Optimization of Antioxidant Extraction from Dandelion (Taraxacum officinale) Leaves Using BBD-RSM (BBD-RSM을 이용한 민들레로부터 항산화성분의 추출공정 최적화)

  • In this study, an antioxidant was extracted from dandelion leaves using traditional hot water and ultrasonic extraction methods. In order to optimize the extraction yield and total flavonoid, an antioxidant, Box-Behnken design (BBD) model among response surface analysis methods was used. In the case of hot water extraction, the extraction temperature and time as well as the ratio of alcohol/ultrapure water were set as variables, and for the ultrasonic extraction, the ultrasonic survey century and irradiation time and the ratio of alcohol/ultrapure water were variables. Optimum extraction conditions in the hot water extraction method were the extraction temperature and time of $45.76^{\circ}C$ and 1.75 h and the ratio of alcohol/ultrapure water of 41.92 vol.%. While for the ultrasonic extraction method the survey century of 512.63 W, the ratio of alcohol/ultrapure water of 56.97 vol.% and the extraction time of 20.79 min were optimum conditions. Expected reaction yield and flavonoid content values under the optimized condition were calculated as 22.09 wt.% and 28.98 mg QE/mL dw, respectively. In addition, the error value of less than 3% was obtained validating our optimization process.

  • Phytoestrogens are one of quasi-estrogens which are not generated within the endocrine system, but consumed by intaking phytoestrogenic plants. Phytoestrogens, also called as "dietary estrogens", are various botanic chemical compound groups naturally occurring nonsteroidal plant compounds of spontaneous generation. Due to their structural similarities to estrogone, they may provide desirable health benefits for reducing the menopausal symptoms. In this paper, various phytoestrogens were extracted from sophora root, pomegranate and kudzu by ultrasonic extraction process. In this study, the amount of extracted phytoestrogens at various ultrasonic power strengths and extraction times were compared, which could be used as basic data in the study of biology and chemistry related to natural products.

Ultrasonic Effect on the Extraction of Ash-free coal from Low Rank Coal (저등급 석탄으로부터 초청정석탄의 추출과 초음파의 영향)

  • Extraction was performed to prepare ash-free coal from low rank coal under the temperature of $200-430^{\circ}C$ and initial pressure of 0.1MPa. Three kinds of coal samples with different rank were used and N-methyl-2-pyrrolidinone(NMP, polar), 1-methyl naphthalene(I-MN, non-polar), Light Cycle Oil(LCO, non-polar) were used as solvents. Results showed that higher extraction yield could be obtained with NMP than with 1-MN and LCO, but the ash concentration shows minimun in the case of 1-MN. Three operation modes were compared, that is, idle, agitation and ultrasonic extraction mode. From the results, it was found that the extraction yield and ash concentration were 70.09% and 1.03% under the agitation mode, 80.7% and 0.76% under the ultrasonic operation mode respectively in the case of NMP used at the temperature of $350^{\circ}C$. Similar results were obtained with 1-MN. Effect of ultrasonic on the extraction was estimated as 15-20% increase in the yields and 26% reduction in the ash concentration.

Optimization of Iso-flavonoids Extraction Process from Kudge Using Ultrasonic Irradiation Energy (초음파에너지를 이용한 칡으로부터 이소플라보노이드의 추출공정 최적화)

  • In this study, we used the ultrasonic extraction process as a method to extract antioxidant substances from kudzu, and measured the content of iso-flavonoids puerarin, daidzein, daidzin contained in kudzu. The response surface methodology which is a statistical analysis method for optimizing the extraction amount of iso-flavonoids from the kudzu and the process condition for maximizing the yield was applied. It is the final objective of this study to effectively derive the condition of the process that matches the target response with a minimum number of experiments and analyze the effect of each process condition on the response. In the response surface methodology, the central composite design was applied and the optimum condition was analyzed, and the three independent variables were set to ultrasonic irradiation time, volume ratio of ethanol/ultrapure water, ultrasonic irradiation power. Using the response surface methodology, the optimum conditions with the maximum extraction yield and the content of iso-flavonoids were evaluated as ultrasonic irradiation time (24.75 min), ethanol / ultrapure water volume ratio (39.75 vol%), ultrasonic irradiation power (592.36 W). The overall satisfaction level appears as high as 0.8938, which is recognized at a significance level within 5%. As a result of analyzing the optimization process, it was confirmed that the ultrasonic irradiation time is the factor that most affects the responses.

Analytical Method for Dioxin and Organo-Chlorinated Compounds: (Ⅱ) Comparison and Extraction Methods of Dioxins from XAD-2 Adsorbent

  • Supercritical fluid extraction (SFE), ultrasonic extraction (USE), and accelerated solvent extraction (ASE) were compared with the well known Soxhlet extraction for the extraction of polychlorinated biphenyls (PCBs) and polychlorinated dibenzo-p-dioxins(PCDDs) from the XAD-2 resin which was used to adsorb PCDDs in the atmosphere. XAD-2 resin spiked with five PCDDs was chosen as a sample. The optimum conditions for the extraction of PCDDs by SFE were turned out to be the use of CO2 modified with 10% toluene at 100 ℃ and 350 atm, with 5 min static extraction followed by 20 min dynamic extraction. SFE gave a good extraction rate with good reproducibility for PCDDs ranging from 68 to 98%. The ultrasonic extraction of PCDDs from XAD-2 was investigated and compared with other extractions. A probe type method was compared with a bath type. Two extraction solvents, toluene and acetone were compared with their mixture. The use of their mixture in probe type, with 9 minutes of extraction time, was found to be the optimum condition. The average recovery of the five PCDDs for USE was 82-93%. Accelerated solvent extraction (ASE) with a liquid solvent, a new technique for sample preparation, was performed under elevated temperatures and pressures. The effect of tem-perature on the efficiency of ASE was investigated. The extraction time for a 10 g sample was less than 15 min, when the organic solvent was n-hexaneacetone mixture (1 : 1, v/v). Using ASE, the average recoveries of five PCDDs ranged from 90 to 103%. SFE, USE, and ASE were faster and less laborious than Soxhlet extraction. The former three methods required less solvent than Soxhlet extraction. SFE required no concentration of the solvent extracts. SFE and ASE failed to perform simultaneous parallel extractions because of instrumental limitations.

Optimum Extraction of Tricin and Tricin 4'-O-(threo-β-guaiacylglyceryl) Ether (TTGE) from Rice Hull (Oryza sativa L.) (왕겨에서 Tricin과 Tricin 4'-O-(threo-β-guaiacylglyceryl) Ether(TTGE) 추출 조건의 최적화)

  • This study was conducted to evaluate the effects of extraction conditions on tricin and tricin 4'-O-(threo-${\beta}$-guaiacylglyceryl) ether (TTGE) extracted from rice hull (Oryza sativa L.). Extraction conditions were an ethanol concentration of extraction solvent of 50~90%, extraction time of 0.5~48 h, and ultrasonic-assisted and agitated extraction as extraction methods. The total tricin and TTGE contents of rice hull were $82.20{\mu}g/g$ and $53.12{\mu}g/g$, respectively. Recovery of tricin and TTGE increased to 76.11% and 71.42% with increasing ethanol concentration until 70% and then decreased above 80%, respectively. In ultrasonic-assisted extraction, highest recovery of tricin was 83.30% after 2 h of extraction and TTGE was 71.80% after 1 h of extraction. In agitated extraction, highest recovery of tricin was 92.34% after 48 h and TTGE was 76.89% after 24 h of extraction. Therefore, optimum extraction conditions for tricin and TTGE of rice hull were 70% ethanol concentration and ultrasonic-assisted extraction for 1 h.

Identification of Curcuminoids from Turmeric (Curcuma longa) Using Ultrasonic Wave and Dipping Method (초음파 및 침적방법을 이용한 강황 (Curcuma longa)으로부터 Curcuminoids의 확인)

  • In this study three major curcuminoids in turmeric curcumin (1), demethoxycurcumin (2) and bismethoxycurcumin (3) were efficiently extracted by optimizing extraction condition and simultaneously identified using a fast and reliable RP-HPLC-UV-MS and TLC method. The analysis by the $C_{18}$ column was performed and the UV wavelength was fixed at 425 nm. In this result, the total extraction yield of turmeric (Curcuma longa) was increased with extraction time from 1 to 7 h. So, optimum extraction time is 4 h. Also, the highest yield of extraction amount 0.433g 8.66% was obtained by ultrasonic waves with quarter frequency kHz and an extraction time of 7 h. The experiment method was consistent with theoretical Value $r^2$ = 0.987 (1), 0.997 (2) and 0.998 (3). Moreover, LC-MS analysis provided efficiently molecular weight information of three major curcuminoids in turmeric extracts and high purity (~95%) of the curcuminoids were obtained. This work offers would be useful for chemical and biological studies of natural plants and its products.

Ultrasonic Signal Processing Algorithm for Crack Information Extraction on the Keyway of Turbine Rotor Disk (터빈 로터 디스크 키웨이의 초음파 신호로부터 균열정보의 추출을 위한 신호처리 알고리즘의 개발)

  • An ultrasonic signal processing algorithm was developed for extracting the information of cracks generated around the keyway of a turbine rotor disk. B-scan images were obtained by using keyway specimens and an ultrasonic scan system with x-y position controller. The B-scan images were used as input images for 2-Dimensional signal processing, and the algorithm was constructed with four processing stages of pre-processing, crack candidate region detection, crack region classification and crack information extraction. It is confirmed by experiments that the developed algorithm is effective for the quantitative evaluation of cracks generated around the keyway of turbine rotor disk.

A Study on the Quantitative Analysis of Damaged Concrete by UNDT (초음파법에 의한 콘크리트 손상의 정량적 해석에 관한 연구)

  • In this paper, we have introduced the feature extraction for the ultrasonic signal analysis of the undamaged and damaged concrete specimens. Since the concrete has the heterogeneous nature by itself, it has been difficult to classify the feature with using ultrasonic signal which is acquired from the undamaged and damaged concrete specimens. Therefore, in this paper, we proposed the combinational analysis which is using both the damage coefficient and the number of zerocrossing for the feature extraction. And the pulse velocity method and the damage coefficient, which was proposed by Suaris, were reviewed. In this experiment, two types of concrete specimen have been considered: 180kg/㎠ and 240kg/㎠. The ultrasonic signals were acquired in normal direction. As a result, it has been that combinational analysis method, which is proposed in this paper, shows the better performance than the traditional ultrasonic pulse velocity method and the damage coefficient using maximum amplitude of the ultrasonic signal in the feature extraction.

Removal Characteristics of TPHs and Heavy Metals in Contaminated Soil with Ultrasonic Washing (초음파세척을 이용한 오염토양 내 TPHs 및 중금속 제거특성)

  • The removal characteristics of total petroleum hydrocarbons (TPHs) and heavy metals in contaminated soils with ultrasonic washing have been studied. The ultrasonic washing was evaluated on a laboratory scale. In this investigation, the effects of factors such as ultrasonic frequency, power intensity, duration of irradiation, contents of the TPHs and heavy metals and mixing ratios between the contaminated soils and water, were considered. Experimental results suggested that the rates for contaminant extraction of the TPHs and heavy metals in the contaminated soil increased considerably with the ultrasonic washing. Therefore, the ultrasonic washing has previously been to be an effective method to remediate the contaminated soils with the TPHs and heavy metals.
