Chemical Properties and Spectroscopic Characteristics of Humic Fractions Isolated from Commercial Organic Fertilizers (국산(國産) 유기질비료(有機質肥料)의 부식조성(腐植組成) 및 분광학적(分光學的) 특성(特性))

  • Humic substances of 17 organic fertilizers available on the market were the objects of study. The list of ingredients for formulation of them comprised fish meal. bone meal, oil-cakes, brewer's grains, peat, sawdust, wood bark, zeolite, soil conditioner, live-stock droppings, amino acid fermentation byproduct, chaff, limestone and others. Humic and fulvic acids were isolated from those substances and given chemical and spectroscopic analyses. Nutritional values of the organic fertilizers showed big diversity. Humification of organic matter was incomplete for some of the fertilizers as indicated by a high C/N ratio. Extractable humic acid percentage was higher, in general, than that of fulvic acid. Also the relative content of humin increased with advanced humification. Total acidity was closely related to phenolic hydroxyl groups. Relationships between carboxyl and hydroxyl groups. and carboxyl and alcoholic hydroxyl groups were very significant. Ultraviolet and visible light absorption spectra of humic and fulvic acids were substantially similar. The types of humic acids were B. P, and Rp. Two humic acids of the 17 samples belonged to B type. 3 to P type and all the rest to Rp type.

Study on Fiber Laser Annealing of p-a-Si:H Deposition Layer for the Fabrication of Interdigitated Back Contact Solar Cells (IBC형 태양전지 제작을 위한 p-a-Si:H 증착층의 파이버 레이저 가공에 관한 연구)

  • Using multi plasma enhanced chemical vapor deposition system (Multi-PECVD), p-a-Si:H deposition layer as a $p^+$ region which was annealed by laser (Q-switched fiber laser, $\lambda$ = 1064 nm) on an n-type single crystalline Si (100) plane circle wafer was prepared as new doping method for single crystalline interdigitated back contact (IBC) solar cells. As lots of earlier studies implemented, most cases dealt with the excimer (excited dimer) laserannealing or crystallization of boron with the ultraviolet wavelength range and $10^{-9}$ sec pulse duration. In this study, the Q-switched fiber laser which has higher power, longer wavelength of infrared range ($\lambda$ = 1064 nm) and longer pulse duration of $10^{-8}$ sec than excimer laser was introduced for uniformly deposited p-a-Si:H layer to be annealed and to make sheet resistance expectable as an important process for IBC solar cell $p^+$ layer on a polished n-type Si circle wafer. A $525{\mu}m$ thick n-type Si semiconductor circle wafer of (100) plane which was dipped in a buffered hydrofluoric acid solution for 30 seconds was mounted on the Multi-PECVD system for p-a-Si:H deposition layer with the ratio of $SiH_4:H_2:B_2H_6$ = 30:120:30, at $200^{\circ}C$, 50 W power, 0.2 Torr pressure for 20 minutes. 15 mm $\times$ 15 mm size laser cut samples were annealed by fiber laser with different sets of power levels and frequencies. By comparing the results of lifetime measurement and sheet resistance relation, the laser condition set of 50 mm/s of mark speed, 160 kHz of period, 21 % of power level with continuous wave mode of scanner lens showed the features of small difference of lifetime and lowering sheet resistance than before the fiber laser treatment with not much surface damages. Diode level device was made to confirm these experimental results by measuring C-V, I-V characteristics. Uniform and expectable boron doped layer can play an important role to predict the efficiency during the fabricating process of IBC solar cells.

Effect of Food Additives on Heat Sensitivity of Listeria monocytogenes H-12 and Decontamination of Kitchen Utensils (식품 첨가물이 Listeria monocytogenes H-12의 내열성에 미치는 영향 및 오염된 조리기구 제균)

  • Effect of food additives on the heat sensitivity of listeria monocytogenes H-12 inoculated into Pollack surimi was investigated and also confirmed the effectiveness of various decontamination method such as tap water washing, chlorination, ultraviolet irradiation and heat treatment haying been applied on cooking utensils. Food additives such as polyphosphate, chitosan, and potassium sorbate increased heat sensitivity of t monocytogenes H-12 and polyphosphate showed the strongest synergistic effect. The tested strain was not detected from stainless steel and plastic cutting board contaminated with $10^4{\~}10^5/cm^2$ of L monocytogenes H-12 after tap water washing for 10 seconds or 1 minute, but washing effect was not found in wooden cutting board. The chlorination of stainless steel and plastic cutting board for 10 seconds with $5{\~}50 ppm$ solution eliminated all cells of the contaminated strain, however any change of the viable cell count was not observed in the chlorination of wooden cutting board, W irradiation on stainless steel and plastic cutting board for 5 minutes with 15 W above 30 cm eliminated the contaminated strain, but the tested strain was still found after 60 seconds of irradiation on wooden cutting board. The treatment of hot water on all used cutting boards for 10 seconds at $70^{\circ}C$ resulted in complete loss of viability of the contaminated strain.

Determination of Optimal Storage Condition for Pre-packed Hanwoo Loin

  • The aim of this study was to determine the optimal storage condition of pre-packed Hanwoo beef without freezing. Hanwoo loin was purchased from a local distributor at 48 h after slaughter, then sliced in $1.5{\pm}0.5$ cm thickness, and packed in a polyethylene (PE) tray covered with linear low-density polyethylene (LLDPE) film. The studied factors to set the optimal storage condition were chamber temperature (5, 2.5 and $-1^{\circ}C$ for 14 d), cooling method (direct and indirect cooling system), and ultraviolet (UV) light irradiation for beef surface sterilization (0, 30, 60, and 120 min). The changes of pH, thiobarbituric acid reactive substances (TBARS) and number of aerobic bacteria were measured during storage. Beef samples stored in $-1^{\circ}C$ showed the minimal increasing rate in TBARS and microbial growth. After 15 d of storage, there was no significant difference in pH and TBARS values. However, the microbial population of beef stored in direct type cooling chamber ($4.25{\pm}0.66$ Log CFU/g) was significantly lower than that of beef stored in indirect type chamber ($6.47{\pm}0.08$ Log CFU/g) (p<0.05). After 4 d of storage, 60 or 120 min UV light irradiated beef samples showed significantly lower microbial population, and at 14 d of storage, 60 min UV irradiated beef sample showed significantly lower microbial population ($3.14{\pm}0.43$ Log CFU/g) than control ($4.46{\pm}0.13$ Log CFU/g) (p<0.05). However, TBARS values of 60 or 120 min UV light irradiated beef samples were significantly higher than non-irradiated beef sample after 4 d of storage (p<0.05).

Development of Analytical Methods of Hyperoside from Rosa canina L. (Rosa canina L. 중 hyperoside의 시험법 개발 연구)

  • Rosa canina L. is health functional food materials that can help to temporarily relieve symptoms of arthritis. This study has been conducted to develop and validate analytical methods for hyperoside of Rosa canina L.. Methods based on HPLC with ultraviolet detection (UVD) were established through instrumental analytical conditions, and the examination of data, such as domestic and foreign reliable methods and journals. HPLC UVD analysis using Capcell Pak $C_{18}$ MG II column at 353 nm was determined on test through the column, mobile phase. The validation has been performed on the method to determine linearity, accuracy, limits of quantification (LOQ) and repeatability for hyperoside. The method showed high linearity in the calibration curve at a coefficient of correlation ($R^2$) of 0.999, and the LOQ was $0.393{\mu}g/mL$. Relative standard deviation (RSD) values of data from repeatability precision was between 0.6 and 2.6%. Recovery rate test at hyperoside scored between 98 and 99%. These results indicate that the established HPLC method is very useful for the determination of hyperoside in Rosa canina L. to develop a health functional material.

Physico-Chemical Properties and Antimicrobial Activity of Pyocyanine Produced by Pseudomonase aeruginosa KLP-2 (Pseudomonas aeruginosa KLP-2가 생산한 Pyocyanine의 항균활성 및 생리화학적 성상)

  • The antimicrobial substance produced by Pseudomonas aeruginosa KLP-2 strain was purified and identified. The substance was identified as a pyocyanine by the fast atom bombardment mass(FAB-MS). In physic-chemical properties, the pyocyanine was dark blue needles, and was soluble in various organic solvents such as chlorogorm, methanol, ethanol and ethyl acetae. The pyocyanine possessed a ultraviolet absorbance spectrum in methanol, 0.1 M HCl, and chlorogorm. The maximum absorption peak of the pyocyanine showed at 318 mm in methanol. The molecular formula of the pyocyanine was determined to the $C_{13}$ H$_{10}$ N$_{2}$O and protonate molecular ion species (M+H)$^{+}$ was observed at m/z 211 by FAB-MS. The pyocyanine showed antimicrobial against Bacillus cereus, Bacillus subtilis, Micrococcus luteus, Rodococcus equi, Staphylococcus aureus, Streptococcus faecalis, E. col, Legionella pneumophila, Shigella flexneri Shigella boydii, shgella sonnei, NAG Vibrio cholerae, Vibrio parahaemolyticus, Vibro vulnificus, Yersinia enterocolitica, and Saccharomyces cerevisiae. However, Salmonella spp. Shigela dysenteriae, 3 strains of Pseudomonas aeruginosa, Klebsiela pneumoniae, and Aspergillus niger were resistant to the pyocyanine. The pyocyanine showed the highest antimicrobial activity aganist Legionella pneumophila based on the size of inhibition zone by the disk contained 0.5 $\mu\textrm{g}$ of the pyocyanine.e.

Effects of Plant Extract containing Creams on UVB Radiation-induced Inflammatory Responses in Mice (쥐에서 자외선 B가 유도한 염증 반응에 대한 식물 추출물 함유 크림의 영향)

  • Ultraviolet radiation (UV) is a major cause of photodamages to human skin and the immediate responses of the skin to UV include the erythema and edema. In an attempt to find effective UV-protecting agents to be used in cosmetics, a number of plant extracts were screened in the cell-based assays. Among the total of 38 plant extracts tested, 3 plant extracts derived from Sasa quelpaertensis, Althaea rosea, and Dryopteris crassirhizoma attenuated the UVB-induced cytotoxicity as well as melanin synthesis in cultured human epidermal melanocytes. The anti-inflammatory effects of these plant extracts were further examined in animal models. A control or test cream containing 1% of a plant extract was topically applied to ears of a C57BL/6 mouse or the dorsal skin of a SKH-1 hafirless mouse before and after the exposure to UVB. The change in ear thickness or dorsal skin redness due to UVB exposure was determined to monitor edema and erythema, respectively. All three test creams exhibited anti-inflammatory effects in both experiments. The creams containing Sasa quelpaertensis, Althaea rosea or Dryopteris crassirhizoma extract alleviated the UVB-induced edema response on day 4 by 53.8 %, 56.4 % and 31.1 %, respectively. They also inhibited the erythema formation on day 2 by 45.7 %, 34.1 % and 20.5 %, respectively. This study suggests that the selected plant extracts formulated in cosmetics may attenuate skin inflammation caused by overexposure to UV.

The antibacterial effect of photo-catalytic titanium dioxide on canine skin (광 촉매 이산화 티타늄의 개 피부에 대한 항균효과)

  • Photo-catalytic products have been widely used at home and hospital to prevent bacteria, virus and fungus. Activities of anti-bacteria, anti-viruses and anti-fungi are based upon direct contact of crystals and particles of titanium dioxide with pathogens, into which titanium is catalyzed by photo. Those antimicrobial activities of the photo-catalytic titanium dioxide have been proved in vitro. However, in vivo tests of those activities have not been carried out on dog skin. Aim of this study was to evaluate the antimicrobial activities of the catalytic titanium dioxide in vivo. Ten beagle dogs were divided into two groups. One group was sprayed with 10ml of titanium dioxide(1 mg/ml) whereas the other was not. The treated dogs were exposed under the sunlight for 120 min. A set of three hairs was taken 15, 30, 60 and 120 min after the exposure and the bacteria contaminated in hairs were amplified in, Muller Hilton broth at $35^{\circ}C{\pm}1$ for 3 h. The supernatant of the bacterial culture was diluted 1 : 10 in phosphafe-buffered saline. One milliliter of the diluents was transferred into triphenyltetrazolium medium(TTC) and incubated at $35^{\circ}C{\pm}1$ for 2 days. The number of bacteria was counted. The number of bacteria colonies was decreased compared to control group. To further investigate the longevity effect of titanium dioxide, the dogs were kept in indoor without sun light for 6 and 12 h, 1, 2, 3, 7, 14 days after exposure of the chemical during each 15, 30, 60 min. The number of bacteria colony in 1ml was counted. The number of bacterial colonies was decreased. Treated group is exposured by sun light during 15 min, the longevity effect of titanium dioxide is continued by 1 week. Treated group is exposured by sun light during 30, 60 min, the longevity effect of titanium dioxide is continued over 2 weeks. These data indicated that the photo-catalytic titanium dioxide may be used for prevent bacteria on dog skin.

A Study on the Kinetics and Mechanism of the Hydrolysis of Dihydro-1,4-oxathiin Derivatives (Dihydro-1,4-oxathiin 유도체의 가수분해 Mechanism과 반응속도론적 연구)

  • The kinetics of the hydrolysis of dihydro-1, 4-oxathiin derivatives were investigated by ultraviolet spectrophotometry in H2O at $25^{\circ}C.$ A rate equation which can be applied over a wide pH range was obtained. The substituent effects on the hydrolysis of dihydro-1, 4-oxathiin derivatives were studied and the rate of hydrolysis was shown to be accelerated by electron accepting groups. Final product of the hydrolysis was 2-(2-hydroxyethylthio)acetoacet-anilide enol form. On the basis of rate equations derived and judging from hydrolysis products obtained and from general base effect and substituent effects, plausible mechanism of the hydrolysis in various pH range have been proposed. Below pH 3.5, the hydrolysis was initiated by the protonation and followed by the addition of water to 2-carbon. Above pH 10.0, the hydrolysis was proceeded by the addition of hydroxide to 2-carbon. In the range of pH 4.0∼10.0, the addition of water to dihydro-1,4-oxathiin is rate controlling step.

Changes in Allicin Contents of Garlic via Light Irradiation (광 조사에 의한 마늘의 알리신 함량 변화)

  • Agri-food garlic has been recognized as healthful because of its antibacterial, anticancer and antioxidant effects. As such, its consumption is steadily increasing. This study was conducted to amplify the healthful ingredient of garlic, allicin, using light irradiation. The following conclusions were drawn from the investigation of peeled garlic under various conditions like fluorescent lighting, color (green, yellow, blue and red) and ultraviolet light (UV-A, UV-B and UV-C). The allicin content increased most with the 15-second $0.129W/m^2$ (40 W) UV-B treatment of the garlic at a 700 mm distance from the light source. At the treatment of the garlic with 126 lx (40W) red light for 24 hours at a 700 mm distance from the light source, its allicin content increased from $15.15{\pm}0.25mg/g$ to $15.15{\pm}0.34mg/g$, for a 10-percent amplification effect. Therefore, it is believed that the healthfulness of garlic can be amplified through irradiation processing of its healthful ingredient, allicin, and the development of its processing unit.
