonged to three classes, six orders, and ten genera. Regarding the genus and class levels, the previously unrecorded species belonged to Jiella, Martelella, Rhizobium, Paracoccus, Rhodovulum, and Altererythrobacter of the class Alphaproteobacteria; Mycolicibacterium, Demequina, and Microbacterium of the class Actinobacteria; Confluentibacter of the class Flavobacteria. The twelve species were further characterized by gram staining, colony and cell morphology, biochemical properties, and phylogenetic position.

A report of 20 unrecorded bacterial species in Korea, isolated from soils of coastal areas in 2022

  • To obtain unrecorded bacterial species in Korea, various soils of coastal areas were collected from the Republic of Korea in 2022. After plating the samples on marine agar and incubating aerobically and anaerobically, approximately 1,700 bacterial strains were isolated and identified using 16S rRNA gene sequences. A total of 20 strains showed ≥98.7% 16S rRNA gene sequence similarity with validly published bacterial species but not reported in Korea, indicating they are unrecorded bacterial species in Korea. The unrecorded bacterial strains belonged to four phyla, six classes, 15 orders, 16 families, and 19 genera which were assigned to Blastomonas and Sphingomonas of the class Alphaproteobacteria; Pseudidiomarina, Kushneria, Salinicola, and Salinisphaera of the class Gammaproteobacteria; Evansella, Virgibacillus, and Paenibacillus of the class Bacilli; Cyclobacterium of the class Cytophagia; Pedobacter of the class Sphingobacteriia; and Demequina, Ornithinimicrobium, Blastococcus, Jatrophihabitans, Kineococcus, Glaciihabitans, Aeromicrobium and Streptomyces of the class Actinomycetes.
