Ultrasound-assisted Extraction of Total Flavonoids from Wheat Sprout: Optimization Using Central Composite Design Method (밀싹으로부터 플라보노이드성분의 초음파 추출 : 중심합성계획모델을 이용한 최적화)

  • The process of extracting active ingredients from wheat sprout using ultrasound assisted method was optimized with a central composite design model. The response value of the central composite design model established the extraction yield and the total flavonoids content, main effects and interactive effects were analyzed depending on independent variables such as the extraction time, volume ratio of ethanol to ultrapure water, and ultrasonic irradiation power. The volume ratio of ethanol to ultrapure water and ultrasonic irradiation power were relatively large for the extraction yield and the extraction time was most significantly affected the total flavonoids, Considering both the extraction yield and total flavonoids content, the optimal extraction conditions were as follows: the extraction time of 17.00 min, volume ratio of ethanol to ultrapure water of 50.25 vol%, ultrasonic irradiation power of 551.70 W. In this case, the extraction yield and total flavonoids content were 28.43 wt% and $29.99{\mu}g\;QE/mL\;dw$, respectively. The actual experimental extraction yield and total flavonoids content under this condition were 8.73 wt% and $29.65{\mu}g\;QE/mL\;dw$, respectively with respective error rates of 1.05 and 1.13%.

Extraction of Caffeic Acid and Rosmarinic Acid from Zostera marina Based on Ionic Liquids and Deep Eutectic Solvent (이온성액체와 Deep Eutectic Solvent를 사용한 잘피에서 카페인산과 로즈마리산의 추출)

  • The applicability of the dipping, ultrasonic-assisted, heating methods to the extraction of useful components from Zostera marina was investigated. For the increase of the extraction yield of caffeic acid and rosmarinic acid from Zostera marina, ionic liquids and DES were used as additives in the extraction solvent. The optimum extraction conditions were found: dipping extraction, methanol as extraction solvent, 0.005 g of freeze-dried material powder, 5 mL methanol, 30 min and 0.5 g of DES-3 ($Et_4NCl$ and phenol) as additive. As a result, 0.19 mg/g of caffeic acid and 8.48 mg/g of rosmarinic acid were obtained. This method is simple and sensitive, and has been applied successfully to determine the component of caffeic acid and rosmarinic acid in Zostera marina and these results indicate that DES were used as additives is more suitable than traditional extraction for the extraction of useful components from Zostera marina.

Extraction of Oil from Chlorella vulgaris Using Supercritical Carbon Dioxide and Organic Solvent (초임계 이산화탄소와 유기용매를 이용한 Chlorella vulgaris 오일의 추출)

  • Three different types of extraction processes, which used supercritical carbon dioxide ($SCCO_2$) and organic solvent, were attempted to improve the extraction yield of oil from Chlorella vulgaris: cosolvent-modified $SCCO_2$ extraction, $SCCO_2$ extraction with ultrasonic sample treatment in organic solvent, and static extraction with organic solvent followed by dynamic $SCCO_2$ extraction. Among these, the last $SCCO_2$ extraction process was found to be most effective in the extraction of oil. Compared with pure $SCCO_2$ extraction, the extraction yield of oil was observed to increase about 7 times.

  • To find out the most reasonable analysis conditions of paeoniflorin, different paeoniflorin extraction methods and various UV detector wavelengths were conducted with paeonia radix of 4-year old Euisung local variety. The most reasonable paeoniflorin extraction time by reflux apparatus was 1hr. and by ultrasonic apparatus was 3hrs. and those methods were completed only once. Concentration of paeoniflorin by reflux apparatuses at 1hr. and 2hrs. of extracting time were higher than those of ultrasonic apparatus, and the differences were highly significant. However, the differences of paeoniflorin concentration at 3hrs. and 4hrs. in two methods were not significant. In comparing paeoniflorin concentration of many lines, ultrasonic extracting apparatus was more simple and effective than the reflux apparatus. Paeoniflorin was more reasonable sensitivity at 240nm, and albiflorin was 254nm by HPLC. When paeoniflorin and albiflorin were analyzed simultaneously, 254nm was more stable than any other wavelength.

  • Many technical methods are used for nondestructive testing field for solid materials. Among those, ultrasonic inspection methods are widely used and one of the popular methods involves the extraction of an appropriate set of features followed by the use of a neural network for the classification of the signals in the feature space. This paper describes an approach which uses LMS method to determine the coordinates of the ultrasonic probe followed by the use of SAFT with centroid technique to estimate the location of the ultrasonic reflector. The method is employed for classifying UT-NDE signals from the steam generator tubes in a nuclear power plant. The classification results are presented for the ultrasonic signals from cracks and deposits within steam generator tubes.

  • This paper proposes the optimization of ultrasonic signal recognition in ferrite carbon steel weld zone using neural networks. For these purposes, the ultrasonic signals for defects as porosity, incomplete penetration and slag inclusion in the weld zone are acquired in the type of time series data. And then their applications evaluated feature extraction based on the time-frequency-attractor domain(peak to peak, rise time, rise slope, fall time, fall slope, pulse duration, power spectrum, and bandwidth) and attractor characteristics (fractal dimension and attractor quadrant) etc. The proposed neural networks system in this study can enhances performance of ultrasonic signal recognition.

Extraction and separation of glabridin from licorice by reversed phase high performance liquid chromatography

  • The extraction and separation of glabridin from licorice root by HPLC was performed in this work. First, by investigating the different extraction solvents, extraction methods and extraction times, a one-hour ultrasonic extraction procedure with ethyl acetate as the extraction solvent was optimized. Then the ethyl acetate extraction was applied to RP-HPLC for separation of glabridin. The column efficiencies and resolutions were experimentally investigated with different mobile phase compositions. Baseline separation of glabridin was obtained under the mobile phase composition of 50/50 vol.% (ACN/water). The retention time of glabridin was 20.3 min. The peak of glabridin was collected from the HPLC elution for several times and identified by LC/MS. Under the optimum extraction and HPLC separation methods, 1.26 g of glabridin per kg licorice root could be extracted.
