Influence of Cheongsimyeonja-tang on the Liver Function of Inpatients at the Korean Medicine Hospital (청심연자탕 복용이 한방병원 입원환자의 간 기능에 미치는 영향)

  • Background: Herbal medication is widely used in the Korean Medicine Hospital, and drug-induced liver injury (DILI) in Korea has increased proportionally. Herb-induced liver injury now accounts for approximately 40% of cases of hepatotoxicity in Korea, according to research data. Currently, however, the component responsible for the toxicity is usually unknown or can only be suspected. Objective: To study the hepatotoxicity of Cheongsimyeonja-tang in DILI. Methods: A retrospective review was conducted of 82 inpatients between April 2010 and March 2017 with suspected drug-induced liver injury (n=5). The standard criteria (RUCAM scale) for drug-induced liver injury (DILI) were applied. The electronic medical records (EMRs) were retrospectively reviewed to identify the relevant database. Aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), and total bilirubin (T. Bili) were analyzed in blood samples before and after the administration of Cheongsimyeonja-tang. Results: Five cases out of 82 patients had a criterion-referenced probable (RUCAM) score ranging from 6 to 8 points DILI. However, statistical analysis of the liver function parameters results of the 82 patients did not show a statistically meaningful elevation after taking Cheongsimyeonja-tang. Conclusions: These data suggest a relationship between Cheongsimyeonja-tang and DILI. More studies are needed to validate these observations and to explore their implications.

Nannochloris eucaryotum growth: Kinetic analysis and use of 100% CO2

  • Microalgae are receiving an increasing attention because of their potential use as $CO_2$ capture method and/or as feedstock for biofuels production. On the other hand the current microalgae-based technology is still not widespread since it is characterized by technical and economic constraints that hinder its full scale-up. In such contest the growth kinetics of Nannochloris eucaryotum (a relatively unknown marine strain) in batch and semi-batch photobioreactors is quantitatively investigated with the aim of obtaining the corresponding kinetic parameters suitable for process engineering and its optimization. In particular the maximum growth rate was evaluated to be 1.99 $10^{-3}\;h^{-1}$. Half saturation concentrations for nitrates ($K_N$) and phosphates uptake ($K_P$) were evaluated as 5.4 $10^{-4}\;g_N\;L^{-1}$ and 2.5 $10^{-5}\;g_P\;L^{-1}$, respectively. Yield factors for nitrogen ($Y_N$) and phosphorus ($Y_P$) resulted to be 5.9 $10^{-2}\;g_N\;g^{-1}$ biomass and 6.0 $10^{-3}\;g_P\;g^{-1}{_{biomass}}$, respectively. The possibility of using 100% (v/v) $CO_2$ gas as carbon source is also evaluated for the first time in the literature as far as N. eucaryotum is concerned. The strain showed a good adaptability to high concentrations of dissolved $CO_2$ as well as to low pH. The lipid content under 100% $CO_2$ is about 16.16 %wt $wt^{-1}$ and the fatty acid methyl esters composition of the extracted oil is in compliance with the European regulation for quality biodiesel.

A Kalman filter based algorithm for wind load estimation on high-rise buildings

  • High-rise buildings are generally sensitive to strong winds. The evaluation of wind loads for the structural design, structural health monitoring (SHM), and vibration control of high-rise buildings is of primary importance. Nevertheless, it is difficult or even infeasible to measure the wind loads on an existing building directly. In this regard, a new inverse method for evaluating wind loads on high-rise buildings is developed in this study based on a discrete-time Kalman filter. The unknown structural responses are identified in conjunction with the wind loads on the basis of limited structural response measurements. The algorithm is applicable for estimating wind loads using different types of wind-induced response. The performance of the method is comprehensively investigated based on wind tunnel testing results of two high-rise buildings with typical external shapes. The stability of the proposed algorithm is evaluated. Furthermore, the effects of crucial factors such as cross-section shapes of building, the wind-induced response type, errors of structural modal parameters, covariance matrix of noise, noise levels in the response measurements and number of vibration modes on the identification accuracy are examined through a detailed parametric study. The research outputs of the proposed study will provide valuable information to enhance our understanding of the effects of wind on high-rise buildings and improve codes of practice.

Suppression of Ku80 Correlates with Radiosensitivity and Telomere Shortening in the U2OS Telomerase-negative Osteosarcoma Cell Line

  • Ku70/80 heterodimer is a central element in the nonhomologous end joining (NHEJ) DNA repair pathway, Ku80 playing a key role in regulating the multiple functions of Ku proteins. It has been found that the Ku80 protein located at telomeres is a major contributor to radiosensitivity in some telomerase positive human cancer cells. However, in ALT human osteosarcoma cells, the precise function in radiosensitivity and telomere maintenance is still unknown. The aim of this study was to investigate the effects of Ku80 depletion in the U2OS ALT cell line cell line. Suppression of Ku80 expression was performed using a vector-based shRNA and stable Ku80 knockdown in cells was verified by Western blotting. U2OS cells treated with shRNA-Ku80 showed lower radiobiological parameters (D0, Dq and SF2) in clonogenic assays. Furthermore, shRNA-Ku80 vector transfected cells displayed shortening of the telomere length and showed less expression of TRF2 protein. These results demonstrated that down-regulation of Ku80 can sensitize ALT cells U2OS to radiation, and this radiosensitization is related to telomere length shortening.

  • In the present work, a nonlinear inverse perturbation method which has been used in the structural optimization, is adopted so as to identify the structural damages. Unlike the structural optimization, a larger number of constrained equations than the number of unknown parameters are often required detect structural damage. Therefore, nonlinear least squares method is utilized to solve the problem. Because only a limited number of sensors are available I real situation of damage detection, the determination of sensor location becomes one of the most important issues. Hence, this work concentrates on the issue of sensor placement in the framework of nonlinear inverse perturbation method, and the performances of various methodologies concerning to sensor placement are compared with each other. The comparisons show tat the successive elimination method gets good performance for sensor placement. From the several numerical studies, it is confirmed that the inverse perturbation method, combined with the successive elimination method, is very promising in structural damage detection.

Bayesian Analysis of Dose-Effect Relationship of Cadmium for Benchmark Dose Evaluation (카드뮴 반응용량 곡선에서의 기준용량 평가를 위한 베이지안 분석연구)

  • In this paper, we consider a Bayesian analysis of the dose-effect relationship of cadmium to evaluate a benchmark dose(BMD). For this purpose, two dose-response curves commonly used in the toxicity study are fitted based on Bayesian methods to the data collected from the scientific literature on cadmium toxicity. Specifically, Bayesian meta-analysis and hierarchical modeling build an overall dose-effect relationship that use a piecewise linear model and Hill model, where the inter-study heterogeneity and inter-individual variability of dose and effect such as gender, age and ethnicity are accounted. Estimation of the unknown parameters is made by using a Markov chain Monte Carlo algorithm based user-friendly software WinBUGS. Benchmark dose estimates are evaluated for various cut-offs and compared with different tested subpopulations with with gender, age and ethnicity based on these two Bayesian hierarchical models.

  • The purpose of this study is to develop design equations to calculate optimum specifications and dimensions such as weight, engine horsepower, etc. of the tractor necessary to perform stable rotary tillage. The main results of this study are as follows. 1. A wheel-lug ought to receive a special resistance in downward direction which resists the lug's upward motion on wet sticky soil surface. The authors introduce a new academic name of the "lift resistance(上昇抵抗力, 상승저항력)" for such a force which resists retraction of a wheel lug from the soil in the upward trochoidal motion. This force is composed of the frictional force acting on the trailing and the leading lug side, and the "perpendicular adhesion(鉛直付着力, 연직부착력)" acting on the lug face and the undertread face on adhesive soil. 2. The "lift resistance ratio(上昇抵抗力係數, 상승저항력계수)" and the "perpendicular adhesion ratio(鉛直付着力係數, 연직부착력계수)" were defined, which are something similar to the definition of the motion resistance ratio, the traction coefficient, etc. 3. The design equation of the optimum weight of a rotary tiller mounted on the tractor derived by calaulating the forces acting on the rotary blades. 4. The design equations to calculate optimum specifications and dimensions such as weight, engine horsepower, etc. of the tractor necessary to perform stable rotary tillage were derived. It becomes clear that the optimum weight of a rotary tiller and a tractor can be estimated in planning design by means of putting about 21 design factors of the target into the equation. These equations are useful for planning design to estimate the optimum dimensions and specifications of a rotary tiller as well as a tractor by the use of known and/or unknown design parameters.

The Analysis of Students' Conceptions of Parameter and Development of Teaching-Learning Model (중학생들의 매개변수개념 분석과 교수-학습방안 탐색)

  • In this paper, we analyze nine-grade students' conceptions of parameters, their relation to unknowns and variables and the process of understanding of letters in problem solving of equations and functions. The roles of letters become different according to the letters-used contexts and the meaning of letters Is changed in the process of being used. But, students do not understand the meaning of letters correctly, especially that of parameter. As a result, students operate letters in algebraic expressions according to the syntax without understanding the distinction between the roles. Therefore, the parameter of learning should focus on the dynamic change of roles and the flexible thinking of using letters. We develop a self-regulation model based on the monitoring working question in teaching-learning situations. We expect that this model helps students understand concepts of letters that enable to construct meaning in a concrete context.

Microstructure and Tensile Properties of SS400 Carbon Steel and SUS430 Stainless Steel Butt Joint by Gas Metal Arc Welding

  • The application of SS400 carbon steel and AISI430 ferritic stainless steel joint has been increased in industries because of the advantage of both metals was able to increase the service lifetime of the important structures. Therefore, a fusion welding process that could produce a sound weld and good joint properties should be optimized. This research is aimed to weld a butt joint of SS400 carbon steel and AISI430 ferritic stainless steel using Gas Metal Arc Welding (GMAW) welding process and to study the effects of welding parameters on joint properties. The experimental results were concluded as follows. The optimized welding parameter that produced the tensile strength of 448 MPa was the welding current of 110A, the welding speed of 400 mm/min and the mixed gas of $80%Ar+20%CO_2$. Increase of the welding current affected to increase and decrease the tensile strength of the joint, respectively. Lower welding current produced the incomplete bonding of the metals and indicated the low tensile strength. Microstructure investigation of the welded joint showed a columnar grain in the weld metal and a coarse grain in the heat affected zone (HAZ). The unknown hard precipitated phases were also found at the grain boundaries of the weld metal and HAZ. The hardness profile did not show the difference of the hardness on the joint that was welded by various welding currents but the hardness of the weld metal was higher than that of the other location.

Myoplasmic [$Ca^{2+}$], Crossbridge Phosphorylation and Latch in Rabbit Bladder Smooth Muscle

  • Tonic smooth muscle exhibit the latch phenomenon: high force at low myosin regulatory light chains (MRLC) phosphorylation, shortening velocity (Vo), and energy consumption. However, the kinetics of MRLC phosphorylation and cellular activation in phasic smooth muscle are unknown. The present study was to determine whether $Ca^{2+}$-stimulated MRLC phosphorylation could suffice to explain the agonist- or high $K^+$-induced contraction in a fast, phasic smooth muscle. We measured myoplasmic [$Ca^{2+}$], MRLC phosphorylation, half-time after step-shortening (a measure of Vo) and contractile stress in rabbit urinary bladder strips. High $K^+$-induced contractions were phasic at both $22^{\circ}C$ and $37^{\circ}C$: myoplasmic [$Ca^{2+}$], MRLC phosphorylation, 1/half-time, and contractile stress increased transiently and then all decreased to intermediate values. Carbachol (CCh)-induced contractions exhibited latch at $37^{\circ}C$: stress was maintained at high levels despite decreasing myoplasmic [$Ca^{2+}$], MRLC phosphorylation, and 1/half-time. At $22^{\circ}C$ CCh induced sustained elevations in all parameters. 1/half-time depended on both myoplasmic [$Ca^{2+}$] and MRLC phosphorylation. The steady-state dependence of stress on MRLC phosphorylation was very steep at $37^{\circ}C$ in the CCh- or $K^+$-depolarized tissue and reduced temperature flattend the dependence of stress on MRLC phosphorylation compared to $37^{\circ}C$. These data suggest that phasic smooth muscle also exhibits latch behavior and latch is less prominent at lower temperature.
